| 2 | Composition and phase structure dependence of mechanical and magnetic properties for AlCoCuFeNi_x high entropy alloys显示文摘In this study, the effects of composition and phase constitution on the mechanical properties and magnetic performance of AlCoCuFeNix(x = 0.5, 0.8, 1.0, 1.5, 2.0, 3.0 in molar ratio) high entropy alloys(HEAs)were investigated. The results show that Ni element could lead to the evolution from face centered cubic(FCC), body centered cubic(BCC) and ordered BCC coexisting phase structure to a single FCC phase. The change of phase constitution enhances the plasticity but reduces the hardness and strength. One of the interesting points is the excellent soft magnetic properties of AlCoCuFeNixHEAs. Soft magnetic performance is dependent on composition and phase transition. AlCoCuFeNi1.5 alloy, achieving a better balance of mechanical and magnetic properties, could be applied as structure materials and soft magnetic materials(SMMs). High Curie temperature(>900 K) and strong phase stability below 1350 K of AlCoCuFeNi0.5 alloy confirm its practicability in a high-temperature environment. | 7 | Fibroblast growth factor 9 promotes kidney cell proliferation via WNT signaling-mediated activation of ANXA4显示文摘Fibroblast growth factors(FGFs)play pivotal roles in cell migration and proliferation.However,the identity of the FGF that plays a dominant role in kidney cell proliferation remains unclear.Therefore,in this study,we investigated the dominant FGF among all FGFs.To this end,RNA-sequencing,qRT-PCR,western blotting,and ChIP assays were performed.FGF9 showed the highest expression among all FGFs,and its overexpression significantly promoted proliferation in the mouse kidney cell line C57BL/6 and increased JNK and AKT phosphorylation levels.Further,RNA-seq analysis identified 365 upregulated and 276 downregulated genes in FGF9-overexpressed cells.These differentially expressed genes were classified primarily into 20 biological pathways and were enriched in 31 gene ontology terms.qRT-PCR revealed that the expression of WNT and NF-κB signaling genes,as well as ANXA4 expression patterns,correlated with the RNA-seq data,while FGF9-overexpressed cells accumulated moreβ-catenin,a key WNT signaling protein,compared to control cells.Moreover,downregulation of the gene that encodesβ-catenin or ANXA4 inhibited C57BL/6 cell proliferation.Additionally,the expression of ANXA4 was lower in CTNNB1-knockdown cells than in the control group.Additionally,the ChIP assay revealed that a transcription factor complex containing TCF4 andβ-catenin directly binds to the ANXA4 promoter.Taken together,these results suggest a role of FGF9 in the regulation of kidney cell migration.These findings may prove useful in the development of future therapies. | TING LI XINHUI SUN NANNAN LI HONGMIN GUO | 2021 | BIOCELL2021,45,4: | 1 |

| 14 | Cross-reactivity of anti-programmed death ligand 2 polyclonal antibody in mouse tissues显示文摘The inhibitory co-receptor programmed death 1 (PD-1, encoded by pdcd1) and its two ligands PD-L1 and PD-L2 comprise an important immune inhibitory signaling pathway for defense against microbes and for self-tolerance. Unlike other members of the B7-CD28 family, expression of PD-L1 and PD-L2 is not limited to the immune system. In this study, we determined that a polyclonal antibody (pAb) (R&D Systems) against extracellular domains of mouse PD-L2 (mPD-L2) could recognize anti- gen(s) in diverse mouse tissues, including the anterior and intermediate pituitary gland, olfactory bulbs and olfactory epitheli- um, tongue epithelium, keratinized epithelial cells and skin and whisker hair follicles. These findings differed from previous reports of mPD-L2 localization. Reverse transcription PCR and Western blot analyses, however, were unable to detect any mPD-L2 transcripts or proteins of the 25-kD predicted molecular weight in RNA and protein extracts, respectively, from the above tissues, suggesting that the anti-mPD-L2 pAb cross-reacts with certain novel antigen(s). Developmental studies revealed that the earliest expression of mPD-L2-like antigen was in the olfactory epithelium at embryonic day 12.5 (E12.5). At E14.5, mPD-L2-like antigen was present in the skin, tongue and follicles of the skin and whiskers. The distribution patterns of mPD-L2-like antigen remained similar from E18.5 to adulthood. The results of bioinformatic analysis and other experiments suggested neural cell adhesion molecule and hemicentin-1 as candidate proteins with cross-reactivity to the anti-mPD-L2 pAb. These results demonstrate that care is required in interpreting staining patterns generated when anti-PD-L2 pAb is used to locate PD-L2-expressing cells in the central nervous system and epithelial tissues, such as the olfactory epithelium. | 16 | Identification of prognostic molecular subtypes and model based on CD8+ T cells for lung adenocarcinoma显示文摘Background:Cytotoxic T lymphocytes(CD8+T)cells function critically in mediating anti-tumor immune response in cancer patients.Characterizing the specific functions of CD8+T cells in lung adenocarcinoma(LUAD)could help better understand local anti-tumor immune responses and estimate the effect of immunotherapy.Methods:Gens related to CD8+T cells were identified by cluster analysis based on the single-cell sequencing data of three LUAD tissues and their paired normal tissues.Weighted gene co-expression network analysis(WGCNA),consensus clustering,differential expression analysis,least absolute shrinkage and selection operator(LASSO)and Cox regression analysis were conducted to classify molecular subtypes for LUAD and to develop a risk model using prognostic genes related to CD8+T cells.Expression of the genes in the prognostic model,their effects on tumor cell invasion,and interactions with CD8+T cells were verified by cell experiments.Results:This study defined two LUAD clusters(CD8+0 and CD8+1)based on CD8+T cells,with cluster CD8+0 being significantly associated with the prognosis of LUAD.Three heterogeneous subtypes(clusters 1,2,and 3)differing in prognosis,genome mutation events,and immune status were categorized using 42 prognostic genes.A prognostic model created based on 11 significant genes(including CD200R1,CLEC17A,ZC3H12D,GNG7,SNX30,CDCP1,NEIL3,IGF2BP1,RHOV,ABCC2,and KRT81)was able to independently estimate the death risk for patients in different LUAD cohorts.Moreover,the model also showed general applicability in external validation cohorts.Low-risk patients could benefit more from taking immunotherapy and were significantly related to the resistance to anticancer drugs.The results from cell experiments demonstrated that the expression of CD200R1,CLEC17A,ZC3H12D,GNG7,and SNX30 was significantly downregulated,while that of CDCP1,NEIL3,IGF2BP1,RHOV,ABCC2 and KRT81 was upregulated in LUAD cells.Inhibition of CD200R1 greatly increased the invasiveness of the LUAD cells,but inhibiting CDCP1 expression weakened the invasion ability of LUAD cells.Conclusion:This study defined two prognostic CD8+T cell clusters and classified three heterogeneous molecular subtypes for LUAD.A prognostic model predictive of the potential effects of immunotherapy on LUAD patients was developed. | HONGMIN CAO YING XUE FEI WANG GUANGYAO LI YULAN ZHEN JINGWEN GUO | 2024 | BIOCELL2024,48,3: | 0 |
| 17 | Effects of selective cyclooxygenase-2 inhibitor on C6 glioma cell proliferation and apoptosis显示文摘BACKGROUND:Studies have shown that cyclooxygenase-2 is associated with proliferation and apoptosis of glioma cells. OBJECTIVE:To investigate the effects of selective cyclooxygenase-2 inhibitor celecoxib on proliferation and apoptosis of C6 glioma cells in vitro. DESIGN,TIME AND SETTING:A cellular,molecular,controlled study was performed at the Central Laboratory and Room of Electron Microscope,Medical School,Xi'an Jiaotong University,China from March 2007 to March 2008. MATERIALS:C6 glioma cells during in vitro log phase were assigned to control and experimental groups.Celecoxib(Pfizer,USA),dimethyl sulfoxide(Sigma,USA),and MTT(Sigma,USA) were used for this study. METHODS:The control group was subdivided into blank control and dimethyl sulfoxide control groups.C6 glioma cells in the blank control and dimethyl sulfoxide control groups were incubated in Dulbecco's modified Eagle's medium supplemented with 10%calf serum and 0.3%dimethyl sulfoxide respectively.C6 glioma cells in the experimental group were separately treated with 60,80, and 100μmol/L celecoxib. MAIN OUTCOME MEASURES:Activity of C6 glioma cells was examined by MTT assay.C6 glioma cell cycle and apoptosis were determined by annexin V-fluorescein isothiocyanate/propidium iodide double-staining,followed by flow cytometry.Morphology and ultrastructure of C6 glioma cells were observed with an inverted microscope and a transmission electron microscope,respectively. RESULTS:Compared with the blank control group,cell density was reduced,adherence ability weakened,and irregular nuclei were visible,with the presence of chromatin condensation, margination,and some apoptotic bodies in the experimental group.Activity of C6 glioma cells was significantly decreased(P<0.05),cell number was reduced during S phase,cell number was significantly increased during G_2/M phase(P<0.01),and the apoptotic rate was significantly increased(P<0.05) in the experimental group.These results were displayed in a dose- and time-dependent fashion.The outcomes were obvious in the 100μmol/L celecoxib group following 72 hours of treatment. | 19 | The principle,application,and prospect of using the uranium comminution age to indicate the catchment erosion depth显示文摘Catchment erosion not only shapes various landforms of the earth’s surface but also affects the rates of chemical weathering and CO_(2) absorption by controlling the exposure of rock fragments,and thus acts as a regulator of the global climate.Therefore,studying the occurrence and controlling factors of catchment erosion can improve our understanding of topographic evolution and long-term climate change.This paper introduces the“comminution age”method based on (^(234)U/^(238)U) of sediment detrital components and its applications to indicate the catchment erosion depth,which is widely applied to different depositional environments.In tectonically stable regions,where there is shallow erosion,the sediment output from the catchment is usually accompanied by a low (^(234)U/^(238)U).On the other hand,where there is deep erosion,the (^(234)U/^(238)U) of the downstream sediments is usually high.In tectonically active regions,deep erosion is common,and the sediment erosion process is subject to rapid material transport,such as landslide-induced sediment displacement.Since the sediment exposure history is short,the (^(234)U/^(238)U) of the detrital components is close to 1,thus reaching“secular equilibrium”.The comminution age method is promising in terms of tracing catchment erosion depths at the orbital scale.Furthermore,this method is also sensitive to the erosion changes caused by rapid climate changes and human activities. | Hongmin XU Chao LI Zhifei DUAN Yulong GUO Shouye YANG | 2023 | Science China Earth Sciences2023,66,7: | 0 |
| 20 | Finite element simulation of the micromachining of nanosized-silicon-carbide-particle reinforced composite materials based on the cohesive zone model显示文摘A finite element method based on the cohesive zone model was used to study the micromachining process of nanosized silicon-carbide-particle(SiCp) reinforced aluminum matrix composites. As a hierarchical multiscale simulation method, the parameters for the cohesive zone model were obtained from the stress-displacement curves of the molecular dynamics simulation. The model considers the random properties of the siliconcarbide-particle distribution and the interface of bonding between the silicon carbide particles and the matrix.The machining mechanics was analyzed according to the chip morphology, stress distribution, cutting temperature, and cutting force. The simulation results revealed that the random distribution of nanosized SiCp causes non-uniform interaction between the tool and the reinforcement particles.
