| 1 | Disassembly of the fruit cell wall by the ripening-associated polygalacturonase and expansin influences tomato cracking显示文摘Fruit cracking is an important problem in horticultural crop production.Polygalacturonase(SlPG)and expansin(SlEXP1)proteins cooperatively disassemble the polysaccharide network of tomato fruit cell walls during ripening and thereby,enable softening.A Golden 2-like(GLK2)transcription factor,SlGLK2 regulates unripe fruit chloroplast development and results in elevated soluble solids and carotenoids in ripe fruit.To determine whether SlPG,SlEXP1,or SlGLK2 influence the rate of tomato fruit cracking,the incidence of fruit epidermal cracking was compared between wild-type,Ailsa Craig(WT)and fruit with suppressed SlPG and SlEXP1 expression(pg/exp)or expressing a truncated nonfunctional Slglk2(glk2).Treating plants with exogenous ABA increases xylemic flow into fruit.Our results showed that ABA treatment of tomato plants greatly increased cracking of fruit from WT and glk2 mutant,but not from pg/exp genotypes.The pg/exp fruit were firmer,had higher total soluble solids,denser cell walls and thicker cuticles than fruit of the other genotypes.Fruit from the ABA treated pg/exp fruit had cell walls with less water-soluble and more ionically and covalently-bound pectins than fruit from the other lines,demonstrating that ripening-related disassembly of the fruit cell wall,but not elimination of SlGLK2,influences cracking.Cracking incidence was significantly correlated with cell wall and wax thickness,and the content of cell wall protopectin and cellulose,but not with Ca^(2+)content. | Fangling Jiang Alfonso Lopez Shinjae Jeon Sergio Tonetto de Freitas Qinghui Yu Zhen Wu John M.Labavitch Shengke Tian Ann L.T.Powell Elizabeth Mitcham | 2019 | Horticulture Research2019,6,1: | 13 |
| 2 | Relative position determination of a lunar rover using the biased differential phase delay of same-beam VLBI显示文摘When only data transmission signals with a bandwidth of 1 MHz exist in the rover, the position can be obtained using the differential group delay data of the same-beam very long baseline interferometry (VLBI). The relative position between a lunar rover and a lander can be determined with an error of several hundreds of meters. When the guidance information of the rover is used to determine relative position, the rover's wheel skid behavior and integral movement may influence the accuracy of the determined position. This paper proposes a new method for accurately determining relative position. The differential group delay and biased differential phase delay are obtained from the same-beam VLBI observation, while the modified biased differential phase delay is obtained using the statistic mean value of the differential group delay and the biased phase delay as basis. The small bias in the modified biased phase delay is estimated together with other parameters when the relative position of the rover is calculated. The effectiveness of the proposed method is confirmed using the same-beam VLBI observation data of SELENE. The radio sources onboard the rover and the lander are designed for same-beam VLBI observations. The results of the simulations of the differential delay of the same-beam VLBI observation between the rover and the lander show that the differential delay is sensitive to relative position. An approach to solving the relative position and a strategy for tracking are also introduced. | 3 | Low-latency deep-reinforcement learning algorithm for ultrafast fiber lasers显示文摘The application of machine learning to the field of ultrafast photonics is becoming more and more extensive.In this paper,for the automatic mode-locked operation in a saturable absorber-based ultrafast fiber laser(UFL),a deep-reinforcement learning algorithm with low latency is proposed and implemented.The algorithm contains two actor neural networks providing strategies to modify the intracavity lasing polarization state and two critic neural networks evaluating the effect of the actor networks.With this algorithm,a stable fundamental modelocked(FML)state of the UFL is demonstrated.To guarantee its effectiveness and robustness,two experiments are put forward.As for effectiveness,one experiment verifies the performance of the trained network model by applying it to recover the mode-locked state with environmental vibrations,which mimics the condition that the UFL loses the mode-locked state quickly.As for robustness,the other experiment,at first,builds a database with UFL at different temperatures.It then trains the model and tests its performance.The results show that the average mode-locked recovery time of the trained network model is 1.948 s.As far as we know,it is 62.8%of the fastest average mode-locked recovery time in the existing work.At different temperatures,the trained network model can also recover the mode-locked state of the UFL in a short time.Remote algorithm training and automatic mode-locked control are proved in this work,laying the foundation for long-distance maintenance and centralized control of UFLs. | QIUQUAN YAN QINGHUI DENG JUN ZHANG YING ZHU KE YIN TENG LI DAN WU TIAN JIANG | 2021 | Photonics Research2021,9,8: | 7 |
| 4 | Protective Effect of Holothurian Intestine Against Indomethacin Induced Gastric Mucosal Damage in Rats显示文摘Our study aimed to investigate the protective effects of Holothurian intestines(HI) on NSAIDs-induced gastric mucosal damage and the possible mechanism. At first, 60 male Wistar rats were induced of gastric lesions with indomethacin(IDM, 30 mg kg^(-1)). The rats were pretreated for 15 consecutive days with saline, sucralfate, or HI(0.4g kg^(-1) d-1, 0.8 g kg^(-1) d^(-1) and 1.6 g kg^(-1) d^(-1)) prior to IDM treatment, followed by evaluations of macroscopic damage and microscopic features; and investigation of the levels of inflammatory cytokines, oxidative stress parameters, gastric mucosal prostaglandin E2(PGE2) and total hexosamine in tissues. The expression of COX-1 and COX-2 m RNA in the gastric tissue were determined by quantitative polymerase chain reaction(q PCR). Pathological gastric ulcer indexes, levels of pro-inflammatory cytokines(IL-1β, IL-17, TNF-α) and lipid peroxidation were significantly decreased in HI-treated groups, whereas the levels of protective factors(TGF-β, GSH, SOD activity and PGE2) were significantly elevated especially in the group with HI 1.61 g kg^(-1) d^(-1)(P < 0.05). Furthermore, the expression of COX-2 mRNA decreased significantly in HI groups(P < 0.05). | 5 | SNR periodical variation of Chang'E-3 spacecraft orbiting the Moon显示文摘Chang'E-3 spacecraft was orbiting the Moon from December 6–14, 2013, and very long baseline interferometry(VLBI) observations were performed to improve the accuracy of its orbit determination. In the process of recording VLBI raw data, 2 bits quantization was implemented.Interesting phenomenon was that signal-to-noise ratio(SNR) of each VLBI station experienced periodical change and had large variation on amplitude while in the Moon's orbit, whereas SNR kept in a stable level after Chang'E-3 landed on the Moon. Influence of varying elevation angle on SNR was analyzed and compensation of 2 bits quantization harmonics to SNR calculation was investigated. | 13 | Influences of Echinacea Polysaccharide on Secretion of IL-8mRNA by LPS-injured IEC-6 Cells显示文摘[Objective] The paper aimed to study the effects of Echinacea polysaccharide on secretion of IL-8m RNA by LPS-injured IEC-6 cells,in order to figure out the mechanism of EPS on injured cells. [Method]Total RNA was extracted with TRIzon reagent. IL-8 m RNA was amplified by RT-PCR and detected by agarose gel electrophoresis. Furthermore,electrophoresis and image was analyzed. [Result] The 50 μg/m L EPS could partially inhibit IL-8 m RNA level produced by LPS-stimulated IEC-6; with the increasing concentration of EPS(200 and 500 μg/m L),the inhibitory effect against IL-8 m RNA gradually enhanced. IEC-6 was pretreated by 50,100,200 and 500 μg/m L EPS for 24 h,then stimulated by 10 μg/m L LPS for 1 and 4 h,respectively. RT-PCR analysis showed that the expression of IL-8 m RNA induced by LPS was effectively inhibited by EPS; the inhibitory effect of EPS on expression of IL-8m RNA after stimulated by LPS for 4 h was more intense and the expression concentration was lower; the inhibitory rate at 4 h was higher than that at 1 h. | 15 | A pilot clinical study of developing an External Assist Targeting Device for rapid and precise renal calyx access during percutaneous nephrolithotomy显示文摘Objective:To design a device to increase the accuracy of the targeting process and reduce the radiation exposure to both the patients and the medical staff.Methods:We analyzed the inherent problem and designed the External Assist Targeting Device(EATD)to assist in the alignment of needle targeting on the desired renal calyx under fluoroscopic guidance.The EATD was designed to allow rapid and precise access to calyces at all angles,with a simple two-step puncture protocol developed for puncturing a target renal calyx.We then tested the device in a pilot human trial with four patients.Results:In experiments with phantom models,the time for successful targeting was reduced by 31%using the device.The mean fluoroscopic time was reduced by 40%.In initial human trial,the puncture time was shortened by 66%and the radiation dose was decreased by 65%compared to free-hand technique.No complication was observed during the trial.Conclusion:The EATD was found to be cost effective,portable,simple to set up,and safe to operate for assisting in the percutaneous nephrolithotomy procedures.Our preliminary tests showed high degree of accuracy in gaining precise access to a targeted renal calyx with much shorter time and lesser radiation dose.The EATD also has the potential to be used to access other organs with precision under fluoroscopic guidance. | Qinghui Wu Kesavan Esuvaranathan Teck Kheng Lee Soo Leong Foo Jian Ping Chai Edmund Chiong | 2023 | Asian Journal of Urology2023,10,3: | 0 |
| 16 | Clinical significance of serum IgM and IgG levels in COVID-19 patients in Hubei Province,China显示文摘Background:There have been many studies about coronavirus disease 2019(COVID-19),but the clinical signifi-cance of quantitative serum severe acute respiratory syndrome-coronavirus-2(SARS-CoV-2)-specific IgM and IgG levels of COVID-19 patients have not been exhaustively analyzed.We aimed to investigate the time profiles of these IgM/IgG levels in COVID-19 patients and their correlations with clinical features.Methods:A multicenter clinical study was conducted from February 20 to March 52020.It involved 179 COVID-19 patients(108 males and 71 females)from five hospitals in Huangshi in Hubei Province,China.To detect SARS-CoV-2-specific IgM/IgG,quantitative antibody assays(two-step indirect immunoassays with direct chemilu-minescence technology)based on the nucleocapsid protein(NP)and spike protein 1(S1)were used.For normally distributed data,means were compared using the t-test,��2-test,or exact probability method.For categorical data,medians were compared using Mann-Whitney U test.Results:The median age was 57(44-69)years(58[38-69]for males and 57[49-68]for females).The median duration of positive nucleic acid test was 22.32(17.34-27.43)days.The mortality rate was relatively low(3/179,1.68%).Serum SARS-CoV-2-specific IgG was detected around week 1 after illness onset,gradually increased until peaking in weeks 4 and 5,and then declined.Serum IgM peaked in weeks 2 and 3,then gradually declined and returned to its normal range by week 7 in all patients.Notably,children had milder respiratory symptoms with lower SARS-CoV-2-specific IgM/IgG levels.The duration of positive nucleic acid test in the chronic obstructive pulmonary disease(COPD)group was 30.36(18.99-34.76)days,which was significantly longer than that in the non-COPD group(21.52[16.75-26.51]days;P=0.025).The peak serum SARS-CoV-2-specific IgG was sig-nificantly positively correlated with the duration of positive nucleic acid test.The incidence rate of severe and critical cases in the IgM hi group(using the median IgM level of 29.95 AU/mL as the cutofffor grouping)was about 38.0%(19/50),which was twice as much as that in the IgM lo group(18.4%;9/49).The patients with positive chest imaging and lymphocytopenia(<1×10^(9)/L)had a higher SARS-CoV-2-specific IgM level.Conclusions: Quantitative SARS-CoV-2-specific IgM and IgG levels are helpful for the diagnosis, severity classifi- cation, and management of COVID-19 patients, and they should be monitored in each stage of this disease. | Zhenjiang Bai Qing Li Qinghui Chen Changming Niu Yu Wei Hanpeng Huang Wei Zhao Nian Chen Xin Yao Qiang Zhang Chuanyong Mu Jian Feng Chuanlong Zhu Zhuo Li Ming Ding Binhui Feng Chaochao Jin Xiang Lu Yi Yang Shuiyan Wu Xiaochen Shu Haibo Qiu YingZi Huang | 2022 | Journal of Intensive Medicine2022,2,1: | 0 |
| 17 | A novel lytic phage potentially effective for phage therapy against Burkholderia pseudomallei in the tropics显示文摘Background:Burkholderia pseudomallei is a tropical pathogen that causes melioidosis.Its intrinsic drug-resistance is a leading cause of treatment failure,and the few available antibiotics require prolonged use to be effective.This study aimed to assess the clinical potential of B.pseudomallei phages isolated from Hainan,China.Methods:Burkholderia pseudomallei strain(HNBPoo1)was used as the isolation host,and phages were recovered from domestic environmental sources,which were submitted to the host range determination,lytic property assays,and stability tests.The best candidate was examined via the transmission electron microscope for classification.With its genome sequenced and analyzed,its protective efficacy against B.pseudomallei infection in A549 cells and Caenorhabditis elegans was evaluated,in which cell viability and survival rates were compared using the one-way ANOVA method and the log-rank test.Results:A phage able to lyse 24/25 clinical isolates was recovered.It was classified in the Podoviridae family and was found to be amenable to propagation.Under the optimal multiplicity of infection(MOl)of o.1,an eclipse period of around 20 min and a high titer(io12 PFU/ml)produced within 1 h were demonstrated.This phage was found stabile at a wide range of temperatures(24,37,40,50,and 60 C)and pH values(3-12).After being designated as vB_BpP_HN01,it was fully sequenced,and the 71,398 bp linear genome,containing 93 open reading frames and a tRNA-Asn,displayed a low sequence similarity with known viruses.Additionally,protective effects of applications of vB_BpP_HNo1(MOl=0.1 and MOl=1)alone or in combination with antibiotics were found to improve viability of infected cells(70.6±6.8%,85.8±5.7%,91.9±1.8%,and 96.8±1.8%,respectively).A significantly reduced mortality(10%)and a decreased pathogen load were demonstrated in infected C.elegans following the addition of this phage.Conclusions:As the frst B.pseudomallei phage was isolated in Hainan,China,phage vB_BpP_HNO1 was characterized by promising lytic property,stability,and effciency of bacterial elimination during the in vitro/vivo experiments.Therefore,we can conclude that it is a potential alternative agent for combating melioidosis. | Yanshuang Wang Xuemiao Li David A.B.Dance Han Xia Chen Chen Nini Luo Anyang Li Yanmei Li Qiao Zhu Qinghui Sun Xingyong Wu Yingfei Zeng Lin Chen Shen Tian Qianfeng Xia | 2022 | Infectious Diseases of Poverty2022,11,4: | 0 |
| 18 | Isolation, Identification and Drug Susceptibility Test of a Strain of Escherichia coli Causing Fox Pneumonia显示文摘[Objective] The paper was to determine the pathogen causing fox pneumonia in a breeding factory in Changli County.[Method]Through autopsy, a dominant strain was isolated from the lung of dead foxes, which was then performed Gram staining, 16 S rRNA sequence analysis and biochemical identification.[Result] The strain was negative in Gram staining, and was identified as E. coli through 16 S rRNA sequence analysis and biochemical identification. Drug susceptibility test was conducted using 15 kinds of drug susceptibility papers. | 19 | Correlation Analysis of Drug Resistance and Resistant Genotypes of Pathogenic Klebsiella peneumoniae from Racoon Dog to Fluoroquinolones显示文摘[Objective] The paper was to analyze the correlation between drug resistance and resistant genotypes of pathogenic Klebsiella peneumoniae from racoon dog to fluoroquinolones.[Method] The drug resistance and resistant genes of 24 strains of pathogenic K. peneumoniae from racoon dog to fluoroquinolones were detected by K-B drug susceptibility paper and PCR.[Result] Multiple drug-resistant strains were dominant among 24 strains of pathogenic K. peneumoniae, and the drug resistance to levofloxacin, enrofloxacin, ciprofloxacin and norfloxacin was 70.8%-79.2%;the strains resistant to 4, 3 and 2 antibiotics were 58.3%, 29.2% and 8.3%, respectively. The detection rates of drug-resistant genes TEM, gyrB, parC,parE, gyrA and SHV were 75%-100%, and the number of strains carrying 9, 8, 7, 6, 4 and 3 drug-resistant genes was 4.2%-41.7%.
