2Spatio-temporal variability of phytoplankton assemblages and its controlling factors in spring and summer in the Subei Shoal of Yellow Sea, China显示文摘The Subei Shoal is a special coastal area with complex physical oceanographic properties in the Yellow Sea.In the present study,the distribution of phytoplankton and its correlation with environmental factors were studied during spring and summer of 2012 in the Subei Shoal of the Yellow Sea.Phytoplankton species composition and abundance data were accomplished by Utermohl method.Diatoms represented the greatest cellular abundance during the study period.In spring,the phytoplankton cell abundance ranged from 1.59×10^3 to 269.78×10^3 cell/L with an average of 41.80×10^3 cell/L,and Skeletonema sp.and Paralia sulcata was the most dominant species.In summer,the average phytoplankton cell abundance was 72.59×10^3 cell/L with the range of 1.78×10^3 to 574.96×10^3 cell/L,and the main dominant species was Pseudo-nitzschia pungens,Skeletonema sp.,Dactyliosolen fragilissima and Chaetoceros curvisetus.The results of a redundancy analysis(RDA)showed that turbidity,temperature,salinity,pH,dissolved oxygen(DO),the ratio of dissolved inorganic nitrogen to silicate and SiO4-Si(DIN/SiO4-Si)were the most important environmental factors controlling phytoplankton assemblages in spring or summer in the Subei Shoal of the Yellow Sea.Yuanzi Huo Honghua Shi Jianheng Zhang Qiao Liu Yuanliang Duan Qing He Kefeng Yu Hongsheng Bi Chunlei Fan Peimin He 2019Acta Oceanologica Sinica2019,38,10:4
3A Multifunctional Anti-Proton Electrolyte for High-Rate and Super-Stable Aqueous Zn-Vanadium Oxide Battery显示文摘Large volumetric expansion of cathode hosts and sluggish transport kinetics in the cathode–electrolyte interface,as well as dendrite growth and hydrogen evolution at Zn anode side are considered as the system problems that cause the electrochemical failure of aqueous Zn-vanadium oxide battery.In this work,a multifunctional anti-proton electrolyte was proposed to synchronously solve all those issues.Theoretical and experimental studies confirm that PEG 400 additive can regulate the Zn^(2+) solvation structure and inhibit the ionization of free water molecules of the electrolyte.Then,smaller lattice expansion of vanadium oxide hosts and less associated by-product formation can be realized by using such electrolyte.Besides,such electrolyte is also beneficial to guide the uniform Zn deposition and suppress the side reaction of hydrogen evolution.Owing to the integrated synergetic modifica-tion,a high-rate and ultrastable aqueous Zn-V_(2)O_(3)/C battery can be constructed,which can remain a specific capacity of 222.8 m Ah g^(-1)after 6000 cycles at 5 A g^(-1),and 121.8 m Ah g^(-1) even after 18,000 cycles at 20 A g^(-1),respectively.Such“all-in-one”solution based on the electrolyte design provides a new strategy for developing high-performance aqueous Zn-ion battery.Yangwu Chen Dingtao Ma Kefeng Ouyang Ming Yang Sicheng Shen Yanyi Wang Hongwei Mi Lingna Sun Chuanxin He Peixin Zhang 2022Nano-Micro Letters2022,14,9:3
4Efficacy of rigosertib, a small molecular RAS signaling disrupter for the treatment of KRAS-mutant colorectal cancer显示文摘Objective:Mutant KRAS,the principal isoform of RAS,plays a pivotal role in the oncogenesis of colorectal cancer by constitutively activating the RAF/MEK/ERK and PI3K/AKT pathways.Effective targeted therapies are urgently needed.We investigated whether rigosertib,a benzyl styryl sulfone RAS signaling disruptor,could selectively kill KRAS-mutant colorectal cancer cells.Methods:CCK-8 was used to determine the cell viability.Patient-derived tumor and cancer cell xenograft models were used to detect the inhibitory efficacy of rigosertib.Flow cytometry was used to evaluate the apoptosis and cell cycle progression.Apoptosis and cell cycle arrest markers were detected by Western blot.DCFH-DA was used to determine the reactive oxygen species.Immunohistochemistry staining and Western blot were performed to characterize RAS signaling markers in colorectal cancer tissues and cells.Results:Rigosertib(RGS)exhibited a cytotoxic effect against colorectal cancer cells,which was greater in KRAS-mutant cells.Furthermore,RGS induced mitotic arrest and oxidative stress-dependent apoptosis in KRAS-mutant DLD1 and HCT116 cells.Besides,RGS disrupted RAS signaling,and the inhibition of RAS/MEK/ERK was independent of cellular oxidative stress.Using patient-derived xenograft models,the response and tumor inhibition of RGS were significantly higher in the KRAS-mutant subgroup,while p-MEK,p-ERK,and p-AKT levels of RGS-treated tumors were significantly decreased.Finally,in a KRAS-mutant,chemotherapy-resistant patient-derived xenograft model,RGS showed a stronger therapeutic effect than the combination standard therapy involving fluoropyrimidine+oxaliplatin/irinotecan+bevacizumab.Conclusions:These data showed that targeting RAS signaling using RGS could be a therapeutic treatment for KRAS-mutant colorectal cancer patients.Xinyi Zhou Qian Xiao Dongliang Fu Haochen Zhang Yang Tang Jinjie He Yeting Hu Xiangxing Kong Fei Teng Xiangrui Liu Ying Yuan Kefeng Ding 2022Cancer Biology & Medicine2022,19,2:2

10Hierarchical Capillary Coating to Biofunctionlize Drug-Eluting Stent for Improving Endothelium Regeneration显示文摘The drug-eluting stent(DES)has become one of the most successful and important medical devices for coronary heart disease,but yet suffers from insufficient endothelial cell(EC)growth and intima repair,eventually leading to treatment failure.Although biomacromolecules such as vascular endothelial growth factor(VEGF)would be promising to promote the intima regeneration,combining hydrophilic and vulnerable biomacromolecules with hydrophobic drugs as well as preserving the bioactivity after harsh treatments pose a huge challenge.Here,we report on a design of hierarchical capillary coating,which composes a base solid region and a top microporous region for incorporating rapamycin and VEGF,respectively.The top spongy region can guarantee the efficient,safe,and controllable loading of VEGF up to 1μg/cm^(2) in 1 minute,providing a distinctive real-time loading capacity for saving the bioactivity.Based on this,we demonstrate that our rapamycin-VEGF hierarchical coating impressively promoted the competitive growth of endothelial cells over smooth muscle cells(ratio of EC/SMC~25)while relieving the adverse impact of rapamycin to ECs.We further conducted the real-time loading of VEGF on stents and demonstrate that the hierarchical combination of rapamycin and VEGF showed remarkable endothelium regeneration while maintaining a very low level of in-stent restenosis.This work paves an avenue for the combination of both hydrophobic and hydrophilic functional molecules,which should benefit the next generation of DES and may extend applications to diversified combination medical devices.Jing Wang Yunfan Xue Jun Liu Mi Hu He Zhang Kefeng Ren Yunbing Wang Jian Ji 2020Research2020,,1:1

14Screening and engineering of high-activity promoter elements through transcriptomics and red fluorescent protein visualization in Rhodobacter sphaeroides显示文摘The versatile photosyntheticα-proteobacterium Rhodobacter sphaeroides,has recently been extensively engineered as a novel microbial cell factory(MCF)to produce pharmaceuticals,nutraceuticals,commodity chemicals and even hydrogen.However,there are no well-characterized high-activity promoters to modulate gene transcription during the engineering of R.sphaeroides.In this study,several native promoters from R.sphaeroides JDW-710(JDW-710),an industrial strain producing high levels of co-enzyme Q10(Q10)were selected on the basis of transcriptomic analysis.These candidate promoters were then characterized by using gusA as a reporter gene.Two native promoters,Prsp_7571 and Prsp_6124,showed 620%and 800%higher activity,respectively,than the tac promoter,which has previously been used for gene overexpression in R.sphaeroides.In addition,a Prsp_7571-derived synthetic promoter library with strengths ranging from 54%to 3200%of that of the tac promoter,was created on the basis of visualization of red fluorescent protein(RFP)expression in R.sphaeroides.Finally,as a demonstration,the synthetic pathway of Q10 was modulated by the selected promoter T334*in JDW-710;the Q10 yield in shake-flasks increased 28%and the production reached 226 mg/L.These well-characterized promoters should be highly useful in current synthetic biology platforms for refactoring the biosynthetic pathway in R.sphaeroides-derived MCFs.Tong Shi Lu Zhang Mindong Liang Weishan Wang Kefeng Wang Yue Jiang Jing Liu Xinwei He Zhiheng Yang Haihong Chen Chuan Li Dongyuan Lv Liming Zhou Biqin Chen Dan Li Li-Xin Zhang Gao-Yi Tan 2021Synthetic and Systems Biotechnology2021,6,4:0

16A rapid method for the detection of humic acid based on the poly(thymine)-templated copper nanoparticles显示文摘This work described a new method for the detection of humic acid(HA) based on the poly(thymine)(polyT)-templated copper nanoparticles(CuNPs). Without the presence of HA, the formation of poly Ttemplated CuNPs could take place, resulting in strong fluorescence emission peaks at 610 nm(upon excitation at 340 nm). On the other hand, when HA was present, strong interaction between HA and Cu^(2+)took place, which then hampered the effective formation of fluorescent CuNPs, leading to the decrease in fluorescence intensity. Furthermore, under the optimal experimental conditions, the method exhibited a high specificity to HA with a detection limit of 0.4 mg/L. This work has demonstrated a low-cost and convenient method that could be accomplished within 10 min.
