2Host metabolism dysregulation and cell tropism identification in human airway and alveolar organoids upon SARS-CoV-2 infection显示文摘The coronavirus disease 2019(COVID-19)pandemic is caused by infection with the severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),which is spread primary via respiratory droplets and infects the lungs.Currently widely used cell lines and animals are unable to accurately mimic human physiological conditions because of the abnormal status of cell lines(transformed or cancer cells)and species differences between animals and humans.Organoids are stem cell-derived selforganized three-dimensional culture in vitro and model the physiological conditions of natural organs.Here we showed that SARS-CoV-2 infected and extensively replicated in human embryonic stem cells(hESCs)-derived lung organoids,including airway and alveolar organoids which covered the complete infection and spread route for SARS-CoV-2 within lungs.The infected ceils were ciliated,club,and alveolar type 2(AT2)cells,which were sequentially located from the proximal to the distal airway and terminal alveoli,respectively.Additionally,RNA-seq revealed early cell response to virus infection including an unexpected downregulation of the metabolic processes,especially lipid metabolism,in addition to the well-known upregulation of immune response.Further,Remdesivir and a human neutralizing antibody potently inhibited SARS-CoV-2 replication in lung organoids.Therefore,human lung organoids can serve as a pathophysiological model to investigate the underlying mechanism of SARS-CoV-2 infection and to discover and test therapeutic drugs for COVID-19.Rongjuan Pei Jianqi Feng Yecheng Zhang Hao Sun Lian Li Xuejie Yang Jiangping He Shuqi Xiao Jin Xiong Ying Lin Kun Wen Hongwei Zhou Jiekai Chen Zhili Rong Xinwen Chen 2021Protein & Cell2021,12,9:7

4Accelerated evolution of an Lhx2 enhancer shapes mammalian social hierarchies显示文摘Social hierarchies emerged during evolution,and social rank influences behavior and health of individuals.However,the evolutionary mechanisms of social hierarchy are still unknown in amniotes.Here we developed a new method and performed a genome-wide screening for identifying regions with accelerated evolution in the ancestral lineage of placental mammals,where mammalian social hierarchies might have initially evolved.Then functional analyses were conducted for the most accelerated region designated as placental-accelerated sequence 1(PAS1,P=3.15 × 10^-18).Multiple pieces of evidence show that PAS1 is an enhancer of the transcription factor gene Lhx2 involved in brain development.PAS1s isolated from various amniotes showed different c/s-regulatory activity in vitro,and affected the expression of Lhx2 differently in the nervous system of mouse embryos.PAS1 knock-out mice lack social stratification.PAS1 knock-in mouse models demonstrate that PAS1s determine the social dominance and subordinate of adult mice,and that social ranks could even be turned over by mutated PAS1.All homozygous mutant mice had normal huddled sleeping behavior,motor coordination and strength.Therefore,PAS1-Lhx2 modulates social hierarchies and is essential for establishing social stratification in amniotes,and positive Darwinian selection on PAS1 plays pivotal roles in the occurrence of mammalian social hierarchies.Yuting Wang Guangyi Dai Zhili Gu Guopeng Liu Ke Tang Yi-Hsuan Pan Yujie Chen Xin Lin Nan Wu Haoshan Chen Su Feng Shou Qiu Hongduo Sun Qian Li Chuan Xu Yanan Mao Yong Edward Zhang Philipp Khaitovich Yan-Ling Wang Qunxiu Liu Jing-Dong Jackie Han Zhen Shao Gang Wei Chun Xu Naihe Jing Haipeng Li 2020Cell Research2020,30,5:4
5Joining Performance and Microstructure of the 2024/7075 Aluminium Alloys Welded Joints by Vaporizing Foil Actuator Welding显示文摘Vaporizing foil actuator welding(VFAW) was used for joining 2024-T3 and 7075-T6 aluminum alloy sheets, and the resulting joint microstructure was analyzed. 2024/7075 aluminum alloy pairs with suitable processing parameters can be prepared by using VFAW. Dynamic preform addresses the poor formability problem of target material and advantage of VFAW on dissimilar materials in some conditions. But with standoff sheet inserting in the flyer and target, 2024/7075 welded pairs gets the better weld strength, compared with flyer preformed method. The microstructure of the circular weld area of the welded joint showed a wave interface, in which a thin melt layer formed at the center and edge parts. The crystal grains near the bonding interface were remarkably elongated and refined. 7Natural variations in grain length 10(GL10)regulate rice grain size显示文摘Grain size is an important determinant of grain weight and yield in rice.Although several genes related to grain size have been identified,natural variations in these genes that affect grain size are poorly characterized.Here,we describe the grain length QTL GL10,encoding MADS56,which positively regulates grain length and grain weight.A natural allelic variation of NIL-gl10,containing an~1.0-kb deletion in the first exon that abolishes its transcription,results in shorter grain length,lower grain weight and delayed flowering in gl10 plants.The knockout of GL10 in the HJX74 background leads to grain phenotypes similar to that of NIL-gl10,while overexpression of GL10 results in increased grain length and weight and earlier heading date.GL10 regulates grain length by promoting greater longitudinal cell growth in the grain glume.Additionally,GL10 participates in the regulation of gibberellic acid(GA)signaling pathway genes in young panicle tissues.Analysis of GL10 haplotypes shows obvious divergence between the japonica and indica lineages.Our findings reveal an allelic variation of GL10 that may explain differences in grain length among modern cultivars and could be used to breed rice varieties with optimized grain shape.Penglin Zhan Shuaipeng Ma Zhili Xiao Fangping Li Xin Wei Shaojun Lin Xiaoling Wang Zhe Ji Yu Fu Jiahao Pan Mi Zhou Yue Liu Zengyuan Chang Lu Li Suhong Bu Zupei Liu Haitao Zhu Guifu Liu Guiquan Zhang Shaokui Wang 2022Journal of Genetics and Genomics2022,49,5:3
8Noninvasive Ultrasound Stimulation of Ventral Tegmental Area Induces Reanimation from General Anaesthesia in Mice显示文摘Evidence in animals suggests that deep brain stimulation or optogenetics can be used for recovery from disorders of consciousness(DOC).However,these treatments require invasive procedures.This report presents a noninvasive strategy to stimulate central nervous system neurons selectively for recovery from DOC in mice.Through the delivery of ultrasound energy to the ventral tegmental area,mice were aroused from an unconscious,anaesthetized state in this study,and this process was controlled by adjusting the ultrasound parameters.The mice in the sham group under isoflurane-induced,continuous,steady-state general anaesthesia did not regain their righting reflex.On insonation,the emergence time from inhaled isoflurane anaesthesia decreased(sham.13.63±0.53 min,ultrasound.1.5±0.19 min,p<0.001).Further,the induction time(sham.12.0±0.6 min,ultrasound.17.88±0.64 min,p<0.001)and the concentration for 50%of the maximal effect(EC50)of isoflurane(sham.0.6%,ultrasound.0.7%)increased.In addition,ultrasound stimulation reduced the recovery time in mice with traumatic brain injury(sham.30.38±1.9 min,ultrasound.7.38±1.02 min,p<0.01).This noninvasive strategy could be used on demand to promote emergence from DOC and may be a potential treatment for such disorders.Tianyuan Bian Wen Meng Meihong Qiu Zhigang Zhong Zhengrong Lin Junjie Zou Yibo Wang Xiaowei Huang Lisheng Xu Tifei Yuan Zhili Huang Lili Niu Long Meng Hairong Zheng 2021Research2021,,1:2

15Distribution,Seismic Characteristics and Controlling Factors of Carbonate Platforms on the Northwestern Margin of the South China Sea显示文摘Newly-acquired seismic data reveal widespread carbonate deposits covering a large part of the northwestern South China Sea margin.Three carbonate platforms are identified to have developed on the topographic highs inherited from tectonic deformation and volcanic accretion.Across the carbonate platforms,the Miocene strata are characterized by high-amplitude seismic reflections and distinct platform architecture that overlaps older strata.The Guangle and Xisha carbonate platforms grew on faulted blocks due to South China Sea continental rifting,while the Zhongjian carbonate platform developed on a structural high induced by early Miocene volcanism.During the late Miocene,partial drowning resulted in the inhibition of platform growth,eventual platform drowning and termination of most carbonate deposition.The drowning of the Guangle and Zhongjian carbonate platforms is shown by the supply of siliciclastic sediments during the late Miocene and seems to be closely linked to late Neogene volcanic activity,whilst the drowning of the Xisha carbonate platform is primarily related to relative eustatic changes.Our results imply that tectonic activity,volcanism and eustasy are the dominant controls on the evolution of carbonate platforms on the northwestern margin of the South China Sea.LI Lin WANG Bin YANG Taotao YANG Zhili LI Li LU Yintao WANG Xuefeng WU Jingwu SUN Guozhong 2021Acta Geologica Sinica(English Edition)2021,95,1:1
16Ultra-compact tunable graphene-based plasmonic multimode interference power splitter in mid infrared frequencies显示文摘In this paper, we propose graphene-based plasmonic multimode interference power splitters with ultra-compact size working in mid infrared range. Further, the arbitrary-ratio 1×2 power splitter with a size of 140 nm×232 nm, where the splitting ratio can be tuned continuously from 1:1 to 100:0, is numerically demonstrated. Meanwhile, the graphene-based arbitrary-ratio 1×2 power splitters with different frequencies and chemical potentials are also investigated. 19Graphene binding on black phosphorus enables high on/off ratios and mobility显示文摘Graphene is one of the most promising candidates for integrated circuits due to its robustness against short-channel effects,inherent high carrier mobility and desired gapless nature for Ohmic contact,but it is difficult to achieve satisfactory on/off ratios even at the expense of its carrier mobility,limiting its device applications.Here,we present a strategy to realize high back-gate switching ratios in a graphene monolayer with well-maintained high mobility by forming a vertical heterostructure with a black phosphorus multi-layer.By local current annealing,strain is introduced within an established area of the graphene,which forms a reflective interface with the rest of the strain-free area and thus generates a robust off-state via local current depletion.Applying a positive back-gate voltage to the heterostructure can keep the black phosphorus insulating,while a negative back-gate voltage changes the black phosphorus to be conductive because of hole accumulation.Then,a parallel channel is activated within the strain-free graphene area by edge-contacted electrodes,thereby largely inheriting the intrinsic carrier mobility of graphene in the on-state.As a result,the device can provide an on/off voltage ratio of>10^(3)as well as a mobility of∼8000 cm^(2)V^(−1)s^(−1)at room temperature,meeting the low-power criterion suggested by the International Roadmap for Devices and Systems.Fanrong Lin Zhonghan Cao Feiping Xiao Jiawei Liu Jiabin Qiao Minmin Xue Zhili Hu Ying Liu Huan Lu Zhuhua Zhang Jens Martin Qingjun Tong Wanlin Guo Yanpeng Liu 2024National Science Review2024,11,2:0
20Facet-induced coordination competition for highly ordered CsPbBr_(3) nanoplatelets with strong polarized emission显示文摘Controllable anisotropic growth of perovskite nanocrystals(NCs)is challenging since it is difficult to separate the nucleation and growth processes.Here,a two-step nucleation strategy is proposed to control the binding interaction between surface ligands and NCs,resulting in facet-induced coordination competition.Oleic acid as surface activated ligand leads to the formation of defective lead bromine octahedron,and the binding interaction between 4-dodecylbenzenesulfonic acid and lead atoms promotes the formation of two kinds of binding interactions.Based on this strategy,the anisotropic growth of CsPbBr_(3) nanoplatelet(NPLs)with adjusted length from 11.4 to 24 nm,and the evolution of NPLs from stacked to tongue-shaped have been realized.Elemental line scan reveals the sulfur atoms mainly distribute at the edge of NPLs.Furthermore,binding energy calculation and experimental results illustrate the coordination competition of different binding interaction on specific facets induces the anisotropic growth of NPLs.Importantly,strong emission anisotropy of highly ordered NPLs with polarization ratio up to 0.58 is illustrated.This work not only deepens our understanding of the controllable synthesis of perovskite NCs,but also provides a reference for the regulation of light emitting diode and soler cells.Dandan Yang Xiaoming Li Yuelei Li Bo Cai Lin Su Shengli Zhang Zhili Juan Cuifang Meng Dongling Geng Jiaxin Chen Feng Xu Haibo Zeng 2022Nano Research2022,15,1:0
