| 1 | The establishment of polypeptide PSMA-targeted chimericantigen receptor-engineered natural killer cells forcastration-resistant prostate cancer and the induction of ferroptosis-related cell death显示文摘Background:The mortality of castration-resistant prostate cancer(CRPC)is high due to lack of an effective treatment.Chimeric antigen receptor(CAR)-based therapy is a promising immunotherapeutic strategy.Here,we aimed to design a novel CAR-natural killer(NK)cells with a clinically significant tumoricidal effect on CRPC.Methods:We constructed novel CAR-NK92MI cells with a CD244-based recombinant lentiviral vector.Different intracellular segments(CD244,NKG2D,or CD3ζ)were screened to identify the best candidate according to cell lysis assay and CD107a expression levels.To enhance the affinity of the CAR to the tumor antigen,we compared an antibody specific for prostate-specific membrane antigen(anti-PSMA)with PSMA-targeted polypeptide(p-PSMA),which was screened by phage display combinatorial library.Then,CAR-NK92MI cells with both a high affinity for PSMA and a strong tumoricidal capacity were generated.In addition,we verified their tumor-killing effect in vitro and in vivo.The release of cytokine by NK92MI cells was compared with that by CAR-NK92MI cells through flow cytometry and enzyme-linked immunosorbent assay.Moreover,ferroptosis-related cell death was explored as a possible underlying mechanism.Results:Three different CAR intracellular regions CAR1(CD244),CAR2(CD244,NKG2D)and CAR3(CD244,NKG2D,and CD3ζ)were constructed.CAR2 was chosen to confer a stronger tumoricidal ability on CAR-NK92MI cells.Compared with anti-PSMA,p-PSMA exhibited enhanced affinity for the tumor antigen.Thus,p-PSMA-CAR-NK92MI cells,which expressed CAR with a polypeptide-based antigen-binding region,an intracellular CD244 and a NKG2D costimulatory domain,were generated.They could selectively and successfully kill PSMA+target cells and exhibited specific lysis rate of 73.19%for PSMA-positive C4-2 cells and 33.04%for PSMA-negative PC3 cells.Additionally,p-PSMA-CAR-NK92MI cells had significantly higher concentrations of IFN-γ,TNF-αand granzyme B than NK92MI cells.In a CRPC cancer xenograft model,p-PSMA-CAR-NK92MI cells significantly inhibited tumor growth and exerted a more consistent killing effect than NK92MI cells.Moreover,ferroptosis is a potential mechanism through which CAR-NK92MI cells attack cancer cells,and is triggered by IFN-γ.Conclusions:p-PSMA-CAR-NK92MI cells can effectively kill CRPCPSMA+cells in vitro and in vivo.This strategy may provide additional treatment options for patients with CRPC. | Liyuan Wu Fei Liu Le Yin Fangming Wang Hui Shi Qinxin Zhao Feiya Yang Dong Chen Xiying Dong Yuchun Gu Nianzeng Xing | 2022 | Cancer Communications2022,42,8: | 2 |

| 5 | Autonomous bus services:current research status and future recommendations显示文摘Implementing autonomous bus services in several cities has garnered substantial research attention worldwide.However,the benefits and challenges of this emerging mode remain insufficiently understood.Consequently,VOSviewer was employed for a bibliometric analysis involving 300 publications,investigating the associations among authors,journals,and keywords.Subsequently,we comprehensively reviewed the current state of research on two topics and proposed future recommendations.Results indicate that the first document related to autonomous bus services was published in 2009.Most user attitude-related research data are obtained via questionnaires and analyzed using statistical techniques.Autonomous bus services are expected to benefit passengers regarding travel time,cost,safety,etc.,while passenger preferences are inconsistent.However,integrating the service into existing bus systems requires careful consideration of the schedule sequences.Notably,modular autonomous bus services present a new opportunity for the further optimization of bus services.In future studies,standardized data acquisition procedures should be developed to achieve comparable results.Regarding traveler choice behavior,the effect of specific autonomous bus service policies over time and the heterogeneity due to cultural or social contexts across regions should be assessed.To further promote autonomous bus services,based on fluctuating travel demands,the effects of vehicle capacity,speed,and cost of fleet composition should be evaluated comprehensively to optimize the bus network and schedule sequence.Owing to the protracted nature of the transition from conventional to fully autonomous buses,one should prioritize semi-autonomous bus services.Another essential future research direction is to integrate modular autonomous bus assembly or disassembly strategies with different fine-grained operation optimization techniques in various scenarios. | Jinxing Shen Qinxin Liu Zi Ye Wenfeng Jiang Changxi Ma | 2023 | Digital Transportation and Safety2023,2,3: | 1 |

| 11 | Iridium-Doped 10H-Phase Perovskite BaCo_(0.8)Fe_(0.15)Ir_(0.05)O_(3-δ)as an Efficient Oxygen Evolution Reaction Catalyst显示文摘Designing low-cost and high-performance catalysts of electrocatalytic oxygen evolution reaction(OER)has always been one of the hot topics in current research.Hexagonal perovskite materials show great application potential in this field.Here,we synthesized 5%Iridium(Ir)-doped hexagonal phase perovskite BaCo_(0.8)Fe_(0.15)Ir_(0.05)O_(3–δ)(BCFI).Using the Rietveld method to refine the structure,combining with the spherical aberration corrected high-angle annular dark-field scanning transmission electron microscopy(HAADF-STEM)atomic imaging,we found that BCFI is a ten-layer hexagonal(10H)structure,which belongs to the Purn:x-wiley:1001604X:media:cjoc202200200:cjoc202200200-math-0001m2 space group.In addition,BCFI shows excellent OER catalytic effect and good stability in 1 mol/L KOH solution,and the overpotential reaches 294 mV at a current density of 10 mA cm^(–2).The reduction of Cobalt(Co)valence state induced by Ir doping significantly enhances the OER activity of BCFI.Meanwhile,density functional theory(DFT)calculations show that the 2p-band center of oxygen(O)in BCFI is closer to the Fermi level than that in undoped BaCo_(0.8)Fe_(0.15)Ir_(0.05)O_(3–δ)(BCF),which is beneficial to the OER.This work reveals a novel 10H-BCFI material with excellent OER performance,and provides a basis for the design of hexagonal perovskite materials in OER and other energy conversion fields. | Huimin Liu Qinxin Luo Jiaping Hu Lianwei Wei Wanqun Zhang Hui Zheng Shusheng Wu Kaibin Tang | 2022 | Chinese Journal of Chemistry2022,40,19: | 0 |

| 13 | Expression,purification and identification of LBD domain of human PPARδ in E.coli显示文摘Peroxisome proliferator-activated receptors(PPARs) are ligand-activated transcription factors known to play a pivotal role in regulations of metabolism.In order to yield soluble ligand binding domain of PPARδ(PPARδLBD) for screening ligands,the cDNA was amplified using total RNA from HepG2 cells by RT-PCR.Then the enzyme-digested product was inserted downstream of the malE gene in the vector pMAL-p2x,which encoded maltose-binding protein(MBP),resulting in the expression of an MBP-PPARδLBD fusion protein.The recombinant plasmid was transformed into E.coli TB1 that was cultured shakily at 30 °C,200 r/min and induced by 0.4 mmol/L IPTG for 6 h.The cells were harvested by centrifugation and broken by sonication.The expressed fusion protein was soluble and accounted for 0.31 of the total protein in the supernatant.Western blot analysis showed that the expressed MBP-PPARδLBD could bind to anti-MBP-antibody.The MBP-PPARδLBD fusion protein of 77 kDa and the PPARδLBD protein of 34 kDa were obtained by amylose-resin affinity chromatography without or with digestion of Factor Xa.They were both homogeneity,judged by SDS-PAGE.The recombinant MBP-PPARδLBD and PPARδLBD protein with high purity is obtained,which provides the necessary material for screening and researching PPARδ ligands. | Liu Hua Li Changqing Ling Baodong Zhou Qinxin | 2009 | Journal of Medical Colleges of PLA(China)2009,24,2: | 0 |
| 14 | Bacterial community diversity of meltwater runoff and soil in Midre Lovénbreen glacier in Ny-Alesund,Arctic显示文摘Glacial meltwater runoff is a dynamic ecosystem.On the one hand,nutrient concentration changes as it flows from upstream to downstream,and on the other hand,bacterial community structure changes due to its contact with nearby soil during the flow process.We studied meltwater and soil in the Midre Lovénbreen glacier region,to explore changes in bacterial diversity as meltwater flows,and the relationship between meltwater and soil bacterial diversity.As glacial meltwater flows from upstream to downstream,the relative abundance of dominant bacterial groups changes.In addition,we found that during the flowing process,nutrient exchange and bacterial contact had occurred between the meltwater runoff and the soil.As a result,the distribution patterns of some bacteria in the meltwater are very similar to those in the soil.Finally,we combined distance-based redundancy analysis and weighted correlation network analysis to show that NO_(3)^(-)-N and NO_(2)^(-)-N are the most two significant factors affecting glacial meltwater and soil,respectively.Our results suggest that in such a close-knit ecosystem,the interaction of glacial meltwater with soil,as well as environmental factors,together determine bacterial community composition. | LIN Lidong WANG Nengfei HAN Wenbing ZHANG Botao ZANGJiaye LI Qinxin QIN Yiling WANG Long ZHANG Fang LIU Jie | 2022 | Advances in Polar Science2022,33,2: | 0 |
