| 1 | New generation of multi-scale NWP system (GRAPES):general scientific design显示文摘A new generation of numerical prediction system GRAPES (a short form of Global/Regional Assimilation and PrEdiction System) was set up in China Meteorological Administration (CMA). This paper focuses on the scientific design and preliminary results of the numerical prediction model in GRAPES, including basic idea and strategy of the general scientific design, multi-scale dynamic core, physical package configuration, architecture and parallelization of the codes. A series of numerical experiments using the real data with horizontal resolutions from 10 to 280 km and idealized experiments with very high resolution up to 100 m are conducted, giving encouraging results supporting the multi-scale application of GRAPES. The results of operational implementation of GRAPES model in some NWP centers are also presented with stress at evaluations of the capability to predict the main features of precipitation in China. | 2 | Long-term follow-up of the effects of fecal microbiota transplantation in combination with soluble dietary fiber as a therapeutic regimen in slow transit constipation显示文摘As some studies have reported that strategies targeting the gut microbiota such as fecal microbiota transplantation(FMT) with or without other microecological therapy might have efficacy in treating slow transit constipation(STC), we conducted a singlecenter, open-label trial to study the long-term effect of FMT combined with soluble dietary fiber(pectin) on STC. Thirty-one adult patients with STC were enrolled into the trial. Patients received 6-day FMT procedures repeatedly for the first 3 months and soluble dietary fiber(pectin) daily during the follow-up. The rate of clinical remission and improvement, stool consistency, the Wexner constipation scale, and assessment of constipation-related symptoms were evaluated at week 4 and 1 year later. The clinical remission and improvement rates at week 4 were 69.0%(20/29) and 75.9%(22/29), respectively. At the end of the study,48.3%(14/29) of patients continued to have at least three complete spontaneous bowel movements per week and 58.6%(17/29)of patients showed clinical improvements. Stool consistency, the Wexner constipation scale, and constipation symptoms improved both at short-term and long-term follow-up. | 3 | Genetic diversity of rice cultivars (Oryza sativa L.) in China and the temporal trends in recent fifty years显示文摘To understand geographical distribution of the genetic diversity of rice cultivars {Oryza sativa L.) and its trends in recent fifty years in China, 453 ’ accessions were analyzed by 36 microsatellites loci and 42 phenotypic traits. Results revealed that the genetic diversity by SSRs is highly consistent with that by phenotypic traits and the genetic diversity of indica cultivars was higher than that of japonica cultivars; the genetic diversity of cultivars declined from 1950s to 1980s and then increased greatly; among the six rice ecological zones (REZs), genetic diversity of REZTl was the highest and those of REZV and REZVT were the lowest at both DNA and phenotypic level. Jiangsu and Jiangxi provinces in the middle and lower reaches of Yangtze River and Sichuan province in southwest of China were the areas with the highest genetic diversity. | 4 | ~4.1 Ga xenocrystal zircon from Ordovician volcanic rocks in western part of North Qinling Orogenic Belt显示文摘High-precision U-Pb dating by in situ LA-ICP-MS yields an age of 4079±5 Ma for a xenocrystal zircon from Ordovician volcanics of the Caotangou Group in western part of the North Qinling Orogenic Belt. As a result, the North Qinling Orogenic Belt becomes one of a few localities in the world that contain Hadean age records (4276±6 Ma and 4404±8 Ma detrital zircons from Jack Hill of the Yilgarn craton, 4016 Ma Acasta gneisses of the Wopmay Orogeny and Burang quartzite with detrital zircon of 4103 Ma in Tibet). It is also the first report of the Hadean age in Phanerozoic volcanics. The finding of the 4.1 Ga xenocrystal zircon provides not only the geochronological record of the oldest crustal materials in China, but also the condition for further search for rocks forming in the region during the early time of the Earth’s evolution. Thirty-six zircon U-Pb dates from the Ordovician volcanic rocks are subgrouped into seven generations that represent different tectono-magmatic events in the North Qinling Orogenic Belt. | 8 | Complement C3 activation regulates the production of tRNA-derived fragments Gly-tRFs and promotes alcohol-induced liver injury and steatosis显示文摘Complement is known to play a role in alcoholic fatty liver disease (AFLD), but the underlying mechanisms are poorly understood, thereby constraining the development of a rational approach for therapeutic intervention in the complement system. C3 deficiency has been shown to impart protective effects against ethanol-induced hepatic steatosis and inflammation. Here we demonstrate a protection effect in wild-type mice by treatment with CR2-Crry, a specific inhibitor of C3 activation. The expression of glycine transfer (t) RNA-derived fragments (Gly-tRFs) is upregulated in ethanol-fed mice and inhibition of Gly-tRFs in vivo decreases chronic ethanol feeding-induced hepatosteatosis without affecting inflammation. The expression of Gly-tRF was downregulated in C3-deficient or CR2-Crry-treated mice, but not in C5-deficient mice;Gly-tRF expression was restored by the C3 activation products C3a or Asp (C3a-des-Arg) via the regulation of CYP2E1. Transcriptome profiling of hepatic tissues showed that Gly-tRF inhibitors upregulate the expression of sirtuin1 (Sirt1) and subsequently affect downstream lipogenesis and β-oxidation pathways. Mechanistically, Gly-tRF interacts with AGO3 to downregulate Sirt1 expression via sequence complementarity in the 3′ UTR. Notably, the expression levels of C3d, CYP2E1 and Gly-tRF are upregulated, whereas Sirt1 is decreased in AFLD patients compared to healthy controls. Collectively, our findings suggest that C3 activation products contribute to hepatosteatosis by regulating the expression of Gly-tRF. | 9 | Verification of GRAPES unified global and regional numerical weather prediction model dynamic core显示文摘During the past few years, most of the new developed numerical weather prediction models adopt the strategy of multi-scale technique. Therefore, China Meteorological Administration has devoted to developing a new generation of global and regional multi-scale model since 2003. In order to validate the performance of the GRAPES (Global and Regional Assimilation and PrEdiction System) model both for its scientific design and program coding, a suite of idealized tests has been proposed and conducted, which includes the density flow test, three-dimensional mountain wave and the cross-polar flow test. The density flow experiment indicates that the dynamic core has the ability to simulate the fine scale nonlinear flow structures and its transient features. While the three-dimensional mountain wave test shows that the model can reproduce the horizontal and vertical propagation of internal gravity waves quite well. Cross-polar flow test demonstrates the rationality of both for the semi-Lagrangian departure point calculation and the discretization of the model near the poles. | 10 | Cardiac autonomic nerve fiber regeneration in chronic heart failure Do Akt gene-transduced mesenchymal stem cells promote repair?显示文摘BACKGROUND: Transplantation of Akt-over-expressing mesenchymal stem cells (Akt-MSCs) has been shown to repair infarcted myocardium and improve cardiac function. However, little is known about the therapeutic effects of Akt-MSCs on cardiac autonomic neuropathy in chronic heart failure (CHF). OBJECTIVE: The present study used adriamycin-induced CHF rat models to observe the effect of Akt-MSCs on cardiac autonomic nervous regeneration and the factors mediating this effect. DESIGN, TIME AND SETTING: A randomized, controlled animal experiment was performed at the Central Laboratory of Basic Medical College, China Medical University, between September 2008 and April 2009. MATERIALS: Rabbit anti-choline acetyltransferase (ChAT), growth associated protein-43 (GAP-43), synaptophysin (SYN) polyclonal antibodies and the secondary antibody (goat anti-rabbit IgG) were purchased from Boster, China. Cat-A-Kit assay system was provided by Amersham, USA. METHODS: (1) Adult rat MSCs were isolated and cultured for the preparation of Akt-MSCs. (2) Forty male Wistar rats were intramyocardially administered adriamycin at 2 mg/kg over 3 days for a total of five times and once a week for additional five times thereafter to establish CHF models. At 2 weeks after final adriamycin treatment, 34 successful CHF rat models were randomized to three groups: Akt-MSCs (n = 11), simple MSCs (s-MSCs, n =11), and control (n = 12). Each group was intravenously administered Akt-MSCs (2×106 cells in 100 μL PBS), s-MSCs (2×106 cells in 100 μL PBS) or equal volume of phosphate buffered saline, once a day for a total of three times. MAIN OUTCOME MEASURES: At 4 weeks after final adriamycin treatment, myocardial norepi- nephrine (NE) content was detected using a Cat-A-Kit assay system. Myocardial ChAT, SYN and GAP-43 were performed by immunohistochemistry and Western blot analysis. Prior to, 2 and 4 weeks after adriamycin treatment, echocardiographic examination was performed and left ventricular ejection fraction (LVEF) was determined. RESULTS: Myocardial NE content, as well as SYN-positive and GAP-43-positive nerve fiber density and expression, and LVEF, was the greatest in the Akt-MSCs group, followed by the s-MSCs group, and lastly the control group (P < 0.05 or P < 0.01). ChAT expression was similar between Akt-MSCs and s-MSCs groups, but it was higher compared with the control group (P < 0.05). NE contents were negatively correlated to LVEF (r = -0.64, P = 0.015). | 13 | Overexpression of a dominant-negative mutant of SIRT1 in mouse heart causes cardiomyocyte apoptosis and early-onset heart failure显示文摘SIRT1,a mammalian ortholog of yeast silent information regulator 2(Sir2),is an NAD+-dependent protein deacetylase that plays a critical role in the regulation of vascular function.The current study aims to investigate the functional significance of deacetylase activity of SIRT1 in heart.Here we show that the early postnatal hearts expressed the highest level of SIRT1deacetylase activity compared to adult and aged hearts.We generated transgenic mice with cardiac-specific expression of a dominant-negative form of the human SIRT1(SIRT1H363Y),which represses endogenous SIRT1 activity.The transgenic mice displayed dilated atrial and ventricular chambers,and died early in the postnatal period.Pathological,echocardiographic and molecular phenotype confirmed the presence of dilated cardiomyopathy.Terminal deoxynucleotidyl transferase-mediated dUTP nick-end-labeling analysis revealed a greater abundance of apoptotic nuclei in the hearts of transgenic mice.Furthermore,we show that cardiomyocyte apoptosis caused by suppression of SIRT1 activity is,at least in part,due to increased p53acetylation and upregulated Bax expression.These results indicate that dominant negative form of SIRT1(SIRT1H363Y)overexpression in mouse hearts causes cardiomyocyte apoptosis and early-onset heart failure,suggesting a critical role of SIRT1 in preserving normal cardiac development during the early postnatal period. | MU WenLi ZHANG QingJun TANG XiaoQiang FU WenYan ZHENG Wei LU YunBiao LI HongLiang WEI YuSheng LI Li SHE ZhiGang CHEN HouZao LIU DePei | 2014 | Science China(Life Sciences)2014,57,9: | 12 |

The package's component integrality is comparative with the other advanced models physics. A three-level structure of connecting GRAPES physics and dynamic frame has been constructed. The friendly interface is designed for users to plug in their own physics packages. Phenomenon of grid-point storm rainfall in numerical prediction is analyzed with the numerical tests. The scheme of air vertical velocity calculation is improved. Optimizing tests of physics schemes are performed with the correlative parameters adjusting. The results show that the false grid-point storm rainfall is removed by precipitation scheme improving. | 15 | Adipocyte-derived microvesicles from obese mice induce M1 macrophage phenotype through secreted miR-155显示文摘The pro-inflammatory profile of M1 macrophage accumulation in adipose tissue is a central event leading to the metabolic complications of obesity.However,the mechanisms by which M1 macrophages are enriched in adipose tissue during weight gain remain incompletely understood.Here,we investigated the effects of adipocyte-derived microvesicles(ADM)on modulating macrophage phenotype in mice and explored the involved molecular signalling pathways.We found that,compared with ADM from lean mice(SD ADM),ADM from obese mice(HFD ADM)significantly enhanced M1 marker expression.The quantitative RTPCR assay demonstrated that miR-155 was upregulated in both HFD ADM and HFD ADM-treated macrophages.By depleting miR-155 expression in HFD ADM and increasing miR-155 level in SD ADM,we further illustrated that miR-155 in ADM-induced M1 macrophage polarization.Functionally,in contrast to SD ADM,HFD ADM significantly decreased the protein level of SOCS1,a proven miR-155 target,leading to activation of STAT1,and suppression of STAT6 signalling;these effects were reversed by silencing miR-155 in HFD ADM.Furthermore,the supernatant of bone marrow-derived macrophages pre-stimulated with miR-155-bearing ADM interfered with insulin signalling and insulin-induced glucose uptake in adipocytes.Collectively,these results provide the first evidence that M1 macrophage polarization can be mediated by miR-155-bearing ADM,which reciprocally regulates insulin signalling and glucose uptake in adipocytes.Our study reveals a novel mechanism through which obesity induces an imbalance in the M1-to-M2 macrophage ratio in adipose tissue,thus causing chronic inflammation and local insulin resistance. | Yaqin Zhang Hongliang Mei Xiaoai Chang Fang Chen Yunxia Zhu Xiao Han | 2016 | Journal of Molecular Cell Biology2016,8,6: | 11 |
| 16 | Cell multiplication, apoptosis and p-Akt protein expression of bone mesenchymal stem cells of rat under hypoxia environment显示文摘Objective: To elucidate whether cell multiplication, apoptosis, glucose intake and p-Akt protein expression of bone Mesenchy-mal Stem Cells(MSCs) of rats is influenced by a hypoxic environment ex vivo. Methods: Passage 3 of bone marrow MSCs takenfrom Wistar rats, were cultured in a culturing chamber with 94%N2, 1%O2, 5%CO2 at 37℃. At different hypoxia time points, 0, 0.5,1, 4 and 8 h, glucose uptake was assayed by using radiation isotope 3H-G, Apoptotic Rate(AR) and dead rate(DR) were analyzedby flow cytometry(FCM) after Annexin V/PI staining, cell multiplication(by MTT methods) and p-Akt protein by immunocyto-chemistry and western blot. Results: Assay for CD29+, CD44+, CD71+, CD34- , Tn T+(after 5-azacytidine agent inducing) andALP+(after bone differentiation agent inducing) suggested these bone-derived cells were MSCs. The 3H-G intaking ratio(CPM/flask value: 157 ±11, 110 ±11, 107 ±13, 103 ±10, 100 ±9 and 98 ±10) of MSCs at different hypoxia time points, significantlydecreased compared to that of normoxia(P < 0.01) and tended to descend slowly with hypoxia time duration, for which there wasno statistical significance(P > 0.05). The AR(0.09 ±2.03%, 12.9 ±1.72%, 13.7 ±2.26%, 13.8 ±3.01%, 14.1 ±2.78% and 14.7 ±4.01% at 0, 0.5, 1, 4 and 8 h, respectively, P < 0.01) and DR (0.04 ±1.79%, 0.93 ±1.85%, 3.11 ±2.14%, 4.09 ±2.36%, 4.72 ±2.05% and 4.91 ±3.72% at 0, 0.5, 1, 4 and 8 h, respectively, P < 0.05) at different hypoxia time points significantly increasedcompared to those time in normoxia; The AR further went up with time (P < 0.05), however there was no statistical significance(P > 0.05) for the DR. Optical absorption value of MTT methods at different hypoxia time points significantly decreased com-pared to those with a corresponding normoxia time (P < 0.01) and degraded with time (in an hypoxic environment - P < 0.01).IOD of p-Akt protein of MSCs at different hypoxia time points significantly increased (0.367 ±0.031, 0.556 ±0.023, 0.579 ±0.013, 0.660 ±0.024, 0.685 ±0.039 and 0.685 ±0.011, respectively) compared to their equivalents in normoxia (P<0.05), how-ever, there was no statistical significance (P > 0.05) for different hypoxia time points. Hypoxia may result in ultramicrostructurechanges, such as defluvium of Microvilli, apoptotic body, 'margination'and so on and are further aggravated with hypoxia timestretching. | 17 | A new-generation sand and dust storm forecasting system GRAPES_CUACE/Dust:Model development,verification and numerical simulation显示文摘Based on the new-generation Global/Regional Assimilation and PrEdiction System(GRAPES) developed by the Numerical Prediction Research Center,China Meteorological Administration and the Chinese Unified Atmospheric Chemistry Environment for Dust Atmospheric Chemistry Module(CUACE/Dust) developed by the Centre for Atmosphere Watch and Services(CAWAS) of the Chinese Academy of Meteorological Sciences(CAMS) ,the China sand and dust storm forecasting system GRAPES /CUACE-Dust model has been established.The latest data of land desertification,optical properties of China sand and dust aerosols,daily soil moisture and snow cover over China main land was introduced in this system.The system showed good performance in mass conservation.The comparisons of real-time prediction outputs with surface observations and aerosol indices derived from TOMS ozone spectrophotometers(TOMS AI) indicate that the model can predict the outbreak,development,transport and depletion processes of sand and dust storms accurately over China and the East Asian region.The system makes real-time quantitative prediction of a series of elements including sand and dust injection from the surface,dry and wet deposition amount,dust concentration and optical depth.We selected 7 major dust storms occurring in April 2006 and analyzed the spatiotemporal distribution patterns of dust emission,dry and wet deposition and atmospheric load in this paper.The results showed that about 225 million tons of dust were emitted into the atmosphere from the deserts over east Asia in that month.The 3 major sand and dust sources were just as the following:The deserts in northern Inner Mongolia and the boundary area around China-Mongolia were the first dust sources with a contribution of 153 million tons accounting for 68%of the total emission.The Taklimakan Desert ranked second and contributed approximately 40 million tons accounting for 17%of the dust emission.The Onqin Daga Sandland emitted about 15 million tons or 7%of the total emission.The contributions from other deserts,sandy lands and abandoned farmlands were about only 8%of the total dust emission.The total deposition over the East Asian region in April 2006 was 136 million tons.The regional distribution of dust deposition showed that the 3 major sources were also the major deposition areas.The deposition amount in the 3 major sources accounted for 78%of the total with about 135 million tons falling back to the source regions.The secondary deposition area was the China mainland downriver,where more than 2 million tons deposited accounting for 16%of the total deposition.The deposition over the region east of 120°E including China off-shore regions,Korean Peninsula,Japan and the West Pacific took only 6%of the total deposition,which was about 850000 tons.The analysis on dry and wet deposition showed that dry deposition dominated in the process,accounting for 94%of the total sand and dust depositions in the period and wet deposition only was around for 6%,since it was generally dry with less rainfall over northern China in April. | WANG Hong GONG ShanLing ZHANG HongLiang CHEN Yong SHEN XueShun CHEN DeHui XUE JiShan SHEN YuanFang WU XiangJun JIN ZhiYan | 2010 | Chinese Science Bulletin2010,55,7: | 10 |
| 18 | Pre-stack inversion for caved carbonate reservoir prediction:A case study from Tarim Basin,China显示文摘The major storage space types in the carbonate reservoir in the Ordovician in the TZ45 area are secondary dissolution caves.For the prediction of caved carbonate reservoir,post-stack methods are commonly used in the oilfield at present since pre-stack inversion is always limited by poor seismic data quality and insufficient logging data.In this paper,based on amplitude preserved seismic data processing and rock-physics analysis,pre-stack inversion is employed to predict the caved carbonate reservoir in TZ45 area by seriously controlling the quality of inversion procedures.These procedures mainly include angle-gather conversion,partial stack,wavelet estimation,low-frequency model building and inversion residual analysis.The amplitude-preserved data processing method can achieve high quality data based on the principle that they are very consistent with the synthetics.Besides,the foundation of pre-stack inversion and reservoir prediction criterion can be established by the connection between reservoir property and seismic reflection through rock-physics analysis.Finally,the inversion result is consistent with drilling wells in most cases.It is concluded that integrated with amplitude-preserved processing and rock-physics,pre-stack inversion can be effectively applied in the caved carbonate reservoir prediction. | Zhang Yuanyin Sam Zandong Sun Yang Haijun Wang Haiyang HanJianfa Gao Hongliang Luo Chunshu Jing Bing | 2011 | Petroleum Science2011,8,4: | 9 |

| 20 | Assessment of the sources and transformations of nitrogen in a plain river network region using a stable isotope approach显示文摘The great spatial and temporal variability in hydrological conditions and nitrogen(N)processing introduces large uncertainties to the identification of N sources and quantifying N cycles in plain river network regions. By combining isotopic data with chemical and hydrologic measurements, we determined the relative importance of N sources and biogeochemical N processes in the Taige River in the East Plain Region of China. The river was polluted more seriously by anthropogenic inputs in winter than in summer. Manure and urban sewage effluent were the main nitrate(NO-3) sources, with the nitrification of N-containing organic materials serving as another important source of NO-3. In the downstream, with minor variations in hydrological conditions, nitrification played a more important role than assimilation for the decreasing ammonium(NH+4-N) concentrations.The N isotopic enrichment factors(ε) during NH+4utilization ranged from- 13.88‰ in March to- 29.00‰ in July. The ratio of the increase in δ18O and δ15N of river NO-3in the downstream was 1.04 in January and 0.92 in March. This ratio indicated that NO-3assimilation by phytoplankton was responsible for the increasing δ15N and δ18O values of NO-3in winter.
