1A life support-based comprehensive treatment regimen dramatically lowers the in-hospital mortality of patients with fulminant myocarditis: a multiple center study显示文摘Fulminant myocarditis(FM) has unacceptable high mortality. This study aimed to evaluate the therapeutic efficacy of a life support-based comprehensive treatment regimen(LSBCTR), a completely novel treatment regimen, for FM. A total of 169 FM patients recruited from January 2008 to December 2018 were divided into two groups: patients receiving LSBCTR(81 cases),which includes(i) mechanical life support(positive pressure respiration, intra-aortic balloon pump with or without extracorporeal membrane oxygenation),(ii) immunomodulation therapy using sufficient doses of glucocorticoids and immunoglobulins, and(iii) application of neuraminidase inhibitors, and those receiving conventional treatment(88 cases). The endpoints were in-hospital death and heart-transplantation. Of all the population, 44 patients(26.0%) died in hospitals. Inhospital mortality was 3.7%(3/81) for LSBCTR group and 46.6%(41/88) for traditional treatment(P<0.001). Early application of LSBCTR, mechanical life support, neuraminidase inhibitors, and immunomodulation therapy significantly contributed to reduction in in-hospital mortality. This study describes a novel treatment regimen for FM patients that dramatically reduces inhospital mortality. 2Latitudinal pattern and the driving factors of leaf functional traits in 185 shrub species across eastern China显示文摘Aims To explore the pattern of the leaf functional traits of shrub species along a latitudinal gradient in eastern China and determine the driv-ing factors of leaf trait variation at a large scale.Methods We investigated the leaf thickness(LT),leaf area(LA),specific leaf area(SLA)and leaf dry mass content(LDMC)of 185 shrub species from 13 sites across eastern China.The trends of these four-leaf traits were ana-lyzed with respect to latitude,and the differences between different life forms(e.g.,evergreen and deciduous)and habitats(e.g.,understory and typical)were compared.We quantified the effects of the plant life forms and environmental factors on the leaf traits via mixed-model analyses.Important Findings The LT and LA decreased,whilst and the LDMC increased,as the latitude increased,and significant differences in these traits were observed between the different plant life forms.The LT and LA were smaller,whereas the SLA and LDMC were larger in decidu-ous shrubs than in evergreen shrubs.Among the different habitats,the LA and SLA were larger,while the LDMC was smaller in under-story shrubs than in typical shrub species.These results indicate that typical shrub species are better adapted to drier environments,as indicated by a reduced LT and increased LDMC.Furthermore,general linear models showed that variations in the four-leaf traits with respect to latitude were mainly caused by a shift in plant life forms.Yongkai Luo Huifeng Hu Mengying Zhao He Li Shangshi Liu Jingyun Fang 2019Journal of Plant Ecology2019,12,1:7

4Meso-Cenozoic Evolution of Earth Surface System under the East Asian Tectonic Superconvergence显示文摘The East Asian geological setting has a long duration related to the superconvergence of the Paleo-Asian, Tethyan and Paleo-Pacific tectonic domains. The Triassic Indosinian Movement contributed to an unified passive continental margin in East Asia. The later ophiolites and I-type granites associated with subduction of the Paleo-Pacific Plate in the Late Triassic, suggest a transition from passive to active continental margins. With the presence of the ongoing westward migration of the Paleo-Pacific Subduction Zone, the sinistral transpressional stress field could play an important role in the intraplate deformation in East Asia during the Late Triassic to Middle Jurassic, being characterized by the transition from the E-W-trending structural system controlled by the Tethys and Paleo-Asian oceans to the NE-trending structural system caused by the Paleo-Pacific Ocean subduction. The continuously westward migration of the subduction zones resulted in the transpressional stress field in East Asia marked by the emergence of the Eastern North China Plateau and the formation of the Andean-type active continental margin from late Late Jurassic to Early Cretaceous(160-135 Ma), accompanied by the development of a small amount of adakites. In the Late Cretaceous(135-90 Ma), due to the eastward retreat of the Paleo-Pacific Subduction Zone, the regional stress field was replaced from sinistral transpression to transtension. Since a large amount of late-stage adakites and metamorphic core complexes developed, the Andean-type active continental margin was destroyed and the Eastern North China Plateau started to collapse. In the Late Cretaceous, the extension in East Asia gradually decreased the eastward retreat of the Paleo-Pacific subduction zones. Futhermore, a significant topographic inversion had taken place during the Cenozoic that resulted from a rapid uplift of the Tibet Plateau resulting from the India-Eurasian collision and the formation of the Bohai Bay Basin and other basins in the East Asian continental margin. The inversion caused a remarkable eastward migration of deformation, basin formation and magmatism. Meanwhile, the basins that mainly developed in the Paleogene resulted in a three-step topography which typically appears to drop eastward in altitude. In the Neogene, the basins underwent a rapid subsidence in some depressions after basin-controlled faulting, as well as the intracontinental extensional events in East Asia, and are likely to be a contribution to the uplift of the Tibetan Plateau.DMITRIENKO Liudmila V. 627-Hydroxycholesterol-induced EndMT acts via STAT3 signaling to promote breast cancer cell migration by altering the tumor microenvironment显示文摘Objective:The endothelial to mesenchymal transition(EndMT)plays a major role in cancer metastasis by regulating the complexity of the tumor microenvironment(TME).Here,we investigated whether 27-hydroxycholesterol(27 HC)induces EndMT in endothelial cells(ECs).Methods:EndMT markers in the human microvascular endothelial cell-1(HMEC-1)cell line and human umbilical vein endothelial cells(HUVECs)stimulated with 27 HC were evaluated with Western blot.Epithelial to mesenchymal transition(EMT)markers in breast cancer(BC)cells cultured in conditioned medium were investigated with quantitative real time polymerase chain reaction(qRT-PCR).The MMP-2 and MMP-9 mRNA expression and activity were detected with qRT-PCR and gelatin zymography assays,respectively.The effect of activated STAT3 on 27 HC-induced EndMT was validated by Western blot,immunofluorescence staining,and cell transfection assays.The migration ability of BC cells was evaluated with Transwell assays.Results:We found that 27 HC induced EndMT in HMEC-1 and HUVECs,and 27 HC-induced EndMT facilitated EMT and BC cell migration.The 27 HC-induced EMT of BC cells also promoted EndMT and HUVEC migration.Investigation of the underlying molecular mechanisms revealed that STAT3 knockdown repressed EndMT in HUVECs as well as migration in BC cells induced with 27 HC.In addition,C646 and resveratrol,inhibitors of STAT3 acetylation,repressed the expression of Ac-STAT3,p-STAT3,and EndMT markers in HUVECs exposed to 27 HC;these HUVECs in turn attenuated the migration ability of BC cells in 27 HC-induced EndMT.Conclusions:Cross-talk between 27 HC-induced EndMT and EMT was observed in the TME.Moreover,activation of STAT3 signaling was found to be involved in 27 HC-induced EndMT.Kailin Jiao Jing Zhen Maoxuan Wu Mengying Teng Keke Yang Qian Zhou Chunyan Hu Ming Zhou Yuan Li Zhong Li 2020Cancer Biology & Medicine2020,17,1:3

8Effect of Land Use on Soil Properties in Debris Flow Bottomland:A Case Study at Xiaojiang Basin, Yunnan显示文摘In order to understand the effect of different land use on soil physics and nutrients properties of the debris flow bottomland, a case study at Daqing gully (in Xiaojiang Basin, Yunnan) was conducted in 2004. Soil samples were taken at depth of 0-10,10-20,20-40,40-60,60-80 cm under three land use patterns crop bottomland(CL), forest bottomland(FL), and barren bottomland(BL). The results showed that the developing bottomland to CL promoted soil toaccumulate total phosphorus (TP) and available phosphorus (AP), pH value transferred from neutral to alkalescency, and organic matter decreased significantly. Furthermore, the contents of total nitrogen (TN) and available nitrogen (AN) in CL were lower than that of FL and BL because the growth of crops consumed more nutrients in soil. 9Overexpression of Sirt6 ameliorates sleep deprivation induced-cognitive impairment by modulating glutamatergic neuron function显示文摘Sleep benefits the restoration of energy metabolism and thereby suppo rts neuronal plasticity and cognitive behaviors.Sirt6 is a NAD+-dependent protein deacetylase that has been recognized as an essential regulator of energy metabolism because it modulates various transcriptional regulators and metabolic enzymes.The aim of this study was to investigate the influence of Sirt6 on cerebral function after chronic sleep deprivation(CSD).We assigned C57BL/6J mice to control or two CSD groups and subjected them to AAV2/9-CMV-EGFP or AAV2/9-CMV-Sirt6-EGFP infection in the prelimbic cortex(PrL).We then assessed cerebral functional connectivity(FC) using resting-state functional MRI,neuron/astrocyte metabolism using a metabolic kinetics analysis;dendritic spine densities using sparse-labeling;and miniature excitato ry postsynaptic currents(mEPSCs) and action potential(AP) firing rates using whole-cell patchclamp recordings.In addition,we evaluated cognition via a comprehensive set of behavioral tests.Compared with controls,Sirt6 was significantly decreased(P<0.05) in the PrL after CSD,accompanied by cognitive deficits and decreased FC between the PrL and accumbens nucleus,piriform cortex,motor co rtex,somatosensory co rtex,olfactory tubercle,insular cortex,and cerebellum.Sirt6 ove rexpression reve rsed CSD-induced cognitive impairment and reduced FC.Our analysis of metabolic kinetics using [1-13C] glucose and [2-13C] acetate showed that CSD reduced neuronal Glu4and GABA2synthesis,which could be fully restored via forced Sirt6 expression.Furthermore,Sirt6 ove rexpression reversed CSD-induced decreases in AP firing rates as well as the frequency and amplitude of mEPSCs in PrL pyramidal neurons.These data indicate that Sirt6 can improve cognitive impairment after CSD by regulating the PrL-associated FC network,neuronal glucose metabolism,and glutamatergic neurotransmission.Thus,Sirt6 activation may have potential as a novel strategy for treating sleep disorder-related diseases.Jinpiao Zhu Chang Chen Zhen Li Xiaodong Liu Jingang He Ziyue Zhao Mengying He Binbin Nie Zili Liu Yingying Chen Kuanpin Su Xiang Li Juxiang Chen Hongbing Xiang Fuqiang Xu Kangguang Lin Zongze Zhang Jie Wang 2023Neural Regeneration Research2023,18,11:1

13Adoptive transfer of metabolically reprogrammed macrophages for atherosclerosis treatment in diabetic ApoE^(-/-) mice显示文摘Atherosclerosis is characterized by inflammation in the arterial wall,which is known to be exacerbated by diabetes.Therapeutic repression of inflammation is a promising strategy for treating atherosclerosis.In this study,we showed that diabetes aggravated atherosclerosis in apolipoproteinE knockout(ApoE^(-/-))mice,in which increased expression of long-chain acyl-CoA synthetase 1(Acsl1)in macrophages played an important role.Knockdown of Acsl1 in macrophages(Mφ^(shAcsl1))reprogrammed macrophages to an anti-inflammatory phenotype,especially under hyperglycemic conditions.Injection of Mφ^(shAcsl1) reprogrammed macrophages into streptozotocin(STZ)-induced diabetic ApoE^(-/-) mice(ApoE^(-/-)+STZ)alleviated inflammation locally in the plaque,liver and spleen.Consistent with the reduction in inflammation,plaques became smaller and more stable after the adoptive transfer of reprogrammed macrophages.Taken together,our findings indicate that increased Acsl1 expression in macrophages play a key role in aggravated atherosclerosis of diabetic mice,possibly by promoting inflammation.Adoptive transfer of Acsl1 silenced macrophages may serve as a potential therapeutic strategy for atherosclerosis.Tingting Wang Yan Dong Li Yao Fan Lu Chenxi Wen Zhuo Wan Li Fan Zhelong Li Te Bu Mengying Wei Xuekang Yang Yi Zhang 2022Bioactive Materials2022,7,10:1

17Kinetically regulated one-pot synthesis of cationic gold nanoparticles and their size-dependent antibacterial mechanism显示文摘Cationic gold nanoparticles(cAuNPs)have been regarded as promising candidates for antibacterial applications due to their high surface charge density,favorable biocompatibility,and controllable surface chemistry.Nevertheless,the complicated fabrication process and unclear antibacterial mechanism have greatly hindered the further biomedical application of cAuNPs.Herein,we have developed a simple and controllable strategy for synthesizing cAuNPs with tailored size and antibacterial behavior by kinetically modulating the reaction process.Specifically,a functional ligand,(11-mercaptoundecyl)-N,N,Ntrimethylammonium bromide(MUTAB),was chosen to chemically manipulate the positive surface charge of cAuNPs via a one-step strategy.The size of cAuNPs could be flexibly adjusted from 1.1 to 14.8 nm by simply elevating the stirring speed of the reaction from 0 to 1500 rpm.Further studies revealed that the antibacterial effect of cAuNPs was strongly correlated with the particle size.MUTAB-protected ultrasmall gold nanoclusters(MUTAB-AuNCs)were able to eradicate E.coli at a concentration as low as 1.25μg mL^(-1),while the minimum inhibitory concentration of MUTAB-AuNPs with a large size for E.coli was 5μg mL^(-1).Mechanistic investigation revealed that MUTAB-AuNPs were able to damage the bacterial membrane and stimulate the production of reactive oxygen species more effectively than MUTAB-AuNCs.Conversely,MUTAB-AuNCs were more active in inducing membrane depolarization in contrast to MUTAB-AuNPs,suggesting the unique size-dependent antibacterial manner of cAuNPs.This study presents a new strategy for the controlled preparation of cAuNPs with distinct sizes and antibacterial behavior,laying a valuable foundation for developing efficient cationic NP-based bactericidal agents.Chenqiang Shen Yumeng Xue Yixiao Li Mengying Wei Mengyao Wen Lianbing Zhang Li Shang 2023Journal of Materials Science & Technology2023,,31:1
