1STAT3 mediates resistance of CD44^+CD24^(-/low) breast cancer stem cells to tamoxifen in vitro显示文摘We sought to determine whether STAT3 mediated tamoxifen resistance of breast cancer stem cells in vitro.The capacities for mammosphere formation and STAT3 expression of CD44+CD24-/low MCF-7 and MCF-7 were observed.The CD44+CD24-/low subpopulation ratio and its sensitivity to adriamycin were analyzed in MCF-7 and TAM resistant(TAM-R) cells.Cell cycle,apoptosis,STAT3 and phospho-STAT3 changes were observed af-ter treatment with tamoxifen.Small interference RNA-mediated knockdown of STAT3 in TAM-R cells was also performed.CD44+CD24-/low MCF-7 showed higher capacities for mammosphere formation and STAT3 expression than total MCF-7.The CD44+CD24-/low subpopulation was also upregulated in TAM-R cells with less sensitivity to adriamycin than MCF-7.Cell cycle changes,anti-apoptotic effects and STAT3 changes were also found.Mean-while,the knock-down of STAT3 in TAM-R resulted in an increase in sensitivity to tamoxifen.It is concluded that STAT3 plays an essential role in breast cancer stem cells,which correlated with tamoxifen resistance.Xiaoyan Wang Guozhu Wang Yi Zhao Xiaoan Liu Qiang Ding Jingping Shi Yin Ding Shui Wang 2012The Journal of Biomedical Research2012,26,5:6
2Research on allocation efficiency of the redistributed pseudo inverse algorithm显示文摘Allocation efficiency is an important performance index to measure the quality of the allocation algorithm. In order to compute the efficiency, the volume of the subset of attainable moments must be solved. The efficiency of the redistributed pseudo inverse (RPI) algorithm depends on the choice of the pseudo-inverse matrix. The subset of attainable moments of RPI is a complex non-convex polyhedron. By analyzing twodimensional and three-dimensional allocation problems with a 'micro-element' method, here we propose an approximate calculation algorithm to compute the volume of the non-convex polyhedron. In order to improve the allocation efficiency of RPI, genetic algorithm is used to find the best pseudo-inverse matrix. 3Two-dimensional Pd-based nanomaterials for bioapplications显示文摘Noble metal nanomaterials have been extensively explored in cancer diagnostic and therapeutic applications owing to their unique physical and chemical properties, such as facile synthesis, straightforward surface functionalization, strong photothermal effect, and excellent biocompatibility. Herein, we summarize the recent development of two-dimensional(2D) Pd-based nanomaterials and their applications in cancer diagnosis and therapy. Different synthetic strategies for Pd nanosheets and the related nanostructures, including Pd@Au, Pd@Ag nanoplates and mesocrystalline Pd nanocorolla, are first discussed.Together with their unique properties, the potential bioapplications of these 2D Pd nanomaterials are then demonstrated. With strong absorption in near-infrared(NIR) region, these nanomaterials have great potentials in cancer photothermal therapy(PTT). They also readily act as contrast agents in photoacoustic(PA) imaging or X-ray computed tomography(CT) to achieve image-guided cancer therapy. Moreover,significant efforts have been devoted to studying the combination of PTT and other treatment modalities(e.g., chemotherapy or photodynamic therapy) based on Pd nanomaterials. The remarkable synergistic or collaborative effects to achieve better therapeutic efficacy are discussed as well. Additionally, the biosafety of 2D Pd-based nanomaterials in vitro and in vivo was evaluated. 4Effect of glutathione on in vivo biodistribution and clearance of surface-modified small Pd nanosheets显示文摘Plasmonic Pd nanosheets have been emerging as promising materials for application in near-infrared(NIR) photothermal therapy(PTT) of cancer. However, animal in mice studies indicated that the original synthesized poly(vinylpyrrolidone)(PVP)-protected small Pd nanosheets(Pd-PVP) and some further surface-modified small Pd nanosheets such as Pd-PEG(SH) easily accumulated in reticuloendothelial system(RES) organs(liver, spleen, etc.) and were difficult to be cleared from these organs quickly. In the work, we surprisingly found that glutathione(GSH) could promote the clearance of surface-modified small Pd nanosheets(e.g. Pd-PVP, Pd-PEG(SH) and Pd-GSH) from the RES organs efficiently. The effects of GSH on the biodistribution and clearance of different surface-modified Pd nanosheets were investigated. Our results indicated that these surface-modified Pd nanosheets with or without GSH added caused no morbidity at target primary organs, and GSH can promote the clearance of different surface-modified Pd nanosheets in the order of Pd-PVP≈Pd-PEG(SH)>Pd-GSH. 7Effects of chronic peripheral pretreatment with an angiotensin II type-1 receptor blocker on apoptosis-related molecules in rats with cerebral ischemia/reperfusion injury显示文摘Chronic systemic treatment with blockers of angiotensin II type-1(AT1) receptors inhibits ischemia-induced apoptosis and reduces ischemic neuronal damage.However,the molecular mechanisms of AT1 receptor blockers in modulating neuronal apoptosis remain poorly understood.Pretreatment with irbesartan significantly suppressed cell apoptosis at 1-7 days following cerebral ischemia/reperfusion,increased levels of brain-derived neurotrophic factor,and elevated the ratios of Bcl-2/Bax and phosphorylated cyclic adenosine monophosphate response element-binding protein(pCREB)/CREB in the ischemic cortex at 1 day after reperfusion,as well as suppressing caspase-3 activation.Cerebral ischemia increased the mRNA expression of AT1 and AT2 receptors in the ischemic cortex,whereas irbesartan blocked this increase in AT1 expression but potentiated the expression of AT2.Therefore,this AT1 receptor blocker was neuroprotective by increasing the ratios of Bcl-2/Bax and pCREB/CREB,increasing brain-derived neurotrophic factor levels,inhibiting caspase-3 activation,and modulating AT receptor expression.Jingping Shi Jingde Dong Jie Lu Yingdong Zhang 2010Neural Regeneration Research2010,5,15:2

13Continuous activation of polymorphonuclear myeloid-derived suppressor cells during pregnancy is critical for fetal development显示文摘The maternal immune system is vital in maintaining immunotolerance to the semiallogeneic fetus for a successful pregnancy.Although studies have shown that myeloid-derived suppressor cells(MDSCs)play an important role in maintaining feto-maternal tolerance,little is known about the role of MDSCs in pregnancies with intrauterine growth retardation(IUGR).Here,we reported that the activation of polymorphonuclear myeloid-derived suppressor cells(PMN-MDSCs)during pregnancy was closely associated with fetal growth.In humans,class E scavenger receptor 1(SR-E1),a distinct marker for human PMN-MDSCs,was used to investigate PMN-MDSC function during pregnancy.Continuous activation of SR-E1+PMN-MDSCs was observed in all stages of pregnancy,accompanied by high cellular levels of ROS and arginase-1 activity,mediated through STAT6 signaling.However,SR-E1+PMN-MDSCs in pregnancies with IUGR showed significantly lower suppressive activity,lower arginase-1 activity and ROS levels,and decreased STAT6 phosphorylation level,which were accompanied by an increase in inflammatory factors,compared with those in normal pregnancies.Moreover,the population of SR-E1+PMN-MDSCs was negatively correlated with the adverse outcomes of newborns from pregnancies with IUGR.In mice,decreases in cell population,suppressive activity,target expression levels,and STAT6 phosphorylation levels were also observed in the pregnancies with IUGR compared with the normal pregnancies,which were rescued by the adoptive transfer of PMN-MDSCs from pregnant mice.Interestingly,the growth-promoting factors(GPFs)secreted by placental PMN-MDSCs in both humans and mice play a vital role in fetal development.These findings collectively support that PMN-MDSCs have another new role in pregnancy,which can improve adverse neonatal outcomes.Mengyu Shi Ziyang Chen Meiqi Chen Jingping Liu Jing Li Zhe Xing Xiaogang Zhang Shuaijun Lv Xinyao Li Shaowen Zuo Shi Feng Ying Lin Gang Xiao Liping Wang Yumei He 2021Cellular & Molecular Immunology2021,18,7:1

15Hope and Academic Procrastination in Adolescents: A Moderated Mediation Model显示文摘Academic procrastination among adolescents is an increasingly prominent problem. It is important to look forinfluences behind academic procrastination in the adolescent population. The present study aimed to revealthe explanatory mechanisms underlying the association between hope and academic procrastination behaviorsamong Chinese adolescents by testing the mediating role of attentional control and the moderating role of traitmindfulness. Participants in the current study were 1156 Chinese adolescents who completed self-report questionnaireson hope, attentional control, academic procrastination, and trait mindfulness. The results indicatedthat adolescent hope was negatively related to academic procrastination and that attentional control partiallymediated this association. In addition, trait mindfulness moderated the direct association between hope and academicprocrastination in the mediated model. 16A potential threat from biodegradable microplastics: mechanism of cadmium adsorption and desorption in the simulated gastrointestinal environment显示文摘It has been demonstrated that microplastics (MPs) can accumulate heavy metals from the environment and transfer them into organisms via the food chain. However, adsorption and desorption capacities for biodegradable MPs relative to those for conventional MPs remain poorly understood. In this study, cadmium (Cd(II)) adsorption and desorption characteristics of polylactic acid (PLA), a typical biodegradable MP, were investigated. Two conventional MPs, i.e., polypropylene (PP) and polyamide (PA) were used for comparison. The maximum Cd(II) adsorption capacities of the MPs studied in the adsorption experiments decreased in the order PA (0.96 ± 0.07 mg/g) > PLA (0.64 ± 0.04 mg/g) > PP (0.22 ± 0.03 mg/g). The Pseudo-second-order kinetic model and Freundlich isothermal model described the Cd(II) adsorption behaviors of PLA MPs well. X-ray photoelectron spectroscopy and two-dimensional Fourier transform infrared correlation spectroscopy analysis indicated that oxygen functional groups were the major and preferential binding sites of PLA MPs, which contributed to their high Cd(II) adsorption capacities. Simulated gastric and intestinal fluids both significantly enhanced the desorption capacities of the examined MPs. Notably, degradation of the PLA MPs during in vitro human digestion made the Cd(II) on the PLA MPs more bioaccessible (19% in the gastric phase and 62% in the intestinal phase) than Cd(II) on the PP and PA MPs. 17Influences of the Fresh Air Volume on the Removal of Cough-Released Droplets in a Passenger Car of a High-Speed Train Using CFD显示文摘The spread and removal of pollution sources,namely,cough-released droplets in three different areas(front,middle,and rear areas)of a fully-loaded passenger car in a high-speed train under different fresh air flow volume were studied using computational fluid dynamics(CFD)method.In addition,the structure of indoor flow fields was also analysed.The results show that the large eddies are more stable and flow faster in the air supply under Mode 2(fresh air volume:2200m3/h)compared to Mode 1(fresh air volume:1100m3/h).By analysing the spreading process of droplets sprayed at different locations in the passenger car and with different particle sizes,the removal trends for droplets are found to be similar under the two air supply modes.However,when increasing the fresh air flow volume,the droplets in the middle and front areas of the passenger car are removed faster.When the droplets had dispersed for 60s,Mode 2 exhibited a removal rate approximately 1%–3%higher than Mode 1 for small and medium-sized droplets with diameters of 10 and 50μm.While those in the rear area,the situation is reversed,with Mode 1 slightly surpassing Mode 2 by 1%–3%.For large droplets with a diameter of 100μm,both modes achieved a removal rate of over 96%in all three regions at the 60 s.The results can provide guidance for air supply modes of passenger cars of high-speed trains,thus suppressing the spread of virus-carrying droplets and reducing the risk of viral infection among passengers.Jun Xu Kai Bi Yibin Lu TiantianWang Hang Zhang Zeyuan Zheng Fushan Shi Yaxin Zheng Xiaoying Li Jingping Yang 2024Computer Modeling in Engineering & Sciences2024,138,3:0
18Transcriptome analysis reveals tumor antigen and immune subtypes of melanoma显示文摘Purpose:To screen potential tumor antigens for melanoma vaccine development and identify different immune subtypes.Methods:Transcriptional data(HTSEQ-FPKM)and clinical information of a 472 Melanoma cohort GDC TCGA Melanoma(SKCM)were downloaded from the UCSC XENA website(http://gffzz797b50e9334d4ed4hcxupocunov6q6vvu.ffgz.tsg.suse.edu.cn/).Subsequently,transcriptome data and clinical information of 210 melanoma cohort GSE65904 were downloaded from Gene Expression Omnibus(GEO),a large global public database.All the transcriptome expression data matrices were log2 transformed for subsequent analysis.GEPIA,TIMER,and IMMPORT databases are also used for analysis.Cell function experiments were performed to validate the role of the IDO1 gene in melanoma cell line A375.Results:Our study provides potential tumor antigens for vaccine development in melanoma patients:GZMB,GBP4,CD79A,APOBEC3F,IDO1,JCHAIN,LAG3,PLA2G2D,XCL2.In addition,we divide melanoma patients into two immune subtypes that have significant differences in tumor immunity and may have different responses to vaccination.In view of the unclear role of IDO1 in melanoma,we selected IDO1 for cell assay validation.Cell function assay showed that IDO1 was significantly overexpressed in the melanoma A375 cell line.After IDO1 knockdown,the activity,invasion,migration and healing ability of A375 cell lines were significantly decreased.Conclusion:Our study could provide a reference for the development of vaccines for melanoma patients.JIAHENG XIE MENGMENG OU PAN YU DAN WU QIKAI TANG YUAN CAO JING HANG LU YIN TINGHONG XIANG MING WANG JINGPING SHI 2023Oncology Research2023,31,3:0
19Study on Evaluation Method of Teaching Quality Based on Cloud Model and D-S Theory显示文摘The Evaluation of teaching quality is a complicated project. It’s a typical problem of uncertainty. A design scheme of teaching quality evaluation method based on cloud model and D-S theory is proposed with the school teaching evaluation index. With the experiences and knowledge of experts, evaluation index was described by cloud model and judgment matrix of AHP was revised by D-S theory. The weight of evaluation indexes was determined reasonably and objectively.
