| 4 | Clinical study on treatment of chronic viral cholestatic hepatitis with Chishaodanpi decoction显示文摘OBJECTIVE: To observe the therapeutic effect of Chishaodanpi decoction(CSDPD) on chronic viral cholestatic hepatitis.METHODS: A total of 107 subjects with chronic viral cholestatic hepatitis were enrolled in our hospital from March 2007 to November 2012. Patients were randomly divided into treatment(54 cases)and control groups(53 cases). The control group was treated with potassium magnesium aspartate,diammonium glycyrrhizinate, glucurolactone, vitamin C, and lamivudine, once a day. The treatment group was treated with modified CSDPD, 100 m L a time, twice a day, in addition to the treatment given to the control group. The patients in both groups were treated for 8 weeks. The main symptoms and signs were recorded every day throughout the clinical trial. Before and after the trial,changes in liver function including total bilirubin(TBil), direct bilirubin(DBil), total bile acid(TBA),and the activities of alkaline phosphatase(ALP), alanine aminotransferase(ALT), aspartate aminotransferase(AST), and γ-glutamyl transferase(γ-GT),were all detected. Adverse reactions were also recorded.RESULTS: There were no differences in gender, age,disease duration, symptoms, signs, or laboratory findings between the two groups(P>0.05). After an8-week treatment, improvements in jaundice, weakness, poor appetite, abdominal distention, and skin itching were significantly better in the treatment group than in the control group(P<0.05). In the treatment group, 43 patients had a significant response to the treatment, seven patients had a response, and four patients had no response, with 21,12, and 20 patients in the control group, respectively. The total effective rate was 92.6% in the treatment group and 62.3% in the control group, which was a significant difference(P<0.05). The levels of TBil, DBil, TBA, ALP, ALT, AST, and γ-GT in both groups were significantly lower after treatment,and were significantly different between the two groups(P<0.05). A few patients in the treatment group had mild adverse effects such as increased bowel movement frequency and mild stomachache. | 11 | Scientific objectives of the Hot Universe Baryon Surveyor(HUBS) mission显示文摘The Hot Universe Baryon Surveyor(HUBS) is a proposed space-based X-ray telescope for detecting X-ray emissions from the hot gas content in our universe. With its unprecedented spatially-resolved high-resolution spectroscopy and large field of view,the HUBS mission will be uniquely qualified to measure the physical and chemical properties of the hot gas in the interstellar medium, the circumgalactic medium, the intergalactic medium, and the intracluster medium. These measurements will be valuable for two key scientific goals of HUBS, namely to unravel the AGN and stellar feedback physics that governs the formation and evolution of galaxies, and to probe the baryon budget and multi-phase states from galactic to cosmological scales. In addition to these two goals, the HUBS mission will also help us solve some problems in the fields of galaxy clusters, AGNs, difuse X-ray backgrounds, supernova remnants, and compact objects. | 14 | Glutamate drives local Ca^(2+)release'in cardiac pacemaker cells显示文摘The sinoatrial node(SAN)is the origin of the electrical signals for rhythmic heartbeats in mammals.The spontaneous fring of SAN pacemaker cells(SANPCS)triggers cardiac contraction.'Local Ca^(2+)release'(LCR),a unique cellular activity,acts as the'engine'of the spontaneous fring of SANPCs.However,the mechanism of LCR initiation remains unclear.Here,we report that endogenous glutamate drives LCRs in SANPCs.Using a glutamate sensor,we unraveled a tight correlation between glutamate accumulation and LCR occurrence,indicating a potential relationship between glutamate and LCRs.Intracellular application of glutamate significantly enhanced the LCRs in both intact and permeabilized SANPCs.Mechanitically,we revealed that mitochondrial excitatory amino acid transporter 1(EAAT 1)-dependent mitochondrial glutamate import promoted ROS generation,which in turn led to the oxidation of Ca^(2+)-handling proteins,ultimately resulting in enhanced LCRs.Importantly,EAAT1 depletion reduced both the spontaneous fring rates of isolated SANPCs and the heart rate in vitro and in vivo,suggesting the central role of EAAT1 as a glutamate transporter in the regulation of cardiac autonomic rhythm.In conclusion,our results indicate that glutamate serves as an LCR igniter in SANPCs,adding a potentially important element to the coupled clock theory that explains the origin of spontaneous firing.These findings shed new light on the future prevention and treatment of cardiac pacemaker cell-related arrhythmias. | Duanyang Xie Ke Xiong Xuling Su Guanghua Wang Qicheng Zou Luxin Wang Caihong Zhang Yuting Cao Beihua Shao Yixin Zhang Peidong Zhang Dandan Liang Yi Liu Yi-Han Chen | 2022 | Cell Research2022,32,9: | 0 |

| 17 | Transmission of Carbapenem Resistance Between Human and Animal NDM-Positive Escherichia coli Strains显示文摘Although carbapenem use is prohibited in animals in China,carbapenem-resistant Escherichia coli(CREC),especially New Delhi metallo-β-lactamase(NDM)-producing strains,are widely prevalent in foodproducing animals.At present,the impact of livestock-associated CREC strains on human populations at the national level is unknown.Here,we conduct a retrospective cross-sectional study to investigate the prevalence of CREC from clinical settings across 22 Chinese provinces or municipalities and analyze anthropogenic factors associated with their presence.We also ascertain the blaNDMand blaKPCabundance among pig and chicken farms and present a detailed genomic framework for CREC of animal and human origin.Overall,631/29799(2.1%)clinical Escherichia coli(E.coli)isolates were identified as CREC.Multivariable analysis revealed that being male,an age below 1,an age between 13 and 18,provinces with greater chicken production,and provinces with higher pig production were associated with higher odds of CREC infection.In general,73.8%(n=45/61)of pig farms and 62.2%(n=28/45)of chicken farms had a blaNDMabundance of 1×10^(-5)to 1×10^(-3)and 1×10^(-3)to 1×10^(-2),respectively.Among all the Chinese NDM-positive E.coli(n=463)available at the National Center for Biotechnology Information(NCBI),the genomic analysis revealed that blaNDM-5and Inc X3 were the predominant carbapenemase gene-plasmid combination,while a highly homogeneous relationship between NDM-positive isolates from humans and animals was demonstrated at the plasmid and core genome levels.All the findings suggest frequent CREC transmission between humans and animals,indicating that further discussions on the use of antibiotics in animals and humans are needed,both in China and across the globe. | Yingbo Shen Fupin Hu Yongqiang Wang Dandan Yin Lu Yang Yiqiang Chen Chunyan Xu Jiyun Li Junyao Jiang Xueyang Wang Yulin Fu Dongyan Shao Dejun Liu Tengfei Ma Chang Cai Zhangqi Shen Shaolin Wang Juan Li Rong Zhang Yuebin Ke Congming Wu Jianzhong Shen Timothy R.Walsh Yang Wang | 2022 | Engineering2022,8,8: | 0 |
