1Stenting for symptomatic intracranial arterial stenosis in China:1-year outcome of a multicentre registry study显示文摘background and purpose A multicentre prospective registry study of individually tailored stenting for a patient with symptomatic intracranial atherosclerotic stenosis(ICAS)combined with poor collaterals in China showed that the short-term safety and efficacy of stenting was acceptable.However,it remained uncertain whether the low event rate could be of a long term.We reported the 1-year outcome of this registry study to evaluate the long-term efficacy of individually tailored stenting for patients with severe symptomatic ICAS combined with poor collaterals.Methods Patients with symptomatic ICAS caused by 70%-99% stenosis located at the intracranial internal carotid,middle cerebral,intracranial vertebral or basilar arteries combined with poor collaterals were enrolled.Balloon-mounted stent or balloon plus self-expanding stent were selected based on the ease of vascular access and lesion morphology determined by the operators.The primary outcome was the rate of 30-day stroke,transient ischaemic attack and death,and 12-month ischaemic stroke within the same vascular territory,haemorrhagic stroke and vascular death after stenting.results From September 2013 to January 2015,300 patients(ages 58.3±9.78 years)were recruited.Among them,159 patients were treated with balloon-mounted stent and 141 with balloon plus self-expanding stent.During the 1-year follow-up,25 patients had a primary end point event.The probability of primary outcome at 1 year was 8.1%(95% CI 5.3% to 11.7%).In 76 patients with digital subtraction angiography follow-up,27.6%(21/76)had re-stenosis≥50% and 18.4%(14/76)had re-stenosis≥70%.No baseline characteristic was associated with the primary outcome.Conclusion The event rate remains low over 1 year of individually tailored stenting for patients with severe symptomatic ICAS combined with poor collaterals.Further randomised trial of comparing individually tailored stenting with best medical therapy is needed.Ning Ma Yong Zhang Jie Shuai Changchun Jiang Qiyi Zhu Kangning Chen Li Liu Baomin Li Xiangqun Shi Lianbo Gao Yajie Liu Feng Wang Yongli Li Tieyan Liu Hongbo Zheng Dapeng Mo Feng Gao Yilong Wang Yongjun Wang Lei Feng Zhongrong Miao 2018Stroke & Vascular Neurology2018,3,3:29

3Influence of Weave Structures on the Tribological Properties of Hybrid Kevlar/PTFE Fabric Composites显示文摘The existing research of the woven fabric self-lubricating liner mainly focus on the tribological performance improvements and the service life raised by changing different fiber type combinations, adding additive modification, and performing fiber surface modification. As fabric composites, the weave structures play an important role in the mechanical and tribological performances of the liners. However, hardly any literature is available on the friction and wear behavior of such composites with different weave structures. In this paper, three weave structures (plain, twill 1/3 and satin 8/5) of hybrid Kevlar/PTFE fabric composites are selected and pin-on-flat linear reciprocating wear studies are done on a CETR tester under different pressures and different frequencies. The relationship between the tensile strength and the wear performance are studied. The morphologies of the worn surfaces under the typical test conditions are analyzed by means of scanning electron microscopy (SEM). The analysis results show that at 10 MPa, satin 8/5 performs the best in friction-reduction and antiwear performance, and plain is the worst. At 30 MPa, however, the antiwear performance is reversed and satin 8/5 does not even complete the 2 h wear test at 16 Hz. There is no clear evidence proving that the tensile strength has an influence on the wear performance. So the different tribological performance of the three weave structures of fabric composites may be attributed to the different PTFE proportions in the fabric surface and the different wear mechanisms. The fabric composites are divided into three regions: the lubrication region, the reinforced region and the bonding region. The major mechanisms are fatigue wear and the shear effects of the friction force in the lubrication region. In the reinforced region fiber-matrix de-bonding and fiber breakage are involved. 5Staged angioplasty versus regular carotid artery stenting in patients with carotid artery stenosis at high risk of hyperperfusion:a randomised clinical trial显示文摘Background and purpose Hyperperfusion(HP)is a devastating complication associated with carotid artery stenting(CAS)or endarterectomy.The efficacy and safety of staged angioplasty(SAP)in patients with CAS at high risk of HP remains unclear.We sought to determine whether SAP is superior to regular CAS in patients with high risk of HP.Methods A randomised,multicentre open-label clinical trial with blinded outcome assessment(STEP)was conducted.Patients with severe carotid stenosis at high risk of HP were randomly assigned(1:1)to the SAP or regular CAS group.The primary endpoint was hyperperfusion syndrome(HPS)and intracerebral haemorrhage(ICH)within 30 days after the procedure.Results From November 2014 to January 2017,a total of 64 patients were enrolled in 11 centres.33 patients were allocated to the SAP group and 31 to the regular CAS group.At 30 days,the rate of primary endpoint was 0.0%(0/33)in the SAP group and 9.7%(3/31)in the regular CAS group(absolute risk reduction(ARR),9.7%;95%CI−20.1% to 0.7%;p=0.11).As one of the secondary endpoints,the incidence of HP phenomenon(HPP)was lower in the SAP group than the regular CAS group(0.0%vs 22.6%,ARR,−22.6%;95% CI−36.8%to−10.2%;p=0.04).Conclusion The rate of HPS and ICH was not significantly lower in SAP group;the extended secondary endpoint of HPP,however,significantly reduced,which suggested that SAP may be a safe and effective carotid revascularisation procedure to prevent HP.Dapeng Mo Baixue Jia Huaizhang Shi Yaxuan Sun Qingan Liu Chengzhe Fan Jianping Deng Jinglin Yuan Wei Wu Changchun Jiang Guilian Zhang Hanjun Du Ning Ma Feng Gao Xuan Sun Ligang Song Lian Liu Guangge Peng Yongjun Wang Yilong Wang Zhongrong Miao the STEP Study Group 2021Stroke & Vascular Neurology2021,6,1:3
6Dietary fats suppress the peritoneal seeding of colorectal cancer cells through the TLR4/Cxcl10 axis in adipose tissue macrophages显示文摘Peritoneal carcinomatosis(PC)of colorectal cancer(CRC)is a terminal phase of malignancy with no effective strategies for the prevention of this condition.Here we established PC models in mice by intraperitoneal engraftment of CRC cells and revealed an unexpected role for a high-fat diet(HFD)in preventing metastatic seeding in the visceral fat.Mechanistically,the HFD stimulated the activation of adipose tissue macrophages(ATMs)toward an M1-like phenotype and enhanced ATM tumor phagocytosis in a TLR4-dependent manner.Furthermore,the TLR4–Cxcl10 axis in ATMs promoted T cell recruitment,and M1-like macrophages stimulated T cell activation in tumor-seeded fats.The inhibitory effect of the HFD on tumor seeding was abolished with the ablation of macrophages,inactivation of T cells,or blockade of the TLR4–Cxcl10 axis in macrophages.Finally,we showed that a HFD and conventional chemotherapeutic agents(oxaliplatin or 5-fluorouracil)synergistically improved the survival of tumor-seeded mice.Collectively,our findings demonstrate that peritoneal seeding of CRC can be suppressed by short-term treatment with a HFD in the early phase,providing a novel concept for the management of these patients in the clinic.Wei Xiang Rongchen Shi Dapeng Zhang Xia Kang Lili Zhang Jing Yuan Xuan Zhang Hongming Miao 2020Signal Transduction and Targeted Therapy2020,5,1:3
7The Agpat4/LPA axis in colorectal cancer cells regulates antitumor responses via p38/p65 signaling in macrophages显示文摘Lipid metabolic reprogramming plays an essential role in regulating the progression of colorectal cancer(CRC).However,the effect of lysophosphatidic acid(LPA)metabolism on CRC development is incompletely characterized.Here,we compared the mRNA levels of human CRC tissues to those of paracarcinoma tissues and focused on the notably enriched LPA metabolic pathways.We identified and verified that 1-acylglycerol-3-phosphate O-acyltransferase 4(Agpat4)was aberrantly expressed in CRC tissues and predicted poor survival in CRC patients.Manipulating Agpat4 expression in CRC cells did not affect the growth or migration of CRC cells in vitro,whereas Agpat4 silencing suppressed CRC cell growth in subcutaneous and peritoneal xenograft models.Mechanistically,Agpat4 silencing-induced LPA release from CRC cells and polarized macrophages to an M1-like phenotype through LPA receptors 1 and 3.This M1 activation,characterized by elevated p38/p65 signaling and increased proinflammatory cytokines,promoted the infiltration and activation of CD4+and CD8+T cells in the tumor microenvironment.Modulation of the Agpat4/LPA/p38/p65 axis regulated macrophage polarization,T-cell activity and CRC progression.Notably,combined therapy with LPA and regular chemotherapy drugs synergistically suppressed CRC development.Taken together,our results showed that the Agpat4/LPA axis in CRC cells regulated p38/p65 signaling-dependent macrophage polarization,T-cell activation,and CRC progression.The Agpat4/LPA/p38/p65 axis might represent a potential target for therapy in the clinic.Dapeng Zhang Rongchen Shi Wei Xiang Xia Kang Bo Tang Chuan Li Linfeng Gao Xuan Zhang Lili Zhang Rongyang Dai Hongming Miao 2020Signal Transduction and Targeted Therapy2020,5,1:3
8mTOR regulates TLR-induced c-fos and Th1 responses to HBV and HCV vaccines显示文摘Although IL-12 plays a critical role in priming Th1 and cytotoxic T lymphocyte(CTL) responses, Toll-like receptor(TLR) signaling only induces low amounts of IL-12 in dendritic cells and macrophages, implying the existence of stringent regulatory mechanisms. In this study, we sought to uncover the mechanisms underlying TLR-induced IL-12 expression and the Th1 response. By systemic screening, we identified a number of protein kinases involved in the regulation of TLRinduced IL-12 expression. In particular, PI3 K, ERK, and m TOR play critical roles in the TLR-induced Th1 response by regulating IL-12 and IL-10 production in innate immune cells. Moreover, we identified c-fos as a key molecule that mediates m TOR-regulated IL-12 and IL-10 expression in TLR signaling. Mechanistically, m TOR plays a crucial role in c-fos expression, thereby modulating NFκB binding to promoters of IL-12 and IL-10. By controlling the expression of a special innate gene program, m TOR can specifically regulate the TLR-induced T cell response in vivo. Furthermore, blockade of m TOR by rapamycin efficiently boosted TLR-induced antigen-specific T and B cell responses to HBV and HCV vaccines. 10Development of a real time PCR assay for rapid detection of Vibrio parahaemolyticus from seafood显示文摘A real time PCR assay for the detection of Vibrio para-haemolyticus in seafood samples was developed using a novel specific target and a competitive internal ampli-fication control(IAC).The specificity of this assay was evaluated using 390 bacterial strains including V.parahaemolyticus,and other strains belonging to Vibrio and non-Vibrio species.The real time PCR assay un-ambiguously distinguished V.parahaemolyticus with a detection sensitivity of 4.8 fg per PCR with purified genomic DNA or 1 CFU per reaction by counting V.parahaemolyticus colonies.The assays of avoiding interference demonstrated that,even in the presence of 2.1μg genomic DNA or 107 CFU background bacteria,V.parahaemolyticus could still be accurately detected.In addition,the IAC was used to indicate false-negative results,and lower than 94 copies of IAC per reaction had no influence on the detection limit.Ninety-six sea-food samples were tested,of which 58(60.4%)were positive,including 3 false negative results.Conse-quently,the real time PCR assay is effective for the rapid detection of V.parahaemotyticus contaminants in seafood.Bin Liu Xiaohua He Wanyi Chen Shuijing Yu Chunlei Shi Xiujuan Zhou Jing Chen Dapeng Wang Xianming Shi 2012Protein & Cell2012,3,3:2
11Self-assembled core-shell structured organic nanofibers fabricated by single-nozzle electrospinning for highly sensitive ammonia sensors显示文摘Electrospinning is a unique method to prepare 1-dimentional nanofiber for largescalemanufacturing. Here in, we demonstrated chemical sensors based on thesemiconducting nanofibers with core-shell structure by simple single-nozzleelectrospinning with the spontaneous phase separation. The core-shell structurenanofiber has large active sites which make it highly sensitive to chemical analytes.The thickness of the sensing shell can be tuned by controlling the mass ratio ofcore and shell components. As a demonstration, the nanofiber-based sensorexhibits high sensitivity to low-concentration ammonia (ppb level), as well as stabilityand reversibility. 12Study on Fishery Ecological Environment and Fish Species Diversity in Yantan Water Area显示文摘Through analysis on water quality,planktonic organism,fish resources,and fish species diversity in Yantan water area,this paper evaluated current situation of quality of fishery ecological environment in Yantan water area. The survey recorded all 52365 fishes and 1410. 2kg catches obtained by fishermen in half a year,and more than 98% catches are small fishes. The Shannon-Weiner diversity index of Yantan water area is 0. 162,Wilhm improvement index is 1. 814,DG- Findex is 0. 083,and the index of fish species diversity is far lower than other water areas. The average quantity per unit of phytoplankton is 1. 0134 million ind. / L,and the average quantity per unit of organisms is 1. 1151 mg / L. The average quantity per unit of zooplankton is 459. 6 ind. / L,and the average quantity per unit of organisms is 0. 6422 mg / L. Evaluation results indicate that water quality and planktonic organism in Yantan water area are basically normal,but fish resources are increasingly exhausted,fish resource composition is not reasonable,and fish species diversity is extremely low. 16Engineering of a NIR-activable hydrogel-coated mesoporous bioactive glass scaffold with dual-mode parathyroid hormone derivative release property for angiogenesis and bone regeneration显示文摘Osteogenesis,osteoclastogenesis,and angiogenesis play crucial roles in bone regeneration.Parathyroid hormone(PTH),an FDA-approved drug with pro-osteogenic,pro-osteoclastogenic and proangiogenic capabilities,has been employed for clinical osteoporosis treatment through systemic intermittent administration.However,the successful application of PTH for local bone defect repair generally requires the incorporation and delivery by appropriate carriers.Though several scaffolds have been developed to deliver PTH,they suffer from the weaknesses such as uncontrollable PTH release,insufficient porous structure and low mechanical strength.Herein,a novel kind of NIR-activable scaffold(CBP/MBGS/PTHrP-2)with dual-mode PTHrP-2(a PTH derivative)release capability is developed to synergistically promote osteogenesis and angiogenesis for high-efficacy bone regeneration,which is fabricated by integrating the PTHrP-2-loaded hierarchically mesoporous bioactive glass(MBG)into the N-hydroxymethylacrylamide-modified,photothermal agent-doped,poly(N-isopropylacrylamide)-based thermosensitive hydrogels through assembly process.Upon on/off NIR irradiation,the thermoresponsive hydrogel gating undergoes a reversible phase transition to allow the precise control of on-demand pulsatile and long-term slow release of PTHrP-2 from MBG mesopores.Such NIR-activated dual-mode delivery of PTHrP-2 by this scaffold enables a well-maintained PTHrP-2 concentration at the bone defect sites to continually stimulate vascularization and promote osteoblasts to facilitate and accelerate bone remodeling.In vivo experiments confirm the significant improvement of bone reparative effect on critical-size femoral defects of rats.This work paves an avenue for the development of novel dual-mode delivery systems for effective bone regeneration.Shi Liu Zhengzhe Han Ji-Na Hao Dapeng Zhang Xianglong Li Yuanyuan Cao Jinghuan Huang Yongsheng Li 2023Bioactive Materials2023,,8:1

18Does a large group of pedestrians follow the evacuation signs?An experimental study显示文摘Guidance signs are generally considered to be key in improving the efficiency of emergency building evacuations.During a group evacuation,how obedient is the group to the evacuation signs?Would different evacuation signs affect the judgment of evacuated groups?Evacuation experiments were conducted to explore the influence of evacuation signs on the path selection of evacuation groups.Fourteen classes with 514 participants participated in these experiments,and each class participated in one experiment.Six classes conducted the experiments without guidance signs,and the rest conducted the experiments with guidance signs.The influences of the sign’s color(red and green),pattern(arrow and running man),and status(whether it flashes)on the route selection were studied.The results show that(1)the evacuation signs significantly affect the route choices of the evacuated groups.(2)The herding effect during the evacuation process does not influence the guiding effect of evacuation signs.(3)Evacuation signs of different colors and patterns have different guiding effects,and the Green running man sign has the best effect.(4)The constant bright,or flashing evacuation sign corresponding to the flashing Red running man has a significant impact on the route selection of the evacuated group.The results may provide a reference for improving the emergency evacuation guidance model of office buildings or other buildings with a large flow of people and optimize ideas for the design and layout of emergency evacuation signs.Jiguang Shi Dapeng Dong Ning Ding Chang Sun Zhenyu Fan 2022Journal of Safety Science and Resilience2022,3,4:1
