2Magnetic properties of high-road-side pine tree leaves in Beijing and their environ-mental significance显示文摘This is a report on magnetic properties of highroad-side tree (Pinus pumila Regel) leaves collected along an expressway linking Beijing City and the Capital International Airport and further focus on their environmental contributions. A series of rock magnetic experiments show that the primary mag-netic mineral of leaf samples was identified to be magnetite,in the pseudo-single domain (PSD) grain size range 0.2―5.0 μm. On the other hand,magnet-ite concentration and grain size in leaves are ascer-tained to decrease with increasing of sampling dis-tance to highroad asphalt surface,suggesting that high magnetic response to traffic pollution is localized within a distance of some two meters away from highroad asphalt surface. Although magnetic suscep-tibility is ordinarily regarded as a simple,rapid and low-cost method for monitoring traffic pollution,saturation isothermal remanent magnetization (SIRM) can be treated as a valid proxy for monitoring air par-ticulate matter (PM) when samples are magnetically weak. 3The characteristic differences of tropical cyclones forming over the western North Pacific and the South China Sea显示文摘The best track dataset of tropical cyclones in the western North Pacific (WNP) and the South China Sea (SCS) from 1977 to 2005 during the satellite era, the NCEP/NCAR reanalysis dataset and the extended reconstructed sea surface temperature dataset are employed in this study. The main climatological characteristics of tropical cyclone formation over the WNP and the SCS are compared. It is found that there is obviously different for the locations of tropical cyclone origins, achieving the lowest central pressure and termination points between over the WNP and over the SCS. The annual number of tropical cyclones forming over the SCS is obviously less than over the WNP, and there is a significant negative correlation with the correlation coefficient being -0.36 at the 5% significance level between over the WNP and over the SCS. The mean speed of tropical cyclone moving is 6.5 m /s over the WNP and 4.6 m /s over the SCS. The mean lowest central pressure of tropical cyclones is obviously weaker over the SCS than over the WNP. The tropical cyclone days per year, mean total distance and total displacement of tropical cyclone traveled over the WNP are all obviously longer than those over the SCS. Tropical cyclone may intensify to Saffir-Simpson hurricane scale 5 over the WNP, but no tropical cyclone can intensify to Saffir-Simpson hurricane scale 3 over the SCS. The changing ranges of the radii(R15,R16) of the 15.4 m /s winds them and the 25.7 m /s winds over the WNP are obviously wider than those over the SCS, and the median values of the radii over the WNP are also larger than those over the SCS. For the same intensity of tropical cyclones, both radii have larger medians over the WNP than over the SCS. The correlations of annual mean tropical cyclone size parameters between over the WNP and over the SCS are not significant. 4Brain structural changes and their correlation with vascular disease in type 2 diabetes mellitus patients: a voxel-based morphometric study显示文摘Voxel-based morphometry has been used in the study of alterations in brain structure in type 1 diabetes mellitus patients. These changes are associated with clinical indices. The age at onset, pathogenesis, and treatment of type 1 diabetes mellitus are different from those for type 2 diabetes mellitus. Thus, type 1 and type 2 diabetes mellitus may have different impacts on brain structure. Only a few studies of the alterations in brain structure in type 2 diabetes mellitus patients using voxel-based morphometry have been conducted, with inconsistent results. We detected subtle changes in the brain structure of 23 cases of type 2 diabetes mellitus, and demonstrated that there was no significant difference between the total volume of gray and white matter of the brain of type 2 diabetes mellitus patients and that in controls. Regional atrophy of gray matter mainly occurred in the right temporal and left occipital cortex, while regional atrophy of white matter involved the right temporal lobe and the right cerebellar hemisphere. The ankle-brachial index in patients with type 2 diabetes mellitus strongly correlated with the volume of brain regions in the default mode network. The ankle-brachial index, followed by the level of glycosylated hemoglobin, most strongly correlated with the volume of gray matter in the right temporal lobe. These data suggest that voxel-based morphometry could detect small structural changes in patients with type 2 diabetes mellitus. 9High-Resolution Genome-wide Association Study Identifies Genomic Regions and Candidate Genes for Important Agronomic Traits in Wheat显示文摘Wheat(Triticum aestivum)is a major staple food crop worldwide.Genetic dissection of important agronomic traits is essential for continuous improvement of wheat yield to meet the demand of the world's growing population.We conducted a large-scale genome-wide association study(GWAS)using a panel of 768 wheat cultivars that were genotyped with 327609 single-nucleotide polymorphisms generated by genotyping-by-sequencing and detected 395 quantitative trait loci(QTLs)for 12 traits under 7 environments.Among them,273 QTLs were delimited to≤1.0-Mb intervals and 7 of them are either known genes(Rht-D,Vrn-B1,and Vrn-D1)that have been cloned or known QTLs(TaGA2ox8,APO1,TaSus1-7B,and Rht12)that were previously mapped.Eight putative candidate genes were identified for three QTLs that enhance spike seed setting and grain size using gene expression data and were validated in three bi-parental populations.Protein sequence analysis identified 33 putative wheat orthologs that have high identity with rice genes in QTLs affecting similar traits.Large r^2 values for additive effects observed among the QTLs for most traits indicated that the phenotypes of these identified QTLs were highly predictable.Results from this study demonstrated that significantly increasing GWAS population size and marker density greatly improves detection and identification of candidate genes underlying a QTL,solidifying the foundation for large-scale QTL fine mapping,candidate gene validation,and developing functional markers for genomics-based breeding in wheat.Yunlong Pang Chunxia Liu Danfeng Wang Paul St.Amand Amy Bernardo Wenhui Li Fang He Linzhi Li Liming Wang Xiufang Yuan Lei Dong Yu Su Huirui Zhang Meng Zhao Yunlong Liangi Hongze Jia Xitong Shen Yue Lu Hongming Jiang Yuye Wu Anfei Li Honggang Wang Lingrang Kong Guihua Bai Shubing Liu 2020Molecular Plant2020,13,9:8

11Evaluation of the Global and Regional Assimilation and Prediction System for Predicting Sea Fog over the South China Sea显示文摘In the South China Sea, sea fog brings severe disasters every year, but forecasters have yet to implement an effective seafog forecast. To address this issue, we test a liquid-water-content-only(LWC-only) operational sea-fog prediction method based on a regional mesoscale numerical model with a horizontal resolution of about 3 km, the Global and Regional Assimilation and Prediction System(GRAPES), hereafter GRAPES-3 km. GRAPES-3 km models the LWC over the sea, from which we infer the visibility that is then used to identify fog. We test the GRAPES-3 km here against measurements in 2016 and 2017 from coastal-station observations, as well as from buoy data, data from the Integrated Observation Platform for Marine Meteorology, and retrieved fog and cloud patterns from Himawari-8 satellite data. For two cases that we examine in detail, the forecast region of sea fog overlaps well with the multi-observational data within 72 h. Considering forecasting for0–24 h, GRAPES-3 km has a 2-year-average equitable threat score(ETS) of 0.20 and a Heidke skill score(HSS) of 0.335,which is about 5.6%(ETS) and 6.4%(HSS) better than our previous method(GRAPES-MOS). 12Hijacking antibody-induced CTLA-4 lysosomal degradation for safer and more effective cancer immunotherapy显示文摘It remains unclear why the clinically used anti-CTLA-4 antibodies, popularly called checkpoint inhibitors, have severe immunotherapy-related adverse effects (irAEs) and yet suboptimal cancer immunotherapeutic effects (CITE). Here we report that while irAE-prone Ipilimumab and TremeIgG1 rapidly direct cell surface CTLA-4 for lysosomal degradation, the non-irAE-prone antibodies we generated, HL12 or HL32, dissociate from CTLA-4 after endocytosis and allow CTLA-4 recycling to cell surface by the LRBA-dependent mechanism. Disrupting CTLA-4 recycling results in robust CTLA-4 downregulation by all anti-CTLA-4 antibodies and confers toxicity to a non-irAE-prone anti-CTLA-4 mAb. Conversely, increasing the pH sensitivity of TremeIgG1 by introducing designed tyrosine-to-histidine mutations prevents antibody-triggered lysosomal CTLA-4 downregulation and dramatically attenuates irAE. Surprisingly, by avoiding CTLA-4 downregulation and due to their increased bioavailability, pH-sensitive anti-CTLA-4 antibodies are more effective in intratumor regulatory T-cell depletion and rejection of large established tumors. 13Uncommon EGFR mutations in a cohort of Chinese NSCLC patients and outcomes of first-line EGFR-TKIs and platinum-based chemotherapy显示文摘Objective: Data on the clinical activity of epidermal growth factor receptor(EGFR) tyrosine kinase inhibitors(TKIs) in patients with non-small-cell lung cancer(NSCLC) and uncommon EGFR mutations remain insufficient.This study aimed to investigate the effect of first-line EGFR-TKIs or platinum-based chemotherapy in NSCLC patients with uncommon EGFR mutations.Methods: We retrospectively enrolled 504 patients with EGFR-mutant NSCLC.The clinical characteristics and treatment outcomes were collected and compared between patients with common and uncommon EGFR-mutant NSCLC.Results: Seventy patients(13.9%) harboring uncommon EGFR mutations were included.Thirty of these patients received EGFR-TKIs and 40 received platinum-based chemotherapy as first-line therapy.The objective response rate(ORR) and median progression-free survival(m PFS) of patients treated with TKIs in the uncommon mutation group was significantly inferior to that in the common mutation group(ORR: 23.3% vs.51.8%,P=0.003; m PFS:7.1 vs.10.9 months,P<0.001).In the uncommon group,m PFS was similar between first-line EGFR-TKIs treatment and platinum-based chemotherapy(7.1 vs.6.1 months,P=0.893).In patients with EGFR G719 X or L861 Q mutations,the m PFS was longer in the first-line EGFR-TKIs treatment group than in the chemotherapy group,but the difference was not statistically significant(G719 X: 8.2 vs.5.8 months,P=0.061; L861 Q: 7.6 vs.4.1 months,P=0.872).Multivariate analyses identified adenocarcinoma(P=0.003) as the independent predictive factor for PFS in patients with uncommon EGFR mutations who were treated with first-line EGFR-TKIs.Conclusions: The current study demonstrated that the effect of first-line EGFR-TKIs was similar to that of platinum-based chemotherapy in patients with uncommon EGFR-mutant NSCLC.Adenocarcinoma was the independent predictive factor for PFS in uncommon EGFR-mutant NSCLC patients treated with first-line EGFRTKIs.Jinpeng Shi Hui Yang Tao Jiang Xuefei Li Chao Zhao Limin Zhang Sha Zhao Xiaozhen Liu Yijun Jia Yan Wang Lei Xi Shijia Zhang Chunxia Su Shengxiang Ren Caicun Zhou 2017Chinese Journal of Cancer Research2017,29,6:6

15Transcription factor ZmNAC126 plays an important role in transcriptional regulation of maize starch synthesis-related genes显示文摘Maize(Zea mays L.)is one of the most important food crops in the world,and starch is the main component of its endosperm.Transcriptional regulation plays a vital role in starch biosynthesis.However,it is not well understood in maize.We report the identification of the transcription factor ZmNAC126 and its role in regulation of starch synthesis in maize.Transcriptional expression of ZmNAC126 was higher in maize endosperm and kernels than in roots or stems.ZmNAC126 shared a similar expression pattern with starch synthesis genes during seed development,and its expression pattern was also consistent with the accumulation of starch.ZmNAC126 is a typical transcription factor with a transactivation domain between positions 201 and 227 of the amino acid sequence,is located in the nucleus,and binds to CACG repeats in vitro.Yeast one-hybrid assay revealed that ZmNAC126 bound the promoters of ZmGBSSI,ZmSSIIa,ZmSSIV,ZmISA1,and ZmISA2.Transient overexpression of ZmNAC126 in maize endosperm increased the activities of promoters pZmSh2,pZmBt2,pZmGBSSI,pZmSSIIIa,and pZmBT1 but inhibited the activities of pZmISA1 and pZmISA2.ZmNAC126 thus acts in starch synthesis by transcriptionally regulating targeted starch synthesis-related genes in maize kernels.Qianlin Xiao Yayun Wang Hui Li Chunxia Zhang Bin Wei Yongbin Wang Huanhuan Huang Yangping Li Guowu Yu Hanmei Liu Junjie Zhang Yinghong Liu Yufeng Hu Yubi Huang 2021The Crop Journal2021,9,1:5

17Single copper sites dispersed on hierarchically porous carbon for improving oxygen reduction reaction towards zinc-air battery显示文摘The demand for high-performance non-precious-metal electrocatalysts to replace the noble metal-based catalysts for oxygen reduction reaction(ORR)is intensively increasing.Herein,single-atomic copper sites supported on N-doped three-dimensional hierarchically porous carbon catalyst(Cu_(1)/NC)was prepared by coordination pyrolysis strategy.Remarkably,the Cu_(1)/NC-900 catalyst not only exhibits excellent ORR performance with a half-wave potential of 0.894 V(vs.RHE)in alkaline media,outperforming those of commercial Pt/C(0.851 V)and Cu nanoparticles anchored on N-doped porous carbon(CuNPs/NC-900),but also demonstrates high stability and methanol tolerance.Moreover,the Cu_(1)/NC-900 based Zn-air battery exhibits higher power density,rechargeability and cyclic stability than the one based on Pt/C.Both experimental and theoretical investigations demonstrated that the excellent performance of the as-obtained Cu_(1)/NC-900 could be attributed to the synergistic effect between copper coordinated by three N atoms active sites and the neighbouring carbon defect,resulting in elevated Cu d-band centers of Cu atoms and facilitating intermediate desorption for ORR process.This study may lead towards the development of highly efficient non-noble metal catalysts for applications in electrochemical energy conversion.Wenjie Wu Yan Liu Dong Liu Wenxing Chen Zhaoyi Song Ximin Wang Yamin Zheng Ning Lu Chunxia Wang Junjie Mao Yadong Li 2021Nano Research2021,14,4:4
18Research on performance of complex networks based on principal component analysis显示文摘For complex networks, their effectiveness and invulnerability are extremely important. With the development of complex networks, how to evaluate the effectiveness and invulnerability of these networks becomes an important research topic. The relationship among many influencing factors is very complicated, so it is essential to confirm the weighting coefficient of these influencing factors. Principal component analysis(PCA) is proposed to evaluate the performance of complex networks. It can improve one-sidedness of the single evaluation index and select different evaluation models according to different complex networks, which make the evaluation result more accurate. Performance of complex networks can be predicted according to comprehensive evaluation model. To verify the rationality and validity of this method, several small-world networks with different probability values and scale-free network are chosen to evaluate the network performance. 19LncRNA NORAD promotes the progression of myocardial infarction by targeting the miR-22-3p/PTEN axis显示文摘NORAD is a newly identified long non-coding RNA(lncRNA)that plays an important role in cancers.NORAD has been found to be highly expressed in the mouse model of acute myocardial infarction(AMI).However,the role of NORAD in the regulation of AMI remains unknown.In the present study,we aimed to investigate the function of NORAD in AMI and explore the potential regulatory mechanisms.A mouse model of AMI was established and NORAD was knocked-down.The infarcted size of heart tissues and the cardiac function were evaluated.In addition,two cardiomyocyte cell lines were treated with hypoxia/re-oxygenation(H/R)to mimic AMI in vitro.Luciferase reporter assay,RNA pull-down assay,fluorescence in situ hybridization,qRT-PCR,and western blot analysis were performed.Apoptotic cells and the levels of L-lactate dehydrogenase(LDH)and malondialdehyde(MDA)were detected.Our results show that downregulation of NORAD efficiently attenuates heart damage in the AMI mouse model.NORAD interacts with miR-22-3p.Knock-down of NORAD inhibits H/R-induced cell apoptosis and reduces LDH and MDA levels,while its effects are abolished by miR-22-3p inhibitor.MiR-22-3p interacts with PTEN and inhibits its expression.Overexpression of miR-22-3p inhibits H/R-induced cell apoptosis and reduces LDH and MDA levels,while its effects are abolished by overexpression of PTEN.Finally,overexpression of NORAD inhibits the AKT/mTOR signaling pathway,and its effects are attenuated by overexpression of miR-22-3p.Taken together,our study reveals that NORAD promotes the progression of AMI by regulating the miR-22-3p/PTEN axis,and the AKT/mTOR signaling may also be involved in the regulatory processes.Chunxia Li Lihui Zhang Xingpeng Bu Guofang Chu Xiaofang Zhao Yaru Liu 2022Acta Biochimica et Biophysica Sinica2022,54,4:3
20Development and validation of an HPLC–MS/MS method to determine clopidogrel in human plasma显示文摘A quantitative method for clopidogrel using online-SPE tandem LC–MS/MS was developed and fully validated according to the well-established FDA guidelines.The method achieves adequate sensitivity for pharmacokinetic studies,with lower limit of quantifications(LLOQs)as low as 10 pg/m L.Chromatographic separations were performed on reversed phase columns Kromasil Eternity-2.5-C18-UHPLC for both methods.Positive electrospray ionization in multiple reaction monitoring(MRM)mode was employed for signal detection and a deuterated analogue(clopidogrel-d_4)was used as internal standard(IS).Adjustments in sample preparation,including introduction of an online-SPE system proved to be the most effective method to solve the analyte back-conversion in clinical samples.Pooled clinical samples(two levels)were prepared and successfully used as real-sample quality control(QC)in the validation of back-conversion testing under different conditions.The result showed that the real samples were stable in room temperature for 24 h.Linearity,precision,extraction recovery,matrix effect on spiked QC samples and stability tests on both spiked QCs and real sample QCs stored in different conditions met the acceptance criteria.This online-SPE method was successfully applied to a bioequivalence study of75 mg single dose clopidogrel tablets in 48 healthy male subjects.Gangyi Liu Chunxia Dong Weiwei Shen Xiaopei Lu Mengqi Zhang Yuzhou Gui Qinyi Zhou Chen Yu 2016Acta Pharmaceutica Sinica B2016,6,1:3
