1Effect of human umbilical cord-derived mesenchymal stem cells on lung damage in severe COVID-19 patients:a randomized,double-blind,placebo-controlled phase 2 trial显示文摘Treatment of severe Coronavirus Disease 2019(COVID-19)is challenging.We performed a phase 2 trial to assess the efficacy andsafety of human umbilical cord-mesenchymal stem cells(UC-MScs)to treat severe coViD-19 patients with lung damage,based onour phase 1 data.In this randomized,double-blind,and placebo-controlled trial,we recruited 101 severe coVID-19 patients withlung damage.They were randomly assigned at a 2:1 ratio to receive either UC-MSCs(4×10^(7)cells per infusion)or placebo on day 0,3,and 6.The primary endpoint was an altered proportion of whole lung lesion volumes from baseline to day 28.Other imagingoutcomes,6-minute walk test(6-MWT),maximum vital capacity,diffusing capacity,and adverse events were recorded and analyzed.In all,100 COVID-19 patients were finally received either UC-MSCs in=65)or placebo(n=35).UC-MSCs administrationexerted numerical improvement in whole lung lesion volume from baseline to day 28 compared with the placebo(the mediandifference was-13.31%,95%Cl-29.14%,2.13%,P=0.08).UC-MSCs significanty reduced the proportions of solid componentlesion volume compared with the placebo(median difference:-15.45%;95%CI-30.82%,-0.39%;P=0.043).The 6-MWT showedan increased distance in patients treated with UC-MSCs(difference:27.00 m;95%CI 0.00,57.00;P=0.057).The incidence of adverseevents was similar in the two groups.These results suggest that UC-MSCs treatment is a safe and potentially effective therapeuticapproach for COVID-19 patients with lung damage.A phase 3 trial is required to evaluate effects on reducing mortality andpreventing long-term pulmonary disability.Lei Shi Hai Huang Xuechun Lu Xiaoyan Yan Xiaojing Jiang Ruonan Xu Siyu Wang Chao Zhang Xin Yuan Zhe Xu Lei Huang Jun-Liang Fu Yuanyuan Li Yu Zhang Wei-Qi Yao Tianyi Liu Jinwen Song Liangliang Sun Fan Yang Xin Zhang Bo Zhang Ming Shi Fanping Meng Yanning Song Yongpei Yu Jiqiu Wen Qi Li Qing Mao Markus Maeurer Alimuddin Zumla Chen Yao Wei-Fen Xie Fu-Sheng Wang 2021Signal Transduction and Targeted Therapy2021,6,3:13
2Pm67, a new powdery mildew resistance gene transferred from Dasypyrum villosum chromosome 1V to common wheat(Triticum aestivum L.)显示文摘Powdery mildew, caused by the biotrophic fungus Blumeria graminis f. sp. tritici(Bgt), is a global disease that poses a serious threat to wheat production. To explore additional resistance gene, a wheatDasypyrum villosum 1 V#5(1 D) disomic substitution line NAU1813(2 n = 42) with high level of seedling resistance to powdery mildew was used to generate the recombination between chromosomes 1 V#5 and1 D. Four introgression lines, including t1 VS#5 ditelosomic addition line NAU1815, t1 VL#5 ditelosomic addition line NAU1816, homozygous T1 DL·1 VS#5 translocation line NAU1817, and homozygous T1 DS·1 VL#5 translocation line NAU1818 were developed from the selfing progenies of 1 V#5 and 1 D double monosomic line that derived from F1 hybrids of NAU1813/NAU0686. All of them were characterized by fluorescence in situ hybridization, genomic in situ hybridization, 1 V-specific markers analysis, and powdery mildew tests at different developmental stages. A new powdery mildew resistance gene named Pm67 was physically located in the terminal bin(FL 0.70–1.00) of 1 VS#5. Lines with Pm67 exhibited seedling stage immunity and tissue-differentiated reactions at adult plant stage. The sheaths, stems, and spikes of the Pm67 line were still immune, but the leaves showed a low degree of susceptibility.Microscopic observation showed that most penetration attempts were stopped in association with papillae on the sheath, and colonies cannot form conidia on the susceptible leaf of Pm67 line at adult plant stage, suggesting that the defence layers of the Pm67 line is tissue-differentiated. 3Bio-ecological remediation of freshwater aquaculture environments:A systematic review and bibliometric analysis显示文摘Freshwater aquaculture plays an important role in the global aquaculture industry.The success of freshwater aquaculture relies on the integration of bio-ecological remediation technologies to ensure its sustainable development.This paper systematically reviews the global literatures on bio-ecological remediation of freshwater aquaculture environments from 1990 to 2021,and discusses in detail the research progress and status quo in this field using both bibliometric analysis and content analysis.Our results indicate that the number of publications per year has increased year by year,with a particularly rapid increase after 2015.Institutions and researchers from China and the USA publish the greatest number of papers and contribute the most to this field.By analyzing the research progress in this field,we also find that the protection and restoration of freshwater aquaculture environments has received attention since as early as 1990,such as the application of constructed wetlands and recirculating aquaculture systems.Recently,researchers have increasingly focused on the use of microbial communities in remediation.Furthermore,vanRijn,J's papers are of great value to the research in this field.In content analysis,the application of primary technologies,including plant selection,aquatic animals,microorganisms,and biological filtration and purification equipment,as well as ecological engineering technologies with combinations of various types of organisms,is described to improve the effectiveness and efficiency of remediation.The performance,advantages,disadvantages,or limitations and prospects of these technology applications are analyzed in detail.By comparison,ecological engineering is shown to be the best remediation technology,being most comprehensive,and well-established.Taken together,this paper composes and summarizes existing work in this field,and provides guidance for future research on the improvement of existing technologies and the development of new technologies,with the aim of enhancing remediation capability,improving ecological sustainability and ensuring economic viability.Qiaohong Zhou Kaixuan Wu Lu Yao Ruonan Chen Shouzhuang Liu Hao Xing Linmei Nie Zhenbin Wu 2024Water Biology and Security2024,3,1:0

5A systematic identification of cold tolerance genes in peanut using yeast functional screening system显示文摘Peanut(Arachis hypogaea L.)is a thermophilic crop,and low temperature leads to a significant reduction in annual yields.Despite a few cold tolerant germplasms or cultivars have been discovered and developed,molecular mechanisms governing peanut cold tolerance is poorly understood.Identification of keys genes involved in cold tolerance is the first step to address the underlying mechanism.In this study,we isolated and characterized 157 genes with potentials to confer cold tolerance in peanut by using a yeast functional screening system.GO(Gene ontology)and KEGG(Kyoto encyclopedia of genes and genomes)enrichment analysis of these genes revealed that ribosome and photosynthesis proteins might play essential roles in peanut cold response.Transcriptome results indicated that 60 cold tolerance candidate genes were significantly induced or depressed by low temperature.qRT-PCR analysis demonstrated that several candidate genes could be also regulated by salt or drought stress.Individual overexpression of two UDP-glycosyltransferases(AhUGT2 and AhUGT268)in transgenic yeast cells could enhance their tolerance to multiple abiotic stress.In conclusion,this study advances our understanding of the mechanisms associated with the cold stress responses in peanut,and offers valuable gene resources for genetic improvement of abiotic stress tolerance in crops.Xin Wang Yue Liu Ruonan Yao Lei Ouyang Tingting Yu Liying Yan Yuning Chen Dongxin Huai Zhihui Wang Yanping Kang Qianqian Wang Huifang Jiang Yong Lei Boshou Liao 2023Oil Crop Science2023,8,3:0
6Simultaneously enhancing mechanical and tribological properties of carbon fiber composites by grafting SiC hexagonal nanopyramids for brake disk application显示文摘Extensive attention has been drawn to the development of carbon fiber composites for their application in brake disks due to the increasing demand for brake disks with high mechanical strength and better tribological properties.Herein,we design SiC hexagonal nanopyramids modified carbon/carbon(SiCNPsC/C)composites,in which SiCNPs are radially grafted on the carbon fibers by the combined sol-gel and carbothermal reduction method,and pyrolytic carbon(Py C)matrix is deposited on nucleation sites including carbon fibers and SiCNPs by isothermal chemical vapor infiltration(ICVI).Benefiting from the special structure,SiCNPs-C/C composites exhibit superior mechanical and frictional performance.Compared with C/C composites,SiCNPs-C/C composites have 147%,90.3%,70.6%,and 117.9%improvement in the hardness,interlaminar shear strength,and out-of-plane and in-plane compressive strength,respectively,which is attributed to the optimized fiber/matrix(F/M)interfaces bonding and the enhanced cohesion strength of Py C matrix.In addition,the friction coefficient of SiCNPs-C/C composites increases by 25.5%,and the wear rate decreases by 38.0%.This work provides an optional design thought for the nanomaterials and enlightens the mechanical and frictional modification of composites in the field of the brakes.Yao Guo Leilei Zhang Qiang Song Ruonan Zhang Fei Zhao Wei Li Hongchao Sheng Xianghui Hou Hejun Li 2022Journal of Materials Science & Technology2022,,26:0
