| 1 | Chromosome-scale genome assembly provides insights into the evolution and flavor synthesis of passion fruit (Passiflora edulis Sims)显示文摘Passion fruit(Passiflora edulis Sims)is an economically valuable fruit that is cultivated in tropical and subtropical regions of the world.Here,we report an~1341.7Mb chromosome-scale genome assembly of passion fruit,with 98.91%(~1327.18Mb)of the assembly assigned to nine pseudochromosomes.The genome includes 23,171 protein-coding genes,and most of the assembled sequences are repetitive sequences,with long-terminal repeats(LTRs)being the most abundant.Phylogenetic analysis revealed that passion fruit diverged after Brassicaceae and before Euphorbiaceae.Ks analysis showed that two whole-genome duplication events occurred in passion fruit at 65 MYA and 12 MYA,which may have contributed to its large genome size.An integrated analysis of genomic,transcriptomic,and metabolomic data showed that‘alpha-linolenic acid metabolism’,‘metabolic pathways’,and‘secondary metabolic pathways’were the main pathways involved in the synthesis of important volatile organic compounds(VOCs)in passion fruit,and this analysis identified some candidate genes,including GDP-fucose Transporter 1-like,Tetratricopeptide repeat protein 33,protein NETWORKED 4B isoform X1,and Golgin Subfamily A member 6-like protein 22.In addition,we identified 13 important gene families in fatty acid pathways and eight important gene families in terpene pathways.Gene family analysis showed that the ACX,ADH,ALDH,and HPL gene families,especially ACX13/14/15/20,ADH13/26/33,ALDH1/4/21,and HPL4/6,were the key genes for ester synthesis,while the TPS gene family,especially PeTPS2/3/4/24,was the key gene family for terpene synthesis.This work provides insights into genome evolution and flavor trait biology and offers valuable resources for the improved cultivation of passion fruit. | Zhiqiang Xia Dongmei Huang Shengkui Zhang Wenquan Wang Funing Ma Bin Wu Yi Xu Bingqiang Xu Di Chen Meiling Zou Huanyu Xu Xincheng Zhou Rulin Zhan Shun Song | 2021 | Horticulture Research2021,8,1: | 15 |

| 3 | Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. | 5 | A Discussion on the Origin of Shale Reservoir Inter-Laminar Fractures in the Shahejie Formation of Paleogene, Dongying Depression显示文摘This paper was conducted on the shale reservoir in the Dongying depression in the Shahejie Formation of Paleogene. Based on detailed core description, rock thin sections, argon ion polishing, scanning electron microscopy(SEM) analysis, CT scanning and carbon-oxygen isotopic test, the formation mechanism of the inter-laminar fractures(or seams) and their development conditions were discussed by comprehensive analysis of the diagenetic evolution features and the reservoir space evolution characteristics. The results show that the laminated composite pattern in Dongying depression was dominated by vertical distribution of laminated calcite and clay minerals. Contrasted to the traditional understanding, development degree of inter-laminar fractures was much lower. Pure fluid between layers cannot exist without framework minerals, and the pre-existing well connected pore or horizontal overpressure micro seam was favorable paths of fluid migration. From the points of inter-laminar fractures origin mechanism and its evolution, the so called seams, were much more complex than traditional understanding that hydrocarbon generation raised the pore pressure and led to the parallel microfractures. But the inter-laminar fractures had little relationship with that whether the lamination was developed or bedding boundary was clear or not. In fact, the shale reservoir inter-laminar fractures were not rigorous fracture. They were controlled by pre-existing inter-granular pore(or seam) or dissolved pore, which distributed along carbonate laminae boundary and were connected by later super pressure, dissolution and the seepage force. And the development conditions were burial depth(bigger than 3 000 m) and the Ro value(>0.6%). Numerous fine parallel second phases in Mg–2Ce alloy are obtained, as well as a large number of dislocations around them, but few dislocations appear around the reticular second phase in the Mg–1Ce alloy. Among the three alloys, two internal friction peaks(P_1 and P_2) are detected at about 78 and 167?C in both the Mg–0.5Ce and Mg–1Ce alloys.In addition, the alloy with special parallel second phase structure exhibits excellent damping capacity in both strain amplitude and temperature-dependent regions. These results may be ascribed to the stress concentration and the formation of abundant parallel and uniform dislocation configurations in the ?-Mg matrix without the influence of crystal orientation.
