3Genome Engineering in Rice Using Cas9 Variants that Recognize NG PAM Sequences显示文摘CRISPR/Cas9 genome editing relies on sgRNA-target DNA base pairing and a short downstream PAM sequence to recognize target DNA. The strict protospacer adjacent motif (PAM) requirement hinders applications of the CRISPR/Cas9 system since it restricts the targetable sites in the genomes. xCas9 and SpCas9-NG are two recently engineered SpCas9 variants that can recognize more relaxed NG PAMs, implying a great potential in addressing the issue of PAM constraint. Here we use stable transgenic lines to evaluate the efficacies of xCas9 and SpCas9-NG in performing gene editing and base editing in rice. We found that xCas9 can efficiently induce mutations at target sites with NG and GAT PAM sequences in rice. However, base editors containing xCas9 failed to edit most of the tested target sites. SpCas9-NG exhibited a robust editing activity at sites with various NG PAMs without showing any preference for the third nucleotide after NG. Moreover, we showed that xCas9 and SpCas9-NG have higher specificity than SpCas9 at the CGG PAM site. We further demonstrated that different forms of cytosine or adenine base editors containing SpCas9-NG worked efficiently in rice with broadened PAM compatibility. 6Low-dose decitabine enhances the effect of PD-1 blockade in colorectal cancer with microsatellite stability by re-modulating the tumor microenvironment显示文摘PD-1 blockade has demonstrated impressive clinical outcomes in colorectal cancers that have high microsatellite instability.However,the therapeutic efficacy for patients with tumors with low microsatellite instability or stable microsatellites needs further improvement.Here,we have demonstrated that low-dose decitabine could increase the expression of immune-related genes such as major histocompatibility complex genes and cytokine-related genes as well as the number of lymphocytes at the tumor site in CT26 colorectal cancer-bearing mice.A more significant inhibition of tumor growth and a prolongation of survival were observed in the CT26 mouse model after treatment with a combination of PD-1 blockade and decitabine than in mice treated with decitabine or PD-1 blockade alone.The anti-tumor effect of the PD-1 blockade was enhanced by low-dose decitabine.The results of RNA sequencing and whole-genome bisulfite sequencing of decitabine-treated CT26 cells and tumor samples with microsatellite stability from the patient tumor-derived xenograft model have shown that many immune-related genes,including antigen-processing and antigen-presenting genes,were upregulated,whereas the promoter demethylation was downregulated after decitabine exposure.Therefore,decitabine-based tumor microenvironment re-modulation could improve the effect of the PD-1 blockade.The application of decitabine in PD-1 blockade-based immunotherapy may elicit more potent immune responses,which can provide clinical benefits to the colorectal cancer patients with low microsatellite instability or stable microsatellites.Ganjun Yu Yanfeng Wu Wenying Wang Jia Xu Xiaoping Lv Xuetao Cao Tao Wan 2019Cellular & Molecular Immunology2019,16,4:14

8Status and development of high-power laser facilities at the NLHPLP显示文摘In this paper, we review the status of the multifunctional experimental platform at the National Laboratory of High Power Laser and Physics(NLHPLP). The platform, including the SG-II laser facility, SG-II 9th beam, SG-II upgrade(SG-II UP) facility, and SG-II 5 PW facility, is operational and available for interested scientists studying inertial confinement fusion(ICF) and a broad range of high-energy-density physics. These facilities can provide important experimental capabilities by combining different pulse widths of nanosecond, picosecond, and femtosecond scales. In addition, the SG-II UP facility, consisting of a single petawatt system and an eight-beam nanosecond system, is introduced including several laser technologies that have been developed to ensure the performance of the facility. 9Rechargeable solid-state Li-air batteries: a status report显示文摘The theoretical specific energy of lithium-air battery is as high as 3436 Wh·kg^(-1), and the possible achieved value may reach 600–700 Wh·kg^(-1), which enables this energy storage system as an important propulsion power sources for electric vehicles with the driving range of 500–800 km. Currently, Li-air batteries are facing main challenges at stability, efficiency, applicability and safety. In particular, from a practical view of point, the Li-air batteries should be operated directly in ambient air. Solid-state battery system is the best avenue to eventually solve these main issues. At the heart of the solid state, Li-air technology is the solid-state Li^+-conducting ceramic material. Developing solid-state lithium-air batteries(SSLAB) can solve the problem of applicability fundamentally and circumvent the safety issues completely, and it is also an important avenue to improve the stability of the battery system. In this paper, we provide a systematical review of the progress in the cell construction,the regulation of the electrode/electrolyte interface, the cell assembly, the electrochemical performance and the mechanism for the SSLAB. In every section, the contributions of the recent research progress in the main challenges and the remained questions will be commented. 10Disruption of MIR396e and MIR396f improves rice yield under nitrogen-deficient conditions显示文摘The microRNA miR396 directly represses GROWTH-REGULATING FACTORs(OsGRFs)and has been implicated in regulating rice yield and in nitrogen assimilation.Overexpressing the miR396 targets OsGRF4 and OsGRF6 improves rice yield via increased grain size and panicle branching,respectively.Here,we used CRISPR/Cas9 to assess the function of miR396 genes in rice.Knockout of MIR396 ef(MIR396e and MIR396f),but not other isoforms,enhanced both grain size and panicle branching,resulting in increased grain yield.Importantly,under nitrogen-deficient conditions,mir396ef mutants showed an even higher relative increase in grain yield as well as elevated above-ground biomass.Furthermore,we identified OsGRF8 as a new target of miR396,in addition to the known targets OsGRF4 and OsGRF6.Disruption of the miR396-targeting site in OsGRF8 was sufficient to both enlarge grain size and elongate panicles.Our results suggest that rice-seed and panicle development are regulated by miR396ef-GRF4/6/8-GIF1/2/3 modules and that mi R396ef are promising targets of genome editing for breeding environmentally friendly rice varieties that require less nitrogen fertilization.Jinshan Zhang Zhenyu Zhou Jinjuan Bai Xiaoping Tao Ling Wang Hui Zhang Jian-Kang Zhu 2020National Science Review2020,7,1:7
11Cross-cultural adaption and psychometric properties of the Chinese version of the Diabetes Behavior Rating Scale: a pilot study显示文摘Self-care behavior plays a major role in diabetic management. However, in China, a satisfactory instrument has not yet been developed to evaluate the compliance of self-care behavior for young patients with type 1 diabetes mellitus(T1 DM). The Diabetes Behavior Rating Scale(DBRS) has a potential to be the first mature instrument. The purpose of this study is to cross-culturally adapt the DBRS, and preliminarily evaluate its psychometric properties. The instrument translation included translation, back translation and culture adaptation. Psychometric properties were assessed in a sample of 116 young patients with T1 DM adapting insulin injection therapy. The Chinese version of the DBRS was divided to four subscales. Cronbach's α for the total scale was 0.92. The mean inter-item and item-total correlations were 0.35 and 0.54 respectively. Test-retest reliability showed good temporal stability(r=0.81, P=0.001). Negative correlations were found between DBRS scores with the Diabetes Distress Scale scores(r=.0.32, P=0.003) and hemoglobin A1 c(HbA1 c) levels(r=.0.36, P=0.002). Higher DBRS scores correlated with better glycemic control. The Chinese insulin injection therapy version of the DBRS is well translated and culturally adapted. 12Phenformin alone or combined with gefitinib inhibits bladder cancer via AMPK and EGFR pathways显示文摘Background:In previous studies,we have shown that the combination of metformin and gefitinib inhibits the growth of bladder cancer cells.Here we examined whether the metformin analogue phenformin,either used alone or in combination with gefitinib,could inhibit growth of bladder cancer cells.Methods:The growth-inhibitory effects of phenformin and gefitinib were tested in one murine and two human bladder cancer cell lines using MTT and clonogenic assays.Effects on cell migration were assessed in a wound healing assay.Synergistic action between the two drugs was assessed using CompuSyn software.The potential involvement of AMPK and EGFR pathways in the effects of phenformin and gefitinib was explored using Western blotting.Results:In MTT and clonogenic assays,phenformin was>10-fold more potent than metformin in inhibiting bladder cancer cell growth.Phenformin also potently inhibited cell migration in wound healing assays,and promoted apop-tosis.AMPK signaling was activated;EGFR signaling was inhibited.Phenformin was synergistic with gefitinib,with the combination of drugs showing much stronger anticancer activity and apoptotic activation than phenformin alone.Conclusions:Phenformin shows potential as an effective drug against bladder cancer,either alone or in combination with gefitinib.Yanjun Huang Sichun Zhou Caimei He Jun Deng Ting Tao Qiongli Su Kwame Oteng Darko Mei Peng Xiaoping Yang 2018Cancer Communications2018,38,1:5
13Edge biomarkers for classification and prediction of phenotypes显示文摘In general,a disease manifests not from malfunction of individual molecules but from failure of the relevant system or network,which can be considered as a set of interactions or edges among molecules.Thus,instead of individual molecules,networks or edges are stable forms to reliably characterize complex diseases.This paper reviews both traditional node biomarkers and edge biomarkers,which have been newly proposed.These biomarkers are classified in terms of their contained information.In particular,we show that edge and network biomarkers provide novel ways of stably and reliably diagnosing the disease state of a sample.First,we categorize the biomarkers based on the information used in the learning and prediction steps.We then briefly introduce conventional node biomarkers,or molecular biomarkers without network information,and their computational approaches.The main focus of this paper is edge and network biomarkers,which exploit network information to improve the accuracy of diagnosis and prognosis.Moreover,by extracting both network and dynamic information from the data,we can develop dynamical network and edge biomarkers.These biomarkers not only diagnose the immediate pre-disease state but also detect the critical molecules or networks by which the biological system progresses from the healthy to the disease state.The identified critical molecules can be used as drug targets,and the critical state indicates the critical point of disease control.The paper also discusses representative biomarker-based methods.ZENG Tao ZHANG WanWei YU XiangTian LIU XiaoPing LI MeiYi LIU Rui CHEN LuoNan 2014Science China(Life Sciences)2014,57,11:5
14The efficacy and safety of nimodipine in acute ischemic stroke patients with mild cognitive impairment: a double-blind, randomized,placebo-controlled trial显示文摘Nimodipine might be effective in subcortical vascular dementia(VaD). Its benefit in preventing further cognitive decline in patients with acute ischemic stroke(AIS) and vascular mild cognitive impairment(VaMCI) remains to be established. In this multicenter, double-blind trial, we randomly assigned 654 eligible patients to nimodipine 30 mg three times a day or placebo. The primary outcome was any cognitive decline defined by the changes on the Mini-Mental State Examination(DMMSE à3) or vascular AD assessment scale cognitive subscale(DADAS-cog ! 4) at 6 months. Secondary outcomes included any distribution shift of DADAS-cog, DMMSE or cognitive improvement defined by DADAS-cog à2, or DMMSE ! 0. The primary outcome in the nimodipine group and placebo group were similar for DMMSE à3(4.18% and 7.22%, respectively, P = 0.15) and DADAS-cog ! 4(8.36% and 8.93% respectively,P = 0.88). The distribution shift of DADAS-cog and DMMSE differed significantly between the two groups(P = 0.03 and P = 0.05 respectively). Cognitive improvement occurred in 55.4% in the nimodipine group and 43.6% in the placebo group measured by DADAS-cog à2(Odds Ratio, 1.54; 95% confidence interval[CI] 1.10–2.14, P < 0.01) or 84.0% and 74.6% respectively by DMMSE ! 0(Odds Ratio, 1.79; 95% CI 1.18–2.70, P < 0.01). Nimodipine was associated with better cognitive function in the memory domain. The adverse events rate was similar in two groups. This study is registered with ClinicalTrials.gov,NCT01220622. 15Protective effect of oxysophoridine on cerebral ischemia/reperfusion injury in mice显示文摘Oxysophoridine, a new alkaloid extracted from Sophora alopecuroides L., has been shown to have a protective effect against ischemic brain damage. In this study, a focal cerebral ischemia/reperfusion injury model was established using middle cerebral artery occlusion in mice. Both 62.5, 125, and 250 mg/kg oxysophoridine, via intraperitoneal injection, and 6 mg/kg nimodipine, via intragastric administration, were administered daily for 7 days before modeling. After 24 hours of reperfusion, mice were tested for neurological deficit, cerebral infarct size was assessed and brain tissue was collected. Results showed that oxysophoridine at 125, 250 mg/kg and 6 mg/kg nimodipine could reduce neurological deficit scores, cerebral infarct size and brain water content in mice. These results provided evidence that oxysophoridine plays a protective role in cerebral ischemia/reperfusion injury. In addition, oxysophoridine at 62.5, 125, and 250 mg/kg and 6 mg/kg nimodipine increased adenosine-triphosphate content, and decreased malondialdehyde and nitric oxide content. These compounds enhanced the activities of glutathione-peroxidase, superoxide dismutase, catalase, and lactate dehydrogenase, and decreased the activity of nitric oxide synthase. Protein and mRNA expression levels of N-methyl-D-aspartate receptor subunit NR1 were markedly inhibited in the presence of 250 mg/kg oxysophoridine and 6 mg/kg nimodipine. 16Integration of small world networks with multi-agent systems for simulating epidemic spatiotemporal transmission显示文摘This study proposes an integrated model based on small world network (SWN) and multi-agent system (MAS) for simulating epidemic spatiotemporal transmission. In this model, MAS represents the process of spatiotemporal interactions among individuals, and SWN describes the social relation network among agents. The model is composed of agent attribute definitions, agent movement rules, neighborhoods, construction of social relation network among agents and state transition rules. The construction of social relation network and agent state transition rules is essential for implementing the proposed model. The decay effects of infection 'memory', distance and social relation between agents are introduced into the model, which are unavailable in traditional models. The proposed model is used to simulate the transmission process of flu in Guangzhou City based on the swarm software platform. The integration model has better performance than the traditional SEIR model and the pure MAS based epidemic model. This model has been applied to the simulation of the transmission of epidemics in real geographical environment. 19Upconversion nanoparticles@AgBiS_(2) core-shell nanoparticles with cancer-cell-specific cytotoxicity for combined photothermal and photodynamic therapy of cancers显示文摘UCNPs@AgBiS_(2) core-shell nanoparticles that AgBiS_(2) coated on the surface of upconversion nanoparticles (UCNPs) was successfully prepared through an ion exchange reaction. The photothermal conversion efficiency of AgBiS_(2) can be improved from 14.7% to 45% due to the cross relaxation between Nd ions and AgBiS_(2). The doping concentration of Nd ions played a critical role in the production of reactive oxygen species (ROS) and enhanced the photothermal conversion efficiency. The NaYF4:Yb/Er/Nd@NaYF4:Nd nanoparticles endows strong upcon-version emissions when the doped concentration of Nd ions is 1% in the inner core, which excites the AgBiS_(2) shell to produce ROS for photodynamic therapy (PDT) of cancer cells. As a result, the as-prepared NaYF4:Yb/Er/ Nd@NaYF4:Nd@AgBiS_(2) core-shell nanoparticles showed combined photothermal/photodynamic therapy (PTT/ PDT) against malignant tumors. 20Characterization of the unprecedented polynya events north of Greenland in 2017/2018 using remote sensing and reanalysis data显示文摘Based on an ice concentration threshold of 90%,it has been identified that two polynya events occurred in the region north of Greenland during the 2017/2018 ice season.The winter event lasted from February 20 to March 3,2018 and the summer event persisted from August 2 to September 5,2018.The minimum ice concentration derived from Advanced Microwave Scanning Radiometer 2(AMSR2)observations was 72%and 65%during the winter and summer events,respectively.The occurrence of both events can be related to strengthened southerly winds associated with an increased east-west zonal surface level air pressure gradient across the north Greenland due to perturbation of mid-troposphere polar vortex.The relatively warm air temperature during the 2017/2018 freezing season in comparison with previous years,together with the occurrence of the winter polynya,formed favourable pre-conditions for ice field fracturing in summer,which promoted the formation of the summer polynya.Diminished southerly winds and increased cover of new ice over the open water were the dominant factors for the disappearance of the winter polynya,whereas increased ice inflow from the north was the primary factor behind the closure of the summer polynya.Sentinel-1 Synthetic Aperture Radar(SAR)images were found better suited than AMSR2 observations for quantification of a new ice product during the polynya event because the SAR images have high potential for mapping of different sea ice regimes with finely spatial resolution.The unprecedented polynya events north of Greenland in 2017/2018 are important from the perspective of Arctic sea ice loss because they occurred in a region that could potentially be the last“Arctic sea ice refuge”in future summers.Ruibo Lei Dawei Gui Zhouli Yuan Xiaoping Pang Ding Tao Mengxi Zhai 2020Acta Oceanologica Sinica2020,39,9:3
