2Hypoxia-stressed cardiomyocytes promote early cardiac differentiation of cardiac stem cells through HIF-1α/Jagged1/Notch1 signaling显示文摘Hypoxia is beneficial for the differentiation of stem cells transplanted for myocardial injury,but mechanisms underlying this benefit remain unsolved. Here, we report the impact of hypoxia-induced Jagged1 expression in cardiomyocytes(CMs) for driving the differentiation of cardiac stem cells(CSCs).Forced hypoxia-inducible factor 1α(HIF-1α) expression and physical hypoxia(5% O_2) treatment could induce Jagged1 expression in neonatal rat CMs. Pharmacological inhibition of HIF-1α by YC-1 attenuated hypoxia-promoted Jagged1 expression in CMs. An ERK inhibitor(PD98059), but not inhibitors of JNK(SP600125), Notch(DAPT), NF-κB(PTDC), JAK(AG490), or STAT3(Stattic) suppressed hypoxiainduced Jagged1 protein expression in CMs. c-Kit^+ CSCs isolated from neonatal rat hearts using a magnetic-activated cell sorting method expressed GATA4, SM22α or vWF, but not Nkx2.5 and cTnI.Moreover, 87.3% of freshly isolated CSCs displayed Notch1 receptor expression. Direct co-culture of CMs with BrdU-labeled CSCs enhanced CSCs differentiation, as evidenced by an increased number of BrdU^+/Nkx2.5^+ cells, while intermittent hypoxia for 21 days promoted co-culture-triggered differentiation of CSCs into CM-like cells. 3A Hybrid Electrode of Co_3O_4@PPy Core/Shell Nanosheet Arrays for High-Performance Supercapacitors显示文摘Herein, combining solverthermal route and electrodeposition, we grew unique hybrid nanosheet arrays consisting of Co_3O_4 nanosheet as a core, PPy as a shell. Benefiting from the PPy as conducting polymer improving an electron transport rate as well as synergistic effects from such a core/shell structure, a hybrid electrode made of the Co_3O_4@PPy core/shell nanosheet arrays exhibits a large areal capacitance of 2.11 F cm-2at the current density of 2 m A cm^(-2), a *4-fold enhancement compared with the pristine Co_3O_4electrode; furthermore, this hybrid electrode also displays good rate capability(*65 % retention of the initial capacitance from 2 to 20 m A cm^(-2)) and superior cycling performance(*85.5 % capacitance retention after 5000 cycles). In addition, the equivalent series resistance value of the Co_3O_4@PPy hybrid electrode(0.238 X) is significantly lower than that of the pristine Co_3O_4electrode(0.319 X). 4Influence of order degree of coaly graphite on its structure change during preparation of graphene oxide显示文摘Coaly graphite is an important natural graphite resource that derived from coal under a natural process that associated with igneous intrusion.Flake graphite is usually used for the chemical synthesis of graphene oxide(GO),the main precursor for preparation of graphene,but few papers pay attention to preparing GO using coaly graphite.In this paper,four kinds of natural coaly graphite with different graphitization degrees were exposed to a modified Hummer’s oxidation method to prepare GO.The flake graphite sample was also used for comparison.The results showed that the structural change process from graphite to GO were significantly affected by the graphitization degree of the original coaly graphite.The relationship between yields of GO and graphitization degrees of the coaly graphite was explored.The mechanism of why the graphite with low graphitization degrees cannot be totally oxidized was proposed.Coaly graphite with a graphitization degree of higher than 80% was easier to be oxidized and yielded the same amount of GO as the flake graphite did,suggesting it is the potential substitute for the flake graphite to produce GO in bulk quantities.Dandan Hou Kuo Li Rujia Ma Qinfu Liu 2020Journal of Materiomics2020,6,3:2
5A Novel Photothermal Nanocrystals of Cu_7S_4 Hollow Structure for Efficient Ablation of Cancer Cells显示文摘Cu2-x S nanocrystals(NCs), characterized by low cost, low toxicity, high stability and high photothermal conversion efficiency, provide promising platforms as photothermal agents. 9Super enhancers targeting ZBTB16 in osteogenesis protect against osteoporosis显示文摘As the major cell precursors in osteogenesis, mesenchymal stem cells(MSCs) are indispensable for bone homeostasis and development. However, the primary mechanisms regulating osteogenic differentiation are controversial. Composed of multiple constituent enhancers, super enhancers(SEs) are powerful cis-regulatory elements that identify genes that ensure sequential differentiation. The present study demonstrated that SEs were indispensable for MSC osteogenesis and involved in osteoporosis development. Through integrated analysis, we identified the most common SE-targeted and osteoporosis-related osteogenic gene,ZBTB16. ZBTB16, positively regulated by SEs, promoted MSC osteogenesis but was expressed at lower levels in osteoporosis.Mechanistically, SEs recruited bromodomain containing 4(BRD4) at the site of ZBTB16, which then bound to RNA polymerase IIassociated protein 2(RPAP2) that transported RNA polymerase Ⅱ(POL Ⅱ) into the nucleus. The subsequent synergistic regulation of POL Ⅱ carboxyterminal domain(CTD) phosphorylation by BRD4 and RPAP2 initiated ZBTB16 transcriptional elongation, which facilitated MSC osteogenesis via the key osteogenic transcription factor SP7. 10Roles of SET7/9 and LSD1 in the Pathogenesis of Arsenicinduced Hepatocyte Apoptosis显示文摘Background and Aims:Multiple regulatory mechanisms play an important role in arsenic-induced liver injury.To investigate whether histone H3 lysine 4(H3K4)methyltransferase(SET7/9)and histone H3K4 demethyltransferase(LSD1/KDM1A)can regulate endoplasmic reticulum stress(ERS)-related apoptosis by modulating the changes of H3K4 methylations in liver cells treated with arsenic.Methods:Apoptosis,proliferation and cell cycles were quantified by flow cytometry and real-time cell analyzer.The expression of ERS-and epigenetic-related proteins was detected by Western blot analysis.The antisense SET7/9 expression vector and the overexpressed LSD1 plasmid were used for transient transfection of LO_(2) cells.The effects of NaAsO_(2) on the methylation of H3 in the promoter regions of 78 kDa glucose-regulated protein,activating transcription factor 4 and C/EBP-homologous protein were evaluated by chromatin immunoprecipitation assay.Results:The protein expression of LSD1(1.25±0.08 vs.1.77±0.08,p=0.02)was markedly decreased by treatment with 100μM NaAsO_(2),whereas the SET7/9(0.68±0.05 vs.1.10±0.13,p=0.002)expression level was notably increased,which resulted in increased H3K4me1/2(0.93±0.64,1.19±0.22 vs.0.71±0.13,0.84±0.13,p=0.03 and p=0.003).After silencing SET7/9 and overexpressing LSD1 by transfection,apoptosis rate(in percentage:3.26±0.34 vs.7.04±0.42,4.80±0.32 vs.7.52±0.38,p=0.004 and p=0.02)was significantly decreased and proliferation rate was notably increased,which is reversed after inhibiting LSD1(in percentage:9.31±0.40 vs.7.52±0.38,p=0.03).Furthermore,the methylation levels of H3 in the promoter regions of GRP78(20.80±2.40 vs.11.75±2.47,20.46±2.23 vs.14.37±0.91,p=0.03 and p=0.01)and CHOP(48.67±4.04 vs.16.67±7.02,59.33±4.51 vs.20.67±3.06,p=0.004 and p=0.001)were significantly increased in LO_(2) cells exposed to 100μM NaAsO_(2) for 24 h.Conclusions:Histone methyltransferase SET7/9 and histone demethyltransferase LSD1 jointly regulate the changes of H3K4me1/me2 levels in arsenic-induced apoptosis.NaAsO_(2) induces apoptosis in LO_(2) cells by activating the ERS-mediated apoptotic signaling pathway,at least partially by enhancing the methylation of H3 on the promoter regions of ERS-associated genes,including GRP78 and CHOP.Bing Han Yi Yang Lei Tang Qin Yang Rujia Xie 2021Journal of Clinical and Translational Hepatology2021,9,3:1

14Design of a unified rendering shader for mobile device显示文摘Real-time graphics processing is all along a crucial task of mobile device, and it is conventionally supported by programmable graphics processing unit(GPU). These GPUs are designed to flexibly support vertex and pixel processing with classic techniques such as on-chip cache and dynamic programmable pipelining. However, it is difficult for the vertex shader and pixel shader to achieve high utilization of hardware resources, even though there is a certain balance by reasonable processor quantity ratio. In this paper, a unified render shader with a very long instruction word(VLIW) processor was designed. The viewport transformation algorithm and the Mipmap mapping algorithm are respectively mapped on the shader, with the purpose of providing an energy-efficient and flexible hardware platform for graphics processing in mobile device. 15Accelerated deterioration mechanism of 316L stainless steel in NaCl solution under the intermittent tribocorrosion process显示文摘In this research,the tribocorrosion behavior of 316L stainless steel in simulated seawater was investigated under continuous and intermittent sliding at open circuit potential.The tribocorrosion mechanism was discussed in terms of wear morphologies,mechanical property as well as chemical composition.Meanwhile,microstructure evolution inside the wear track and open circuit potential recorded after sliding were analyzed to quantify the repassivation kinetics and evaluate the impact of the regenerated passive film on wear.The results showed that the wear rate increased under intermittent sliding when the pause time is long enough to repassivate after sliding.Repeated sliding promoted the refinement of the grain inside the sliding area,which was beneficial to the generation of the thicker and more compact passive film inside the wear track.The ruptured passive film often acted as abrasives during subsequent sliding.Therefore,the accelerated material loss under intermittent sliding was attributed to the periodic mechanical removal of the thickened passive film and the enhanced abrasive wear inside the wear track.Yingrui Liu Linlin Liu Shuyu Li Rujia Wang Peng Guo Aiying Wang Peiling Ke 2022Journal of Materials Science & Technology2022,,26:1
16Construction of Ultrathin Layered MXene-TiN Heterostructure Enabling Favorable Catalytic Ability for High-Areal-Capacity Lithium-Sulfur Batteries显示文摘Catalysis has been regarded as an effective strategy to mitigate sluggish reaction kinetics and serious shuttle effect of Li-S batteries.Herein,a spherical structure consists of ultrathin layered Ti_(3)C_(2)T_(x)-TiN heterostructures(MX-TiN)through in-situ nitridation method is reported.Through controllable nitridation,highly conductive TiN layer grew on the surface and close coupled with interior MXene to form unique 2D heterostructures.The ultrathin heterostructure with only several nanometers in thickness enables outstanding ability to shorten electrons diffusion distance during electrochemical reactions and enlarge active surface with abundant adsorptive and catalytic sites.Moreover,the(001)surface of TiN is dominated by metallic Ti-3d states,which ensures fast transmitting electrons from high conductive MX-TiN matrix and thus guarantees efficient catalytic performance.Calculations and experiments demonstrate that polysulfides are strongly immobilized on MX-TiN,meanwhile the bidirectional reaction kinetics are catalytically enhanced by reducing the conversion barrier between liquid LiPSs and solid Li_(2)S_(2)/Li_(2)S.As a result,the S/MX-TiN cathode achieves excellent long-term cyclability with extremely low-capacity fading rate of 0.022%over 1000 cycles and remarkable areal capacity of 8.27 mAh cm^(−2) at high sulfur loading and lean electrolytes.Hao Wang Zhe Cui Shu-Ang He Jinqi Zhu Wei Luo Qian Liu Rujia Zou 2022Nano-Micro Letters2022,14,11:1

18Effects of ambient humidity and thermal aging on properties of Nomex insulation in mining dry‐type transformer显示文摘Temperature and humidity are the main factors that cause the decomposition of Nomex paper widely used as turn‐to‐turn insulation material of mining dry‐type transformers. In this study, for understanding the effect of ambient humidity and thermal aging on the properties of Nomex paper, specimens with the initial moisture content of 7% were aged in the oven for 168, 504, 672 h at 180°C, respectively. After thermal aging, corona discharge experiments of unaged and aged specimens with different aging degrees were carried out at different relative humidity (RH) levels from 10 to 70%. The characteristic parameters of corona discharge, such as the average discharge amplitude and the number of discharges, were measured. Then, the surface conductivity and the surface potential decay rate were measured. The morphology and attenuated total internal reflectance Fourier transform infrared spectroscopy were also analysed for explaining the difference of properties be-tween unaged and aged specimens. The experiment results indicate that the average discharge amplitude decreases with the increase of RH or the deepening of the aging degree of specimens, but the number of discharges shows the reversal tendency. 19Real-Time and Spatial Electroanalysis of Biomolecules in One Living Cell Using Liquid-Phase Modified Nanopipette显示文摘The well-developed solid-phase modified strategy at the electrode has enabled the preparation of biosensors for the detection of multiple analytes,even in single living cells.However,limited assay elements can be modified at the solid surface,restricting the types of molecules that can be analyzed and the sensitivity of detection.Here,a novel liquid-phase modified strategy at the tip of a nanopipette is designed to realize real-time and local analysis of biomolecules inside the cell that are barely detectable using solid-phase modified nanoelectrodes.This design utilizes the nanotip structure at a platinized carbon open nanopipette to stably retain a nanodroplet that contains the required reagents with high reactivity for the assay of the target analyte.The generated hydrogen peroxide is electrochemically quantified at the Pt layer to carry out the real-time measurement in a living cell with a spatial resolution of 70 nm.Taking advantage of highly spatial and real-time detection,uneven distribution of sphingomyelinase(SMase)in the living CT26 cell is unprecedentedly shown to exhibit the significance in the establishment of liquid-phase modified nanopipette.This new modification strategy opens up a new direction for sensor design and consequently advances the development of biosensors in the chemical and biological research.Kang Liu Rujia Liu Dengchao Wang Rongrong Pan Hong-Yuan Chen Dechen Jiang 2023CCS Chemistry2023,5,6:0
