| 2 | Soybean AP1 homologs control flowering time and plant height显示文摘Flowering time and plant height are key agronomic traits that directly affect soybean(Glycine max)yield.APETALA1(AP1)functions as a class A gene in the ABCE model for floral organ development,helping to specify carpel,stamen,petal,and sepal identities.There are four AP1 homologs in soybean,all of which are mainly expressed in the shoot apex.Here,we used clustered regularly interspaced short palindromic repeats(CRISPR)–CRISPR-associated protein 9 technology to generate a homozygous quadruple mutant,gmap1,with loss-of-function mutations in all four GmAP1 genes.Under short-day(SD)conditions,the gmap1 quadruple mutant exhibited delayed flowering,changes in flower morphology,and increased node number and internode length,resulting in plants that were taller than the wild type.Conversely,overexpression of GmAP1a resulted in early flowering and reduced plant height compared to the wild type under SD conditions.The gmap1 mutant and the overexpression lines also exhibited altered expression of several genes related to flowering and gibberellic acid metabolism,thereby providing insight into the role of GmAP1 in the regulatory networks controlling flowering time and plant height in soybean.Increased node number is the trait with the most promise for enhancing soybean pod number and grain yield.Therefore,the mutant alleles of the four AP1 homologs described here will be invaluable for molecular breeding of improved soybean yield. | Liyu Chen Haiyang Nan Lingping Kong Lin Yue Hui Yang Qingsong Zhao Chao Fang Haiyang Li Qun Cheng Sijia Lu Fanjiang Kong Baohui Liu Lidong Dong | 2020 | Journal of Integrative Plant Biology2020,62,12: | 9 |
| 3 | Parallel selection of distinct Tof5 alleles drove the adaptation of cultivated and wild soybean to high latitudes显示文摘Photoperiod responsiveness is a key factor limiting the geographic distribution of cultivated soybean and its wild ancestor.In particular,the genetic basis of the adaptation in wild soybean remains poorly understood.In this study,by combining whole-genome resequencing and genome-wide association studies we identified a novel locus,Time of Flowering 5(Tof5),which promotes flowering and enhances adaptation to high latitudes in both wild and cultivated soybean.By genomic,genetic and transgenic analyses we showed that Tof5 en-codes a homolog of Arabidopsis thaliana FRUITFULL(FUL).Importantly,further analyses suggested that different alleles of Tof5 have undergone parallel selection.The Tof5H1 allele was strongly selected by humans after the early domestication of cultivated soybean,while Tof5H2 allele was naturally selected in wild soybean,and in each case facilitating adaptation to high latitudes.Moreover,we found that the key flowering repressor E1 suppresses the transcription of Tof5 by binding to its promoter.In turn,Tof5 physically associates with the promoters of two important FLOWERING LOCUS T(FT),FT2a and FT5a,to upregulate their transcription and promote flowering under long photoperiods.Collectively,ourfindings provide insights into how wild soybean adapted to high latitudes through natural selection and indicate that cultivated soybean underwent changes in the same gene but evolved a distinct allele that was artificially selected after domestication. | Lidong Dong Qun Cheng Chao Fang Lingping Kong Hui Yang Zhihong Hou Yongli Li Haiyang Nan Yuhang Zhang Qingshan Chen Chunbao Zhang Kun Kou Tong Su Lingshuang Wang Shichen Li Haiyang Li Xiaoya Lin Yang Tang Xiaohui Zhao Sijia Lu Baohui Liu Fanjiang Kong | 2022 | Molecular Plant2022,15,2: | 5 |
| 4 | Structural shifts in the intestinal microbiota of rats treated with cyclosporine A after orthotropic liver transplantation显示文摘Understanding the effect of immunosuppressive agents on intestinal microbiota is important to reduce the mortality and morbidity from orthotopic liver transplantation (OLT). We investigated the relationship between the commonly used immunosuppressive agent cyclosporine A (CSA) and the intestinal microbial variation in an OLT model. The rat samples were divided as follows:(1) N group (normal control);(2) I group (isograft LT, Brown Norway [BN] rat to BN);(3) R group (allograft LT, Lewis to BN rat);and (4) CSA group (R group treated with CSA). The intestinal microbiota was assayed by denaturing gradient gel electrophoresis profiles and by using real-time polymerase chain reaction. The liver histopathology and the alanine/aspartate aminotransferase ratio after LT were both ameliorated by CSA. In the CSA group, the numbers of rDNA gene copies of Clostridium cluster I, Clostridium cluster XIV, and Enterobacteriaceae decreased, whereas those of Faecalibacterium prausnitzii increased compared with the R group. Cluster analysis indicated that the samples from the N, I, and CSA groups were clustered, whereas the other clusters contained the samples from the R group. | 6 | Severity of early allograft dysfunction following donation after circulatory death liver transplantation:a multicentre study显示文摘Background:Early allograft dysfunction(EAD)is associated with decreased graft and patient survival rates.This study aimed to identify the severity of EAD and develop a predictive model for EAD after donation after circulatory death(DCD)liver transplantation(LT).Furthermore,the influence of operative time on EAD incidence was also evaluated.Methods:In this retrospective,multicentre cohort study,nomograms were established based on a single-centre training cohort(n=321)and validated in a 3-center validation cohort(n=501).Results:The incidence rate of EAD was 46.4%(149/321)in the training cohort and 40.5%(203/501)in the validation cohort.Of the 149 EAD patients in the training cohort,77 patients with either elevated alanine aminotransferase(ALT)or aspartate aminotransferase(AST)were classified as having EAD type A,and the rest of the EAD patients were classified as having EAD type B.Recipients with EAD type B had lower graft and patient survival rates than recipients with EAD type A(P=0.043 and 0.044,respectively).We further developed a nomogram to predict EAD(graft weight,cold ischemia time,donor age,model for end-stage liver disease(MELD)score)and another nomogram to predict EAD type B(graft weight,cold ischemia time,MELD score).The nomograms for the prediction of EAD and EAD type B had good discrimination[concordance index(C-index)=0.712(0.666-0.758),0.707(0.641-0.773)]and calibration[Hosmer-Lemeshow(HL)P=0.384,P=0.425]in the validation cohort.An increased operative time(>6 h)was associated with increased EAD and EAD type B incidence in the high-risk group(P=0.005,P=0.020,respectively).Conclusions:EAD type B was associated with decreased graft and patient survival rates.The novel nomograms effectively predicted the incidence of EAD and EAD type B in DCD LT patients. | Kun Wang Di Lu Yuhui Liu Wangyao Li Li Zhuang Zhenyu Ma Qinfen Xie Binhua Pan Yichao Wu Junli Chen Lidan Lin Xiaowen Feng Qiang Wei Xuyong Wei Haiyang Xie Zhengxin Wang Shusen Zheng Xiao Xu | 2021 | Hepatobiliary Surgery and Nutrition2021,10,1: | 4 |

| 8 | SARS-CoV-2 spike protein receptor-binding domain perturbates intracellular calcium homeostasis and impairs pulmonary vascular endothelial cells显示文摘Exposure to the spike protein or receptor-binding domain(S-RBD)of SARS-CoV-2 significantly influences endothelial cells and induces pulmonary vascular endotheliopathy.In this study,angiotensin-converting enzyme 2 humanized inbred(hACE2 Tg)mice and cultured pulmonary vascular endothelial cells were used to investigate how spike protein/S-RBD impacts pulmonary vascular endothelium.Results show that S-RBD leads to acute-to-prolonged induction of the intracellular free calcium concentration([Ca^(2+)]i)via acute activation of TRPV4,and prolonged upregulation of mechanosensitive channel Piezo1 and store-operated calcium channel(SOCC)key component Orai1 in cultured human pulmonary arterial endothelial cells(PAECs).In mechanism,S-RBD interacts with ACE2 to induce formation of clusters involving Orai1,Piezo1 and TRPC1,facilitate the channel activation of Piezo1 and SOCC,and lead to elevated apoptosis.These effects are blocked by Kobophenol A,which inhibits the binding between S-RBD and ACE2,or intracellular calcium chelator,BAPTA-AM.Blockade of Piezo1 and SOCC by GsMTx4 effectively protects the S-RBDinduced pulmonary microvascular endothelial damage in hACE2 Tg mice via normalizing the elevated[Ca^(2+)]i.Comparing to prototypic strain,Omicron variants(BA.5.2 and XBB)of S-RBD induces significantly less severe cell apoptosis.Transcriptomic analysis indicates that prototypic S-RBD confers more severe acute impacts than Delta or Lambda S-RBD.In summary,this study provides compelling evidence that S-RBD could induce persistent pulmonary vascular endothelial damage by binding to ACE2 and triggering[Ca^(2+)]i through upregulation of Piezo1 and Orai1.Targeted inhibition of ACE2-Piezo1/SOCC-[Ca^(2+)]i axis proves a powerful strategy to treat S-RBD-induced pulmonary vascular diseases. | Kai Yang Shiyun Liu Han Yan Wenju Lu Xiaoqian Shan Haixia Chen Changlei Bao Huazhuo Feng Jing Liao Shuxin Liang Lei Xu Haiyang Tang Jason X-J.Yuan Nanshan Zhong Jian Wang | 2023 | Signal Transduction and Targeted Therapy2023,8,8: | 2 |

It is important to recognise how groundwater changes affect the riparian forest ecosystem. This information will not only help us to understand the ecological and hydrological process of the riparian forest but also provide support for ecological recovery of riparian forests and water-resources management of arid inland river basins. This study aims to estimate the suitability of the Water Vegetation Energy and Solute Modelling(WAVES) model to simulate the Ejina Desert riparian forest ecosystem changes,China, to assess effects of groundwater-depth change on the canopy leaf area index(LAI) and water budgets, and to ascertain the suitable groundwater depth for preserving the stability and structure of desert riparian forest. Results demonstrated that the WAVES model can simulate changes to ecological and hydrological processes. The annual mean water consumption of a Tamarix chinensis riparian forest was less than that of a Populus euphratica riparian forest, and the canopy LAI of the desert riparian forest should increase as groundwater depth decreases. Groundwater changes could significantly influence water budgets for T. chinensis and P. euphratica riparian forests and show the positive and negative effects on vegetation growth and water budgets of riparian forests. Maintaining the annual mean groundwater depth at around 1.7-2.7 m is critical for healthy riparian forest growth. | 16 | Study on the Bonding Performance of the Moso Bamboo Dowel Welded to a Poplar Substrate Joint by High-Speed Rotation显示文摘The wood friction welding technique with its high bonding strength,low cost,high efficiency,and without any adhesive has been increasing concern in China.Moso bamboo(Phyllostachys pubescens)and poplar(Populus sp.)are widely planted and used in the furniture industry,interior decoration,and wood structure construction in China.The aim of this work was to investigate the bonding performance of moso bamboo dowel rotation welded joints with different dowel/receiving hole diameter ratios.The results indicated that the ratio of dowel/receiving hole diameter was an important parameter that influenced the welding performance.The bonding strength of the bamboo-to-poplar welded joints at the optimal ratio of 10/7 was as high as 7.50 MPa,which was higher than that of the beech(Fagus sylvatica,L.),schima(Schima superba)dowels and PVAc glued joints.The temperature measurement results showed a peak temperature of bamboo dowel welding as high as 350–360°C.Some differences in the temperature curves between each dowel/hole diameter ratio group were observed at the three different hole depths,such as the friction time,peak temperatures,and stabilization time at the maximum temperature,which could explain the differences in welding strengths between different ratios.The SEM results showed the temperature-induced softening,melting and flowing of cell-interconnected polymer material in the wood and bamboo structure.In addition,the bamboo fibers(mainly vascular bundles)were wrapped to form a dense continuous bonding layer,similar to the reinforced concrete,thus producing a good bonding effect.The Fourier transform-infrared spectroscopy(FT-IR)analyses showed that the high temperature resulted in the increase of the lignin relative content due to the degradation reaction of cellulose in the welding zone,which improved the bonding properties. | Suxia Li Haiyang Zhang Biqing Shu Liangsong Cheng Zehui Ju Xiaoning Lu | 2021 | Journal of Renewable Materials2021,9,7: | 1 |

| 19 | Secure coordinated direct and untrusted relay transmissions via interference engineering显示文摘This paper investigates the physical layer security issue of the coordinated direct and untrusted relay transmissions, considering both uplink and downlink scenarios. To effectively prevent the untrusted relay from eavesdropping on the confidential information it receives, two novel interference-assisted jamming schemes are proposed, where the inter-user interference and the artificial noise are intelligently engineered to degrade the signal reception quality at the untrusted relay, therefore enhancing the physical layer security for the uplink and downlink coordinated transmission, respectively. For each of the two proposed jamming schemes, a lower bound for the ergodic secrecy sum rate(ESSR) is derived in closed form to evaluate the secrecy performance. To obtain more system design insights, we further provide an asymptotic analysis for the ESSR scaling laws over the regimes of the high signal-to-noise ratio and a large number of antennas at the base-station. We carry out computer simulations to confirm the accuracy of the derived theoretical analysis.Our simulation results show that the proposed jamming schemes achieve a similar asymptotic performance limit to the case of using a trusted relay.
