2Microcystis aeruginosa/Pseudomonas pseudoalcaligenes interaction effects on off-flavors in algae/bacteria co-culture system under different temperatures显示文摘We conducted an experiment to study the interaction effects of Microcystis aeruginosa and Pseudomonas pseudoalcaligenes on off-flavors in an algae/bacteria co-culture system at three temperatures(24, 28 and 32℃). Gas chromatography–mass spectrometry was applied to measure off-flavor compounds dimethyl sulfide(DMS), dimethyl trisulfide(DMTS),2-methylisoborneol, geosmin(GEO) and β-cyclocitral. During the lag phase of co-cultured M. aeruginosa(first 15 days), P. pseudoalcaligenes significantly increased the production of DMS, DMTS and β-cyclocitral at all three temperatures. In the exponential phase of co-cultured M. aeruginosa(after 15 days), M. aeruginosa became the main factor on off-flavors in the co-culture system, and β-cyclocitral turned to the highest off-flavor compound. These results also indicated that DMS, DMTS and β-cyclocitral were the main off-flavor compounds in our M. aeruginosa/P. pseudoalcaligenes co-culture system. Univariate analysis was applied to investigate the effects of M. aeruginosa and P. pseudoalcaligenes on the production of off-flavors. The results demonstrated that both M. aeruginosa and P. pseudoalcaligenes could increase the production of DMS and DMTS, while β-cyclocitral was mainly determined by M. aeruginosa. 3Identification and characterization of genes involved in the jasmonate biosynthetic and signaling pathways in mulberry(Morus notabilis)显示文摘Jasmonate(JA) is an important phytohormone regulating growth, development, and environmental response in plants, particularly defense response against herbivorous insects. Recently, completion of the draft genome of the mulberry(Morus notabilis) in conjunction with genome sequencing of silkworm(Bombyx mori) provides an opportunity to study this unique plant‐herbivore interaction. Here, we identified genes involved in JA biosynthetic and signaling pathways in the genome of mulberry for the first time, with the majority of samples showing a tissue‐biased expression pattern. The analysis of the representative genes 12‐oxophytodienoic acid reductase(OPRs) and jasmonate ZIM‐domain(JAZs) was performed and the results indicated that the mulberry genome contains a relatively small number of JA biosynthetic and signaling pathway genes. A gene encoding animportant repressor, MnNINJA, was identified as an alternative splicing variant lacking an ethylene‐responsive element binding factor‐associated amphiphilic repression motif. 4Improving the Extreme Rainfall Forecast of Typhoon Morakot(2009) by Assimilating Radar Data from Taiwan Island and China's Mainland显示文摘This study examined the impact of an improved initial field through assimilating ground-based radar data from China's Mainland and Taiwan Island to simulate the long-lasting and extreme rainfall caused by Morakot(2009). The vortex location and the subsequent track analyzed through the radial velocity data assimilation(VDA) are generally consistent with the best track. The initial humidity within the radar detecting region and Morakot's northward translation speed can be significantly improved by the radar reflectivity data assimilation(ZDA). As a result, the heavy rainfall on both sides of Taiwan Strait can be reproduced with the joint application of VDA and ZDA. Based on sensitivity experiments, it was found that, without ZDA, the simulated storm underwent an unrealistic inward contraction after 12-h integration, due to underestimation of humidity in the global reanalysis, leading to underestimation of rainfall amount and coverage. Without the vortex relocation via VDA, the moister(drier) initial field with(without) ZDA will produce a more southward(northward) track, so that the rainfall location on both sides of Taiwan Strait will be affected. It was further found that the improvement in the humidity field of Morakot is mainly due to assimilation of high-value reflectivity(strong convection) observed by the radars in Taiwan Island, especially at Kenting station. 6Functional identification of lncRNAs in sweet cherry(Prunus avium)pollen tubes via transcriptome analysis using single-molecule long-read sequencing显示文摘Sweet cherry(Prunus avium)is a popular fruit with high nutritional value and excellent flavor.Although pollen plays an important role in the double fertilization and subsequent fruit production of this species,little is known about its pollen tube transcriptome.In this study,we identified 16,409 transcripts using single-molecule sequencing.After filtering 292 transposable elements,we conducted further analyses including mRNA classification,gene function prediction,alternative splicing(AS)analysis,and long noncoding RNA(lncRNA)identification to gain insight into the pollen transcriptome.The filtered transcripts could be matched with 3,438 coding region sequences from the sweet cherry genome.GO and KEGG analyses revealed complex biological processes during pollen tube elongation.A total of 2043 AS events were predicted,7 of which were identified in different organs,such as the leaf,pistil and pollen tube.Using BLASTnt and the Coding-Potential Assessment Tool(CPAT),we distinguished a total of 284 lncRNAs,among which 154 qualified as natural antisense transcripts(NATs).As the NATs could be the reverse complements of coding mRNA sequences,they might bind to coding sequences.Antisense transfection assays showed that the NATs could regulate the expression levels of their complementary sequences and even affect the growth conditions of pollen tubes.In summary,this research characterizes the transcripts of P.avium pollen and lays the foundation for elucidating the physiological and biochemical mechanisms underlying sexual reproduction in the male gametes of this species.Yang Li Chuanbao Wu Chunsheng Liu Jie Yu Xuwei Duan Wenqi Fan Jing Wang Xiaoming Zhang Guohua Yan Tianzhong Li Kaichun Zhang 2019Horticulture Research2019,6,1:1

12Bio-clickable mussel-inspired peptides improve titanium-based material osseointegration synergistically with immunopolarization-regulation显示文摘Upon the osteoporotic condition,sluggish osteogenesis,excessive bone resorption,and chronic inflammation make the osseointegration of bioinert titanium(Ti)implants with surrounding bone tissues difficult,often lead to prosthesis loosening,bone collapse,and implant failure.In this study,we firstly designed clickable mussel-inspired peptides(DOPA-N3)and grafted them onto the surfaces of Ti materials through robust catechol-TiO2 coordinative interactions.Then,two dibenzylcyclooctyne(DBCO)-capped bioactive peptides RGD and BMP-2 bioactive domain(BMP-2)were clicked onto the DOPA-N3-coated Ti material surfaces via bio-orthogonal reaction.We characterized the surface morphology and biocompatibility of the Ti substrates and optimized the osteogenic capacity of Ti surfaces through adjusting the ideal ratios of BMP-2/RGD at 3:1.In vitro,the dual-functionalized Ti substrates exhibited excellent promotion on adhesion and osteogenesis of mesenchymal stem cells(MSCs),and conspicuous immunopolarization-regulation to shift macrophages to alternative(M2)phenotypes and inhibit inflammation,as well as enhancement of osseointegration and mechanical stability in osteoporotic rats.In summary,our biomimetic surface modification strategy by bio-orthogonal reaction provided a convenient and feasible method to resolve the bioinertia and clinical complications of Ti-based implants,which was conducive to the long-term success of Ti implants,especially in the osteoporotic or inflammatory conditions.Jie Sun Yingkang Huang Huan Zhao Junjie Niu Xuwei Ling Can Zhu Lin Wang Huilin Yang Zhilu Yang Guoqing Pan Qin Shi 2022Bioactive Materials2022,7,3:0
13Rapid and sensitive detection of urinary KIM-1 using fully printed photonic crystal microarrays显示文摘Urinary kidney injury molecule 1(uKIM-1)serves as a reliable marker for the early diagnosis of acute kidney injury(AKI).The rapid and facile detection of changes in uKIM-1 is essential for early AKI diagnosis,ultimately improving the prognosis of patients.In this study,we developed a fully printed photonic crystal-integrated microarray with photonic crystal-enhanced fluorescence properties,which can detect uKIM-1 levels at the point-of-care.We confirmed its efficacy in the early diagnosis of AKI using clinical urine specimens.Direct quantitative detection of uKIM-1 was achieved within 10 min.The lowest limit of detection is 8.75 pg·mL^(-1) with an accuracy of 94.2%.The diagnostic efficacy was validated using 86 clinical urine samples,highlighting the high sensitivity and stability of the photonic crystal microarray.Consequently,a facile and reliable immunoassay was designed and prepared for the rapid quantitative detection of uKIM-1,which is crucial for the early identification and convenient detection of AKI in hospital or community settings.Rapid,convenient,cost-effective,and long-term monitoring of changes in uKIM-1 levels can assist clinicians in making timely adjustments to treatment regimens,preventing the transition from AKI to chronic kidney disease(CKD),improving the quality of life of patients with AKI,and reducing healthcare costs.It highlights the advantages of utilizing urine samples as a noninvasive and easily accessible medium for early detection and monitoring of kidney-related conditions.Yang Liu Xuwei He Zewei Lian Qian Guo Jimei Chi Xiaoxue Lin Liyue Zhang Zheng Liu Yingyuan Liu Meng Su Keyu Wang Qiangguo Ao Qingli Cheng 2024Nano Research2024,17,5:0
14Experimental investigation on the permeability of gap-graded soil due to horizontal suffusion considering boundary effect显示文摘The boundary condition is a crucial factor affecting the permeability variation due to suffusion.An experimental investigation on the permeability of gap-graded soil due to horizontal suffusion considering the boundary effect is conducted,where the hydraulic head difference(DH)varies,and the boundary includes non-loss and soil-loss conditions.Soil samples are filled into seven soil storerooms connected in turn.After evaluation,the variation in content of fine sand(ΔR_(f))and the hydraulic conductivity of soils in each storeroom(C_(i))are analyzed.In the non-loss test,the soil sample filling area is divided into runoff,transited,and accumulated areas according to the negative or positive ΔR_(f) values.ΔR_(f) increases from negative to positive along the seepage path,and Ci decreases from runoff area to transited area and then rebounds in accumulated area.In the soil-loss test,all soil sample filling areas belong to the runoff area,where the gentle-loss,strengthened-loss,and alleviated-loss parts are further divided.ΔR_(f) decreases from the gentle-loss part to the strengthened-loss part and then rebounds in the alleviated-loss part,and C_(i) increases and then decreases along the seepage path.The relationship between ΔR_(f) and Ci is different with the boundary condition.Ci exponentially decreases with ΔR_(f) in the non-loss test and increases with ΔR_(f) generally in the soil-loss test.Xuwei Wang Yeshuang Xu 2024Journal of Rock Mechanics and Geotechnical Engineering2024,16,3:0
15Detection and targeted control of regional stress field for coal burst prevention显示文摘As coal burst normally occurs in the area of high stress concentration,it is of significance to study the features of regional stress field in coal mines.Primitive stress field,mining-induced stress field and their coupling effect are investigated through the methods of theoretical analysis,field measurement,numerical simulation and wave velocity CT inversion,and the relationship among regional stress field,high-energy seismic events and coal bursts are analyzed.Investigation of the 3#mining district in Xingcun coal mine shows that:(1)Though stress concentration changes in the mining process,several special areas witnesses stress concentration in the whole mining process,such as the rise pillar area,several syncline axis areas and large fault areas;(2)Coal burst occurrence and high-energy seismic events have a close relationship with regional stress field.Coal bursts and high-energy seismic events tend to occur in areas of stable stress concentration,such as the rise area,several syncline axis areas and large fault areas.Targeted control of stress field for coal burst prevention is developed based on stress field detection.The process of targeted control of stress field for coal burst prevention is:detection of regional stress field based mainly on wave velocity CT,identification of stress concentration,implementation of destress measures to control the identified stress concentration.This method of stress field control was applied to LW3306 working face in Xingcun coal mine and coal burst hazards were effectively controlled in LW3306.Linming Dou Zhenyu Sun Xuwei Li Quan Liu Siyuan Gong Chao Wang 2023Geohazard Mechanics2023,1,1:0

18Piezo1-targeted aerosol inhalation nanoparticles for acute lung injury显示文摘Inhaled drug-containing nanocarriers have been well applied as an important strategy in pulmonary dis-eases.Acute lung injury(ALI)is characterized by abnormal lung tension and complex pathogenesis,with high mortality because of the limitations of targeted intervention.Accordingly,in the present study,we first identified that the mechanosensitive ion channel Piezo1 participated in the ALI-associated processes induced by lipopolysaccharide(LPS)in vitro and in vivo.Then,chitosan-stabilized bovine serum albu-min nanoparticles(NCs)emulsified with the Piezo1 inhibitor GsMTx4(NC-GsMTx4)were generated and exhibited excellent biocompatibility and biological function.Through aerosol inhalation,NC-GsMTx4 im-proved lung injury and inhibited cell apoptosis in LPS-stressed ALI mice and alleviated pulmonary fibrosis during the later stage of ALI.Mechanistically,GsMTx4 could regulate inflammation and apoptosis in lung epithelial cells via NF-κB and ERK1/2 signaling.In summary,our findings provide new insights into the pathological mechanisms of Piezo1 in ALI progression,and nebulized inhalation of NC-GsMTx4 offers a prospective platform for targeting Piezo1 to treat ALI efficiently and conveniently.Xinxin Liu Xuwei Ling Jiachen He Xingzhi Liu Lin Wang Huiling Liu Chang Liu Yi Wang Xiao Lin Chunhua Ling Qin Shi 2023Journal of Materials Science & Technology2023,,10:0
