1A life support-based comprehensive treatment regimen dramatically lowers the in-hospital mortality of patients with fulminant myocarditis: a multiple center study显示文摘Fulminant myocarditis(FM) has unacceptable high mortality. This study aimed to evaluate the therapeutic efficacy of a life support-based comprehensive treatment regimen(LSBCTR), a completely novel treatment regimen, for FM. A total of 169 FM patients recruited from January 2008 to December 2018 were divided into two groups: patients receiving LSBCTR(81 cases),which includes(i) mechanical life support(positive pressure respiration, intra-aortic balloon pump with or without extracorporeal membrane oxygenation),(ii) immunomodulation therapy using sufficient doses of glucocorticoids and immunoglobulins, and(iii) application of neuraminidase inhibitors, and those receiving conventional treatment(88 cases). The endpoints were in-hospital death and heart-transplantation. Of all the population, 44 patients(26.0%) died in hospitals. Inhospital mortality was 3.7%(3/81) for LSBCTR group and 46.6%(41/88) for traditional treatment(P<0.001). Early application of LSBCTR, mechanical life support, neuraminidase inhibitors, and immunomodulation therapy significantly contributed to reduction in in-hospital mortality. This study describes a novel treatment regimen for FM patients that dramatically reduces inhospital mortality. 2Latitudinal pattern and the driving factors of leaf functional traits in 185 shrub species across eastern China显示文摘Aims To explore the pattern of the leaf functional traits of shrub species along a latitudinal gradient in eastern China and determine the driv-ing factors of leaf trait variation at a large scale.Methods We investigated the leaf thickness(LT),leaf area(LA),specific leaf area(SLA)and leaf dry mass content(LDMC)of 185 shrub species from 13 sites across eastern China.The trends of these four-leaf traits were ana-lyzed with respect to latitude,and the differences between different life forms(e.g.,evergreen and deciduous)and habitats(e.g.,understory and typical)were compared.We quantified the effects of the plant life forms and environmental factors on the leaf traits via mixed-model analyses.Important Findings The LT and LA decreased,whilst and the LDMC increased,as the latitude increased,and significant differences in these traits were observed between the different plant life forms.The LT and LA were smaller,whereas the SLA and LDMC were larger in decidu-ous shrubs than in evergreen shrubs.Among the different habitats,the LA and SLA were larger,while the LDMC was smaller in under-story shrubs than in typical shrub species.These results indicate that typical shrub species are better adapted to drier environments,as indicated by a reduced LT and increased LDMC.Furthermore,general linear models showed that variations in the four-leaf traits with respect to latitude were mainly caused by a shift in plant life forms.Yongkai Luo Huifeng Hu Mengying Zhao He Li Shangshi Liu Jingyun Fang 2019Journal of Plant Ecology2019,12,1:7

6Cardiomyocyte-targeted anti-inflammatory nanotherapeutics against myocardial ischemia reperfusion (IR) injury显示文摘Myocardial ischemia reperfusion(IR)injury is closely related to the overwhelming inflammation in the myocardium.Herein,cardiomyocyte-targeted nanotherapeutics were developed for the reactive oxygen species(ROS)-ultrasensitive co-delivery of dexamethasone(Dex)and RAGE small interfering RNA(siRAGE)to attenuate myocardial inflammation.PPTP,a ROSdegradable polycation based on PGE2-modified,PEGylated,ditellurium-crosslinked polyethylenimine(PEI)was developed to surface-decorate the Dex-encapsulated mesoporous silica nanoparticles(MSNs),which simultaneously condensed siRAGE and gated the MSNs to prevent the Dex pre-leakage.Upon intravenous injection to IR-injured rats,the nanotherapeutics could be efficiently transported into the inflamed cardiomyocytes via PGE2-assisted recognition of over-expressed E-series of prostaglandin(EP)receptors on the cell membranes.Intracellularly,the over-produced ROS degraded PPTP into small segments,promoting the release of siRAGE and Dex to mediate effective RAGE silencing(72%)and cooperative antiinflammatory effect.As a consequence,the nanotherapeutics notably suppressed the myocardial fibrosis and apoptosis,ultimately recovering the systolic function.Therefore,the current nanotherapeutics represent an effective example for the codelivery and on-demand release of nucleic acid and chemodrug payloads,and might find promising utilities toward the synergistic management of myocardial inflammation.Min Lan Mengying Hou Jing Yan Qiurong Deng Ziyin Zhao Shixian Lv Juanjuan Dang Mengyuan Yin Yong Ji Lichen Yin 2022Nano Research2022,15,10:2
7Yield and quality properties of silage maize and their influencing factors in China显示文摘Silage maize(Zea mays L.) is one of the most important forages in the world, and its yield and quality properties are critical parameters for livestock production and assessment of forage values. However, relationships between its yield and quality properties and the controlling factors are not well documented. In this study, we collected 5,663 observations from 196 publications across the country to identify the relationships between yield and quality properties of silage maize and to assess the impact of management practices and climatic factors on its yield and quality in China. The average dry matter yield of silage maize was(19.98±6.93) Mg ha^(-1), and the average value of crude protein, ether extract, crude ash, crude fiber, acid detergent fiber, neutral detergent fiber, nitrogen-free extract, and relative feed value was 7.86%±1.71%, 2.53%±1.01%, 5.05%±1.66%,23.97%±6.34%, 27.62%±7.12%, 51.60%±9.85%, 59.68%±7.72%, and 131.17±31.49, respectively. In general, its nutritive value decreased as its yield increased. Increasing planting density could increase the yield but inhibit the nutritive values, while increasing fertilization could benefit the nutritive values. Geographically, the yield increased and the nutritive value decreased from warm(south) to cold(north) regions. The length of growth duration was a major controlling factor for the patterns of these properties. 8SbMYB3 transcription factor promotes root-specific f lavone biosynthesis in Scutellaria baicalensis显示文摘Scutellaria baicalensis Georgi produces abundant root-specific f lavones(RSFs),which provide various benefits to human health.We have elucidated the complete biosynthetic pathways of baicalein and wogonin.However,the transcriptional regulation of f lavone biosynthesis in S.baicalensis remains unclear.We show that the SbMYB3 transcription factor functions as a transcriptional activator involved in the biosynthesis of RSFs in S.baicalensis.Yeast one-hybrid and transcriptional activation assays showed that SbMYB3 binds to the promoter of flavone synthase II-2(SbFNSII-2)and enhances its transcription.In S.baicalensis hairy roots,RNAi of SbMYB3 reduced the accumulation of baicalin and wogonoside,and SbMYB3 knockout decreased the biosynthesis of baicalein,baicalin,wogonin,and wogonoside,whereas SbMYB3 overexpression enhanced the contents of baicalein,baicalin,wogonin,and wogonoside.Transcript profiling by qRT–PCR demonstrated that SbMYB3 activates SbFNSII-2 expression directly,thus leading to more abundant accumulation of RSFs.This study provides a potential target for metabolic engineering of RSFs.Yumin Fang Jie Liu Minmin Zheng Sanming Zhu Tianlin Pei Mengying Cui Lijing Chang Hanwen Xiao Jun Yang Cathie Martin Qing Zhao 2023Horticulture Research2023,10,2:2
9Overexpression of Sirt6 ameliorates sleep deprivation induced-cognitive impairment by modulating glutamatergic neuron function显示文摘Sleep benefits the restoration of energy metabolism and thereby suppo rts neuronal plasticity and cognitive behaviors.Sirt6 is a NAD+-dependent protein deacetylase that has been recognized as an essential regulator of energy metabolism because it modulates various transcriptional regulators and metabolic enzymes.The aim of this study was to investigate the influence of Sirt6 on cerebral function after chronic sleep deprivation(CSD).We assigned C57BL/6J mice to control or two CSD groups and subjected them to AAV2/9-CMV-EGFP or AAV2/9-CMV-Sirt6-EGFP infection in the prelimbic cortex(PrL).We then assessed cerebral functional connectivity(FC) using resting-state functional MRI,neuron/astrocyte metabolism using a metabolic kinetics analysis;dendritic spine densities using sparse-labeling;and miniature excitato ry postsynaptic currents(mEPSCs) and action potential(AP) firing rates using whole-cell patchclamp recordings.In addition,we evaluated cognition via a comprehensive set of behavioral tests.Compared with controls,Sirt6 was significantly decreased(P<0.05) in the PrL after CSD,accompanied by cognitive deficits and decreased FC between the PrL and accumbens nucleus,piriform cortex,motor co rtex,somatosensory co rtex,olfactory tubercle,insular cortex,and cerebellum.Sirt6 ove rexpression reve rsed CSD-induced cognitive impairment and reduced FC.Our analysis of metabolic kinetics using [1-13C] glucose and [2-13C] acetate showed that CSD reduced neuronal Glu4and GABA2synthesis,which could be fully restored via forced Sirt6 expression.Furthermore,Sirt6 ove rexpression reversed CSD-induced decreases in AP firing rates as well as the frequency and amplitude of mEPSCs in PrL pyramidal neurons.These data indicate that Sirt6 can improve cognitive impairment after CSD by regulating the PrL-associated FC network,neuronal glucose metabolism,and glutamatergic neurotransmission.Thus,Sirt6 activation may have potential as a novel strategy for treating sleep disorder-related diseases.Jinpiao Zhu Chang Chen Zhen Li Xiaodong Liu Jingang He Ziyue Zhao Mengying He Binbin Nie Zili Liu Yingying Chen Kuanpin Su Xiang Li Juxiang Chen Hongbing Xiang Fuqiang Xu Kangguang Lin Zongze Zhang Jie Wang 2023Neural Regeneration Research2023,18,11:1

11iRGD-reinforced, photo-transformable nanoclusters toward cooperative enhancement of intratumoral penetration and antitumor efficacy显示文摘Insufficient intratumoral penetration greatly hurdles the anticancer performance of nanomedicine. To realize highly efficient tumor penetration in a precisely and spatiotemporally controlled manner, far-red light-responsive nanoclusters (NCs) capable of size shrinkage and charge conversion were developed and co-administered with iRGD to synergistically improve the intratumoral penetration and the anticancer efficacy. The NCs were constructed using the singlet oxygen-sensitive (SOS) polyethylene glycolpolyurethane-polyethylene glycol (PEG-(1O2)PU-PEG) triblock copolymer to encapsulate the doxorubicin (DOX)-loaded, chlorin e6 (Ce6)-conjugated polyamindoamine (PAMAM) dendrimer (DCD) via the double-emulsion method. Co-administration of iRGD notably increased the permeability of NCs within tumor vasculature and tumor tissues. In addition, upon far-red light irradiation (660 nm) of tumors at low optical density (10 mW/cm2), the generated 1O2 could disintegrate the NCs and release the DCD with positive surface charge and ultra-small size (~ 5 nm), which synergized with iRGD to enable deep intratumoral penetration. Consequently, the local 1O2 at lethal concentrations along with the released DOX efficiently and cooperatively eradicated tumor cells. 14Macrophage-targeting gene silencing orchestrates myocardial microenvironment remodeling toward the anti-inflammatory treatment of ischemia-reperfusion (IR) injury显示文摘Ischemia-reperfusion (IR) injury represents a major cause of myocardial dysfunction after infarction and thrombolytic therapy, and it is closely related to the free radical explosion and overwhelming inflammatory responses. Herein, macrophage-targeting nanocomplexes (NCs) are developed to mediate efficient co-delivery of siRNA against MOF (siMOF) and microRNA-21 (miR21) into myocardial macrophages, cooperatively orches-trating the myocardial microenvironment against IR injury. Bioreducible, branched poly(β-amino ester) (BPAE-SS) is designed to co-condense siMOF and miR21 into NCs in a multivalency-reinforced approach, and they are surface-decorated with carboxylated mannan (Man-COOH) to shield the positive surface charges and enhance the serum stability. The final MBSsm NCs are efficiently internalized by myocardial macrophages after systemic administration, wherein BPAE-SS is degraded into small segments by intracellular glutathione to promote the siMOF/miR21 release, finally provoking efficient gene silencing. Thus, cardiomyocyte protection and macro-phage modulation are realized via the combined effects of ROS scavenging, inflammation inhibition, and autophagy attenuation, which ameliorates the myocardial microenvironment and restores the cardiac function via positive cellular crosstalk. 18Impact of the national hepatitis B immunization program in China:a modeling study显示文摘Background:Elimination of hepatitis B virus(HBV)is a striking challenge for countries with high or moderate disease burden.Therefore,using China as a practical case to share experiences for similar countries may accelerate the achievement of the WHO 2030 target of 90%reduction in HBV-related incidence.We aim to evaluate the impact of national HBV immunization strategies in China;and the feasibility to achieve WHO 2030 targets under diferent scenarios.Methods:We constructed an expanded Susceptible-Exposed-Infectious-Recovered(SEIR)model and decision treeMarkov model to estimate the epidemic of HBV in China,assess the feasibility of 2030 Elimination Goals through the projections and conduct the economic analysis.Least square method was used to calibrate the expanded SEIR model by yearly data of laboratory-confrmed HBV cases from 1990 to 2018.Two models were separately used to evaluate the impact and cost-efectiveness of HBV vaccine by comparing prevalence of chronic HBV infections,qualityadjusted life-years(QALYs),incremental cost efectiveness ratio and beneft–cost ratio(BCR)under various intervention options,providing a basis for exploring new containment strategies.Results:Between 1990 and 2020,the number of chronic HBV infections decreased by 33.9%.The current status quo would lead to 55.73 million infections(3.95%prevalence)in 2030,compared to 90.63 million(6.42%prevalence)of the“Without the NIP”scenario(NIP:National Immunization Program),114.78 million(8.13%prevalence)without any interventions.The prevention of mother to child transmission(PMTCT)strategy showed a net beneft as 12,283.50 dollars per person,with BCR as 12.66,which is higher than that of universal vaccination at 9.49.Compared with no screening and no vaccination,the PMTCT strategy could save 7726.03 dollars for each QALY increase.Conclusions:Our fndings proved the HBV vaccination has demonstrated a substantial positive impact on controlling the epidemic of HBV in terms of efectiveness and economy after about 30 years of implementation of the national hepatitis B immunization program which also provided containment experience for high or medium burden countries.As for China,the next step should focus on exploring strategies to improve diagnosis and treatment coverage to reduce the burden of HBV-related deaths and ultimately eliminate HBV.Zhixi Liu Mengying Li David W.Hutton Abram L.Wagner Ye Yao Wenlong Zhu Lingsheng Cao Shenglan Tang Jinhua Pan Yesheng Wang Qi Zhao Hong Ren Ying Wang Weibing Wang 2022Infectious Diseases of Poverty2022,11,5:0
19Gap-free genome assembly and CYP450 gene family analysis reveal the biosynthesis of anthocyanins in Scutellaria baicalensis显示文摘Scutellaria baicalensis Georgi,a member of the Lamiaceae family,is a widely utilized medicinal plant.The flavones extracted from S.baicalensis contribute to numerous health benefits,including anti-inflammatory,antiviral,and anti-tumor activities.However,the incomplete genome assembly hinders biological studies on S.baicalensis.This study presents the first telomere-to-telomere(T2T)gap-free genome assembly of S.baicalensis through the integration of Pacbio HiFi,Nanopore ultra-long and Hi-C technologies.A total of 384.59 Mb of genome size with a contig N50 of 42.44 Mb was obtained,and all sequences were anchored into nine pseudochromosomes without any gap or mismatch.In addition,we analysed the major cyanidin-and delphinidin-based anthocyanins involved in the determination of blue-purple flower using a widely-targeted metabolome approach.Based on the genome-wide identification of Cytochrome P450(CYP450)gene family,three genes(SbFBH1,2,and 5)encoding flavonoid 3′-hydroxylases(F3′Hs)and one gene(SbFBH7)encoding flavonoid 3′5′-hydroxylase(F3′5′H)were found to hydroxylate the B-ring of flavonoids.Our studies enrich the genomic information available for the Lamiaceae family and provide a toolkit for discovering CYP450 genes involved in the flavonoid decoration.Tianlin Pei Sanming Zhu Weizhi Liao Yumin Fang Jie Liu Yu Kong Mengxiao Yan Mengying Cui Qing Zhao 2023Horticulture Research2023,10,12:0
20An intra-articular injectable phospholipids-based gel for the treatment of rheumatoid arthritis显示文摘Rheumatoid arthritis(RA)is a chronic inflammatory and destructive arthropathy with a high deformity rate.Despite numerous studies and clinical trials,no curative treatment is available for large weight-bearing joints.Intra-articular(IA)injections could deliver high concentrations of drug to the afflicted joint and improve the drug efficacy while reducing systemic toxicity.However,free drugs are rapidly cleared from synovial fluid and do not significantly halt the progression of joint disease.Herein,a phospholipids-based controlledrelease gel was prepared for sustained IA delivery of celastrol(CEL)and the therapeutic efficiencywas evaluated in a rheumatoid arthritis rabbitmodel.The CEL-loaded gel(CEL-gel)contained up to 70%phospholipids yetwas easy to inject.After injecting into the joint cavity,CEL-gel achieved sol to gel phase transition without special stimuli and gelling agent.In vitro release and in vivo pharmacokinetic studies evidenced the stable and sustained release action of CEL-gel.A single IA injection of CEL-gel could maintain therapeutic efficiency for about 25 d and showed much better anti-arthritic efficacy compared to repeated injections of free drug solution(CEL-sol).Furthermore,the IA injection of CEL-gel greatly reduced the systemic toxicity of CEL.With good biocompatibility and biodegradability,CEL-gel might be a promising IA drug delivery system.Yuping Yang Shiqin Luo Xiong Peng Ting Zhao Qin He Mengying Wu Wei Zhang Tao Gong Zhirong Zhang 2023Asian Journal of Pharmaceutical Sciences2023,18,1:0
