2Annual 30-m land use/land cover maps of China for 1980–2015 from the integration of AVHRR, MODIS and Landsat data using the BFAST algorithm显示文摘Annual land use land cover(LULC)change information at medium spatial resolution(i.e.at 30 m)is required in numerous subjects,such as biophysical modelling,land management and global change studies.Annual LULC information,however,is usually not available at continental or national scale due to reasons such as insufficient remote sensing data coverage or lack of computational capabilities.Here we integrate high temporal resolution and coarse spatial resolution satellite images(i.e.,Moderate Resolution Imaging Spectroradiometer(MODIS)and Global Inventory Modelling and Mapping Studies(GIMMS)normalized difference vegetation index(NDVI))with high spatial resolution datasets(China’s Land-Use/cover Datasets(CLUDs)derived from 30-meter Landsat TM/ETM+/OLI)to generate reliable annual nominal 30 m LULC maps for the whole of China between 1980 and 2015.We also test the performance of a statistical based change detection algorithm(Breaks for Additive Seasonal and Trend),originally designed for tracking forest change,in classifying all-type LULC change.As a result,a nominal 30 m annual land use/land cover datasets(CLUD-A)from 1980 to 2015 was developed for the whole China.The mapping results were assessed with a change sample dataset,a regional annual validation sample set and a three-year China sample set.Of the detected change years,75.61%matched the exact time of conversion within±1 year.Annual mapping results provided a detail process of urbanization,deforestation,afforestation,water and cropland dynamics over the past 36 years.The consistent characterization of land change dynamics for China can be further used in scientific research and to support land management for policy-makers.Yidi XU Le YU Dailiang PENG Jiyao ZHAO Yuqi CHENG Xiaoxuan LIU Wei LI Ran MENG Xinliang XU Peng GONG 2020Science China Earth Sciences2020,63,9:12
3Effective suppression of stimulated Raman scattering in half 10 kW tandem pumping fiber lasers using chirped and tilted fiber Bragg gratings显示文摘The average power of fiber lasers has been scaled deeply into the kW regime in the past years. However, stimulated Raman scattering(SRS) is still a major factor limiting further power scaling. Here, we have demonstrated for the first time, to the best of our knowledge, the suppression of SRS in a half 10 kW tandem pumping fiber amplifier using chirped and tilted fiber Bragg gratings(CTFBGs). With specially self-designed and manufactured CTFBGs inserted between the seed laser and the amplifier stage, a maximum SRS suppression ratio of >15 dB in spectrum is observed with no reduction in laser efficiency. With one CTFBG, the effective output power is improved to3.9 kW with a beam quality M2 factor of ~1.7 from <3.5 k W with an M2 factor of >2; with two CTFBGs, the effective laser power reaches 4.2 kW with an increasing ratio of 20% and an M2 factor of ~1.8, and further power improvement is limited by the power and performance of the 1018 nm pump sources. 4Regulation of Rice Tillering by RNA-Directed DNA Methylation at Miniature Inverted-Repeat Transposable Elements显示文摘Tillering is a major determinant of rice plant architecture and grain yield.Here,we report that depletion of rice OsNRPD1a and OsNRPD1b,two orthologs of the largest subunit of RNA polymerase IV,leads to a high-tillering phenotype,in addition to dwarfism and smaller panicles.OsNRPD1a and OsNRPD1b are required for the production of 24-nt small interfering RNAs that direct DNA methylation at transposable elements(TEs)including miniature inverted-repeat TEs(MITEs).Interestingly,many genes are regulated either positively or negatively by TE methylation.Among them,OsMIR156d and OsMIR156j,which promote rice tillering,are repressed by CHH methylation at two MITEs in the promoters.By contrast,D14,which suppresses rice tillering,is activated by CHH methylation at an MITE in its downstream.Our findings reveal regulation of rice tillering by RNA-directed DNA methylation at MITEs and provide potential targets for agronomic trait enhancement through epigenome editing.Le Xu Kun Yuan Meng Yuan Xiangbing Meng Min Chen Jianguo Wu Jiayang Li Yijun Qi 2020Molecular Plant2020,13,6:9
5Mitigation of stimulated Raman scattering in kilowatt-level diode-pumped fiber amplifiers with chirped and tilted fiber Bragg gratings显示文摘The average power of diode-pumped fiber lasers has been developed deeply into the kW regime in the past years.However, stimulated Raman scattering(SRS) is still a major factor limiting the further power scaling. Here, we have demonstrated the mitigation of SRS in kilowatt-level diode-pumped fiber amplifiers using a chirped and tilted fiber Bragg grating(CTFBG) for the first time. The CTFBG is designed and inscribed in large-mode-area(LMA) fibers, matching with the operating wavelength of the fiber amplifier. With the CTFBG inserted between the seed laser and the amplifier stage, an SRS suppression ratio of ~10 dB is achieved in spectrum at the maximum output laser power of 2.35 kW,and there is no reduction in laser slope efficiency and degradation in beam quality. 7Protective Effects of Combination of Radix Astragali and Radix Salviae Miltiorrhizae on Kidney of Spontaneously Hypertensive Rats and Renal Intrinsic Cells显示文摘Objective: To evaluate the effects of combination of Radix Astragali(RA) and Radix Salviae Miltiorrhizae(RS) on kidney of spontaneously hypertensive rats(SHRs) and renal intrinsic cells. Methods: SHRs were intragastrically administrated with RA(5.09 g/kg) and RS(2.55 g/kg) either alone or with combination for 4 weeks;valsartan(13.35 mg/kg) was used as a positive control. Blood pressure and renal ultrasonography were monitored periodically. The biomarkers [microalbumin(m ALB), cystatin C, angiotensin Ⅱ(Ang Ⅱ), interleukin-1 beta(IL-1β), and β2-microglobulin(β2-Mg), etc.] in serum and urine were measured by enzyme-linked immunosorbent assay(ELISA). The protein expressions [phosphorylated adenosine 5’-monophosphate-activated protein kinase-α1(p-AMPKα1), sestrin-β, calcium/calmodulin-dependent protein kinase kinase-β(Ca MKK-β), phosphoinositide 3-kinases(PI3 K), serine-threonine protein kinase 1(AKT1), and vascular endothelial growth factor receptor 2(VEGFR2)] in renal cortex were determined by Western blot. In vitro, the hypertensive cellular model was established by applying 2×10^-6 mol/L Ang Ⅱ. The primary human podocytes, human glomerular endothelial cells(HRGECs), and human proximal tubular epithelial cells(HK-2 s) were pre-incubated with sulfotanshinone sodium(Tan, 10 μg/m L) and/or calycosin-7-O-β-D-glucoside(Cal, 5 μg/m L). The cellular viability and apoptosis were assayed by 3-(4, 5-Dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide(MTT) and Annexin V/PI staining, respectively. The level of endothelial nitric oxide synthase(eNOS) in culture supernatant was determined by ELISA. Results: RA+RS significantly decreased the diastolic blood pressure, renal vascular resistance index, and parenchymal thickness, increased 24 h urinary volume as well as lowered the levels of urine m ALB and serum cystatin C, IL-1β and β2-Mg of SHRs(P<0.05 vs. SHRs). The decreased protein levels of p-AMPKα1, sestrinβ and Ca MKK-β and the increased protein levels of PI3 K, AKT1 and VEGFR2 in renal cortex of SHRs were normalized after RA+RS treatment(P<0.05). In vitro, Tan and Cal attenuated the Ang Ⅱ-induced abnormal proliferation and increased the apoptosis of HRGECs and HK-2 s and improved the level of e NOS in culture supernatant. Whereas, neither of them showed powerful effect on podocyte. 9First genetic assessment of brackish water polychaete Tylorrhynchus heterochaetus: mitochondrial CO/sequences reveal strong genetic differentiation and population expansion in samples collected from southeast China and north Vietnam显示文摘Tylorrhynchus heterochaetus is a widespread benthic polychaete worm found in coastal brackish waters of the west Pacific.It has high ecological and economic value as a biomarker of water quality and as a high-quality feed in aquaculture and fisheries and is considered a delicacy in some areas of Asia.However,it has experienced a marked reduction in recent years due to overexploitation as well as changes in the environment and climate.Here,to comprehensively understand its genetic background and thus provide insights for better conservation and utilization of this species,we assessed the genetic variability and demographic history of T.heterochaetus individuals sampled from eight locations along the coasts of southeast China and north Vietnam based on mitochondrial cytochrome c oxidase I(COI)sequences.We observed high haplotype diversity(Hd),with an average of 0.926,but relatively low nucleotide diversity(π),with a mean of 0.032 across all samples.A total of 94 polymorphic sites and 85 haplotypes were identified among 320 individuals.The pairwise genetic distances among haplotypes ranged from 0.001 to 0.067,with the high intraspecific divergence possibly reflecting geographic isolation and gene pool fragmentation.Significant genetic structures were revealed among the studied locations;specifically,the eight locations could be treated as six genetically different populations based on pairwiseΦST results(0.026–0.951,P<0.01).A significant pattern of isolation-by-distance was detected between the genetic and geographic distances(r=0.873,P=0.001).Three geographic lineages were defined based on phylogenetic tree and network analyses of COI haplotypes.AMOVA results indicated that genetic variations mainly occurred among the three lineages(89.96%).Tests of neutrality and mismatch distribution suggested that T.heterochaetus underwent recent population expansion.These results provide the first report on the genetic status of T.heterochaetus and will be valuable for the management of genetic resources and better understanding of the ecology and evolution in this species.Xing-Han Chen Sen Yang Wei Yang Yuan-Yuan Si Rui-Wen Xu Bin Fan Le Wang Zi-Ning Meng 2020Zoological Research2020,41,1:3

11Bispecific antibody simultaneously targeting PD1 and HER2 inhibits tumor growth via direct tumor cell killing in combination with PD1/PDL1 blockade and HER2 inhibition显示文摘Immune checkpoint blockade has shown significant clinical benefit in multiple cancer indications,but many patients are either refractory or become resistant to the treatment over time.HER2/neu oncogene overexpressed in invasive breast cancer patients associates with more aggressive diseases and poor prognosis.Anti-HER2 mAbs,such as trastuzumab,are currently the standard of care for HER2-overexpressing cancers,but the response rates are below 30%and patients generally suffer relapse within a year.In this study we developed a bispecific antibody(BsAb)simultaneously targeting both PD1 and HER2 in an attempt to combine HER2-targeted therapy with immune checkpoint blockade for treating HER2-positive solid tumors.The BsAb was constructed by fusing scFvs(anti-PD1)with the effector-functional Fc of an IgG(trastuzumab)via a flexible peptide linker.We showed that the BsAb bound to human HER2 and PD1 with high affinities(EC50 values were 0.2 and 0.14 nM,respectively),and exhibited potent antitumor activities in vitro and in vivo.Furthermore,we demonstrated that the BsAb exhibited both HER2 and PD1 blockade activities and was effective in killing HER2-positive tumor cells via antibody-dependent cellular cytotoxicity.In addition,the BsAb could crosslink HER2-positive tumor cells with T cells to form PD1 immunological synapses that directed tumor cell killing without the need of antigen presentation.Thus,the BsAb is a new promising approach for treating late-stage metastatic HER2-positive cancers.Chang-ling Gu Hai-xia Zhu Lan Deng Xiao-qing Meng Kai Li Wei Xu Le Zhao Yue-qin Liu Zhen-ping Zhu Hao-min Huang 2022Acta Pharmacologica Sinica2022,43,3:3
12Thin-layered MoS_2/polyaniline nanocomposite for highly sensitive electrochemical detection of chloramphenicol显示文摘In this study, we synthesized molybdenum disulfide/polyaniline(MoS_2/PANI) nanocomposite via in situ polymerization of aniline in the presence of thin-layered MoS_2. The as-prepared MoS_2/PANI nanocomposite obtained an improved electrochemical performance due to the physisorption interaction between aromatic aniline and the basal plane of MoS_2. Furthermore, we constructed a new kind of electrochemical sensor based on MoS_2/PANI nanocomposite for the detection of chloramphenicol, which showed an excellent performance. 13Target serum concentration of vancomycin may be reached earlier with a loading dose显示文摘Background:Vancomycin treatment failure against vancomycin-susceptible gram-positive cocci is not rare in the intensive care unit(ICU).One of the reasons for this is the substandard drug trough concentration.We aimed to examine the hypothesis that the target serum concentration could be reached earlier with a loading dose of vancomycin.Methods:This retrospective cohort study was conducted at our ICU between June 2018 and June 2020 and involved patients who were suspected of having,or confirmed to have,gram-positive cocci infection and treated with vancomycin.One group of the patients was administered a loading dose of vancomycin(loading group)and compared with the group that did not receive a loading dose(control group).The baseline characteristics,vancomycin serum concentrations,and clinical outcomes were collected and analyzed.Results:Fifty-five patients were finally included,of which 29 received a loading dose of vancomycin.The serum concentration of vancomycin before the second dose was significantly higher for the loading group than for the control group(10.3±6.1 mg/Lvs.5.7±4.4 mg/L,P=0.002).The results for both groups were similar before the fifth dose(12.4±7.3 mg/Lvs.10.3±6.3 mg/L in the loading and the control groups,respectively;P=0.251).The 28-day mortality was lower for the loading group than for the control group(6.7%vs.34.6%in the loading and control groups,respectively;P=0.026).No significant differences were observed in serum creatinine(Cr)concentrations of the two groups.Conclusion:With the loading dose of vancomycin,the target serum concentration of vancomycin may be reached earlier without increasing the risk of acute kidney injury.Yanxia Huang Le He Yunxin Deng Renjing Zhang Mei Meng Jiao Liu Dechang Chen 2022Chinese Medical Journal2022,,3:2
14Machine learning-based real-time visible fatigue crack growth detection显示文摘Many large-scale and complex structural components are applied in the aeronautics and automobile industries.However,the repeated alternating or cyclic loads in service tend to cause unexpected fatigue fractures.Therefore,developing real-time and visible monitoring methods for fatigue crack initiation and propagation is critically important for structural safety.This paper proposes a machine learning-based fatigue crack growth detection method that combines computer vision and machine learning.In our model,computer vision is used for data creation,and the machine learning model is used for crack detection.Then computer vision is used for marking and analyzing the crack growth path and length.We apply seven models for the crack classification and find that the decision tree is the best model in this research.The experimental results prove the effectiveness of our method,and the crack length measurement accuracy achieved is 0.6 mm.Furthermore,the slight machine learning models help us realize real-time and visible fatigue crack detection.Le Zhang Zhichen Wang Lei Wang Zhe Zhang Xu Chen Lin Meng 2021Digital Communications and Networks2021,7,4:2
15Synthesis of 2,3-dihydroquinazolin-4(1H)-ones catalyzed by a-chymotrypsin显示文摘We discovered that a-chymotrypsin has a promiscuous ability to catalyze the cyclocondensation of aromatic and aliphatic aldehydes with 2-aminobenzamides to afford the corresponding 2,3-dihydroquinazolin-4(1H)-ones successfully in high yields(90%–98%) under alcohol solvent. The catalytic activity of a-chymotrypsin was evaluated through investigating the temperature, the enzyme loading and the ratio of substrates in the enzyme-catalyzed reactions. 17China’s Urbanization Strategy and Policy During the 14th Five-Year Plan Period显示文摘During the 13th Five-Year Plan period,China’s urbanization construction has obtained remarkable achievements.Its urbanization continues to advance rapidly.The gap between different regions is constantly narrowed.City clusters have become the strategic core areas to promote urbanization and economic development.Currently,the key issue in China’s urbanization is not about the level or speed,but the quality.Tough challenges faced by China in its urbanization include:(1)granting permanent urban residency to people originally from rural areas(citizenization)is severely lagging-behind;(2)the key links in integrated urban–rural development system and mechanism are blocked and(3)the resources and environment costs in advancing urbanization are excessively high,etc.During the 14th Five-Year Plan period,China’s urbanization will continue to advance rapidly,but the speed will further slow down.By 2025,China’s urbanization rate is expected to reach about 65.5%,among which the eastern,central,western and northeastern regions will reach 73.0%,63.1%,61.2%and 66.7%,respectively.To this end,China’s urbanization should be people-centered and led by high-quality integrated urban and rural development;thus,the urbanization quality can be improved in an all-round way.It should be a path of highquality urbanization with Chinese characteristics featuring moderate and reasonable advancing speed,effectively improved quality in citizenization,balanced and orderly urbanization pattern,deeply integrated urban and rural development,and green,healthy and sustainable development.In terms of specific measures,the government should strengthen the leading,demonstrating and radiating role of central cities and promote the formation of four-tier central city system,namely,the global central cities,national central cities,regional central cities and local central cities;plan and build 34 high-end metropolitan areas at the national level,making them the core areas of advancing new-type urbanization in the new era;accelerate the process of establishing cities,encourage the transformation of large towns into cities,and strictly control the transformation of counties into urban districts;implement differentiated policies for expanding,stable and shrinking cities and towns,respectively;connect the channels between homesteads exiting the market and collective construction land for commercial use entering the market.WEI Houkai LI Le NIAN Meng 2021Chinese Journal of Urban and Environmental Studies2021,9,1:1
