1Analysis of in vivo patterns of caspase 3 gene expression in primary hepatocellular carcinoma and its relationship to p21^(WAF1) expression and hepatic apoptosis显示文摘AIM To detect the expression of caspase 3gene in primary human hepatocellular carcinoma(HCC)and investigate its relationship to p21WAF1gene expression and HCC apoptosis.METHODS In situ hybridization was employedto determine caspase 3 and p21WAF1expression inHCC.In situ end-labeling was used to detecthepatocytic apoptosis in HCC.RESULTS Twenty-one of 39(53.8%)cases ofHCC were found to express caspase 3transcripts,while 45.2% of HCC failed toexpress caspase 3.Non-cancerous adjacent livertissues showed more positive caspase 3(87.5%,7/8)as compared with HCC(P<0.05).The expression of caspase 3 is correlated withHCC differentiation,72.2%(13/18)ofmoderately to highly differentiated HCC showedcaspase 3 transcripts positive,while only 38.1%of poorly differentiated HCC harbored caspase 3transcripts(P<0.05).No relationship wasfound between caspase 3 expression and tumorsize or grade or metastasis,although 52.5%(5/8)of HCC with metastasis were caspase 3positive and a little higher than that with nometastasis(51.6%,P>0.05).Expression of caspase 3 alone did not affect the apoptosisindex(AI)of HCC.The AI was 7.12%o in caspase3-positive tumors(n=21),while in caspase 3-negative cases(n=18)6.59%0(P>0.05).Expression of caspase 3 clearly segregated withp21WAF1positive tumors as compared withp21WAF1-negative cases(16 of 23,69.6% versus5 of 16,31.3%)with statistical significance(P=0.017).In the cases with positive caspase 3and negative p21WAF1,the Al was found slightlyhigher,but with no statistical significance,thanthat with expression of p21WAF1and caspase 3(7.21‰ vs 6.98‰,P>0.05).CONCLUSION Loss of caspase 3 expressionmay contribute to HCC carcinogenesis,althoughthe expression of caspase 3 does not correlatewell with cell apoptosis in HCC.p21WAF1may bemerely one of the inhibitors which can reducecaspase 3 mediated cell apoptosis in HCCs.Bao Hua Sun Jun Zhang Bao JǜWang Xi Ping Zhao You Kun Wang Zhi Qun Yu Dong Liang Yang Lian Jie Hao Department of Clinical Immunology,Tongji Hospital,Tongji Medical University,Wuhan 430030,Hubei Province,China 2000World Journal of Gastroenterology2000,6,3:65

5Scale dependence of plant species richness and vegetation-environment relationship along a gradient of dune stabilization in Horqin Sandy Land, Northern China显示文摘Ecological patterns and processes in dune ecosystems have been a research focus in recent years, however the information on how dune stabilization influences the spatial scale dependence of plant diversity is still lacking. In this study, we measured the plant species richness, soil properties and altitude across four spatial scales(1, 10, 100 and 1,000 m2) at three different dune stabilization stages(mobile dune, semi-fixed dune and fixed dune) in Horqin Sandy Land, Northern China. We also examined the relationships between plant species richness, community composition and environmental factors along the gradient of dune stabilization. Our results showed that plant species richness increased with the increase of spatial scales in each dune stabilization stage, as well as with the increase of dune stabilization degrees. Canonical correspondence analysis(CCA) showed that plant distribu- tions in the processes of dune stabilization were determined by the combined environmental gradient in relation to soil organic carbon(SOC), total nitrogen(TN), carbon/nitrogen(C/N), pH, electrical conductivity(EC), soil water content(SWC), fine sand(FS), very fine sand(VFS), silt and clay(SC), and altitude. Plant species richness was significantly and positively correlated to SOC and TN in mobile dune, and significantly and positively correlated to SOC, TN, C/N, VFS and SC in semi-fixed dune. However, no significant correlation between plant species richness and environmental factors was observed in fixed dune. In addition, plant species richness in different dune stabili- zation stages was also determined by the combined gradient of soil properties and altitude. These results suggest that plant species richness has obvious scale dependence along the gradient of dune stabilization. 7Accumulation and potential sources of heavy metals in soils of the Hetao area,Inner Mongolia,China显示文摘Soil contamination by heavy metals is a problem in agricultural irrigation systems.To assess the accumulation and sources of heavy metals in the Yongji irrigation district of the Hetao area,Inner Mongolia,China,195 soil samples from 39 sites(0–100 cm)were collected,and Zn,Cu,Pb,Cr,and Cd concentrations were analyzed.The mean concentrations were 107.17,32.48,12.31,53.53,and 0.22 mg kg-1,respectively,with no significant differences between soil depths(P>0.05).Concentrations of Zn,Cu,and Cd were higher than the background levels,with moderate accumulation;the contamination factor(CF)values were 1.9,1.7,and 1.9,respectively,and the geoaccumulation index(Igeo)was>0.Concentrations of Pb and Cr were lower than,or close to,the background levels(CF<1,Igeo<0),indicating that they originated from a natural source.The monomial potential ecological risk index(Eri)for Zn,Cu,Pb,and Cr was low;Eri for Cd was 55.73,implying a moderate risk.The grade of potential ecological risk index of the five heavy metals(RI)was low,declining from south to north.The studied soils were contaminated with Zn,Cu,and Cd;principal component(PC)analysis implicated the enrichment of Cd and partial Cu(high loading in PC 2)was related to agricultural activities;Zn and partial Cu,closely associated with PC 3,may have originated from irrigation water from the Yellow River.Future agricultural development should focus on fertilizer and pesticide application and the quality of irrigation water.Yangchun ZHU Lijin WANG Xueyong ZHAO Jie LIAN Zhenhua ZHANG 2020Pedosphere2020,30,2:12
8Chinese herbal medicine reduces mortality in patients with severe and critical coronavirus disease 2019: a retrospective cohort study显示文摘This study aimed to evaluate the efficacy of Chinese herbal medicine(CHM)in patients with severe/critical coronavirus disease 2019(COVID-19).In this retrospective study,data were collected from 662 patients with severe/critical COVID-19 who were admitted to a designated hospital to treat patients with severe COVID-19 in Wuhan before March 20,2020.All patients were divided into an exposed group(CHM users)and a control group(non-users).After propensity score matching in a 1:1 ratio,156 CHM users were matched by propensity score to 156 non-users.No significant differences in seven baseline clinical variables were found between the two groups of patients.All-cause mortality was reported in 13 CHM users who died and 36 non-users who died.After multivariate adjustment,the mortality risk of CHM users was reduced by 82.2%(odds ratio 0.178,95%CI 0.076–0.418;P<0.001)compared with the non-users.Secondly,age(odds ratio 1.053,95%CI 1.023–1.084;P<0.001)and the proportion of severe/critical patients(odds ratio 0.063,95%CI 0.028–0.143;P<0.001)were the risk factors of mortality.These results show that the use of CHM may reduce the mortality of patients with severe/critical COVID-19.Guohua Chen Wen Su Jiayao Yang Dan Luo Ping Xia Wen Jia Xiuyang Li Chuan Wang Suping Lang Qingbin Meng Ying Zhang Yuhe Ke An Fan Shuo Yang Yujiao Zheng Xuepeng Fan Jie Qiao Fengmei Lian Li Wei Xiaolin Tong 2020Frontiers of Medicine2020,14,6:10
9Reversal of multidrug resistance in gastric cancer cells by CDX2 downregulation显示文摘AIM: To explore the role of CDX2 in the multi-drug resistance (MDR) process of gastric cancer in vitro and in vivo . METHODS: A cisplatin-resistant gastric cancer cell line with stable downregulation of CDX2 was established. mRNA and protein expression levels of CDX2, survivin, cyclin D1, and c-Myc were detected by western blotting and semi-quantitative reverse-transcriptase polymerase chain reaction (RT-PCR). The influence of downregulation of CDX2 on MDR was assessed by measuring IC50 of SGC7901/DDP cells to cisplatin, doxorubicin, and 5-fluorouracil, rate of doxorubicin efflux, apoptosis, and cell cycle progression detected by flow cytometry. In addition, we determined the in vivo effects of CDX2 small interfering RNA (siRNA) on tumor size, and apoptotic cells in tumor tissues were detected by deoxynucleotidyl transferase-mediated dUTP-biotin nick end labeling and hematoxylin and eosin staining. RESULTS: CDX2 siRNA led to downregulation of endogenous CDX2 mRNA (0.31 ± 0.05 vs 1.10 ± 0.51, 0.31 ± 0.05 vs 1.05 ± 0.21, P = 0.003) and protein (0.12 ± 0.08 vs 0.51 ± 0.07, 0.12 ± 0.08 vs 0.55 ± 0.16, P = 2.57 × 10 -4) expression. It significantly promoted the sensitivity of SGC7901/DDP cells to cisplatin (0.12 ± 0.05 vs 0.33 ± 0.08, 0.12 ± 0.05 vs 0.39 ± 0.15, P = 0.001), doxorubicin (0.52 ± 0.13 vs 4.11 ± 1.25, 0.52 ± 0.13 vs 4.05 ± 1.44, P = 2.81 × 10-4), and 5-fluorouracil (0.82 ± 0.13 vs 2.81 ± 0.51, 0.82 ± 0.13 vs 3.28 ± 1.03, P = 1.71 × 10-4). Flow cytometry confirmed that the percentage of apoptotic cells increased after CDX2 downregulation (32.15% ± 2.15% vs 17.63% ± 3.16%, 32.15% ± 2.15% vs 19.3% ± 2.25%, P = 1.73 × 10-6). This notion was further supported by the observation that downregulation of CDX2 blocked entry into the S-phase of the cell cycle (31.53% ± 3.78% vs 65.05% ± 7.25%, 31.53% ± 3.78% vs 62.27% ± 5.02%, P = 7.55 × 10-7). Furthermore, downregulation of CDX2 significantly increased intracellular accumulation of doxorubicin (0.21 ± 0.06 vs 0.41 ± 0.11, 0.21 ± 0.06 vs 0.40 ± 0.08, P = 0.003). In molecular studies, semiquantitative RT-PCR and western blotting revealed that CDX2 downregulation could inhibit expression of c-Myc, survivin and cyclin D1. 10Comorbidity Burden of Dementia:A Hospital-Based Retrospective Study from 2003 to 2012 in Seven Cities in China显示文摘Dementia is increasing dramatically and imposes a huge burden on society. To date, there is a lack of data on the health status of patients with dementia in China. In an attempt to investigate the comorbidity burden of dementia patients in China at the national level, we enrolled 2,938 patients with Alzheimer's disease(AD), vascular dementia(Va D), or other types of dementia, who were admitted to tertiary hospitals in seven regions of China from January2003 to December 2012. The Charlson Comorbidity Index(CCI) was used to evaluate the comorbidity burden of the patients with dementia. Among these patients, 53.4% had AD, 26.3% had Va D, and 20.3% had other types of dementia. The CCI was 3.0 ± 1.9 for all patients,3.4 ± 1.8 for those with Va D, and 3.0 ± 2.1 for those with AD. The CCI increased with age in all patients, andthe length of hospital stay and daily expenses rose with age and CCI. Males had a higher CCI and a longer stay than females. Moreover, patients admitted in the last 5 years of the study had a higher CCI than those admitted in the first 5 years. We found that the comorbidity burden of patients with dementia is heavy. 11A new paleoenvironmental index for anoxic events—Mo isotopes in black shales from Upper Yangtze marine sediments显示文摘This paper investigates the high-solution of Mo isotopes and uses trace-element analyses for fresh and representative black shales and siliceous shales collected from the transition between the Late Ordovician and the Early Silurian at the Wangjiawan section in Yichang and the Late Permian Dalong Formation in the Shangsi Section of Sichuan. The applicability of different geochemical parameters used as paleo-oxygenation indices are also compared. The preliminary results show that V/(V+Ni), U auth (auth U), V/Cr, Ce anom and U/Th have a scattered variation range, but most samples plot within the suboxic-anoxic fields. The suboxic-anoxic environment was dominant during the deposition and formation of the two anoxic facies. These redox indicators show little correspondence to the δ 98 Mo values. The U/Mo ratio can be used as a potential proxy for the paleo-redox conditions due to the possibility that Mo is enriched relative to U at different redox gradients during early diagenesis. This evidence is more significant for the euxinicity condition and corresponds to positive δ 98 Mo (>1.5‰) values with low U/Mo ratios. This evidence is likely related to the depositional conditions near the boundary between anoxic and euxinic environments, which are characterised by low bioturbation or water circulation. Other samples reveal a wide scatter of U/Mo ratios and δ 98 Mo <1.5‰. 12Clinical applications of MARSALA for preimplantation genetic diagnosis of spinal muscular atrophy显示文摘Conventional PCR methods combined with linkage analysis based on short tandem repeats(STRs) or Karyomapping with single nucleotide polymorphism(SNP) arrays, have been applied to preimplantation genetic diagnosis(PGD) for spinal muscular atrophy(SMA), an autosome recessive disorder. However, it has limitations in SMA diagnosis by Karyomapping, and these methods are unable to distinguish wildtype embryos with carriers effectively. Mutated allele revealed by sequencing with aneuploidy and linkage analyses(MARSALA) is a new method allowing embryo selection by a one-step next-generation sequencing(NGS) procedure, which has been applied in PGD for both autosome dominant and X-linked diseases in our group previously. In this study, we carried out PGD based on MARSALA for two carrier families with SMA affected children. As a result, one of the couples has given birth to a healthy baby free of mutations in SMA-causing gene. It is the first time that MARSALA was applied to PGD for SMA, and we can distinguish the embryos with heterozygous deletion(carriers) from the wild-type(normal) ones accurately through this NGS-based method. In addition, direct mutation detection allows us to identify the affected embryos(homozygous deletion), which can be regarded as probands for linkage analysis, in case that the affected family member is absent. 13Sini powder ameliorates the inflammatory response in rats with stress-induced non-alcoholic fatty liver disease by inhibiting the nuclear factor kappa-B/pyrin domain-containing protein 3 pathway显示文摘OBJECTIVE: To evaluate the effectiveness of Sini powder for the treatment of non-alcoholic fatty liver disease(NAFLD) in rats and the molecular mechanisms involved.METHODS: A rat model of stress-induced NAFLD was established by a combination of long-term tethering and feeding of a high-fat, high-calorie diet. These rats were then intragastrically administered with either simvastatin, Sini powder, or vehicle for 1 week. The body mass and field test scores for each group were recorded weekly. Serum aspartate aminotransferase and alanine aminotransferase activities, and triglyceride, total cholesterol,and free fatty acid concentrations were measured.Liver tissue histopathology was examined on hematoxylin and eosin-stained paraffin sections and oil red O-stained frozen sections. The hepatic m RNA expression of nuclear factor kappa-B(NF-κB),NOD-, LRR-, and pyrin domain-containing protein 3(NLRP3), apoptosis-associated speck-like protein containing CARD(ASC), and caspase-1 were measured by reverse transcription-polymerase chain reaction(RT-PCR). The hepatic protein concentrations of NF-κB and NLRP3, ASC, caspase-1, interleukin-1β(IL-1β), interleukin-6(IL-6), and the serum concentrations of IL-1β and IL-6 were measured by enzyme-linked immunosorbent assay.RESULTS: Compared with the Blank group, rats in the Compound model group showed significant pathologic manifestations of NAFLD, and the expression of NF-κB, NLRP3, ASC, caspase-1, IL-1βand IL-6 were significantly higher(all P < 0.01). Both simvastatin and Sini powder significantly ameliorated the NAFLD pathology and the abnormal expression of NF-κB, NLRP3, ASC, caspase-1, IL-1β, and IL-6(all P < 0.01).CONCLUSION: Sini powder inhibits the inflamma-tory response in rats with NAFLD, which is mediated by NF-κB/NLRP3, IL-1β, and IL-6, reduces the effects of psychological stress, and improves lipid metabolism. 14The effects of Trichoderma on preventing cucumber fusarium wilt and regulating cucumber physiology显示文摘In our previous studies, we identified 3 Trichoderma strains with anti-Fusarium oxysporum activity, including T. asperellum 525, T. harzianum 610, and T. pseudokoningii 886. Here, we evaluated the effects of these 3 Trichoderma strains on preventing cucumber fusarium wilt through pot culture and greenhouse culture experiments. All 3 Trichoderma strains demonstrated higher control effects toward cucumber fusarium wilt than previous studies, with efficacies over 78%. Additionally, inoculation with the 3 Trichoderma strains significantly promoted the quality and yield of cucumbers. Among the 3 strains, Trichoderma 866 was the most effective, with disease control efficacy of 78.64% and a cucumber yield increase of 33%. Furthermore, seedlings inoculated with Trichoderma exhibited significantly increased measures of plant height, stem diameter, leaf area, aboveground fresh weight, underground fresh weight, chlorophyll content, and nitric nitrogen content, as well as the activities of several stress-resistance enzymes, including superoxide dismutase(SOD), peroxidase(POD), catalase(CAT), polyphenol oxidase(PPO), and ascorbate oxidase(AAO). In addition, the plants inoculated with Trichoderma showed decreased cell membrane permeability and malondialdehyde(MDA) content in the leaves. Together, our results suggest that T. asperellum 525, T. harzianum 610, and T. pseudokoningii 886 inoculations inhibit F. oxysporum infection, stimulate the metabolism in cucumbers, and enhance the activities of stress-resistance enzymes, which consequently promote the growth of cucumber plants, prevent cucumber fusarium wilt, and improve the yield and quality of cucumbers. T. harzianum is a commonly used biocontrol fungus, while few studies have focused on T. asperellum or T. koningense. 17Medical services for sports injuries and illnesses in the Beijing 2022 Olympic Winter Games显示文摘BACKGROUND:Beijing 2022 Olympic Winter Games was the second Games held amid the COVID-19 pandemic.To a certain extent,it has altered the way sporting activities operate.There is a lack of knowledge on injury risk and illness occurrence in elite winter sport athletes amid the COVID-19 pandemic.This study aimed to describe the incidence of injuries and illnesses sustained during the XXIV Olympic Winter Games in Beijing from February 4 to 20,2022.METHODS:We recorded the daily number of injuries and illnesses among athletes reported by Beijing 2022 medical staff in the polyclinic,medical venues,and ambulance.We calculated injury and illness incidence as the number of injuries or illnesses occurring during competition or training,respectively,with incidence presented as injuries/illnesses per 100 athlete-days.RESULTS:In total,2,897 athletes from 91 nations experienced injury or illness.Beijing 2022 medical staff reported 326 injuries and 80 illnesses,equaling 11.3 injuries and 2.8 illnesses per 100 athletes over the 17-day period.Altogether,11%of the athletes incurred at least one injury and nearly 3%incurred at least one illness.The number of injured athletes was highest in the skating sports(n=104),followed by alpine skiing(n=53),ice track(n=37),freestyle skiing(n=36),and ice hockey(n=35),and was the lowest in the Nordic skiing disciplines(n=20).Of the 326 injuries,14(4.3%)led to an estimated absence from training or competition of more than 1 week.A total of 52 injured athletes were transferred to hospitals for further care.The number of athletes with illness(n=80)was the highest for skating(n=33)and Nordic skiing(n=22).A total of 50 illnesses(62.5%)were admitted to the department of dentistry/ophthalmology/otolaryngology,and the most common cause of illness was other causes,including preexisting illness and medicine(n=52,65%).CONCLUSION:Overall,11%of athletes incurred at least one injury during the Games,which is similar to the findings during the Olympic Winter Games in 2014 and 2018.Regarding illness,2%of athletes were affected,which is approximately one-third of the number affected in the 2018 Olympic Winter Games.Peng-da Han Ding Gao Jie Liu Jing Lou Si-jia Tian Hui-xin Lian Sheng-mei Niu Lu-xi Zhang Yong Wang Jin-jun Zhang 2022World Journal of Emergency Medicine2022,13,6:7
18Floral traits and pollination system of Zygophyllum xanthoxylum in the managed and wild populations in an arid region of Northwest China显示文摘Zygophyllum xanthoxylum, which belongs to Sarcozygium of Zygophyllaceae, is one of the ecologically important species in Northwest China. In order to understand the pollination system of Z. xanthoxylum, we investigated the following characteristics of this species in the Urat Desert-grassland Research Station in western Inner Mongolia of China: flowering dynamics, pollen viability, pollen limitation, floral visitors and breeding system. The results showed that the flowering period and flowering peak were different between the wild and managed populations, being longer in the managed population. Z. xanthoxylum was pollen-limited, and pollen limitation was more intense in the wild population than in the managed population. Chalicodoma deserticola(Hymenoptera) was found to be the most frequent pollinator in the wild population, while Anthophora fulvitarsis(Hymenoptera) was the most frequent and effective visitor in the managed population. 19Toward sustainable desertification reversion:A case study in Horqin Sandy Land of northern China显示文摘Desertification reversion is an interactive process involving climate, land use change, and water processes. In order to reveal the relationship between desertification reversion and these factors, we analyzed historical data on precipitation, air temperature, desertified land changes, underground water tables, and water body changes in Naiman County in the central part of Horqin Sandy Land. Our analysis showed that during 1961-2010 the annual precipitation fluctuated dramatically and has decreased fairly consistently in recent years. The air temperature increased by 0.50-1.25 °C, and the minimum temperature increased more obviously. The desertified land area increased from 42,300 km2 in 1959 to 62,000 km2 in 1985, and then declined to about 50,000 km2 in 2010. The underground water tables have been lowered by about 10 m in the past 30 years, and declined more rapidly in recent years. Desertified land is significantly related to the amount of total cropland, and underground water tables are significantly correlated with annual precipitation and the amount of irrigated cropland.
