1Runoff Responses to Climate Change in Arid Region of Northwestern China During 1960-2010显示文摘Based on runoff, air temperature, and precipitation data from 1960 to 2010, the effects of climate change on water resources in the arid region of the northwestern China were investigated. The long-term trends of hydroclimatic variables were studied by using both Mann-Kendall test and distributed-free cumulative sum (CUSUM) chart test. Results indicate that the mean annual air temperature increases significantly from 1960 to 2010. The annual precipitation exhibits an increasing trend, especially in the south slope of the Tianshan Mountains and the North Uygur Autonomous Region of Xinjiang in the study period. Step changes occur in 1988 in the mean annual air temperature time series and in 1991 in the precipitation time series. The runoff in different basins shows different trends, i.e., significantly increasing in the Kaidu River, the Aksu River and the Shule River, and decreasing in the Shiyang River. Correlation analysis reveals that the runoff in the North Xinjiang (i.e., the Weigan River, the Heihe River, and the Shiyang River) has a strong positive relationship with rainfall, while that in the south slope of the Tianshan Mountains, the middle section of the north slope of the Tianshan Mountains and the Shule River has a strong positive relationship with air temperature. The trends of runoff have strong negative correlations with glacier coverage and the proportion of glacier water in runoff. From the late 1980s, the climate has become warm and wet in the arid region of the northwestern China. The change in runoff is interacted with air temperature, precipitation and glacier coverage. 2Remote Sensing Monitoring of Tobacco Field Based on Phenological Characteristics and Time Series Image―A Case Study of Chengjiang County, Yunnan Province, China显示文摘Using three-phase remote sensing images of China-Brazil Earth Resources Satellite 02B (CBERS02B) and Landsat-5 TM, tobacco field was extracted by the analysis of time series image based on the different phenological characteristics between tobacco and other crops. The spectral characteristics of tobacco and corn in luxuriant growth stage are very similar, which makes them difficult to be distinguished using a single-phase remote sensing image. Field film after tobacco seedlings transplanting can be used as significant sign to identify tobacco. Remote sensing interpre- tation map based on the fusion image of TM and CBERS02B's High-Resolution (HR) camera image was used as stan- dard reference material to evaluate the classification accuracy of Spectral Angle Mapper (SAM) and Maximum Like- lihood Classifier (MLC) for time series image based on full samples test method. SAM has higher classification accu- racy and stability than MLC in dealing with time series image. 3Multiplex genome editing using a dCas9-cytidine deaminase fusion in Streptomyces显示文摘CRISPR/Cas-mediated genome editing has greatly facilitated the study of gene function in Streptomyces. However, it could not be efficiently employed in streptomycetes with low homologous recombination(HR) ability. Here, a deaminase-assisted base editor d Cas9-CDA-ULstr was developed in Streptomyces, which comprises the nuclease-deficient Cas9(dCas9), the cytidine deaminase from Petromyzon marinus(PmCDA1), the uracil DNA glycosylase inhibitor(UGI) and the protein degradation tag(LVA tag). Using d Cas9-CDA-ULstr , we achieved single-, double-and triple-point mutations(cytosine-to-thymine substitutions)at target sites in Streptomyces coelicolor with efficiency up to 100%, 60% and 20%, respectively. This base editor was also demonstrated to be highly efficient for base editing in the industrial strain, Streptomyces rapamycinicus, which produces the immunosuppressive agent rapamycin. Compared with base editors derived from the cytidine deaminase rAPOBEC1, the PmCDA1-assisted base editor dCas9-CDA-ULstr could edit cytosines preceded by guanosines with high efficiency, which is a great advantage for editing Streptomyces genomes(with high GC content). Collectively, the base editor dCas9-CDA-ULstr could be employed for efficient multiplex genome editing in Streptomyces. 15Nucleic acids analysis显示文摘Nucleic acids are natural biopolymers of nucleotides that store, encode, transmit and express genetic information, which play central roles in diverse cellular events and diseases in living things. The analysis of nucleic acids and nucleic acids-based analysis have been widely applied in biological studies, clinical diagnosis, environmental analysis, food safety and forensic analysis.During the past decades, the field of nucleic acids analysis has been rapidly advancing with many technological breakthroughs.In this review, we focus on the methods developed for analyzing nucleic acids, nucleic acids-based analysis, device for nucleic acids analysis, and applications of nucleic acids analysis. The representative strategies for the development of new nucleic acids analysis in this field are summarized, and key advantages and possible limitations are discussed.
