Engineering self-healing adhesive hydrogels with antioxidant properties for intrauterine adhesion prevention显示文摘Intrauterine adhesion (IUA) is the fibrosis within the uterine cavity. It is the second most common cause of female infertility, significantly affecting women’s physical and mental health. Current treatment strategies fail to provide a satisfactory therapeutic outcome for IUA patients, leaving an enormous challenge for reproductive science. A self-healing adhesive hydrogel with antioxidant properties will be highly helpful in IUA prevention. In this work, we prepare a series of self-healing hydrogels (P10G15, P10G20, and P10G25) with antioxidant and adhesive properties. Those hydrogels exhibit good self-healing properties and can adapt themselves to different structures. They possess good injectability and fit the shape of the human uterus. Moreover, the hydrogels exhibit good tissue adhesiveness, which is desirable for stable retention and therapeutic efficacy. The in vitro experiments using P10G20 show that the adhesive effectively scavenges ABTS+, DPPH, and hydroxyl radicals, rescuing cells from oxidative stress. In addition, P10G20 offers good hemocompatibility and in vitro and in vivo biocompatibility. Furthermore, P10G20 lowers down the in vivo oxidative stress and prevents IUA with less fibrotic tissue and better endometrial regeneration in the animal model. It can effectively downregulate fibrosis-related transforming growth factor beta 1 (TGF-β1) and vascular endothelial growth factor (VEGF). Altogether, these adhesives may be a good alternative for the clinical treatment of intrauterine adhesion. | Luyao Feng Liqun Wang Yao Ma Wanglin Duan Sergio Martin-Saldana Ye Zhu Xianpeng Zhang Bin Zhu Chaowei Li Shibo Hu Mingjie Bao Ting Wang Yuan Zhu Fei Yang Yazhong Bu | 2023 | Bioactive Materials2023,,9: | 3
Mechanical properties and frictional resistance of Al composites reinforced with Ti3C2Tx MXene显示文摘Two-dimensional(2D)Ti3C2Tx MXene is an attractive additive not only used in base oil due to its low friction coefficient, but also used in composites due to its high aspect ratio and rich surface functional groups. So far there has been intense research into polymer matrix composites reinforced with Ti3C2Tx,Here we report on the use of 2D Ti3C2Tx to enhance the mechanical and frictional properties of Al matrix composites.Ti3C2Tx/Al composites were designed and prepared by pre s sureless sintering followed by hot extrusion technique. The prepared composites exhibit a homogeneous distribution of Ti3C2Tx.The Vickers hardness and the tensile strength continuously increase with increasing Ti3C2Tx content.A hardness of 0.52 GPa and a tensile strength of 148 MPa were achieved in the 3 wt%Ti3C2Tx/Al composite. The frictional properties of pure Al and the Ti3C2Tx/Al composite were comparably studied under dry sliding.A low friction coefficient of 0.2,twice lower than that of pure Al,was achieved in the 3 wt%Ti3C2Tx/Al composite.Ti3C2Tx acting as a solid lubricant reduces the abrasive wear in the composite, improving the frictional properties of Al matrix composites. | Jie Hu Shibo Li Jing Zhang Qiuying Chang Wenbo Yu Yang Zhou | 2020 | Chinese Chemical Letters2020,31,4: | 3

A non-ACE2 competing human single-domain antibody confers broad neutralization against SARS-CoV-2 and circulating variants显示文摘The current COVID-19 pandemic has heavily burdened the global public health system and may keep simmering for years. The frequent emergence of immune escape variants have spurred the search for prophylactic vaccines and therapeutic antibodies that confer broad protection against SARS-CoV-2 variants. Here we show that the bivalency of an affinity maturated fully human singledomain antibody(n3113.1-Fc)exhibits exquisite neutralizing potency against SARS-CoV-2 pseudovirus, and confers effective prophylactic and therapeutic protection against authentic SARS-CoV-2 in the host cell receptor angiotensin-converting enzyme 2(ACE2)humanized mice. The crystal structure of n3113 in complex with the receptor-binding domain(RBD)of SARS-CoV-2,combined with the cryo-EM structures of n3113 and spike ecto-domain,reveals that n3113 binds to the side surface of up-state RBD with no competition with ACE2.The binding of n3113 to this novel epitope stabilizes spike in up-state conformations but inhibits SARS-CoV-2 S mediated membrane fusion, expanding our recognition of neutralization by antibodies against SARS-CoV-2.Binding assay and pseudovirus neutralization assay show no evasion of recently prevalent SARS-CoV-2 lineages, including Alpha(B.1.1.7),Beta(B.1.351),Gamma(P.1),and Delta(B.1.617.2)for n3113.1-Fc with Y58L mutation, demonstrating the potential of n3113.1-Fc(Y58L)as a promising candidate for clinical development to treat COVID-19. | Zhenlin Yang Yulu Wang Yujia Jin Yuanfei Zhu Yanling Wu Cheng Li Yu Kong Wenping Song Xiaolong Tian Wuqiang Zhan Ailing Huang Shanshan Zhou Shuai Xia Xiaoxu Tian Chao Peng Cuicui Chen Yibing Shi Gaowei Hu Shujuan Du Yuyan Wang Youhua Xie Shibo Jiang Lu Lu Lei Sun Yuanlin Song Tianlei Ying | 2021 | Signal Transduction and Targeted Therapy2021,6,12: | 1

Gaussian-Student's t mixture distribution PHD robust filtering algorithm based on variational Bayesian inference显示文摘Aiming at the problem of filtering precision degradation caused by the random outliers of process noise and measurement noise in multi-target tracking(MTT) system,a new Gaussian-Student’s t mixture distribution probability hypothesis density(PHD) robust filtering algorithm based on variational Bayesian inference(GST-vbPHD) is proposed.Firstly, since it can accurately describe the heavy-tailed characteristics of noise with outliers,Gaussian-Student’s t mixture distribution is employed to model process noise and measurement noise respectively. Then Bernoulli random variable is introduced to correct the likelihood distribution of the mixture probability, leading hierarchical Gaussian distribution constructed by the Gaussian-Student’s t mixture distribution suitable to model non-stationary noise. Finally, the approximate solutions including target weights, measurement noise covariance and state estimation error covariance are obtained according to variational Bayesian inference approach. The simulation results show that,in the heavy-tailed noise environment, the proposed algorithm leads to strong improvements over the traditional PHD filter and the Student’s t distribution PHD filter. | 胡振涛 YANG Linlin HU Yumei YANG Shibo | 2022 | High Technology Letters2022,28,2: | 0
Electrospinning of antibacterial and anti-inflammatory Ag@hesperidin core-shell nanoparticles into nanofibers used for promoting infected wound healing显示文摘Bacterial infection and excessive inflammation are still the main obstacles to wound repair. Thus, antibacterial and antiinflammation nanomaterials are always attracting for infected wound healing. In this work,ultra-uniform(-20 nm)and colloidally stable Ag nanoparticles(Ag-Hes NPs)with core-shell structure were prepared by using hesperidin as reducing and capping agent. The obtained Ag-Hes NPs present effective antibacterial properties on both Staphylococcus aureus and Escherichia coli.Ag-Hes NPs also got high 1,1-diphenyl-1-picrylhydrazyl scavenging capability of 69%.Under the package of polyvinyl alcohol and sodium alginate,Ag-Hes NPs were encapsulated into electro spun nanofibers to form hydrogel(Ag-Hes@H).This strategy provides a moisture environment which could enrich and release Ag-Hes NPs gradually.Cell experiments and animal wound healing investigation proved that Ag-Hes@H could promote the proliferation and migration of human umbilical vein endothelial cells and accelerate infected wound healing.Meanwhile,Ag-Hes@H significantly reduced the expression of inflammatory cytokines, including IL-6,MMP9 and TNF-α.Immunohistochemistry data further suggested that Ag-Hes@H accelerated wound closure by promoting collagen deposition and skin cell proliferation. The designed antibacterial and anti-inflammatory Ag-Hes@H has great potential for promoting infected wound healing. | Xiuli Ren Yanan Hu Linna Chang Shibo Xu Xifan Mei Zhenhua Chen | 2022 | Regenerative Biomaterials2022,9,1: | 0
Synergistic roles of platinum nanoparticles and sodium ions within beta zeolites in N-alkylation of amines with aromatic alcohols显示文摘N-alkylation of amines with alcohols is a promising green route for the synthesis of N-alkylamines, but the designation of highly active and stable heterogeneous catalysts remains one challenge. In this work, we initiated the application of zeolite-encapsulated noble metal nanoparticles(NPs) in the N-alkylation of amines with aromatic alcohols. Platinum(Pt) NPs were directly confined into the framework of BEA zeolite with tunable Si/Al molar ratios and Na^(+) contents. The champion catalyst afforded the extremely high turnover number(TON) of 6,176 and turnover frequency(TOF) of 3,390 h^(-1) plus stable recyclability and broad substrate compatibility. The Na ions modulated the electronic state of Pt NPs to reach a moderate Pt–H strength, which wellbalanced the two crucial steps of dehydrogenation and H-transfer and accounted for the high catalytic performance. The encapsulation of zeolites provided spatial satisfaction for the synergy effect above and enhanced the stabilization of Pt NPs. | Yue Wu Shibo Xi Cailing Chen Qizhi Hu Zhuo Xiong Jianyue Wang Yihu Dai Yu Han Shi Jiang Jun Wang Yu Zhou | 2023 | Science China Chemistry2023,66,9: | 0
Bioinformatic analysis and in vivo validation of angiogenesis related genes in inflammatory bowel disease显示文摘Angiogenesis plays a significant role in the occurrence and development of inflammatory bowel disease(IBD).The aim of this study is to explore potential angiogenesis related genes(ARGs)in IBD through bioinformatics analysis and in vivo experiments.Methods:GSE57945,GSE87466,and GSE36807 were obtained from the Gene Expression Omnibus database.GSE57945 was used as the training set, while GSE87466 and GSE36807 were used as the validation set.The key ARGs associated with IBD were identified using the least absolute shrinkage and selection operator(LASSO)and random forest methods. These identified ARGs were then utilized to construct a diagnostic model for IBD.The Single-Sample Genome Enrichment Analysis,Cibersort, and Xcell methods were used to evaluate the immune infiltration.Expression of amyloid beta precursor protein(APP)was verified in the IBD mouse model induced by dextran sulfate sodium using immunohistochemistry(IHC).Results:The receiver operating curve area of GSE57945 was 0.948.Two distinct clusters were identified using consensus clustering and non-negative matrix factorization clustering.Subsequent analyses revealed significant differences in immune levels and functional enrichment between the two clusters. The successful construction of the animal model for the IBD was evident by hematoxylin and eosin staining, while IHC results showed a high expression of APP in IBD and a low expression in normal tissues. Conclusion:Our findings provide new insights into the diagnosis of IBD by ARGs,and APP could be a potential novel biomarker for IBD and promising therapeutic targets. | ZEPENG DONG CHENYE ZHAO SHIBO HU KUI YANG JUNHUI YU XUEJUN SUN JIANBAO ZHENG | 2023 | BIOCELL2023,47,12: | 0

M2 macrophage-targeted iron oxide nanoparticles for magnetic resonance image-guided magnetic hyperthermia therapy显示文摘Tumor-associated macrophages(TAMs)play an important role in tumor development and progression. In particular,M2 TAMs can promote tumor growth by facilitating tumor progression and malignant behaviors.Selectively targeted elimination of M2 TAMs to inhibit tumor progression is of great significance for cancer treatment.Iron oxide nanoparticles based magnetic hyperthermia therapy(MHT)is a classical approach to destroy tumor tissue with deep penetration depth. In this study,we developed a typical M2 macrophage-targeted peptide(M2pep)functionalized superparamagnetic iron oxide nanoparticle(SPIO)for magnetic resonance imaging(MRI)-guided MHT in an orthotopic breast cancer mouse model. The obtained multifunctional SPIO-M2pep with a hydrodynamic diameter of 20 nm showed efficient targeting capability, high transverse relaxivity(149 mM^(-1) s^(-1))and satisfactory magnetic hyperthermia performance in vitro. In vivo studies demonstrated that the SPIO-M2pep based MRI can monitor the distribution of nanoparticles in tumor and indicate the suitable timing for MHT.The M2 macrophage-targeted MHT significantly reduced the tumor volume and the population of pro-tumoral M2 TAMs in tumor. In addition, the SPIO-M2pep based MHT can remodel the tumor immune microenvironment(TIME).The multifunctional SPIO-M2pep with M2 macrophage-targeting ability, high magnetic hyperthermia efficiency,MR imaging capability and effective role in remodeling the TIME hold great potential to improve clinical cancer therapy outcomes. | Wenshen Wang Fenfen Li Shibo Li Yi Hu Mengran Xu Yuanyuan Zhang Muhammad Imran Khan Shaozhen Wang Min Wu Weiping Ding Bensheng Qiu | 2021 | Journal of Materials Science & Technology2021,,22: | 0
