3Effect of soothing-liver and nourishing-heart acupuncture on early selective serotonin reuptake inhibitor treatment onset for depressive disorder and related indicators of neuroimmunology:a randomized controlled clinical trial显示文摘OBJECTIVE:To observe the effect of soothing-liver and nourishing-heart acupuncture on selective serotonin reuptake inhibitor(SSRIs) treatment effect onset in patients with depressive disorder and related indicators of neuroimmunology.METHODS:Overall,126 patients with depressive disorder were randomly divided into a medicine and acupuncture-medicine group using a random number table.Patients were treated for 6 consecutive weeks.The two groups were evaluated by the Montgomery-Asberg Depression Rating Scale(MADRS) and Side Effects Rating Scale(SERS) to assess the effect of the soothing-liver and nourishing-heart acupuncture method on early onset of SSRI treatment effect.Changes in serum 5-hydroxytryptamine(5-HT) and inflammatory cytokines before and after treatment were recorded and compared between the medicine group and the acupuncture-medicine group.RESULTS:The acupuncture-medicine group had significantly lower MADRS scores at weeks 1,2,4,and 6 after treatment compared with the medicine group(P < 0.01).The acupuncture group had significantly lower SERS scores at weeks 1,2,4,and 6 after treatment compared with the medicine group(P < 0.01).At 6 weeks after treatment,serum 5-HT in the acupuncture-medicine group was significantly higher compared with the medicine group(P <0.01).lnterleukin-6(IL-6) in the acupuncture-medicine group was significantly lower than that in the medicine group(P < 0.01),whereas there was no significant difference in IL-1(3 between the groups(P > 0.05).Anti-inflammatory cytokines IL-4 and IL-10 were significantly higher in the acupuncture-medicine group compared with the medicine group(P < 0.01,P < 0.05,respectively).CONCLUSION:The soothing-liver and nourishing-heart acupuncture method can effectively accelerate the onset of SSRI effects when treating depressive disorder and can significantly reduce the adverse reactions of SSRIs.Moreover,acupuncture can enhance serum 5-HT and regulate the balance of pro-inflammatory cytokines and anti-inflammatory cytokines.Liu Yi Feng Hui Mo Yali Gao Jingfang Mao Hongjing Song Mingfen Wang Shengdong Yin Yan Liu Wenjuan 2015Journal of Traditional Chinese Medicine2015,35,5:13
4SARS-Coronavirus-2 Nsp13 Possesses NTPase and RNA Helicase Activities That Can Be Inhibited by Bismuth Salts显示文摘The ongoing outbreak of Coronavirus Disease 2019(COVID-19)has become a global public health emergency.SARScoronavirus-2(SARS-CoV-2),the causative pathogen of COVID-19,is a positive-sense single-stranded RNA virus belonging to the family Coronaviridae.For RNA viruses,virus-encoded RNA helicases have long been recognized to play pivotal roles during viral life cycles by facilitating the correct folding and replication of viral RNAs.Here,our studies show that SARS-CoV-2-encoded nonstructural protein 13(nsp13)possesses the nucleoside triphosphate hydrolase(NTPase)and RNA helicase activities that can hydrolyze all types of NTPs and unwind RNA helices dependently of the presence of NTP,and further characterize the biochemical characteristics of these two enzymatic activities associated with SARS-CoV-2 nsp13.Moreover,we found that some bismuth salts could effectively inhibit both the NTPase and RNA helicase activities of SARS-CoV-2 nsp13 in a dose-dependent manner.Thus,our findings demonstrate the NTPase and helicase activities of SARS-CoV-2 nsp13,which may play an important role in SARS-CoV-2 replication and serve as a target for antivirals.Ting Shu Muhan Huang Di Wu Yujie Ren Xueyi Zhang Yang Han Jingfang Mu Ruibing Wang Yang Qiu Ding-Yu Zhang Xi Zhou 2020Virologica Sinica2020,35,3:11

6Plasma metabolomic and lipidomic alterations associated with COVID-19显示文摘The pandemic of the coronavirus disease 2019(COVID-19)has become a global public health crisis.The symptoms of COVID-19 range from mild to severe,but the physiological changes associated with COVID-19 are barely understood.In this study,we performed targeted metabolomic and lipidomic analyses of plasma from a cohort of patients with COVID-19 who had experienced different symptoms.We found that metabolite and lipid alterations exhibit apparent correlation with the course of disease in these patients,indicating that the development of COVID-19 affected their whole-body metabolism.In particular,malic acid of the TCA cycle and carbamoyl phosphate of the urea cycle result in altered energy metabolism and hepatic dysfunction,respectively.It should be noted that carbamoyl phosphate is profoundly down-regulated in patients who died compared with patients with mild symptoms.And,more importantly,guanosine monophosphate(GMP),which is mediated not only by GMP synthase but also by CD39 and CD73,is significantly changed between healthy subjects and patients with COVID-19,as well as between the mild and fatal cases.In addition,dyslipidemia was observed in patients with COVID-19.Overall,the disturbed metabolic patterns have been found to align with the progress and severity of COVID-19.This work provides valuable knowledge about plasma biomarkers associated with COVID-19 and potential therapeutic targets,as well as an important resource for further studies of the pathogenesis of COVID-19.Di Wu Ting Shu Xiaobo Yang Jian-Xin Song Mingliang Zhang Chengye Yao Wen Liu Muhan Huang Yuan Yu Qingyu Yang Tingju Zhu Jiqian Xu Jingfang Mu Yaxin Wang Hong Wang Tang Tang Yujie Ren Yongran Wu Shu-Hai Lin Yang Qiu Ding-Yu Zhang You Shang Xi Zhou 2020National Science Review2020,7,7:9
7Applicability and Generality of the Modified Grübler-Kutzbach Criterion显示文摘A generally applicable criterion for all mechanism mobility has been an active domain in mechanism theory lasting more than 150 years. It is stated that the Modified Grübler-Kutzbach criterion for mobility has been successfully used to solve the mobility of many more kinds of mechanisms, but never before has anyone proven the applicability and generality of the Modified Grübler-Kutzbach criterion in theory. In order to fill the gap, the applicability and generality of the Modified Grübler-Kutzbach Criterion of mechanism mobility is systematically demonstrated. Firstly, the mobility research background and the Modified Grübler-Kutzbach criterion are introduced. Secondly, some new definitions, such as half local freedom, non-common constraint space of a mechanism and common motion space of a mechanism, etc, are given to demonstrate the correctness and broad applicability of the Modified Grübler-Kutzbach criterion. Thirdly, the general applicability of the Modified Grübler-Kutzbach criterion is demonstrated based on screw theory. 9Mesoscale fracture behavior of Longmaxi outcrop shale with different bedding angles:Experimental and numerical investigations显示文摘The mechanical properties and fracturing mechanism of shale containing beddings are critically important in shale gas exploitation and wellbore stability.To investigate the effects of shale bedding on crack behavior and fracturing mechanism,scanning electron microscope(SEM)with a loading system was employed to carry out three-point bending tests on Longmaxi outcrop shale.The crack initiation and propagation of Longmaxi shale were observed and recorded by taking photos during loading.The cracking paths were extracted to calculate the crack length through a MATLAB program.The peak load,fracture toughness and fracture energy all increase with the bedding angle from 0°to 90°.The crack length and energy were also found to increase with the bedding angle in the range of 0°-600 and then drop slightly.The fracturing mechanism of shale includes the main crack affected by the bedding angle and disturbed by randomly distributed particles.The main cracking path was accompanied by several microcrack branches which could form an interconnected crack system.When the main crack encounters larger sedimentary particles,it will deflect around the particles and then restore to the initial direction.A numerical technique using extended finite element method(XFEM)coupled with anisotropic cohesive damage criteria was developed,which is able to capture the dependence of crack propagations on bedding angle and sedimentary particles.This study sheds light on understanding and predicting mesoscale fracture behavior of shale with different bedding angles.Jianping Zuo Jingfang Lu Rojin Ghandriz Jintao Wang Yanhong Li Xiaoyan Zhang Jun Li Hongtao Li 2020Journal of Rock Mechanics and Geotechnical Engineering2020,12,2:4
10All-trans retinoic acid reverses malignant biological behavior of hepatocarcinoma cells by regulating miR-200 family members显示文摘As a potential chemo-therapeutic agent,all-trans retinoic acid(ATRA)can signif-icantly reverse epithelial-mesenchymal transition(EMT)of hepal-6 hepatocarcinoma cell line in vitro,but the mechanism is unclear.The expression profile of microRNA-200(miR-200)families is different in hepatocellular carcinoma.In this study,we found that ATRA trea tment could up-regulate the expression of miR-200a-3p,200c-3p,and 141-3p,which were involved in ATRA regulated proliferation and apoptosis of hepal-6 cell,but not colony formation.Meanwhile,miR-200a-3p,200c-3p,and 141-3p could recovery ATRA inhibited migration and invasion abilities of hepal-6 cells at various levels.miR-200a-3p and 200c-3p prevented ATRA from inducing the differentia tion and hepatic functions of hepal-6 cells.An-tagomir specific for miR-200a-3p and 200c-3p down-regulated the expression of CK18,but only miR-200a-3p anta gomir played prominent role in regula ting the expression of these mesenchymal markers,N-Cadherin,Snail and Twist.The transcriptional activities of 8 tran-scription factors were up-regulated and 35 transcription factors were down-regulated by ATRA.Compared with ATRA group,inhibition of miR-200a-3p,200c-3p,and 141-3p significantly strengthened the expression of Fra1/Jun(AP1),Ets1/PEA3,Brn3,and Zeb1/AREB6 at varying degrees.Therefore,this result suggested that ATRA may suppress EMT through down-regulating miR-200a-3p,200c-3p and 141-3p related transcription factors.miR-200 and their downstream genes might be the potentially specific targets for the treat-ment of hepatocarcinoma.Jiejie Cui Mengjia Gong Shuyu Fang Chaoqun Hu Yi Wang Jingfang Zhang Ni Tang Yun He 2021Genes & Diseases2021,8,4:2
11Docking and molecular dynamics studies on CYP2D6显示文摘Drug-metabolizing enzymes,also known as cytochrome P450s,are a superfamily of hemoglobin responsible for metabolizing more than 90% clinical drugs.Cytochrome P450 2D6(CYP2D6) is a significant member of cytochrome P450s for the reason of metabolizing about 20% clinical drugs.In this paper,molecular docking and molecular dynamic simulations are used to investi-gate the active site of CYP2D6,roles of essential amino acids within the active site and time-dependent protein energy changes.The results suggest that amino acids Glu216,Asp301,Ser304 and Ala305 in the active site are likely to form hydrogen bonding interactions with substrates;the benzene ring of Phe120 and aromatic ring in the substrates form Π-Π interactions.In addition,molecular dynamics simulations prove that the catalytic conformation of CYP2D6 without ligands can be obtained by their own atomic fluctuations.The impact of ligands on protein system energy and large conformational shift is not very large.Cytochrome P450s is known for their genetic polymorphisms,which will result in severe adverse drug reactions.Ideally,we hope to use mo-lecular modeling to investigate the differences between the substrates of wild-type and mutants while they are bonded with drugs,and predict the drug metabolizing ability of mutants.Reduce the possibility for people taking drugs that they can not metabolize,therefore reduce the rate of adverse drug reactions,and eventually establish a platform of personalized drugs to largely benefit human health.WANG JingFang ZHANG ChengCheng WEI DongQing LI YiXue 2010Chinese Science Bulletin2010,55,18:2
12Rice FLOURY ENDOSPERM 18 encodes a pentatricopeptide repeat protein required for 5′ processing of mitochondrial nad5 messenger RNA and endosperm development显示文摘Pentatricopeptide repeat(PPR) proteins, composing one of the largest protein families in plants,are involved in RNA binding and regulation of organelle RNA metabolism at the posttranscriptional level. Although several PPR proteins have been implicated in endosperm development in rice(Oryza sativa), the molecular functions of many PPRs remain obscure. Here, we identified a rice endosperm mutant named floury endosperm 18(flo18) with pleiotropic defects in both reproductive and vegetative development.Map-based cloning and complementation tests showed that FLO18 encodes a mitochondriontargeted P-type PPR protein with 15 PPR motifs.Mitochondrial function was disrupted in the flo18 mutant, as evidenced by decreased assembly of Complex I in the mitochondrial electron transport chain and altered mitochondrial morphology. Loss of FLO18 function resulted in defective 5′-end processing of mitochondrial nad5 transcripts encoding subunit 5 of nicotinamide adenine dinucleotide hydrogenase. 13From mandatory icebreaker guiding to a permission regime: changes to the new Russian legislation of the Northern Sea Route显示文摘This article focuses on two issues. The first concerns definitions of the Northern Sea Route(NSR) in old and new Russian legislation, and the second relates to Russian rules on icebreaker guiding. Based on a comprehensive comparative analysis of relevant Russian legal provisions enacted in 2013 and previous laws in this area, we offer the following conclusions.(1) Our legal analysis indicates that Russia's view of the NSR as a historical national transportation route has not changed. However, the new law redefines the scope and coverage of the NSR, which now comprises the internal waters, territorial sea, adjacent zone, and the exclusive economic zone of the Russian Federation. In fact, the new law resolves previous ambiguity regarding extension of the NSR boundary to the high seas.(2) Based on an analysis of the new rules on icebreaker guiding, the article concludes that NSR is transitioning from a mandatory icebreaker guiding regime into a permit regime. This is particularly evident in its provision of a concrete, practical, and predictable clause on permissible or impermissible conditions relating to independent navigation. According to the new rules, it is possible for foreign ships to undertake independent navigation in the NSR. 14Molecular dynamics simulations exploring drug resistance in HIV-1 proteases显示文摘Although HIV-1 subtype B still dominates the epidemic AIDS in developed countries,an increasing number of people in developing countries are suffering from an epidemic of non-subtype B viruses.What is worse,the efficacy of the combinational use of antiretroviral drugs is gradually compromised by the rapid development of drug resistance.To gain an insight into drug resistance, 10-ns MD simulations were simultaneously conducted on the complexes of the TL-3 inhibitor with 4 different proteases(Bwt,Bmut, Fwt and Fmut),among which the complex of the Bwt protease with the TL-3 inhibitor was treated as the control group.Detailed analyses of MD data indicated that the drug resistance of Bmut against TL-3 mainly derived from loss of an important hydrogen bond and that of Fwt was caused by the decrease of hydrophobic interactions in S1/S1'pocket,while both of the two reasons mentioned above were the cause of the Fmut protease's resistance.These results are in good agreement with the previous experiments, revealing a possible mechanism of drug resistance for the aforementioned protease subtypes against the TL-3 inhibitor.Additionally,another indication was obtained that the mutations of M36I,V82A and L90M may induce structural transforms so as to alter the inhibitor's binding mode.GU Hui CHEN HaiFeng WEI DongQing WANG JingFang 2010Chinese Science Bulletin2010,55,24:2
