2Performing optical logic operations by a diffractive neural network显示文摘Optical logic operations lie at the heart of optical computing,and they enable many applications such as ultrahighspeed information processing.However,the reported optical logic gates rely heavily on the precise control of input light signals,including their phase difference,polarization,and intensity and the size of the incident beams.Due to the complexity and difficulty in these precise controls,the two output optical logic states may suffer from an inherent instability and a low contrast ratio of intensity.Moreover,the miniaturization of optical logic gates becomes difficult if the extra bulky apparatus for these controls is considered.As such,it is desirable to get rid of these complicated controls and to achieve full logic functionality in a compact photonic system.Such a goal remains challenging.Here,we introduce a simple yet universal design strategy,capable of using plane waves as the incident signal,to perform optical logic operations via a diffractive neural network.Physically,the incident plane wave is first spatially encoded by a specific logic operation at the input layer and further decoded through the hidden layers,namely,a compound Huygens’metasurface.That is,the judiciously designed metasurface scatters the encoded light into one of two small designated areas at the output layer,which provides the information of output logic states.Importantly,after training of the diffractive neural network,all seven basic types of optical logic operations can be realized by the same metasurface.As a conceptual illustration,three logic operations(NOT,OR,and AND)are experimentally demonstrated at microwave frequencies.Chao Qian Xiao Lin Xiaobin Lin Jian Xu Yang Sun Erping Li Baile Zhang Hongsheng Chen 2020Light(Science & Applications)2020,9,1:8
3Breast cancer stem-like cells can promote metastasis by activating platelets and down-regulating antitumor activity of natural killer cells显示文摘OBJECTIVE:To investigate whether cancer stem cells(CSCs) more efficiently activating platelets and evading immune surveillance than non-CSCs thus promoting metastasis.METHODS:We enriched and identified sphereforming cells(SFCs) and coincubated washed platelets with several platelet activators including collagen,4T1 and SFCs.Platelet-coating tumor cells,platelet activation and TGF-β1 release were analyzed.Then natural kell cells(NK) were incubated with supernatants of different activated platelet samples what we called sample release(SR).The degranulation assay and NKG2 D expression on NK cells were conducted by flow cytometry.Finally tissue factor(TF) expression of SFCs or 4T1 were evaluated by western blot.RESULTS:Breast cancer cell line 4T1 could form spheres in serum-free medium at low adherence.Sphere-forming cells expressed high levels of the CD24-/lowCD44 + stem cell phenotype.Both sphere-forming cells or 4T1 were coated with abundant platelets while sphere-forming cells induced significantly higher expression of platelet activating receptor CD62 p than 4T1 did(P < 0.01).And sphere-forming cells induced platelets to produce more TGF-β1 than 4T1 did(P < 0.01).Furthermore,sample releases induced by sphere-forming cells caused more vigorous inhibition of NK cells antitumor reactivity(P < 0.05) and reduced NKG2 D expression(P < 0.01).The final results showed that sphere-forming cells expressed higher levels of TF than 4T1(P < 0.05).CONCLUSION:Our findings indicate that CSCs could efficiently activate platelets,induce platelets to secrete more TGF-β1,decrease NKG2 D expression and inhibit antitumor activity of NK cell,compared with 4T1.And higher levels of TF expression of CSCs may account for this correlation of CSCs and platelets.Wang Shuo Zhang Ying Cong Weihong Liu Jie Zhang Yuren Fan Huiting XuYonggang Lin Hongsheng 2016Journal of Traditional Chinese Medicine2016,36,4:5
4Spiral Field Generation in Smith-Purcell Radiation by Helical Metagratings显示文摘Moving electrons interacting with media can give rise to electromagnetic radiations and has been emerged as a promising platform for particle detection,spectroscopies,and free-electron lasers.In this letter,we investigate the Smith-Purcell radiation from helical metagratings,chiral structures similar to deoxyribonucleic acid(DNA),in order to understand the interplay between electrons,photons,and object chirality.Spiral feld patterns can be generated while introducing a gradient azimuthal phase distribution to the induced electric dipole array at the cylindrical interface.Experimental measurements show efcient control over angular momentum of the radiated feld at microwave regime,utilizing a phased electromagnetic dipole array to mimic moving charged particles.Te angular momentum of the radiated wave is determined solely by the handedness of the helical structure,and it thus serves as a potential candidate for the detection of chiral objects.Our fndings not only pave a way for design of orbital angular momentum free-electron lasers but also provide a platform to study the interplay between swif electrons with chiral objects.Liqiao Jing Zuojia Wang Xiao Lin Bin Zheng Su Xu Lian Shen Yihao Yang Fei Gao Min Chen Hongsheng Chen 2019Research2019,,1:4

6A novel inhibitor of N^(6)-methyladenosine demethylase FTO induces m RNA methylation and shows anti-cancer activities显示文摘N^(6)-methyladenosine(m^(6)A)modification is critical for m RNA splicing,nuclear export,stability and translation.Fat mass and obesity-associated protein(FTO),the first identified m^(6)A demethylase,is critical for cancer progression.Herein,we developed small-molecule inhibitors of FTO by virtual screening,structural optimization,and bioassay.As a result,two FTO inhibitors namely 18077 and 18097 were identified,which can selectively inhibit demethylase activity of FTO.Specifically,18097 bound to the active site of FTO and then inhibited cell cycle process and migration of cancer cells.In addition,18097 reprogrammed the epi-transcriptome of breast cancer cells,particularly for genes related to P53 pathway.18097 increased the abundance of m^(6)A modification of suppressor of cytokine signaling1(SOCS1)m RNA,which recruited IGF2 BP1 to increase m RNA stability of SOCS1 and subsequently activated the P53 signaling pathway.Further,18097 suppressed cellular lipogenesis via downregulation of peroxisome proliferator-activated receptor gamma(PPARγ),CCAAT/enhancer-binding protein alpha(C/EBPa),and C/EBPβ.Animal studies confirmed that 18097 can significantly suppress in vivo growth and lung colonization of breast cancer cells.Collectively,we identified that FTO can work as a potential drug target and the small-molecule inhibitor 18097 can serve as a potential agent against breast cancer.Guoyou Xie Xu-Nian Wu Yuyi Ling Yalan Rui Deyan Wu Jiawang Zhou Jiexin Li Shuibin Lin Qin Peng Zigang Li Hongsheng Wang Hai-Bin Luo 2022Acta Pharmaceutica Sinica B2022,12,2:4
