| 2 | Discriminated sgRNAs-Based SurroGate System Greatly Enhances the Screening Efficiency of Plant Base-Edited Cells显示文摘The development of CRISPR/Cas9-mediated base editing has made genomic modification more efficient. However, selection of genetically modified cells from millions of treated cells, especially plant cells, is still challenging. In this study, an efficient surrogate reporter system based on a defective hygromycin resistance gene was established in rice to enrich base-edited cells. After step-by-step optimization, the Discriminated sgRNAs-based SurroGate system (DisSUGs) was established by artificially differentiating the editing abilities of a wild-type single guide RNA (sgRNA) targeting the surrogate reporter gene and an enhanced sgRNA targeting endogenous sites. The DisSUGs enhanced the efficiency of screening base-edited cells by 3- to 5-fold for a PmCDA1-based cytosine-to-tyrosine base editor (PCBE), and 2.5- to 6.5-fold for an adenine base editor (ABE) at endogenous targets. These targets showed editing efficiencies of <25% in the conventional systems. The DisSUGs greatly enhanced the frequency of homozygous substitutions and expanded the activity window slightly for both a PCBE and an ABE. Analyses of the total number of single-nucleotide variants from whole-genome sequencing revealed that, compared with the no-enrichment PCBE strategy, the DisSUGs did not alter the frequency of genome-wide sgRNA-independent off-target mutations, but slightly increased the frequency of target-dependent off-target mutations. | 8 | Increases in Anthropogenic Heat Release from Energy Consumption Lead to More Frequent Extreme Heat Events in Urban Cities显示文摘With economic development and rapid urbanization,increases in Gross Domestic Product and population in fastgrowing cities since the turn of the 21st Century have led to increases in energy consumption.Anthropogenic heat flux released to the near-surface atmosphere has led to changes in urban thermal environments and severe extreme temperature events.To investigate the effects of energy consumption on urban extreme temperature events,including extreme heat and cold events,a dynamic representation scheme of anthropogenic heat release(AHR)was implemented in the Advanced Research version of the Weather Research and Forecasting(WRF)model,and AHR data were developed based on energy consumption and population density in a case study of Beijing,China.Two simulations during 1999−2017 were then conducted using the developed WRF model with 3-km resolution with and without the AHR scheme.It was shown that the mean temperature increased with the increase in AHR,and more frequent extreme heat events were produced,with an annual increase of 0.02−0.19 days,as well as less frequent extreme cold events,with an annual decrease of 0.26−0.56 days,based on seven extreme temperature indices in the city center.AHR increased the sensible heat flux and led to surface energy budget changes,strengthening the dynamic processes in the atmospheric boundary layer that reduce AHR heating efficiency more in summer than in winter.In addition,it was concluded that suitable energy management might help to mitigate the impact of extreme temperature events in different seasons. | Bin LIU Zhenghui XIE Peihua QIN Shuang LIU Ruichao LI Longhuan WANG Yan WANG Binghao JIA Si CHEN Jinbo XIE Chunxiang SHI | 2021 | Advances in Atmospheric Sciences2021,38,3: | 2 |
| 9 | Future Precipitation Extremes in China under Climate Change and Their Physical Quantification Based on a Regional Climate Model and CMIP5 Model Simulations显示文摘The atmospheric water holding capacity will increase with temperature according to Clausius-Clapeyron scaling and affects precipitation.The rates of change in future precipitation extremes are quantified with changes in surface air temperature.Precipitation extremes in China are determined for the 21st century in six simulations using a regional climate model,RegCM4,and 17 global climate models that participated in CMIP5.First,we assess the performance of the CMIP5 models and RCM runs in their simulation of extreme precipitation for the current period(RF:1982-2001).The CMIP5 models and RCM results can capture the spatial variations of precipitation extremes,as well as those based on observations:OBS and XPP.Precipitation extremes over four subregions in China are predicted to increase in the mid-future(MF:2039-58)and far-future(FF:2079-98)relative to those for the RF period based on both the CMIP5 ensemble mean and RCM ensemble mean.The secular trends in the extremes of the CMIP5 models are predicted to increase from 2008 to 2058,and the RCM results show higher interannual variability relative to that of the CMIP5 models.Then,we quantify the increasing rates of change in precipitation extremes in the MF and FF periods in the subregions of China with the changes in surface air temperature.Finally,based on the water vapor equation,changes in precipitation extremes in China for the MF and FF periods are found to correlate positively with changes in the atmospheric vertical wind multiplied by changes in surface specific humidity(significant at the p<0.1 level). | Peihua QIN Zhenghui XIE Jing ZOU Shuang LIU Si CHEN | 2021 | Advances in Atmospheric Sciences2021,38,3: | 2 |

| 15 | Atypical Whipple’s disease with special endoscopic manifestations:A case report显示文摘BACKGROUND Whipple’s disease is a rare systemic infection caused by Tropheryma whipplei.Most patients present with nonspecific symptoms,and routine laboratory and imaging examination results also lack specificity.The diagnosis often relies on invasive manipulation,pathological examination,and molecular techniques.These difficulties in diagnosing Whipple’s disease often result in misdiagnosis and inappropriate treatments.CASE SUMMARY This paper reports on the case of a 58-year-old male patient who complained of fatigue and decreased exercise capacity.The results of routine blood tests indicated hypochromic microcytic anemia.Results of gastroscopy and capsule endoscopy showed multiple polypoid bulges distributed in the duodenal and proximal jejunum.A diagnosis of small intestinal adenomatosis was initially considered;hence,the Whipple procedure,a pylorus-preserving pancreaticoduodenectomy,was performed.Pathological manifestations showed many periodic acid-Schiff-positive macrophages aggregated in the intestinal mucosa of the duodenum,upper jejunum,and surrounding lymph nodes.Based on comprehensive analysis of symptoms,laboratory findings,and pathological manifestations,the patient was finally diagnosed with Whipple’s disease.After receiving 1 mo of antibiotic treatment,the fatigue and anemia were significantly improved.CONCLUSION This case presented with atypical gastrointestinal manifestations and small intestinal polypoid bulges,which provided new insight on the diagnosis of Whipple’s disease. | Shuo Chen Yuan-Chen Zhou Shuang Si Hong-Yan Liu Qing-Rui Zhang Teng-Fei Yin Chu-Xi Xie Shu-KunYao Shi-Yu Du | 2022 | World Journal of Clinical Cases2022,10,35: | 1 |
| 16 | Research on a Composite Biomass Insulation Material with Geopolymers as Binders and Forestry Waste as Fillers显示文摘A composite biomass insulation material,which uses geopolymers as adhesives and forestry waste as fillers,was proposed and experimentally tested.The orthogonal experimental method was adopted to analyze the optimum theoretical oxide molar ratios and the mass ratio of mixing water to binder(m_(w2)/m__(B))for preparing geopolymers.The influences of curing regimes(including one-stage and two-stage curing methods)and m_(w2)/m_(B) ratios of the insulation materials on mechanical,thermal,and hydraulic performances were also studied by experiment.The results indicated that the optimum combination scheme of preparing geopolymers was molar ratio x_(SiO_(2))/x_(Na_(2)O)=3.3,x_(SiO_(2))/x_(Al_(2)O_(3))=3.2 and m_(w2)/m_(B)=0.5 with the highest mechanical strength of 34.21 MPa.Besides,the best curing conditions of the composite material were the curing temperatures of 85°C and 70°C under the two-stage curing regime,which could achieve the low heat conductivity of 0.123 and 0.125 W/(m·K),and the high mechanical strength of 1.70 MPa and 1.71 MPa,respectively.The optimum m_(w2)/m_(B)ratios of the biomass material were 0.5 to 0.55 with heat conductivity of 0.114 to 0.125 W/(m·K).This novel composite insulation material has satisfying physical performances,which is helpful for achieving building energy conservation. | LI Hongqiang ZOU Si LIU Tiantian WANG Shuang ZOU Jun ZHANG Xiaofeng LIU Lifang BAI Chengying ZHANG Guoqiang | 2022 | Journal of Thermal Science2022,31,2: | 1 |

The stoichiometry of the complexes and their formation constants have been estimated. The thermodynamic parameters for the formation of complexes were obtained.
