4Divergent Projection Patterns Revealed by Reconstruction of Individual Neurons in Orbitofrontal Cortex显示文摘The orbitofrontal cortex(OFC)is involved in diverse brain functions via its extensive projections to multiple target regions.There is a growing understanding of the overall outputs of the OFC at the population level,but reports of the projection patterns of individual OFC neurons across different cortical layers remain rare.Here,by combining neuronal sparse and bright labeling with a whole-brain florescence imaging system(fMOST),we obtained an uninterrupted three-dimensional whole-brain dataset and achieved the full morphological reconstruction of 25 OFC pyramidal neurons.We compared the wholebrain projection targets of these individual OFC neurons in different cortical layers as well as in the same cortical layer.We found cortical layer-dependent projections characterized by divergent patterns for information delivery.Our study not only provides a structural basis for understanding the principles of laminar organizations in the OFC,but also provides clues for future functional and behavioral studies on OFC pyramidal neurons.Junjun Wang Pei Sun Xiaohua Lv Sen Jin Anan Li Jianxia Kuang Ning Li Yadong Gang Rui Guo Shaoqun Zeng Fuqiang Xu Yu-Hui Zhang 2021Neuroscience Bulletin2021,37,4:5

6Insight into selective removal of copper from high-concentration nickel solutions with XPS and DFT:New technique to prepare 5N-nickel with chelating resin显示文摘An efficient and profitable separation process was proposed to prepare 5N(the purity of the metal solution reaches 99.999%) high-purity nickel from 3N nickel-solutions using Purolite S984. The adsorption performance of this superior resin, especially its selectivity for metal ions,was explored quantitatively. The maximum adsorption capacity for copper was 2.286 mmol/g calculated by the Langmuir model, which was twice as large as that for nickel. In the binary systems, the adsorption capacity for nickel was decreased by 45%, indicating direct competition for the active sites. The infinite separation factor for copper versus nickel exceeded 300, revealing the feasibility of preparing5N-level high-purity nickel solutions, which was further verified using the 800 BV(bed volume) effluent in the column dynamic process.According to the cost–benefit analysis, purification contributed to a profit of approximately 60,000 USD per cycle, and the investment return period was less than 1/3 years. Density functional theory analysis confirmed that four nitrogen atoms would be involved in the coordination complex and thus a structure involving two five-membered rings could be achieved. 10The protective effect of icariin on glucocorticoid-damaged BMECs explored by microfluidic organ chip显示文摘Osteonecrosis of the femoral head(ONFH)is a devastating musculoskeletal disease characterized by the impaired circulation of bone.The purpose of this study was to explore the underlying mechanisms of the protective effect of icariin on the glucocorticoid-induced injury of bone microvascular endothelial cells(BMECs).Normal BMECs were extracted from the femoral heads by enzymatic isolation and magneticactivated cell sorting methods.Dexamethasone and icariin were used to intervene BMECs in microfluidic organ chips,and phalloidin staining was conducted to observe the cell morphology and viability.Then next-generation transcriptome sequencing and real-time quantitative reverse transcription-polymerase chain reaction(q RT-PCR)were performed to identify the differentially expressed genes(DEGs)in different groups.Through the microfluidic organ chip,it can be observed that after dexamethasone intervention,the filamentous structure in cell fibers disappeared and the cell morphology changed from spindle to round until death.Icariin could relieve these changes and showed a protective effect on glucocorticoiddamaged BMECs.In addition,201 DEGs were detected between the icariin protection group and the dexamethasone group,which were significantly enriched in 17 signaling pathways.8 of the top ten selected hub genes(IL6,PTGS2,VEGFA,etc.)were confirmed by q RT-PCR.Transcription factors(TFs)-gene network showed 63 connections between 18 TFs and 12 DEGs.For instance,GATA2 could regulate 5 DEGs.The associations between 92 mi RNA and 12 DEGs were visualized in a mi RNA-gene network.The hub mi RNA,has-mir-335–5 p was predicted to interact with 8 DEGs(PTGS2,VEGFA,etc.).Microfluidic organ chips could provide excellent morphological results for cell experiments,by which it could be observed that icariin showed a protective effect on the glucocorticoid-induced injury of BMECs.Beside,these DEGs,possible regulatory TF(GATA2,FOXC1,etc.)and mi RNA(has-mir-335–5 p)might be dysregulated in the initiation of ONFH and have prospective importance in ONFH diagnosis and therapy.Tengqi Li Qingyu Zhang Fuqiang Gao Yadi Liu Wei Sun Yiyang Dong 2022Chinese Chemical Letters2022,33,6:1

14Review of Recent Progress in Robotic Knee Prosthesis Related Techniques:Structure,Actuation and Control显示文摘As the essential technology of human-robotics interactive wearable devices,the robotic knee prosthesis can provide above-knee amputations with functional knee compensations to realize their physical and psychological social regression.With the development of mechanical and mechatronic science and technology,the fully active knee prosthesis that can provide subjects with actuating torques has demonstrated a better wearing performance in slope walking and stair ascent when compared with the passive and the semi-active ones.Additionally,with intelligent human-robotics control strategies and algorithms,the wearing effect of the knee prosthesis has been greatly enhanced in terms of stance stability and swing mobility.Therefore,to help readers to obtain an overview of recent progress in robotic knee prosthesis,this paper systematically categorized knee prostheses according to their integrated functions and introduced related research in the past ten years(2010−2020)regarding(1)mechanical design,including uniaxial,four-bar,and multi-bar knee structures,(2)actuating technology,including rigid and elastic actuation,and(3)control method,including mode identification,motion prediction,and automatic control.Quantitative and qualitative analysis and comparison of robotic knee prosthesis-related techniques are conducted.The development trends are concluded as follows:(1)bionic and lightweight structures with better mechanical performance,(2)bionic elastic actuation with energy-saving effect,(3)artificial intelligence-based bionic prosthetic control.Besides,challenges and innovative insights of customized lightweight bionic knee joint structure,highly efficient compact bionic actuation,and personalized daily multi-mode gait adaptation are also discussed in-depth to facilitate the future development of the robotic knee prosthesis.Yuanxi Sun Hao Tang Yuntao Tang Jia Zheng Dianbiao Dong Xiaohong Chen Fuqiang Liu Long Bai Wenjie Ge Liming Xin Huayan Pu Yan Peng Jun Luo 2021Journal of Bionic Engineering2021,18,4:1

18Searching for an Optimized Single-objective Function Matching Multiple Objectives with Automatic Calibration of Hydrological Models显示文摘In the calibration of hydrological models, evaluation criteria are explicitly and quantitatively defined as single-or multi-objective functions when utilizing automatic calibration approaches.In most previous studies, there is a general opinion that no single-objective function can represent all important characteristics of even one specific hydrological variable(e.g., streamflow).Thus hydrologists must turn to multi-objective calibration.In this study, we demonstrated that an optimized single-objective function can compromise multi-response modes(i.e., multi-objective functions) of the hydrograph, which is defined as summation of a power function of the absolute error between observed and simulated streamflow with the exponent of power function optimized for specific watersheds.The new objective function was applied to 196 model parameter estimation experiment(MOPEX) watersheds across the eastern United States using the semi-distributed Xinanjiang hydrological model.The optimized exponent value for each watershed was obtained by targeting four popular objective functions focusing on peak flows, low flows, water balance, and flashiness, respectively.Results showed that the optimized single-objective function can achieve a better hydrograph simulation compared to the traditional single-objective function Nash-Sutcliffe efficiency coefficient for most watersheds, and balance high flow part and low flow part of the hydrograph without substantial differences compared to multi-objective calibration.The proposed optimal single-objective function can be practically adopted in the hydrological modeling if the optimal exponent value could be determined a priori according to hydrological/climatic/landscape characteristics in a specific watershed.TIAN Fuqiang HU Hongchang SUN Yu LI Hongyi LU Hui 2019Chinese Geographical Science2019,29,6:0
19Real-time analysis of inflammatory cytokines and regulatory gene expression in tissues surrounding the hematoma after intracerebral hemorrhage显示文摘BACKGROUND:Secondary lesions can occur in tissues surrounding the hematoma following intracerebral hemorrhage,with the presence of inflammatory reactions,cytokine expression and apoptosis. These have been confirmed in animal studies. Our study sought to determine whether these could be detected in human tissues surrounding the hematoma following intracerebral hemorrhage. OBJECTIVE: To investigate expression of inflammatory cytokines,Bax and Bcl-x,and identify neural cell apoptosis in tissues surrounding the hematoma,and to analyze the correlation between them and pathological damage in intracerebral hemorrhage patients. DESIGN,TIME AND SETTING: This histopathology,controlled study was performed at the Department of Neurosurgery,Sichuan People’s Hospital,China,from January 2003 to January 2005. PARTICIPANTS: Brain tissues 1 cm from the hematoma in 30 intracerebral hemorrhage patients served as the experimental group. Brain tissues located away from the hematoma in 7 patients served as the control group. METHODS: TUNEL was used to detect neural cell apoptosis. Immunohistochemistry (labeled dextran polymer) and RT-PCR were used to measure tumor necrosis factor-α,interleukin-1β,interleukin-6,Bax and Bcl-x protein and mRNA expression. Pathological changes in brain tissues surrounding the hematoma were observed following HE staining. MAIN OUTCOME MEASURES: Neural cell apoptosis,inflammatory cytokines,Bax and Bcl-x protein and mRNA expression,pathological changes in brain tissues surrounding the hematoma. RESULTS: Brain tissues surrounding the hematoma were mildly damaged within 6 hours,severely damaged at 24-72 hours,and significantly improved 1 week following intracerebral hemorrhage. Expression of tumor necrosis factor-α protein and mRNA,interleukin-1β and interleukin-6 mRNA was not significant in tissues surrounding the hematoma,which was identical to the control group within 6 hours after intracerebral hemorrhage. This expression was significantly higher compared with the control group from 12-72 hours,and gradually decreased after 72 hours. The number of apoptotic neural cells reached a peak between 12-72 hours. Tumor necrosis factor-α protein and mRNA,interleukin-1β and interleukin-6 mRNA levels were positively correlated with apoptosis,Bax protein and mRNA levels (P<0.01). CONCLUSION: Tumor necrosis factor-α,interleukin-1β,and interleukin-6 levels are highly correlated with apoptosis. 20Acceptance sampling plan of accelerated life testing for lognormal distribution under time-censoring显示文摘Lognormal distribution is commonly used in engineering.It is also a life distribution of important research values.For long-life products follow this distribution,it is necessary to apply accelerated testing techniques to product demonstration.This paper describes the development of accelerated life testing sampling plans(ALSPs)for lognormal distribution under time-censoring conditions.ALSPs take both producer and consumer risks into account,and they can be designed to work whether acceleration factor(AF)is known or unknown.When AF is known,life testing is assumed to be conducted under accelerated conditions with time-censoring.The producer and consumer risks are satisfied,and the size of test sample and the size of acceptance number are optimized.Then sensitivity analyses are conducted.When AF is unknown,two or more predetermined levels of accelerated stress are used.The sample sizes and sample proportion allocated to each stress level are optimized.The acceptance constant that satisfies producer and consumer risk is obtained by minimizing the generalized asymptotic variance of the test statistics.Finally,the properties of the two ALSPs(one for known-AF conditions and one for unknownAF conditions)are investigated to show that the proposed method is correct and usable through numerical examples.Li Xiaoyang Gao Pengfei Sun Fuqiang 2015Chinese Journal of Aeronautics2015,28,3:0
