1Promoter methylation of Wnt/β-Catenin signal inhibitor TMEM88 is associated with unfavorable prognosis of nonsmall cell lung cancer显示文摘Objective:Recent research has indicated that altered promoter methylation of oncogenes and tumor suppressor genes is an important mechanism in lung cancer development and progression.In this study,we investigated the association between promoter methylation of TMEM88,a possible inhibitor of the Wnt/β-Catenin signaling,and the survival of patients with nonsmall cell lung cancer(NSCLC).Methods:Twelve pairs of tumor and adjacent non-tumor samples were used for microarray analyses of DNA methylation and gene expression.For validation,more than two hundred additional samples were analyzed for methylation using bisulfite pyrosequencing and for gene expression using q RT-PCR.Then the cell function were tested by wound healing,transwell,CCK8 and cell cycle assay.Results:Our analysis of patient specimens showed that TMEM88 methylation was higher in NSCLC tumors(82.2%±10.3,P<0.01)compared with the adjacent normal tissues(65.9%±7.2).The survival analysis revealed that patients with high TMEM88methylation had a shorter overall survival(46 months)compared with patients with low TMEM88 methylation(>56 months;P=0.021).In addition,we found that demethylation treatment could inhibit tumor cell proliferation,migration,and invasion,which was supportive of an association between methylation and survival.Conclusions:Based on these consistent observations,we concluded that TMEM88 may play an important role in NSCLC progression and that promoter methylation of TMEM88 may serve as a biomarker for NSCLC prognosis and treatment.Rongna Ma Nannan Feng Xiao Yu Hongyan Lin Xiaohong Zhang Oumin Shi Huan Zhang Shuo Zhang Lei Li Min Zheng Ming Gao Herbert Yu Biyun Qian 2017Cancer Biology & Medicine2017,14,4:6

3Direct conversion of methane to methanol over Cu exchanged mordenite:Effect of counter ions显示文摘Direct conversion of methane(CH4)to methanol(DMTM)is a promising,but very challenging process for the utilization of abundant CH4 as a low carbon resource.In this context,Cu loaded zeolites,mordenite(MOR)in particular,were recognized as the most effective system to perform DMTM.In this work,different Cu salts were used to exchange with MOR,by which the effect of counter ions on the catalytic performance towards DMTM was investigated.The prepared catalysts were characterized and evaluated systematically.It was found that the counter ions affected the speciation of Cu sites,probably due to their capability in extraction of protons from MOR and the influence on the hydrolysis state of the Cu2+in aqueous solution.These behaviors adjusted the association between Cu2+and the exchangeable protons in MOR.As a result,varied DMTM performance was observed.Among the used Cu salts,Cu(CH3COO)2 exchanged MOR showed the highest performance,achieving stable CH3 OH yield of 117±28μmol/g in 5 consecutive cycles,these values are among the highest for Cu loaded MOR zeolites in open publications.Chuang Ma Xianjun Tan Haijiao Zhang Qun Shen Nannan Sun Wei Wei 2020Chinese Chemical Letters2020,31,1:2

6Study on catalytic mechanisms of Fe_(3)O_(4)-rGO_(x) in three typical advanced oxidation processes for tetracycline hydrochloride degradation显示文摘This study explored the catalytic mechanism and performance impacted by the materials ratio of Fe_(3)O_(4)-GO_(x) composites in three typical advanced oxidation processes(AOPs)of O_(3),peroxodisulfate(PDS)and photo-Fenton processes for tetracycline hydrochloride(TCH)degradation.The ratio of GO in the Fe_(3)O_(4)-GO_(x) composites exhibited different trends of degradation capacity in each AOPs based on different mechanisms.Fe_(3)O_(4)-rGO_(20wt%) exhibited the optimum catalytic performance which enhanced the ozone decomposition efficiency from 33.48%(ozone alone)to 51.83%with the major reactive oxygen species(ROS)of O_(2)·-.In PDS and photo-Fenton processes,Fe_(3)O_(4)-rGO_(5wt%) had the highest catalytic performance in PDS and H_(2)O_(2) decomposition for SO_(4)·–,and·OH generation,respectively.Compared with using PDS alone,PDS decomposition rate and TCH degradation rate could be increased by 5.97 and 1.73 times under Fe_(3)O_(4)-rGO_(5wt%) catalysis.In the photo-Fenton system,Fe_(3)O_(4)-rGO_(5wt%) with the best catalyst performance in H_(2)O_(2) decomposition,and TCH degradation rate increased by 2.02 times compared with blank group.Meantime,the catalytic mechanisms in those systems of that the ROS produced by conversion between Fe^(2+)/Fe^(3+)were also analyzed.Heshan Zheng Yunying Hou Shuo Li Jun Ma Jun Nan Nannan Wang 2023Chinese Chemical Letters2023,34,1:1

9Susceptibility of male and female Japanese medaka (Oryzias latipes) to 2,4,6-trichlorophenol-induced micronuclei in peripheral erythrocytes显示文摘2,4,6-trichlorophenol (2,4,6-TCP) is a wide- spread probable human carcinogen and has been proven to have genotoxicity in in vitro assays. However, little genotoxicity information and no micronuclei induction data for 2,4,6-TCP is available from in vivo tests, especially for sex-specific differences. Following a preliminary test, a piscine peripheral erythrocyte micro- nucleus assay was conducted on medaka (Oryzias latipes) after a 28-day exposure to 2,4,6-TCE In the present study, the mean micronuclei (MNC) frequencies of all of the groups increased in a dose-dependent manner, which indicated the potential genotoxicity of 2,4,6-TCE More- over, males were found to be more susceptible compared with females after a 28-day exposure to 2,4,6-TCP in all of the dosed groups above 10 ~tg-L-1. 14Correlation between knowledge of prevention COVID-19 and depression of residents in Henan province,China显示文摘Background:Coronavirus disease 2019(COVID-19) has shown explosive growth in cities in central China in recent days.Residents have the depression due to lack of corresponding prevention knowledge.Methods:A cross-sectional study was investigated 385 residents over 18 years of age.Structured questionnaire and DASS-21 scale used to explore the association between knowledge of prevention COVID-19 and depression among residents.Results:73 respondents (20.3%) suffered varying degrees of depression.The knowledge of the behavior of respondents most concerned (AOR 7.45,95%CI,1.87 ~ 29.77),the main symptoms of the COVID-19 (AOR 9.72,95%CI,1.19 ~ 79.12) and using special ambulances to transport patients or suspected patients (AOR 2.22,95%CI,1.07 ~ 4.62) was found to be significant related to depression.Conclusions:Although residents currently have a lower prevalence of depression,but there is a strong correlation between the knowledge and attitude of prevention COVID-19 and depression.Government agencies need to expand ways to spread the knowledge of prevention COVID-19.Anna Ma Dongyang Wang Nannan Wang ShanShan Zhu Qiong Li Shuangxi Guo 2020中国研究型医院2020,7,2:0

16OPAQUE3, encoding a transmembrane bZIP transcription factor, regulates endosperm storage protein and starch biosynthesis in rice显示文摘Starch and storage proteins are the main components of rice(Oryza sativa L.)grains.Despite their importance,the molecular regulatory mechanisms of storage protein and starch biosynthesis remain largely elusive.Here,we identified a rice opaque endosperm mutant,opaque3(o3),that overaccumulates 57-kDa proglutelins and has significantly lower protein and starch contents than the wild type.The o3 mutant also has abnormal protein body structures and compound starch grains in its endosperm cells.OPAQUE3(O3)encodes a transmembrane basic leucine zipper(bZIP)transcription factor(OsbZIP60)and is localized in the endoplasmic reticulum(ER)and the nucleus,but it is localized mostly in the nucleus under ER stress.We demonstrated that O3 could activate the expression of several starch synthesis-related genes(GBSSI,AGPL2,SBEI,and ISA2)and storage protein synthesis-related genes(OsGluA2,Prol14,and Glb1).O3 also plays an important role in protein processing and export in the ER by directly binding to the promoters and activating the expression of OsBIP1 and PDIL1-1,two major chaperones that assist with folding of immature secretory proteins in the ER of rice endosperm cells.High-temperature conditions aggravate ER stress and result in more abnormal grain development in o3 mutants.We also revealed that OsbZIP50 can assist O3 in response to ER stress,especially under high-temperature conditions.We thus demonstrate that O3 plays a central role in rice grain development by participating simultaneously in the regulation of storage protein and starch biosynthesis and the maintenance of ER homeostasis in endosperm cells.Ruijie Cao Shaolu Zhao Guiai Jiao Yingqing Duan Liuyang Ma Nannan Dong Feifei Lu Mingdong Zhu Gaoneng Shao Shikai Hu Zhonghua Sheng Jian Zhang Shaoqing Tang Xiangjin Wei Peisong Hu 2022Plant Communications2022,3,6:0

18Shaggy regulates tissue growth through Hippo pathway in Drosophila显示文摘The evolutionarily conserved Hippo pathway coordinates cell proliferation,differentiation and apoptosis to regulate organ growth and tumorigenesis.Hippo signaling activity is tightly controlled by various upstream signals including growth factors and cell polarity,but the full extent to which the pathway is regulated during development remains to be resolved.Here,we report the identification of Shaggy,the homolog of mammalian Gsk3β,as a novel regulator of the Hippo pathway in Drosophila.Our results show that Shaggy promotes the expression of Hippo target genes in a manner that is dependent on its kinase activity.Loss of Shaggy leads to Yorkie inhibition and downregulation of Hippo pathway target genes.Mechanistically,Shaggy acts upstream of the Hippo pathway and negatively regulates the abundance of the FERM domain containing adaptor protein Expanded.Our results reveal that Shaggy is functionally required for Crumbs/Slmb-mediated downregulation of Expanded in vivo,providing a potential molecular link between cellular architecture and the Hippo signaling pathway.Honggang Wu Nannan Zhu Jiyong Liu Jun Ma Renjie Jiao 2022Science China(Life Sciences)2022,65,11:0

20The bacterial effector SidN/Lpg1083 promotes cell death by targeting Lamin-B2显示文摘To facilitate survival,replication,and dissemination,the intracellular pathogen Legionella pneumophila relies on its unique type IVB secretion system(T4SS)to deliver over 330 effectors to hijack host cell pathways in a spatiotemporal manner.The effectors and their host targets are largely unexplored due to their low sequence identity to the known proteins and functional redundancy.The T4SS effector SidN(Lpg1083)is secreted into host cells during the late infection period.However,to the best of our knowledge,the molecular characterization of SidN has not been studied.Herein,we identified SidN as a nuclear envelope-localized effector.Its structure adopts a novel fold,and the N-terminal domain is crucial for its specific subcellular localization.Furthermore,we found that SidN is transported by eukaryotic karyopherin Importin-13 into the nucleus,where it attaches to the N-terminal region of Lamin-B2 to interfere with the integrity of the nuclear envelope,causing nuclear membrane disruption and eventually cell death.Our work provides new insights into the structure and function of an L.pneumophila effector protein,and suggests a potential strategy utilized by the pathogen to promote host cell death and then escape from the host for secondary infection.Jiajia Gao Wenwen Xu Feng Tang Minrui Xu Qin Zhou Xingyuan Yang Nannan Zhang Jinming Ma Qi Yang Xiaofang Chen Ximing Qin Honghua Ge 2023Journal of Molecular Cell Biology2023,15,5:0
