| 1 | Microvesicles(MIVs)secreted from adipose-derived stem cells(ADSCs)contain multiple microRNAs and promote the migration and invasion of endothelial cells显示文摘Extracellular vesicles(EVs)such as microvesicles(MIVs)play an important role in intercellular communications.MIVs are small membrane vesicles sized 100e1000 nm in diameter that are released by many types of cells,such as mesenchymal stem cells(MSCs),tumor cells and adipose-derived stem cells(ADSC).As EVs can carry out autocrine and paracrine functions by controlling multiple cell processes,it is conceivable that EVs can be used as delivery vehicles for treating several clinical conditions,such as to improve cardiac angiogenesis after myocardial infarction(MI).Here,we seek to investigate whether ADSC-derived MIVs contain microRNAs that regulate angiogenesis and affect cell migration of endothelial cells.We first characterized the ADSC-derived MIVs and found that the MIVs had a size range of 100 e300 nm,and expressed the MIV marker protein Alix.We then analyzed the microRNAs in ADSCs and ADSC-derived MIVs and demonstrated that ADSC-derived MIVs selectively released a panel of microRNAs,several of which were related to angiogenesis,including two members of the let-7 family.Furthermore,we demonstrated that ADSC-derived MIVs promoted the cell migration and invasion of the HUVEC endothelial cells.The PKH26-labeled ADSC-derived MIVs were effectively uptaken into the cytoplasm of HUVEC cells.Collectively,our results demonstrate that the ADSC-derived MIVs can promote migration and invasion abilities of endothelial cells,suggesting pro-angiogenetic potential.Future studies should focus on investigating the roles and mechanisms through which ADSC-derived MIVs regulate angiogenesis. | Bo Huang Lin-Feng Huang Ling Zhao Zongyue Zeng Xi Wang Daigui Cao Lijuan Yang Zhenyu Ye Xian Chen Bin Liu Tong-Chuan He Xiaozhong Wang | 2020 | Genes & Diseases2020,7,2: | 8 |
| 2 | In vivo evaluation of urokinase-loaded hollow nanogels for sonothrombolysis on suture embolization-induced acute ischemic stroke rat model显示文摘The urokinase-type plasminogen activator(uPA)loaded hollow nanogels(nUK)were synthesized by a one-step reaction of glycol chitosan and aldehyde capped poly(ethylene oxide).The resultant formulation is sensitive to diagnostic ultrasound(US)of 2 MHz.Herein,we evaluated the in vivo sonothrombolysis performance of the nUK on acute ischemic stroke rat model which was established by suture embolization of middle cerebral artery(MCA).Via intravenous(i.v.)administration,the experimental data prove a controlled release of the therapeutic protein around the clots under ultrasound stimulation,leading to enhanced thrombolysis efficiency of the nUK,evidenced from smaller infarct volume and better clinical scores when compared to the i.v.dose of free uPA no matter with or without US intervention.Meanwhile,the preservation ability of the nanogels not only prolonged the circulation duration of the protein,but also resulted in the better blood-brain barrier protection of the nUK formulation,showing no increased risk on the hemorrhagic transformation than the controls.This work suggests that the nUK is a safe sonothrombolytic formulation for the treatment of acute ischemic stroke. | Yuming Teng Haiqiang Jin Ding Nan Mengnan Li Chenghe Fan Yuanyuan Liu Pu Lv Wei Cui Yongan Sun Hongjun Hao Xiaozhong Qu Zhenzhong Yang Yining Huang | 2018 | Bioactive Materials2018,3,1: | 7 |
| 3 | A novel procedure for pollen-based quantitative paleoclimate reconstructions and its application in China显示文摘Traditionally, the evaluation of pollen-based quantitative paleoclimate reconstructions focuses on the ability of calibration sets to infer present climatic conditions and/or the similarity between fossil and modern assemblages. Objective criteria for choosing the most appropriate climate parameter(s) to be reconstructed at a specific site are thus lacking. Using a novel approach for testing the statistical significance of a quantitative reconstruction using random environmental data, in combination with the advantageous large environmental gradients, abundant vegetation types and comprehensive modern pollen databases in China, we describe a new procedure for pollen-based quantitative paleoclimatic reconstructions. First, the most significant environmental variable controlling the fossil pollen assemblage changes is identified. Second, a calibration set to infer changes in this targeted variable is built up, by limiting the modern ranges of other environmental variables. Finally, the pollen-based quantitative reconstruction is obtained and its statistical significance assessed. This novel procedure was used to reconstruct the mean annual precipitation(Pann) from Gonghai Lake in the Lvliang Mountains, and Tianchi Lake in the Liupan Mountains, on the eastern and western fringe of the Chinese Loess Plateau, respectively. Both Pann reconstructions are statistically significant(p<0.001), and a sound and stable correlation relationship exists in their common period, showing a rapid precipitation decrease since 3300 cal yr BP.
