| 5 | Knockdown of long non-coding RNA LCPAT1 inhibits autophagy in lung cancer显示文摘Objective: Long non-coding RNAs(lnc RNAs) are involved in numerous biological processes in lung cancer cells. In our previous studies, we identified a lnc RNA, ENST00000439577, which is highly expressed in lung carcinomas, and termed it lung cancer progression-associated transcript 1(LCPAT1). To characterize the role of LCPAT1 in lung cancer, we conducted the current study.Methods: Expression of LCPAT1 and autophagy-associated markers in tumor tissues and lung cancer cell lines was determined by real-time quantitative polymerase chain reaction(q PCR). Hematoxylin and eosin(HE) staining, q PCR, Western blot, and immunohistochemistry were performed to evaluate xenografted tumor tissues. Autophagy induced by rapamycin was detected by Western blot and immunofluorescence in lung cancer cell lines.Results: Expression of LCPAT1 and microtubule-associated protein 1 light chain 3 beta(LC3B) was positively correlated in lung cancer. Knockdown of LCPAT1 inhibited tumor growth and suppressed cell autophagy in vivo. | 6 | Theoretical aspects and sensing demonstrations of cone-shaped inwall capillary-based microsphere resonators显示文摘In this paper, a detailed theoretical study on the characteristics of cone-shaped inwall capillary-based microsphere resonators is described and demonstrated for sensing applications. The maximum, minimum, slope, contrast, and width of the Fano resonance are analyzed. As the transmission coefficient of the capillary resonator increases, the absolute value of the slope of Fano resonances increases to reach its maximum, which is useful for sensors with an ultra-high sensitivity. There occurs another phenomenon of electromagnetically induced transparency when the reflectivity at the capillary–environment interface is close to 100%. | 7 | Embedded whispering-gallery mode microsphere resonator in a tapered hollow annular core fiber显示文摘We propose and demonstrate a tapered hollow annular core fiber (HACF) coupler for excitation of whisperinggallery modes (WGMs) of an embedded microsphere resonator. The coupler is simply fabricated by fusion splicing of a segment of HACF with the single-mode fiber (SMF), and then improved by tapering the splicing joint to reduce the cone-apex angle. Therefore, the coupling efficiency from the SMF to the HACF is enhanced to excite various WGMs via evanescent field coupling. Normal positive, negative symmetrical Lorentzian and asymmetric Fano line shapes can be obtained by varying the resonator size and location. Another interesting phenomenon is observed that a higher Q-factor mode in a lower Q-factor mode has a contrast as high as 58. Temperature sensing with good stability is also demonstrated. | 10 | Enhanced tumor homing of pathogen-mimicking liposomes driven by R848 stimulation: A new platform for synergistic oncology therapy显示文摘Although multifarious tumor-targeting modifications of nanoparticulate systems have been attempted in joint efforts by our predecessors,it remains challenging for nanomedicine to traverse physiological barriers involving blood vessels,tissues,and cell barriers to thereafter demonstrate excellent antitumor effects.To further overcome these inherent obstacles,we designed and prepared mycoplasma membrane(MM)-fused liposomes(LPs)with the goal of employing circulating neutrophils with the advantage of inflammatory cytokine-guided autonomous tumor localization to transport nanoparticles.We also utilized in vivo neutrophil activation induced by the liposomal form of the immune activator resiquimod(LPsR848).Fused LPs preparations retained mycoplasma pathogen characteristics and achieved rapid recognition and endocytosis by activated neutrophils stimulated by LPs-R848.The enhanced neutrophil infiltration in homing of the inflammatory tumor microenvironment allowed more nanoparticles to be delivered into solid tumors.Facilitated by the formation of neutrophil extracellular traps(NETs),podophyllotoxin(POD)-loaded MM-fused LPs(MM-LPs-POD)were concomitantly released from neutrophils and subsequently engulfed by tumor cells during inflammation.MM-LPs-POD displayed superior suppression efficacy of tumor growth and lung metastasis in a 4T1 breast tumor model.Overall,such a strategy of pathogen-mimicking nanoparticles hijacking neutrophils in situ combined with enhanced neutrophil infiltration indeed elevates the potential of chemotherapeutics for tumor targeting therapy. | Xiaobei Cheng Pei Yu Xiang Zhou Jiale Zhu Yubao Han Chao Zhang Lingyi Kong | 2022 | Acta Pharmaceutica Sinica B2022,12,2: | 2 |

| 12 | PpTCP18 is upregulated by lncRNA5 and controls branch number in peach (Prunus persica) through positive feedback regulation of strigolactone biosynthesis显示文摘Branch number is an important agronomic trait in peach(Prunus persica)trees because plant architecture affects fruit yield and quality.Although breeders can select varieties with different tree architecture,the biological mechanisms underlying architecture remain largely unclear.In this study,a pillar peach(‘Zhaoshouhong’)and a standard peach(‘Okubo’)were compared.‘Zhaoshouhong’was found to have significantly fewer secondary branches than‘Okubo’.Treatment with the synthetic strigolactone(SL)GR24 decreased branch number.Transcriptome analysis indicated that PpTCP18(a homologous gene of Arabidopsis thaliana BRC1)expression was negatively correlated with strigolactone synthesis gene expression,indicating that PpTCP18 may play an important role in peach branching.Yeast one-hybrid,electrophoretic mobility shift,dual-luciferase assays and PpTCP18-knockdown in peach leaf buds indicated that PpTCP18 could increase expression of PpLBO1,PpMAX1,and PpMAX4.Furthermore,transgenic Arabidopsis plants overexpressing PpTCP18 clearly exhibited reduced primary rosette-leaf branches.Moreover,lncRNA sequencing and transient expression analysis revealed that lncRNA5 targeted PpTCP18,significantly increasing PpTCP18 expression.These results provide insights into the mRNA and lncRNA network in the peach SL signaling pathway and indicate that PpTCP18,a transcription factor downstream of SL signaling,is involved in positive feedback regulation of SL biosynthesis.This role of PpTCP18 may represent a novel mechanism in peach branching regulation.Our study improves current understanding of the mechanisms underlying peach branching and provides theoretical support for genetic improvement of peach tree architecture. | Xiaobei Wang Qiuping Wang Lixia Yan Yuhang Hao Xiaodong Lian Haipeng Zhang Xianbo Zheng Jun Cheng Wei Wang Langlang Zhang Xia Ye Jidong Li Bin Tan Jiancan Feng | 2023 | Horticulture Research2023,10,1: | 2 |
