2In situ localization of proteinase inhibitor mRNA in rice plant challenged by brown planthopper显示文摘Proteinase inhibitor (PI) mRNA was localized by in situ hybridization in tissue sections of root, stem and leaf of the resistant rice (B5) plant fed by brown planthopper nymphs. In the rice material without BPH feeding, PI gene was expressed in the root, stem and leaf, while the abundance of PI mRNA was low. In the rice material fed by BPH, PI gene was expressed substantially in the parenchyma of rice stem and leaf, but weakly in the root. The results indicated that the PI gene was up-regulated in the rice plant challenged by brown planthopper. 319世纪80年代以来全球温度变暖趋势一致性进一步加强显示文摘升级了新的1850年以来的全球陆地气温数据集(C-LSAT2.0),结合美国NOAA/NCEI研发的ERSSTv5,将全球表面温度(CMST)观测数据集延长至1854~2019年,为全球气候变化研究提供了一个新的基准数据.对比发现,基于CMST的全球温度变化序列在1880年以前略高于其他几个全球序列,差异主要来源于采用不同海温数据所致,各个序列之间存在结构性不确定性;1880年之后, 5个全球表面温度观测序列的一致性非常高,并有显著一致的变暖趋势,具有高可靠性.基于C-LSAT2.0和CMST,对1880~2019年全球变暖趋势进行了估计,结果表明:近140年, 120年, 60年和40年陆地平均气温增暖趋势分别为:0.103±0.016, 0.115±0.020, 0.252±0.035和0.293±0.055°C/10 a;全球表面温度增暖趋势分别为:0.072±0.010, 0.084±0.011, 0.150±0.019和0.185±0.032°C/10 a.对1900~2018年全球年均温度EOF分析表明,前两个特征向量明显地反映了全球温度变化的主要模态:即全球一致升温模态和与太平洋年代际振荡(IPO)密切相关的模态.说明近120年全球温度变化主要由外部强迫(人类活动)和自然变率(IPO)控制.李庆祥 孙文彬 Boyin Huang 董文杰 Xiaolan Wang 翟盘茂 Phil Jones 2020Science Bulletin2020,65,20:7
4Effect of glutathione on in vivo biodistribution and clearance of surface-modified small Pd nanosheets显示文摘Plasmonic Pd nanosheets have been emerging as promising materials for application in near-infrared(NIR) photothermal therapy(PTT) of cancer. However, animal in mice studies indicated that the original synthesized poly(vinylpyrrolidone)(PVP)-protected small Pd nanosheets(Pd-PVP) and some further surface-modified small Pd nanosheets such as Pd-PEG(SH) easily accumulated in reticuloendothelial system(RES) organs(liver, spleen, etc.) and were difficult to be cleared from these organs quickly. In the work, we surprisingly found that glutathione(GSH) could promote the clearance of surface-modified small Pd nanosheets(e.g. Pd-PVP, Pd-PEG(SH) and Pd-GSH) from the RES organs efficiently. The effects of GSH on the biodistribution and clearance of different surface-modified Pd nanosheets were investigated. Our results indicated that these surface-modified Pd nanosheets with or without GSH added caused no morbidity at target primary organs, and GSH can promote the clearance of different surface-modified Pd nanosheets in the order of Pd-PVP≈Pd-PEG(SH)>Pd-GSH. 5Gas leakage mechanism in bedded salt rock storage cavern considering damaged interface显示文摘During the long-time operation of salt rock storage cavern,between its formations,damaged interfaces induced by discontinuous creep deformations between adjacent layers will possibly lead to serious gas leakage.In this paper,damaged interfaces are considered as main potential leakage path:firstly in meso-level,gas flow rule along the interface is analyzed and the calculation of equivalent permeability is discussed.Then based on porous media seepage theory,gas leakage simulation model including salt rock,cavity interlayers and interface is built.With this strategy,it is possible to overcome the disadvantage of simulation burden with porous-fractured double medium.It also can provide the details of gas flowing along the damaged zones.Finally this proposal is applied to the salt cavern in Qianjian mines(East China).Under different operation pressures,gas distributions around two adjacent cavities are simulated;the evolvement of gas in the interlayers and salt rock is compared.From the results it is demonstrated that the domain of creep damage area has great influence on leakage range.And also the leakage in the interface will accelerate the development of leakage in salt rock.It is concluded that compared with observations,this new strategy provides closer answers.The simulation result proves its validity for the design and reasonable control of operating pressure and tightness evaluation of group bedded salt rock storage caverns.Jun Xiong Xiaolan Huang Hongling Ma 2015Petroleum2015,1,4:3
6Genetic dissection of husk number and length across multiple environments and fine-mapping of a major-effect QTL for husk number in maize(Zea mays L.)显示文摘Husk number(HN)and husk length(HL)influence the mechanical harvesting of maize grain.We investigated the genetic basis of HN and HL using a population of 204 recombinant inbred lines phenotypically evaluated in five environments.The two husk traits showed broad phenotypic variation and high heritability.Nine stable quantitative trait loci(QTL)were identified by single-environment mapping,comprising four QTL for HN and five for HL,and three QTL explained>10%of the phenotypic variation.Joint mapping revealed 22 additive QTL and 46 epistatic QTL.Both additive and epistatic(additive×additive)effects as well as a few large-effect QTL and some minor-effect QTL appeared to contribute to the genetic architecture of HN and HL.The QTL for HN located on chromosome 7,q HN7,which accounted for^20%of phenotypic variation,was detected in all five environments.q HN7 was fine-mapped to a 721.1 kb physical region based on the maize B73 Ref Gen_v3 genome assembly.Within this interval,four genes associated with plant growth and development were selected as candidate genes.The results will be useful for improvement of maize husk traits by molecular breeding and provide a basis for the cloning of q HN7.Guangfei Zhou Yuxiang Mao Lin Xue Guoqing Chen Huhua Lu Mingliang Shi Zhenliang Zhang Xiaolan Huang Xudong Song Derong Hao 2020The Crop Journal2020,8,6:2

8Depression and stress levels increase risk of liver cancer through epigenetic downregulation of hypocretin显示文摘Recent studies suggest that Hypocretin (HCRT, Orexin) are involved in stress regulation of depression through the hypothalamic-pituitary-adrenal (HPA) axis. However, the molecular mechanism by which Hypocretin regulate neurobiological responses is unknown. Herein, the effects of chronic stress on the epigenetic modification of HCRT and its association with depression were explored with regard to a potential role in cancer progression. In the study, Sprague Dawley (SD) rats were used to establish an animal model of cancer with depression by administrating n-nitrosodiethylamine (DEN) and chronic unpredictable mild stress (CUMS). RNA-sequencing was used to detect differentially expressed genes in the hippocampus of rats and quantitative real-time polymerase chain reaction (qRT-PCR) was used to validate the results of RNA-sequencing. The status of HCRT promoter methylation was assessed by methylation specific polymerase chain reaction. Behavioral tests showed that rats exposed to CUMS had significant depressive-like behaviors. The number of liver tumors and tumor load in depressed rats exposed to CUMS was higher than in SD rats without CUMS. RNA-sequencing revealed that HCRT was one of the most siginificantly downregulated gene in the hippocampus of SD rats with CUMS compared to non-stressed group, which was validated by qRT-PCR. HCRT mRNA expression was downregulated and the promoter for HCRT was hyper-methylated in those with depression. These results identified a critical role for chronic psychological stressors in tumorigenesis and cancer progression, via epigenetic HCRT downregulation. 9Observed levels and trends of gaseous SO_2 and HNO_3 at Mt.Waliguan,China:Results from 1997 to 2009显示文摘Long-term measurements of SO2 and HNO3,particularly those from the background sites,are rarely reported.We present for the first time the long-term measurements of SO2 and HNO3 at Waliguan(WLG),the only global baseline station in the back-land of the Eurasian Continent.The concentrations of SO2 and HNO3 were observed at WLG from 1997 to 2009.The observed annual mean concentrations of SO2 and HNO3 at WLG were 1.28±0.41 and 0.22±0.19μg/m3,respectively.The HNO3 concentrations were much higher in warmer seasons than in colder seasons,while the SO2 concentrations showed a nearly reversed seasonal pattern.In most months,the concentration of HNO3 was significantly correlated with that of SO2,suggesting that some common factors influence the variations of both gases and the precursors of HNO3 may partially be from the SO2-emitting sources.The SO2 concentration had a very significant(P < 0.0001) decreasing trend(-0.2μg/(m3·yr)) in 1997-2002,but a significant(P < 0.05) increasing trend(+0.06 μg/(m 3 ·yr)) in 2003-2009.The HNO3 concentration showed no statistically significant trend during 1997-2009.While the decrease of SO2 in 1997-2002 agrees with the trend of global SO2 emissions,the increase in 2003-2009 is not consistent with the decreasing trends in many other regions over the world.Trajectory analysis suggests that the airmasses from the northern Qinghai-Tibetan Plateau and the Takla Makan Desert regions contributed significantly to the increasing trends of SO2 and HNO3 at WLG in 2003-2009,with a rate of +0.13μg/(m3·yr) and +0.007μg/(m3·yr),respectively.Weili Lin Xiaobin Xu Xiaolan Yu Xiaochun Zhang Jianqing Huang 2013Journal of Environmental Sciences2013,25,4:1

11Inhibition of aberrant Hif1αactivation delays intervertebral disc degeneration in adult mice显示文摘The intervertebral disc(IVD) is the largest avascular tissue. Hypoxia-inducible factors(HIFs) play essential roles in regulating cellular adaptation in the IVD under physiological conditions. Disc degeneration disease(DDD) is one of the leading causes of disability, and current therapies are ineffective. This study sought to explore the role of HIFs in DDD pathogenesis in mice. The findings of this study showed that among HIF family members, Hif1α was significantly upregulated in cartilaginous endplate(EP) and annulus fibrosus(AF) tissues from human DDD patients and two mouse models of DDD compared with controls. Conditional deletion of the E3 ubiquitin ligase Vhl in EP and AF tissues of adult mice resulted in upregulated Hif1α expression and age-dependent IVD degeneration. Aberrant Hif1α activation enhanced glycolytic metabolism and suppressed mitochondrial function. On the other hand, genetic ablation of the Hif1α gene delayed DDD pathogenesis in Vhl-deficient mice. Administration of 2-methoxyestradiol(2ME2), a selective Hif1α inhibitor, attenuated experimental IVD degeneration in mice. 18Design of single-phased magnesium alloys with typically high solubility rare earth elements for biomedical applications:Concept and proof显示文摘Rare earth elements(REEs)have been long applied in magnesium alloys,among which the mischmetal-containing WE43 alloy has already got the CE mark approval for clinical application.A considerable amount of REEs(7 wt%)is needed in that multi-phased alloy to achieve a good combination of mechanical strength and corrosion resistance.However,the high complex RE addition accompanied with multiple second phases may bring the concern of biological hazards.Single-phased Mg-RE alloys with simpler compositions were proposed to improve the overall performance,i.e.,“Simpler alloy,better performance”.The single-phased microstructure can be successfully obtained with typical high-solubility REEs(Ho,Er or Lu)through traditional smelting,casting and extrusion in a wide compositional range.A good corrosion resistance with a macroscopically uniform corrosion mode was guaranteed by the homogeneously single-phased microstructure.The bimodal-grained structure with plenty of sub-grain microstructures allow us to minimize the RE addition to<1 wt%,without losing mechanical properties.The single-phased Mg-RE alloys show comparable mechanical properties to the clinically-proven Mg-based implants.They exhibited similar in-vitro and in-vivo performances(without local or systematic toxicity in SD-rats)compared to a high purity magnesium.In addition,metal elements in our single-phased alloys can be gradually excreted through the urinary system and digestive system,showing no consistent accumulation of RE in main organs,i.e.,less burden on organs.The novel concept in this study focuses on the simplification of Mg-RE based alloys for biomedical purpose,and other biodegradable metals with single-phased microstructures are expected to be explored.Dong Bian Xiao Chu Jin Xiao Zhipei Tong He Huang Qinggong Jia Jianing Liu Wenting Li Hui Yu Yue He Limin Ma Xiaolan Wang Mei Li Tao Yang Wenhan Huang Chi Zhang Mengyu Yao Yu Zhang Zhigang Xu Shaokang Guan Yufeng Zheng 2023Bioactive Materials2023,,4:1
