1Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 2Precise base editing of non-allelic acetolactate synthase genes confers sulfonylurea herbicide resistance in maize显示文摘Single-nucleotide polymorphisms contribute to phenotypic diversity in maize. Creation and functional annotation of point mutations has been limited by the low efficiency of conventional methods based on random mutation. An efficient tool for generating targeted single-base mutations is desirable for both functional genomics and precise genetic improvement. The objective of this study was to test the efficiency of targeted C-to-T base editing of two non-allelic acetolactate synthase(ALS) in generating sulfonylurea herbicide-resistant mutants. A CRISPR/Cas9 nickase-cytidine deaminase fused with uracil DNA glycosylase inhibitor(UGI) was employed to achieve targeted conversion of cytosine to thymine in ZmALS1 and ZmALS2. Both protoplasts and recovered mutant plants showed the activity of the cytosine base editor, with an in vivo efficiency of up to 13.8%. Transgene-free edited plants harboring a homozygous ZmALS1 mutation or a ZmALS1 and ZmALS2 double mutation were tested for their resistance at a dose of up to 15-fold the recommended limit of chlorsulfuron, a sulfonylurea herbicide widely used in agriculture. 4Conversion of a normal maize hybrid into a waxy version using in vivo CRISPR/Cas9 targeted mutation activity显示文摘Waxy maize is a specialty maize that produces mainly amylopectin starch with special food or industrial values. The objective of this study was to overcome the limitations of wx mutant allele acquisition and breeding efficiency by conversion of parental lines from normal to waxy maize. The intended mutation activity was achieved by in vivo CRISPR/Cas9 machinery involving desired-target mutation of the Wx locus in the ZC01 background,abbreviated as ZC01-DTM^(wx). Triple selection was applied to segregants to obtain high genome background recovery with transgene-free wx mutations. The targeted mutation was identified, yielding six types of mutations among progeny crossed with ZC01-DTM^(wx).The amylopectin contents of the endosperm starch in mutant lines and hybrids averaged94.9%, while those of the wild-type controls were significantly(P < 0.01) lower, with an average of 76.9%. Double selection in transgene-free lines was applied using the Bar strip test and Cas9 PCR screening. The genome background recovery ratios of the lines were determined using genome-wide SNP data. That of lines used as male parents was as high as98.19% and that of lines used as female parents was as high as 86.78%. Conversion hybrids and both parental lines showed agronomic performance similar to that of their wild-type counterparts. 5Fine mapping of the awn gene on chromosome 4 in rice by association and linkage analyses显示文摘Awnness is a key trait in rice domestication, yet no studies have been conducted on fine mapping or association mapping of the rice awn gene. In this study, we investigated the awnness and genotype of a core collection of 303 cultivated rice varieties and a BC5F2 segregating population of 200 individuals. Combining association and linkage analyses, we mapped the awnness related genes to chromosome 4. Primary association analysis using 24 SSR markers revealed five loci significantly associated with awnness on chromosome 4. The associated markers cover previously identified regions. Fine association mapping was conducted using another 29 markers within a 4-Mb region, covering the associated marker in34, which is close to the awn gene Awn4.1. Seven associated markers were revealed, distributed over an 870-kb region. Combining the fine association mapping and linkage analysis of awnness in the 200 BC5F2 segregating population, we finally identified a 330-kb region as the candidate region for Awn4.1. 6Structure characteristics and microwave dielectric properties of Pr_(2)(Zr_(1-x)Ti_(x))_(3)(MoO_(4))_(9) solid solution ceramic with a stable temperature coefficient显示文摘Pr_(2)(Zr_(1−x)Ti_(x))_(3)(MoO_(4))_(9)(x=0.1-1.0)ceramics were prepared via a conventional solid-state method,the dependence of crystal structure and bond characteristics on microwave dielectric properties was investigated systemically.The X-ray diffraction patterns indicated that the single-phase Pr_(2)Zr_(3)(MoO_(4))_(9)structure was formed in all the specimens.As the Ti^(4+)content increased,the lattice volume gradually decreased,which was ascribed to the fact that the ionic radius of Ti^(4+)was smaller than that of Zr^(4+).Notably,outstanding microwave dielectric properties withεr of 10.73-16.35,Q·f values of 80,696-18,726 GHz and minorτ_(f) values−14.1-−2.6 ppm/℃were achieved in Pr_(2)(Zr_(1−x)Ti_(x))_(3)(MoO_(4))_(9)ceramics.Theε_(r) increased with the rising x values,which was associated with the increase ofα/Vm values.The decreasing Q·f was affected by the decline of lattice energy of[Zr/TiO_(6)]octahedral.Theτf value was dominated by[Zr/TiO_(6)]octahedral distortion,Mo-O bond energy,bond strength and B-site bond valence.Furthermore,infrared reflection spectra suggested that the properties were mainly caused by the absorption of phonon,and the dielectric loss could be further reduced by optimizing the experimental process.Huanrong Tian Jinjie Zheng Lintao Liu Haitao Wu Hideo Kimura Yizhong Lu Zhenxing Yue 2022Journal of Materials Science & Technology2022,,21:3
7Comparing overall survival between first generation EGFR-TKIs and chemotherapy in lung cancer patients with Del19/L858R显示文摘Objective: Combined overall survival(OS) analysis of Lux-Lung 3 and Lux-Lung 6 demonstrated that patients with epidermal growth factor receptor(EGFR) exon 19 deletions(Del19) would benefit from first-line second generation EGFR tyrosine kinase inhibitors(TKIs) afatinib but not for those with L858 R. This study was to investigate the survival difference between first-line first generation EGFR-TKIs and chemotherapy in patients with either Del19 or L858 R, and to directly compare OS in these two mutation groups.Methods: Eligibles were all prospective and retrospective studies comparing EGFR-TKIs with conventional chemotherapy or receiving single agent EGFR-TKIs and demonstrating survival analysis based on mutation types. The primary outcome was OS measured as pooled hazard ratios(HRs). All measures were pooled using randomeffects models and 95% confidential interval(95% CI) was calculated.Results: A total of 14 studies incorporating 1,706 patients with either Del19 or L858 R were included. Enrolling patients with Del19 or L858 R in randomized controlled trials(RCTs), first-line first generation EGFR-TKIs were associated with no OS benefit, compared with chemotherapy(pooled HR_(TKI/Chemo) for Del19: 0.82, 95% CI: 0.64-1.06, P=0.14; pooled HR_(TKI/Chemo) for L858R: 1.15, 95% CI: 0.85-1.56, P=0.38). Direct comparison of Del19 with L858 R receiving with first-line first generation EGFR-TKIs demonstrated no significant survival difference(pooled HR_(19/21): 0.88, 95% CI: 0.67-1.16, P=0.37).Conclusions: Among patients with advanced non-small cell lung cancer(NSCLC) harboring Del19 and L858 R, first-line first generation EGFR-TKIs demonstrated no survival benefit comparing with chemotherapy. 8Advances in Utilization of Agricultural Waste Based on Production of Edible Fungi显示文摘Along with the rapid development of edible fungus industry in China,the traditional mode of production giving priority to wood chips will be severely limited,and using agricultural waste distributed widely,having large yield,and containing high content of organic matter to produce edible fungi has good economic and ecological benefit. In this paper,based on the analysis of characteristics of agricultural waste in China,the present situation of application of agricultural waste in the production of edible fungi at home and abroad was introduced,and the main problems existing in production of edible fungi by using agricultural waste in China at the present stage were pointed out. 9Optimal Design of Novel Electromagnetic-Ring Active Balancing Actuator with Radial Excitation显示文摘Imbalance vibration is a typical failure mode of rotational machines and has significant negative effects on the efficiency,accuracy,and service life of equipment.To automatically reduce the imbalance vibration during the operational process,different types of active balancing actuators have been designed and widely applied in actual production.However,the existing electromagnetic-ring active balancing actuator is designed based on an axial excitation structure which can cause structural instability and has low electromagnetic driving efficiency.In this paper,a novel radial excitation structure and the working principle of an electromagnetic-ring active balancing actuator with a combined driving strategy are presented in detail.Then,based on a finite element model,the performance parameters of the actuator are analyzed,and reasonable design parameters are obtained.Self-locking torque measurements and comparative static and dynamic experiments are performed to validate the self-locking torque and driving efficiency of the actuator.The results indicate that this novel active balancing actuator has sufficient self-locking torque,achieves normal step rotation at 2000 r/min,and reduces the driving voltage by 12.5%.The proposed novel balancing actuator using radial excitation and a combination of permanent magnets and soft-iron blocks has improved electromagnetic efficiency and a more stable and compact structure.Xin Pan Xiaotian He Haiqi Wu Chuanlong Ju Zhinong Jiang Jinji Gao 2021Chinese Journal of Mechanical Engineering2021,34,1:2

11Effect of hypoglycemic anti-deafness capsules in diabetic patients with deafness and toxicological assessment in rats显示文摘OBJECTIVE:Through experiment on animals and clinical trials to explore the safety and efficacy of hypoglycemic anti-deafness capsules on diabetic patients with deafness.METHODS:Total 296 patients with non-insulin dependent diabetes mellitus(NIDDM)were randomly divided into two groups.A treatment group of 164patients(208 ears)was treated with hypoglycemic anti-deafness capsules based onTCM syndrome differentiation.A control group of 132 patients(184ears)was treated with glibenclamide and conventional drug treatment for deafness.The following were observed:hearing,fasting plasma glucose(FPG),2 h postprandial plasma glucose(2hPG),24 h urine glucose(24hUG),improvement of main symptoms,platelet function,and changes in superoxidedismutase(SOD)and lipid peroxide(LPO)levels.In animal studies,Kunming mice,weighing 18-22 g were used.Half of the mice were males and half were females.Wistar rats,weighing 80-120 g were used.Half of the rats were males and half were females.Male Wistar rats,weighing 200-220 g,were also used.Their acute and chronic toxicity was studied.RESULTS:The hearing improvement was 56.7%in the treatment group and 26.6%in the control group.FPG,2hPG,and 24hUG were improved significantly(P<0.05,P<0.01,P<0.01,respectively)in the treatment group and 2hPG and 24hUG improved significantly in the control group(P<0.05,P<0.05).The improvement in 2hPG and 24hUG in the treatment group was significantly greater than that in the control group P<0.01).There was no significant difference in FPG between the two groups(P<0.05).Main symptoms in the treatment group were significantly more improved than those in the control group(P<0.05,P<0.01).In the treatment group,platelet adhesion and aggregation,SOD,and LPO were all significantly improved from before treatment(P<0.05,P<0.01).However,in the control group,except LOP(P<0.05),there were no significant differences from before treatment to after(P<0.05).In animal studies,no obvious acute or long-term toxicity was observed from capsule administration.CONCLUSION:Through experiment on animals and clinical trials,we can found that hypoglycemic anti-deafness capsules could decrease blood glucose and serum triglycerides of alloxan-induced diabetic rats.This herbal capsule is effective for safely treating diabetic patients with deafness.Ruiyu Li Meng Li Kaoshan Guo Bin Li Jinjie Hou Liping Wu Xianyun Liu 2013Journal of Traditional Chinese Medicine2013,33,5:2

15A NOVEL DRIVING STRATEGY FOR DYNAMIC SIMULATION OF HOISTING ROPE WITH TIME-VARYING LENGTH显示文摘A simulation method for investigating the vibration behavior of hoisting rope with time-varying length is improved.By previously creating markers in the MSC.ADAMS software package,the parametric model of the rope wound along helix is established based on the concentrated-mass theory with multi-degree of freedom(multi-DOF).A novel driving strategy,cooperating fixed joints with angle sensors under the control of driving script,is proposed to substitute conventional contact force.Researching on the hoisting rope in the sinking winch mechanism,an equivalent discretization model is obtained with complicated boundary conditions considered.The differential equations of motion of the hoisting system are formulated employing Lagrange’s equation and numerically solved using Runge–Kutta method.The simulation indicates that the horizontal swing is decreased in principle and the simulation with 800 discrete ropes is not performed more than 61 min.Therefore,this feasible strategy could not only guarantee the accuracy but also promote simulation efficiency and stability.The motion curves exported from ADAMS simulation coincide with one in numerical simulation,which validates both the numerical model and the driving strategy.JINJIE WANG GUOHUA CAO YANDONG WANG RONGHUA WU 2013International Journal of Modeling, Simulation, and Scientific Computing2013,4,3:1
16Fracture mechanics of methane clathrate hydrates显示文摘Fundamental mechanics of gas hydrates is of importance to evaluating geomechanical and geotechnical properties of gas hydrate deposits,but it remains largely unexplored yet due to insufficient direct experimental techniques and high-quality of gas hydrate samples.Here,classic molecular dynamic(MD)simulations are used to study the fracture mechanics of three main methane clathrate hydrates of sI,sII and sH types.The results show that the mechanical properties of those three methane clathrate hydrates are intrinsically different and are degraded by the presence of nanocracks.They show brittle facture and different fracture toughness.In terms of energy release rate,they are ranked as sH>sI>sII.Moreover,the three methane clathrate hydrates with nanocracks can be explained by a modified Griffith criterion.Moreover,it is intriguingly identified tip amorphization during the crack propagation process of the three methane clathrate hydrates,and sH methane clathrate hydrate with specific nanocrack exhibits slower crack propagation than other two methane clathrate hydrates.Jinjie Liu Ke Xu Li Yang Yanwen Lin Tong Li Xuezheng Gao Zhisen Zhang Jianyang Wu 2021Acta Mechanica Sinica2021,37,9:1
17Calculation and Test of Hitting Force of Firing Bullet on the Bulletproof Helmet显示文摘In this paper the hit force of firing bullet on bulletproof helmet has been computed and the test device has been described. The device is divided into two parts: 1) The bullet, helmet and mould are in one system, using moment theorem to calculate the hit force; 2) The mould, sensor and support pole are in one system, using the method in reference [1] that measures the dynamic strain and displacement of simulate target of bulletproof clothes. We compute the transfigure energy and momentum energy when hitting the mould, the work done by the sensor and the expend energy of support pole. We get the hit force of helmet using energy balance principle. 18晚期非小细胞肺癌化疗和靶向治疗失败后的挽救性化疗(英文)显示文摘Objective: We conducted a prospective phase II trial of single-agent salvage chemotherapy with docetaxel in patients with advanced non-small cell lung cancer (NSCLC) after failure of chemotherapy and gefitinib to assess the efficacy and toxicity of docetaxel in this setting. Methods: Patients with histologically confirmed NSCLC who were failure of chemotherapy and gefitinib were given docetaxel 75 mg/m2 intravenously for 30 min every 3 weeks until the toxicity was unacceptable or disease progressed. The response evaluation criteria in solid tumors (RECIST) guidelines were used for the evaluation of antitumor activity. Toxicity was graded according to the National Cancer Institute Common Toxicity Criteria version 2.0. Results: In total, 31 patients were enrolled in this phase II trial between February 2004 and December 2006, and 84 cycles (average 2.7 cycles) were given. We observed 4 partial responses (PRs) and 10 stable disease (SD) states in 31 eligible patients. The objective response rate was 12.9%, and the disease control rate was 45.2%. The median survival time (MST) was 10 months (95% CI, 5.05-15.08 months). The 1-year survival rate was 40.6%. The most common toxicities were neutropenia, anemia, and peripheral neuropathy that occurred as follows: 45% of the patients experienced grade 3 or 4 neutropenia, 29% experienced grade 3 anemia, and 25.8% had grade 3 peripheral neuropathy. No patient terminated docetaxel chemotherapy due to toxicity. 19The morphology of hydroxyapatite nanoparticles regulates clathrinmediated endocytosis in melanoma cells and resultant anti-tumor efficiency显示文摘Clathrin-mediated endocytosis plays a critical role for hydroxyapatite nanoparticles(HANPs)to enter tumor cells,induce mitochondrial apoptosis,and inhibit tumor growth.This study was aimed to investigate how the morphology of HANPs impacts the endocytosis of the particles in melanoma cells,and their anti-tumor effect by using in vitro cell experiments and in vivo tumor animal model.Three shapes of HANPs,including granular HANPs(G-HANPs),rod-like HANPs(R-HANPs),and needle-like HANPs(N-HANPs),were successfully prepared by wet chemical method.All the three HANPs could be internalized into A375 melanoma cells as indicated by cellular transmission electron microscopy images.Among these HANPs,only G-HANPs induced morphological change of mitochondria and loss of mitochondrial membrane potential(Δψm),and exhibited the greatest intracellular internalization efficiency in the tumor cells.Furthermore,the results of immunofluorescence staining and western blotting indicated that the level of adaptin-2(AP2)protein was up-regulated by all the HANPs,and highest in G-HANPs treated A375 cells.Moreover,in the tumor-bearing mouse model,we found that tumor growth was delayed by all the three HANPs,of which,G-HANPs delayed tumor growth most efficiently and presented a highest expression level of AP2 protein in tumor tissues.Therefore,this study suggested that the morphology of HANPs regulated their endocytosis efficiency and their effect to inhibit tumor growth.This work facilitates to direct the rational design of nano-materials for tumor therapy.Hongfeng Wu Yuchen Hua Jinjie Wu Qin Zeng Xiao Yang Xiangdong Zhu Xingdong Zhang 2022Nano Research2022,15,7:0
20Changes in Polyphenols and Antioxidant Activities of Yingshan Yunwu Tea during Digestion in Vitro显示文摘[Objectives]To explore the change rule of polyphenol content and antioxidant activity of coarse old leaves of Yingshan Yunwu Tea in the process of human digestion.[Methods]The coarse and old leaves of Yunwu tea in Yingshan,Huanggang,Hubei Province were selected as the research object,and their digestion in vitro was simulated.The total polyphenol content was determined by Folin-phenol reagent colorimetric method,and the DPPH radical scavenging activity and total antioxidant activity were determined.[Results]After simulated gastrointestinal digestion in vitro,the polyphenol content and antioxidant activity of coarse old leaf tea soup showed a downward trend.After gastrointestinal digestion,the polyphenol content in tea infusion separately decreased by 31.8%and 8.5%;the scavenging rate of DPPH free radical was 97%before digestion,decreased to 92%after gastric digestion and 65%after intestinal digestion,which decreased by 5%and 27%,respectively;after gastrointestinal digestion,the total antioxidant capacity of tea soup decreased by 4.7%and 3.1%,respectively.[Conclusions]This study provided a reference for the development and application of coarse old leaves of Yingshan Yunwu tea,and provided a reference for the nutritional value evaluation and comprehensive utilization of coarse old leaves,so as to make the best use of coarse tea leaves and reduce the waste of resources.Ruyi ZHENG Jinjie ZHOU Wenzhuo WU Jianfeng ZHAN Peng WU Ting DING Weixin WANG 2024Medicinal Plant2024,15,2:0
