| 3 | Effect of a single oral dose of rabeprazole on nocturnal acid breakthrough and nocturnal alkaline amplitude显示文摘AIM: To study the effect of rabeprazole (RAB) on nocturnal acid breakthrough (NAB) and nocturnal alkaline amplitude (NAKA) and to compare it with omeprazole (OME) and pantoprazole (PAN).METHODS: By an open comparative study, forty patients with active peptic ulcer were randomly assigned to receive one of the three PPIs (proton pump inhibitor) with a single oral dose. They were divided into RAB group (10 mg), OME group(20 mg) and PAN group (40 mg). Twenty healthy volunteers were enrolled to the control group (without taking any drug). Intragastric pH monitoring was then performed 1 hour before and 24 hours after the dose was given.RESULTS: No clinically undesirable signs and symptoms possibly attributed to the administration of RAB or OME and PAN were recognizable throughout the study period. All subjects completed the study according to the protocol. All data were processed by a computer using the Student ttest or t' test followed by an analysis of covariance. P<0.05 was considered to have statistical significance. The intragastric pH of NAB was significantly higher in RAB group (1.84±0.55) than in either OME group (1.15±0.31) or PAN group (1.10±0.30) (both P<0.01). RAB produced a longer sustaining time (4.65±1.22 h) on NAKA than OME (3.22±1.89 h) (P<0.05),PAN (3.15±1.92 h) (P<0.05), and the sustaining time of NAKA in RAB group was longer than that in the healthy control group (P<0.01) too. | 6 | Accelerated Aging Behaviors and Mechanism of Fluoroelastomer in Lubricating Oil Medium显示文摘The aging behaviors and mechanism of fluoroelastomer(FKM)under lubricating oil(FKM-O)and air(FKM-A,as a comparison)at elevated temperatures were studied from both physical and chemical viewpoints.The obvious changes of mechanical and swelling performances indicate that the coupling effect of lubricating oil and temperature causes more serious deterioration of FKM-O compared to that of FKM-A.Meanwhile,much stronger temperature dependence of both bulk properties and micro-structures for FKM-O is found.Three-stage physical diffusion process is defined in FKM-O due to the competition between oil diffusion and elastic retraction of network.FTIR results reveal that the dehydrofluorination reaction causes the fracture of C-F bonds and produces a large number of C=C bonds in the backbone.The coupling effect of oil medium and high temperature could accelerate the scission of C=C bonds and generate a series of fragments with different molecular sizes.The TGA results,crosslinking density Ve,and glass transition temperature Tg derived from different measurements coherently demonstrate the network destruction in the initial stage and the simultaneous reconstruction occurring at the final stage.The newly formed local network induced by reconstruction cannot compensate the break of the original rubber network and thus only provides lower tensile strength and thermal stability. | Qi-Long Wang Jing-Ke Pei Gao Li Xi He Yan-Hua Niu Guang-Xian Li | 2020 | Chinese Journal of Polymer Science2020,38,8: | 7 |

| 8 | Study on structure-activity relationship of mutation-dependent herbicide resistance acetohydroxyacid synthase through 3D-QSAR and mutation显示文摘Seventy-four sulfonylureas were synthesized and tested for their inhibitory activity against the whole enzyme of E. coli acetohydroxyacid synthase (AHAS, EC 2.2.1.6) isoenzyme II, and 3D-QSAR analyses were performed based on these inhibitory activities. The binding conformation of chlorimuron-ethyl, a commercial herbicide of AHAS, in the crystal structure of AHAS complex was extracted and used as template to build the initial three-dimensional structure of other sulfonylureas, and then all structures were fully geometry optimized. After systematic optimization of the alignment rule, molecular orienta- tion, grid space and attenuation factor, two satisfactory models with excellent performances (CoMFA: q2 = 0.735, r2 = 0.954, n = 7, r 2 pred = 0.832; CoMSIA: q2 = 0.721, r2 = 0.913, n = 8, r 2pred = 0.844) were estab- lished. By mapping the 3D contour maps of CoMFA and CoMSIA models into the possible inhibitory active site in the crystal structure of catalytic subunit of yeast AHAS, a plausible binding model for AHAS, with best fit QSAR in the literature so far, was proposed. Moreover, the results of 3D-QSAR were further utilized to interpret resistance of site-directed mutants. A relative activity index (RAI) for AHAS enzyme mutant was defined for the first time to relate the 3D-QSAR and resistance of mutants. This study, for the first time, demonstrated that combination of 3D-QSAR and enzyme mutation can be used to decipher the molecular basis of ligand-receptor interaction mechanism. | 10 | Microcystis aeruginosa/Pseudomonas pseudoalcaligenes interaction effects on off-flavors in algae/bacteria co-culture system under different temperatures显示文摘We conducted an experiment to study the interaction effects of Microcystis aeruginosa and Pseudomonas pseudoalcaligenes on off-flavors in an algae/bacteria co-culture system at three temperatures(24, 28 and 32℃). Gas chromatography–mass spectrometry was applied to measure off-flavor compounds dimethyl sulfide(DMS), dimethyl trisulfide(DMTS),2-methylisoborneol, geosmin(GEO) and β-cyclocitral. During the lag phase of co-cultured M. aeruginosa(first 15 days), P. pseudoalcaligenes significantly increased the production of DMS, DMTS and β-cyclocitral at all three temperatures. In the exponential phase of co-cultured M. aeruginosa(after 15 days), M. aeruginosa became the main factor on off-flavors in the co-culture system, and β-cyclocitral turned to the highest off-flavor compound. These results also indicated that DMS, DMTS and β-cyclocitral were the main off-flavor compounds in our M. aeruginosa/P. pseudoalcaligenes co-culture system. Univariate analysis was applied to investigate the effects of M. aeruginosa and P. pseudoalcaligenes on the production of off-flavors. The results demonstrated that both M. aeruginosa and P. pseudoalcaligenes could increase the production of DMS and DMTS, while β-cyclocitral was mainly determined by M. aeruginosa. | 12 | Homologous and heterologous interactions between catalytic and regulatory subunits of Escherichia coli acetohydroxyacid synthaseⅠand Ⅲ显示文摘Homologous and heterologous interactions between acetohydroxyacid synthase (AHAS) I and Ⅲ from E. coli have been studied by surface plasmon resonance (SPR). The catalytic and regulatory subunits association for AHAS I (KD = 1.13 × 10-7 M) was stronger than that for AHAS Ⅲ (KD = 5.29 × 10-7 M). A strong heterologous association between regulatory and catalytic subunits and heterologous activation of catalytic subunits were observed. SPR results combined with enzyme kinetics indicate that the reconstituted heterologous enzymes had similar kinetic properties as homologous enzymes, implying that the regulatory subunit of AHAS I could be replaced by the regulatory subunit of AHAS Ⅲ and vice versa. | 13 | From ice superlubricity to quantum friction: Electronic repulsivity and phononic elasticity显示文摘Superlubricity means non-sticky and frictionless when two bodies are set contacting motion.Although this occurrence has been extensively investigated since 1859 when Faraday firstly proposed a quasiliquid skin on ice,the mechanism behind the superlubricity remains uncertain.This report features a consistent understanding of the superlubricity pertaining to the slipperiness of ice,self-lubrication of dry solids,and aqueous lubricancy from the perspective of skin bond-electron-phonon adaptive relaxation.The presence of nonbonding electron polarization,atomic or molecular undercoordination,and solute ionic electrification of the hydrogen bond as an addition,ensures the superlubricity.Nonbond vibration creates soft phonons of high magnitude and low frequency with extraordinary adaptivity and recoverability of deformation.Molecular undercoordination shortens the covalent bond with local charge densification,which in turn polarizes the nonbonding electrons making them localized dipoles.The locally pinned dipoles provide force opposing contact,mimicking magnetic levitation and hovercraft.O:H−O bond electrification by aqueous ions has the same effect of molecular undercoordination but it is throughout the entire body of the lubricant.Such a Coulomb repulsivity due to the negatively charged skins and elastic adaptivity due to soft nonbonding phonons of one of the contacting objects not only lowers the effective contacting force but also prevents charge from being transited between the counterparts of the contact.Consistency between theory predictions and observations evidences the validity of the proposal of interface elastic Coulomb repulsion that serves as the rule for the superlubricity of ice,wet and dry frictions,which also reconciles the superhydrophobicity,superlubricity,and supersolidity at contacts. | Xi ZHANG Yongli HUANG Zengsheng MA Lengyuan NIU Chang Qing SUN | 2015 | Friction2015,3,4: | 3 |
| 14 | Clinical Features and Prognostic Risk Factors of Choroic Plexus Tumors in Children显示文摘Background:Decision-making concerning the treatment ofchoroid plexus tumor (CPT)in pediatric patients remains a topic of considerable debate.The aim of this work was to describe clinical features and prognostic risk factors of CPT in the pediatric population and to provide theoretical opinions regarding clinical decisions for CPT. Methods:The data of 96 patients with CPT and younger than 14 years were retrospectively analyzed.Clinical characteristics such as pathological type of CPTs,rate and severity of hydrocephalus,treatment and outcome,and recurrence were investigated.For categorical variables,the Pearson's Chi-square test was performed.The Mann-Whitney U-test was used for comparisons between nonnormally distributed parameters.Log-rank test was used for progression-free survival (PFS). Results:The study included 70 choroid plexus papilloma (CPP)cases,17 atypical choroid plexus papilioma (aCPP)cases,and 9 choroid plexus carcinoma (CPC)cases.Compared with patients with CPP or aCPP,patients with CPC had a shorter disease course (median:CPP, 4 months;aCPP,2 months;CPC,1 month;H:23.5,P <0.001),higher rate of acute hydrocephalus (CPP,27.1%;aCPP,52.9%;CPC, 77.8%;x^2 =10.9,P <0.05),and lower incidence of cure rate (CPP,85.7%;aCPP,70.5%;CPC,33.3%;x^2 =13.5,P <0.05).The severity of hydrocephalus with tumor in the lateral or third ventricle was significantly higher than that with tumors in the fourth ventricle (severe hydrocephalus:lateral ventricle,51.7%;third ventricle,47.0%;fourth ventricle,11.1%;x^2 =26.0,P <0.001).Patients with gross total surgical resection had no better PFS than those with partial resection because of the use of adjuvant therapy in the latter (x^2 =4.0,P >0.05). Patients with CPC experienced shorter time for recurrence than those with CPP or aCPP (x^2 =40.1,P <0.0001). Their bioactivities against E. coli AHAS II in vitro were tested and the results indicated that the hydroxylation decreased the inhibition activities of sulfonylureas significantly. | 20 | Preparation and in vitro Release Properties of Mercaptopurine Drug-loaded Magnetic Microspheres显示文摘Magnetic Fe3O4 nanoparticles were synthesized by co-precipitation method and the mercaptopurine(MER) drug-loaded magnetic microspheres were obtained through emulsion cross-linking methods. The efficiency of this approach was evaluated in terms of drug loading content(DLC), encapsulation efficiency(EE) and delivery properties in vitro, determined by high performance liquid chromatograph(HPLC). The microspheres showed good DLC values of 11.8%, as well as good EE values of 79.4%. The in vitro drug release study was carried out in phosphate buffer solution(PBS) simulated body fluid, at 37 ℃ with pH=7.4. The release profiles showed an initial fast release rate, which decreased as time progressed and about 84 % had been released after 48 h.
