2Emergence and prevalence of naturally occurring lower virulent African swine fever viruses in domestic pigs in China in 2020显示文摘African swine fever virus(ASFV)has been circulating in China for more than two years,and it is not clear whether the biological properties of the virus have changed.Here,we report on our surveillance of ASFVs in seven provinces of China,from June to December,2020.A total of 22 viruses were isolated and characterized as genotype II ASFVs,with mutations,deletions,insertions,or short-fragment replacement occurring in all isolates compared with Pig/HLJ/2018(HLJ/18),the earliest isolate in China.Eleven isolates had four different types of natural mutations or deletion in the EP402R gene and displayed a nonhemadsorbing(non-HAD)phenotype.Four isolates were tested for virulence in pigs;two were found to be as highly lethal as HLJ/18.However,two non-HAD isolates showed lower virulence but were highly transmissible;infection with 106 TCID50 dose was partially lethal and caused acute or sub-acute disease,whereas 103 TCID50 dose caused non-lethal,sub-acute or chronic disease,and persistent infection.The emergence of lower virulent natural mutants brings greater difficulty to the early diagnosis of ASF and creates new challenges for ASFV control.Encheng Sun Zhenjiang Zhang Zilong Wang Xijun He Xianfeng Zhang Lulu Wang Wenqing Wang Lianyu Huang Fei Xi Haoyue Huangfu Ghebremedhin Tsegay Hong Huo Jianhong Sun Zhijun Tian Wei Xia Xuewu Yu Fang Li Renqiang Liu Yuntao Guan Dongming Zhao Zhigao Bu 2021Science China(Life Sciences)2021,64,5:46
3Comparison of strapdown inertial navigation algorithm based on rotation vector and dual quaternion显示文摘For the navigation algorithm of the strapdown inertial navigation system, by comparing to the equations of the dual quaternion and quaternion, the superiority of the attitude algorithm based on dual quaternion over the ones based on rotation vector in accuracy is analyzed in the case of the rotation of navigation frame. By comparing the update algorithm of the gravitational velocity in dual quaternion solution with the compensation algorithm of the harmful acceleration in traditional velocity solution, the accuracy advantage of the gravitational velocity based on dual quaternion is addressed. In view of the idea of the attitude and velocity algorithm based on dual quaternion, an improved navigation algorithm is proposed, which is as much as the rotation vector algorithm in computational complexity. According to this method, the attitude quaternion does not require compensating as the navigation frame rotates. 4Chinmedomics facilitated quality-marker discovery of Sijunzi decoction to treat spleen qi deficiency syndrome显示文摘Sijunzi decoction (SJZD) is a Chinese classical formula to treat spleen qi deficiency syndrome (SQDS) and has been widely used for thousands of years.However,the quality control (QC) standards of SJZD are insufficient.Chinmedomics has been designed to discover and verify bioactive compounds of a variety of formularapidly.In this study,we used Chinmedomics to evaluate the SJZD's efficacy against SQDS to discover the potential quality-markers (q-markers) for QC.A total of 56 compounds in SJZD were characterized in vitro,and 23 compounds were discovered in vivo.A total of 58 biomarkers were related to SQDS,and SJZD can adjust a large proportion of marker metabolites to normal level and then regulate the metabolic profile to the health status.A total of 10 constituents were absorbed as effective ingredients that were associated with overall efficacy.We preliminarily determined malonyl-ginsenoside Rb2 and ginsenoside Ro as the q-markers of ginseng;dehydrotumulosic acid and dihydroxy lanostene-triene-21-acid as the q-markers of poria;glycyrrhizic acid,isoglabrolide,and glycyrrhetnic acid as the q-markers of licorice;and 2-atractylenolide as the q-marker of macrocephala.According to the discovery of the SJZD q-markers,we can establish the quality standard that is related to efficacy.Qiqi Zhao Xin Gao Guangli Yan Aihua Zhang Hui Sun Ying Han Wenxiu Li Liang Liu Xijun Wang 2020Frontiers of Medicine2020,14,3:7
5Microbial community functional structure in response to micro-aerobic conditions in sulfate-reducing sulfur-producing bioreactor显示文摘Limited oxygen supply to anaerobic wastewater treatment systems had been demonstrated as an effective strategy to improve elemental sulfur(S0) recovery, coupling sulfate reduction and sulfide oxidation. However, little is known about the impact of dissolved oxygen(DO) on the microbial functional structures in these systems. We used a high throughput tool(GeoChip) to evaluate the microbial community structures in a biological desulfurization reactor under micro-aerobic conditions(DO: 0.02–0.33 mg/L). The results indicated that the microbial community functional compositions and structures were dramatically altered with elevated DO levels. The abundances of dsrA/B genes involved in sulfate reduction processes significantly decreased(p < 0.05, LSD test) at relatively high DO concentration(DO: 0.33 mg/L). The abundances of sox and fccA/B genes involved in sulfur/sulfide oxidation processes significantly increased(p < 0.05, LSD test) in low DO concentration conditions(DO: 0.09 mg/L) and then gradually decreased with continuously elevated DO levels. Their abundances coincided with the change of sulfate removal efficiencies and elemental sulfur(S0) conversion efficiencies in the bioreactor. In addition, the abundance of carbon degradation genes increased with the raising of DO levels, showing that the heterotrophic microorganisms(e.g., fermentative microorganisms) were thriving under micro-aerobic condition. 6Traffic Characteristics Based Dynamic Radio Resource Management in Heterogeneous Wireless Networks显示文摘The traffic with tidal phenomenon in Heterogeneous Wireless Networks(HWNs)has radically increased the complexity of radio resource management and its performance analysis.In this paper,a Simplified Dynamic Hierarchy Resource Management(SDHRM)algorithm exploiting the resources dynamically and intelligently is proposed with the consideration of tidal traffic.In network-level resource allocation,the proposed algorithm first adopts wavelet neural network to forecast the traffic of each sub-area and then allocates the resources to those sub-areas to maximise the network utility.In connection-level network selection,based on the above resource allocation and the pre-defined QoS requirement,three typical network selection policies are provided to assign traffic flow to the most appropriate network.Furthermore,based on multidimensional Markov model,we analyse the performance of SDHRM in HWNs with heavy tailed traffic.Numerical results show that our theoretical values coincide with the simulation results and the SDHRM can improve the resource utilization.WEN Juan SHENG Min ZHANG Yan WANG Xijun LI Yuzhou 2014China Communications2014,11,1:4
7m6A RNA methylation-mediated HNF3γreduction renders hepatocellular carcinoma dedifferentiation and sorafenib resistance显示文摘Hepatocyte nuclear factor 3γ(HNF3γ)is a hepatocyte nuclear factor,but its role and clinical significance in hepatocellular carcinoma(HCC)remain unclear.Herein,we report that HNF3γexpression is downregulated in patient HCC and inversely correlated with HCC malignancy and patient survival.Moreover,our data suggested that the HNF3γreduction in HCC could be mediated by METTL14-dependent m6A methylation of HNF3γmRNA.HNF3γexpression was increased during hepatic differentiation and decreased in dedifferentiated HCC cells.Interestingly,HNF3γdelivery promoted differentiation of not only HCC cells but also liver CSCs,which led to suppression of HCC growth.Mechanistic analysis suggested an HNF3γ-centered regulatory network that includes essential liver differentiation-associated transcription factors and functional molecules,which could synergistically facilitate HCC cell differentiation.More importantly,enforced HNF3γexpression sensitized HCC cells to sorafenib-induced growth inhibition and cell apoptosis through transactivation of OATP1B1 and OATP1B3 expression,which are major membrane transporters for sorafenib uptake.Clinical investigation showed that patient-derived HCC xenografts with high HNF3γexpression exhibited a sorafenib response and patients with high HCC HNF3γlevels benefited from sorafenib therapy.Together,these results suggest that HNF3γplays an essential role in HCC differentiation and may serve as a therapeutic target and predictor of sorafenib benefit in patients.Tengfei Zhou Shichao Li Daimin Xiang Junyu Liu Wen Sun Xiuliang Cui Beifang Ning Xiao Li Zhuo Cheng Weiqi Jiang Cheng Zhang Xijun Liang Liang Li Xin Cheng Liu Hui Hongyang Wang Jin Ding 2020Signal Transduction and Targeted Therapy2020,5,1:4
8A simple model of radial nerve injury in the rhesus monkey to evaluate peripheral nerve repair显示文摘Current research on bone marrow stem cell transplantation and autologous or xenogenic nerve transplantation for peripheral nerve regeneration has mainly focused on the repair of peripheral nerve defects in rodents.In this study,we established a standardized experimental model of radial nerve defects in primates and evaluated the effect of repair on peripheral nerve injury.We repaired 2.5-cm lesions in the radial nerve of rhesus monkeys by transplantation of autografts,acellular allografts,or acellular allografts seeded with autologous bone marrow stem cells.Five months after surgery,regenerated nerve tissue was assessed for function,electrophysiology,and histomorphometry.Postoperative functional recovery was evaluated by the wrist-extension test.Compared with the simple autografts,the acellular allografts and allografts seeded with bone marrow stem cells facilitated remarkable recovery of the wrist-extension functions in the rhesus monkeys.This functional improvement was coupled with radial nerve distal axon growth,a higher percentage of neuron survival,increased nerve fiber density and diameter,increased myelin sheath thickness,and increased nerve conduction velocities and peak amplitudes of compound motor action potentials.Furthermore,the quality of nerve regeneration in the bone marrow stem cells-laden allografts group was comparable to that achieved with autografts.The wrist-extension test is a simple behavioral method for objective quantification of peripheral nerve regeneration.Dong Wang Xijun Huang Guo Fu Liqiang Gu Xiaolin Liu Honggang Wang Jun Hu Jianhua Yi Xiaofeng Niu Qingtang Zhu 2014Neural Regeneration Research2014,9,10:3
9Sulfonic acid-based metal organic framework functionalized magnetic nanocomposite combined with gas chromatography-electron capture detector for extraction and determination of organochlorine显示文摘The metal organic framework functionalized with sulfonic acid was combined with magnetic nanoparticles to fabricate a new nanocomposite(denoted as Fe3O4@PDA@Zr-SO3H).By combining with gas chromatography-electron capture detector,the resulting Fe3O4@PDA@Zr-SO3H nanocomposite was successfully used as a high-efficiency adsorbent for pre-concentrating eight organochlorine pesticides from water sample in environment.Apart from the ability of fast separation,the as-prepared Fe3O4@PDA@Zr-SO3H nanocomposite also exhibited high adsorption capacity for organochlorine pesticides.With the use of optimal experimental conditions,the linear relationship can be obtained in the range of 0.05~300μg/L,the correlation coefficient was over 0.9978,and the relative standard deviation was located in 2.5%-7.7%.Moreover,the limit of detection and quantification was between0.005-0.016μg/L and 0.017~0.050μg/L.Finally,the nanocomposite was used for the determination of organochlorine pesticides from environmental water samples,and displayed the recovery of 82%-118%.Ying Wang Qing Ye Menghuan Yu Xijun Zhang Chunhui Deng 2020Chinese Chemical Letters2020,31,7:3

15High-throughput metabolomics reveals the perturbed metabolic pathways and biomarkers of Yang Huang syndrome as potential targets for evaluating the therapeutic effects and mechanism of geniposide显示文摘High-throughput metabolomics can clarify the underlying molecular mechanism of diseases via the qualitative and quantitative analysis of metabolites.This study used the established Yang Huang syndrome(YHS)mouse model to evaluate the efficacy of geniposide(GEN).Urine metabolic data were quantified by ultraperformance liquid chromatography-tandem mass spectrometry.The non-target screening of the massive biological information dataset was performed,and a total of 33 metabolites,including tyramine glucuronide,aurine,and L-cysteine,were identified relating to YHS.These differential metabolites directly participated in the disturbance of phase I reaction and hydrophilic transformation of bilirubin.Interestingly,they were completely reversed by GEN.While,as the auxiliary technical means,we also focused on the molecular prediction and docking results in network pharmacological and integrated analysis part.We used integrated analysis to communicate the multiple results of metabolomics and network pharmacology.This study is the first to report that GEN indirectly regulates the metabolite“tyramine glucuronide”through its direct effect on the target heme oxygenase 1 in vivo.Meanwhile,heme oxygenase-1,a prediction of network pharmacology,was the confirmed metabolic enzyme of phase I reaction in hepatocytes.Our study indicated that the combination of high-throughput metabolomics and network pharmacology is a robust combination for deciphering the pathogenesis of the traditional Chinese medicine(TCM)syndrome.Heng Fang Aihua Zhang Xiaohang Zhou Jingbo Yu Qi Song Xijun Wang 2020Frontiers of Medicine2020,14,5:3
16Identification of New Resistance Loci Against Sheath Blight Disease in Rice Through Genome-Wide Association Study显示文摘Sheath blight(SB) caused by the soil borne pathogen Rhizoctonia solani is one of the most serious global rice diseases. Breeding resistant cultivar is the most economical and effective strategy to control the disease. However, no rice varieties are completely resistant to SB, and only a few reliable quantitative trait loci(QTLs) linked with SB resistance have been identified to date. In this study, we conducted a genome-wide association study(GWAS) of SB resistance using 299 varieties from the rice diversity panel 1(RDP1) that were genotyped using 44 000 high-density single nucleotide polymorphism(SNP) markers. Through artificial inoculation, we found that only 36.5% of the tested varieties displayed resistance or moderate resistance to SB. In particular, the aromatic and aus sub-populations displayed higher SB resistance than the tropical japonica(TRJ), indica and temperate japonica sub-populations. Seven varieties showed similar resistance levels to the resistant control YSBR1. GWAS identified at least 11 SNP loci significantly associated with SB resistance in the three independent trials, leading to the identification of two reliable QTLs, qSB-3 and qSB-6, on chromosomes 3 and 6. Using favorable alleles or haplotypes of significantly associated SNP loci, we estimated that both QTLs had obvious effects on reducing SB disease severity and can be used for enhancing SB resistance, especially in improving SB resistance of TRJ sub-population rice varieties. 17Novel P22-monoclonal antibody based blocking ELISA for the detection of African swine fever virus antibodies in serum显示文摘African swine fever(ASF)is a highly infectious,transboundary viral disease of domestic and wild pigs,and is currently the most serious threat to world swine production,resulting in significant economic loss.In the absence of vaccines and treatments,the control of the disease entirely depends on accurate and early diagnosis accompanied by the culling of infected pigs.Thus,a highly specific and sensitive diagnostic assay is required during an outbreak and surveillance of the disease.In this study,a highly sensitive,specific,rapid and repeatable P22-monoclonal antibody-based blocking enzyme-linked immunosorbent assay(bELISA)assay was developed for the detection of antibodies against genotype I and II African swine fever viruses(ASFVs).A total of 806 pig serum samples were tested to evaluate the performance of the diagnostic assay.To determine the PI(percent Inhibition)cut-off value,receiver-operating characteristic(ROC)analysis was applied.According to the ROC analysis of the data,98.10%specificity and 100%sensitivity were recorded when the threshold cut-off value of PI was established at 47%.In addition,the assay was able to detect ASFV antibodies as early as 9 days post-infection when serum samples from experimentally infected pigs were used.Taking all together,the results of the present study indicated that the P22-mAb based bELISA assay can be used for rapid and accurate detection of antibodies against ASFV,which could play a valuable role in the containment and prevention of ASFV as an alternative to other serological diagnostic methods.Also,this study will assist researchers to further investigate the immunogenic importance of P22 protein in ASFV infection.Ghebremedhin Tsegay Weldu Tesfagaber Yuanmao Zhu Xijun He Wan Wang Zhenjiang Zhang Encheng Sun Jinya Zhang Yuntao Guan Fang Li Renqiang Liu Zhigao Bu Dongming Zhao 2022Biosafety and Health2022,4,4:2
18Investigation of colloidal biogenic sulfur flocculation:Optimization using response surface analysis显示文摘The colloidal properties of biogenic elemental sulfur(S^0)cause solid–liquid separation problems,such as poor settling and membrane fouling.In this study,the separation of S^0 from bulk liquids was performed using flocculation.Polyaluminum chloride(PAC),polyacrylamide(PAM)and microbial flocculant(MBF)were compared to investigate their abilities to flocculate S^0 produced during the treatment of sulfate-containing wastewater.A novel approach with response surface methodology(RSM)was employed to evaluate the effects and interactions of flocculant dose,pH and stirring intensity,on the treatment efficiency in terms of the S^0 flocculation and the supernatant turbidity removal.The dose optimization results indicated that the S^0 flocculation efficiency decreased in the following order PAC>MBF>PAM.Optimum S^0 flocculation conditions were observed at pH 4.73,a stirring speed of 129 r/min and a flocculant dose of 2.42 mg PAC/mg S.During optimum flocculation conditions,the S^0f locculation rate reached 97.53%.Confirmation experiments demonstrated that employing PAC for S^0 flocculation is feasible and RSM is an efficient approach for optimizing the process of S^0 flocculation.The results provide basic parameters and conditions for recovering sulfur during the treatment of sulfate-laden wastewaters.Fan Chen Ye Yuan Chuan Chen Youkang Zhao Wenbo Tan Cong Huang Xijun Xu Aijie Wang 2016Journal of Environmental Sciences2016,28,4:2
19Compound Danshen Dripping Pill inhibits hypercholesterolemia/atherosclerosis-induced heart failure in ApoE and LDLR dual deficient mice via multiple mechanisms显示文摘Heart failure is the leading cause of death worldwide.Compound Danshen Dripping Pill(CDDP)or CDDP combined with simvastatin has been widely used to treat patients with myocardial infarction and other cardiovascular diseases in China.However,the effect of CDDP on hypercholesterolemia/atherosclerosis-induced heart failure is unknown.We constructed a new model of heart failure induced by hypercholesterolemia/atherosclerosis in apolipoprotein E(ApoE)and LDL receptor(LDLR)dual deficient(ApoE^(–/–)LDLR^(–/–))mice and investigated the effect of CDDP or CDDP plus a low dose of simvastatin on the heart failure.CDDP or CDDP plus a low dose of simvastatin inhibited heart injury by multiple actions including anti-myocardial dysfunction and anti-fibrosis.Mechanistically,both Wnt and lysine-specific demethylase 4A(KDM4A)pathways were significantly activated in mice with heart injury.Conversely,CDDP or CDDP plus a low dose of simvastatin inhibited Wnt pathway by markedly up-regulating expression of Wnt inhibitors.While the anti-inflammation and anti-oxidative stress by CDDP were achieved by inhibiting KDM4A expression and activity.In addition,CDDP attenuated simvastatin-induced myolysis in skeletal muscle.Taken together,our study suggests that CDDP or CDDP plus a low dose of simvastatin can be an effective therapy to reduce hypercholesterolemia/atherosclerosis-induced heart failure.Yanfang Yang Ke Feng Liying Yuan Yuxin Liu Mengying Zhang Kaimin Guo Zequn Yin Wenjia Wang Shuiping Zhou He Sun Kaijing Yan Xijun Yan Xuerui Wang Yajun Duan Yunhui Hu Jihong Han 2023Acta Pharmaceutica Sinica B2023,13,3:2
