1Sequencing of Cultivated Peanut, Arachis hypogaea, Yields Insights into Genome Evolution and Oil Improvement显示文摘Cultivated peanut (Arachis hypogaea) is an allotetraploid crop planted in Asia, Africa, and America for edible oil and protein. To explore the origins and consequences of tetraploidy, we sequenced the allotetraploid A. hypogaea genome and compared it with the related diploid Arachis duranensis and Arachis ipaensis genomes. We annotated 39 888 A-subgenome genes and 41 526 B-subgenome genes in allotetraploid peanut. The A. hypogaea subgenomes have evolved asymmetrically, with the B subgenome resembling the ancestral state and the A subgenome undergoing more gene disruption, loss, conversion, and transposable element proliferation, and having reduced gene expression during seed development despite lacking genome-wide expression dominance. Genomic and transcriptomic analyses identified more than 2 500 oil metabolism-related genes and revealed that most of them show altered expression early in seed development while their expression ceases during desiccation, presenting a comprehensive map of peanut lipid biosynthesis. 3Quantifying 3D cell-matrix interactions during mitosis and the effect of anticancer drugs on the interactions显示文摘The mechanical force between cells and the extracellular microenvironment is crucial to many physiological processes such as cancer metastasis and stem cell differentiation. Mitosis plays an essential role in all these processes and thus an in-depth understanding of forces during mitosis gains insight into disease diagnosis and disease treatment. Here, we develop a traction force microscope method based on monolayer fluorescent beads for measuring the weak traction force (tens of Pa) of mitotic cells in three dimensions. We quantify traction forces of human ovarian granulosa (KGN) cells exerted on the extracellular matrix throughout the entire cell cycle in three dimensions. Our measurements reveal how forces vary during the cell cycle, especially during cell division. Furthermore, we study the effect of paclitaxel (PTX) and nocodazole (NDZ) on mitotic KGN cells through the measurement of traction forces. Our results show that mitotic cells with high concentrations of PTX exert a larger force than those with high concentrations of NDZ, which proved to be caused by changes in the structure and number of microtubules. These findings reveal the key functions of microtubule in generating traction forces during cell mitosis and explain how dividing cells regulate themselves in response to anti-mitosis drugs. 4An Ultra-microporous Carbon Material Boosting Integrated Capacitance for Cellulose-Based Supercapacitors显示文摘A breakthrough in advancing power density and stability of carbon-based supercapacitors is trapped by inefficient pore structures of electrode materials.Herein,an ultramicroporous carbon with ultrahigh integrated capacitance fabricated via one-step carbonization/activation of dense bacterial cellulose(BC)precursor followed by nitrogen/sulfur dual doping is reported.The microporous carbon possesses highly concentrated micropores(~2 nm)and a considerable amount of sub-micropores(<1 nm).The unique porous structure provides high specific surface area(1554 m^2 g^-1)and packing density(1.18 g cm^-3).The synergistic effects from the particular porous structure and optimal doping effectively enhance ion storage and ion/electron transport.As a result,the remarkable specific capacitances,including ultrahigh gravimetric and volumetric capacitances(430 F g^-1 and 507 F cm^-3 at 0.5 A g^-1),and excellent cycling and rate stability even at a high current density of 10 A g^-1(327 F g^-1 and 385 F cm^-3)are realized.Via compositing the porous carbon and BC skeleton,a robust all-solid-state cellulose-based supercapacitor presents super high areal energy density(~0.77 mWh cm^-2),volumetric energy density(~17.8 W L^-1),and excellent cyclic stability.Chenfeng Ding Tianyi Liu Xiaodong Yan Lingbo Huang Seungkon Ryu Jinle Lan Yunhua Yu Wei?Hong Zhong Xiaoping Yang 2020Nano-Micro Letters2020,12,5:2

6OsABA8ox2, an ABA catabolic gene, suppresses root elongation of rice seedlings and contributes to drought response显示文摘In rice, OsABA8ox encodes abscisic acid(ABA) 8′-hydroxylase, which catalyzes the committed step of ABA catabolism. The contribution of ABA catabolism in root development remains unclear. We investigated the role of OsABA8ox2 in root growth and development and drought response. GUS staining results showed that OsABA8ox2 was expressed mainly in roots at seedling stage and was strongly expressed in the meristematic zone of the radicle. OsABA8ox2 expression in roots was markedly decreased after 0.5 h polyethylene glycol(PEG) treatment and increased after 0.5 h rehydration, implying that OsABA8ox2 is a drought-responsive gene.OsABA8ox2 knockout mediated by the CRISPR-Cas9 system increased drought-induced ABA and indole-3-acetic acid accumulation in roots, conferred increased ABA sensitivity, and promoted a more vertically oriented root system architecture(RSA) beneficial to drought tolerance.OsABA8ox2 overexpression suppressed root elongation and increased stomatal conductance and transpiration rate. Consequently, OsABA8ox2 knockout dramatically improved rice drought tolerance, whereas OsABA8ox2 overexpression seedlings were hypersensitive to drought stress,suggesting that OsABA8ox2 contributes to drought response in rice. Compared with wild type,functional leaves of OsABA8ox2 knockout seedlings showed higher ABA levels, whereas overexpression lines showed lower ABA levels, suggesting that OsABA8ox2, as an ABA catabolic gene, modulates ABA concentration through ABA catabolism. OsABA8ox2 and OsABA8ox3 were both localized in the endoplasmic reticulum. Together, these results indicate that OsABA8ox2 suppresses root elongation of rice seedlings, increases water transpiration, and contributes to drought response through ABA catabolism, and that OsABA8ox2 knockout dramatically improves rice drought tolerance. They highlight the key role of ABA catabolism mediated by OsABA8ox2 on root growth and development. 14Molecular phylogenomic analysis reveals a single origin of monkeypox virus transmission in Guangzhou,China in June 2023显示文摘The monkeypox(mpox)virus has caused worldwide transmission since its initial report in England in early May 2022.Available data from the World Health Organization(WHO)show that Europe and the Americas experienced a huge wave of mpox virus infection.Now the number of infected cases is on the rise in Asia.Several sporadic infections have been reported in China.In this study,we obtained high‐quality whole viral genomic sequences using a mpox virus‐specific amplicon‐based sequencing strategy.Our analysis of the phylogenomic characteristics indicated that all eight mpox virus sequences from Guangzhou belonged to the clade IIb lineage B.1.3 cluster.However,we could not locate the exact origins where the virus was imported,based on all the available mpox virus sequences from the Global Initiative on Sharing Avian Influenza Data(GISAID)database(http://gffzz25fa773688d24fb7s59p5p5ow0xx96wcc.ffgz.tsg.suse.edu.cn/),except for their closest sequence similarity to that was reported from Japan.Novel amino acid mutations were found among the eight cases,suggesting that a local transmission may have occurred in Guangzhou,China.Mengling Jiang Xizi Deng Qinghong Fan Fei Gu Huiqin Yang Jian Wang Xiaoping Tang Fengyu Hu Yun Lan Feng Li 2023Biosafety and Health2023,5,5:0
15Satellite-based detection of 16.76 MeV γ-ray from H-bomb D-T fusion显示文摘Based on the high energy γ-ray yield from the H-bomb D-T fusion reaction, it brings forward the idea applying the 16.76 MeV γ-ray to judge whether the H-bomb happens or not, and to deduce the explosion TNT equivalent accurately. The Monte Carlo N-Particle was applied to simulate the high energy γ-ray radiation characteristics reaching the geosynchronous orbit satellite, and the CVD diamond detector suit for the requirements was researched. A series of experiments were carried out to testify the capabilities of the diamond detector. 16A Crowdsourcing Recommendation that Considers the Influence of Workers显示文摘In the context of the continuous development of the Internet,crowdsourcing has received continuous attention as a new cooperation model based on the relationship between enterprises,the public and society.Among them,a reasonably designed recommendation algorithm can recommend a batch of suitable workers for crowdsourcing tasks to improve the final task completion quality.Therefore,this paper proposes a crowdsourcing recommendation framework based on workers’influence(CRBI).This crowdsourcing framework completes the entire process design from task distribution,worker recommendation,and result return through processes such as worker behavior analysis,task characteristics construction,and cost optimization.In this paper,a calculation model of workers’influence characteristics based on the ablation method is designed to evaluate the comprehensive performance of workers.At the same time,the CRBI framework combines the traditional open-call task selection mode,builds a new task characteristics model by sensing the influence of the requesting worker and its task performance.In the end,accurate worker recommendation and task cost optimization are carried out by calculating model familiarity.In addition,for recommending workers to submit task answers,this paper also proposes an aggregation algorithm based on weighted influence to ensure the accuracy of task results.This paper conducts simulation experiments on some public datasets of AMT,and the experimental results show that the CRBI framework proposed in this paper has a high comprehensive performance.Moreover,CRBI has better usability,more in line with commercial needs,and can well reflect the wisdom of group intelligence.Zhifang Liao Xin Xu Peng Lan Liu Yang Yan Zhang Xiaoping Fan 2021Computers, Materials & Continua2021,,2:0
17Whole Exome Sequencing Identifies A Novel Pathogenic Bmpr2 Variant in Pulmonary Atresia显示文摘Objective:Pulmonary atresia(PA)is a rare type of complex cyanotic congenital heart defect characterized primarily by an undeveloped pulmonary valve or pulmonary artery.Therefore,defining a disease-causing gene mutation in a pulmonary atresia family is a possible method of genetic counseling,future prenatal diagnosis,and therapeutic approaches for pulmonary atresia.Methods:Blood samples were collected from six PA family members,and genomic DNA was extracted using the QIAamp DNA Blood Mini Kit.Gene detection was performed using a second-generation sequencing gene panel.Results:Genetic testing results indicated that a heterozygous mutation originating from maternal inheritance was detected in the BMPR2 gene of the proband’s genomic DNA.The pathogenic gene was c.2804C>T(p.A935V).The mutation was also detected in the genomic DNA of the proband’s elder brother(III-1),but not in other family members.Conclusion:To the best of our knowledge,this is the first study to report the BMPR2 variant responsible for pulmonary atresia.The frequency of the c.2804C>T(p.A935V)mutation detected in this family is extremely low in the normal population(1/246048).The mutation was highly conserved among different species.Sorting intolerant from tolerant(SIFT)predicts it to be a harmful mutation.Muyu Qi Xiaoping Lan Jia Li Junwen Ge Li Shen Rufang Zhang 2021Congenital Heart Disease2021,16,5:0

19Model for Predicting Climatic Yield of Sugarcane in Nanning City显示文摘According to spatial distribution of climate disasters in Nanning City and physiological and ecological indicator demands of sugarcane,with the aid of HJ- 1 CCD satellite remote sensing images,basic meteorological data and geographic information data,this paper established the model for predicting climatic yield of sugarcane in Nanning City,to predict total yield of sugarcane in Nanning City. Results indicated that the distribution of sugarcane in Nanning City is greatly influenced by drought. In 2010,regions suffered from drought had sugarcane planting area of 346. 20 km2,accounting for 18.88% of the total sugarcane planting area. The influence of frost disaster on distribution of sugarcane in Nanning City is limited. Regions suffered from frost had sugarcane planting area of only 67. 1 km2,taking up 3.75% of the total sugarcane planting area. In 2010,the climatic yield of sugarcane in Nanning City was 8. 8446 million tons. 20Pre-treatment Drug Resistance Could Impact the 96-Week Antiretroviral Efficacy in Treatment-Naive HIV-1–Infected Patients in Guangdong,China显示文摘Background:With the high prevalence of pre-treatment drug resistance(PDR)and the potential impact to the virological inhibition,the detection of PDR was particularly necessary.This study aimed to determine the prevalence of PDR in Guangdong,China,and its impact on antiretroviral therapy(ART)in treatment-naive HIV patients.Methods:A retrospective cohort study was conducted.A total of 1936 HIV-1-infected treatment-naive patients in the clinic of the infectious department,Guangzhou Eighth People’s Hospital,between August 2018 and December 2019 were assayed for PDR mutations before initiating ART.Patients with PDR mutations(PDR arm)were screened and compared with those without drug-resistant mutations(non-PDR arm).The rate of HIV-1 virologic failure(VF)and CD4^(+)T-cell counts of the 2 arms were compared at the 96th week after ART to evaluate the impact of PDR on the efficacy of ART.Results:Pretreatment drug resistance was detected in 125 cases(6.46%)from the 1936 enrolled participants,most of which were resistant to non-nucleoside reverse transcriptase inhibitors(64.00%,80/125).One hundred and eight of 125 completed the follow-up of 96 weeks(PDR arm).In this cohort,52 patients whose ART regimen containing the resistant drug were grouped as con-PDR arm,and the remaining 56 patients whose ART regimen did not contain the resistant drug were grouped as non-con-PDR arm.A total of 125 patients without PDR were randomly selected as the control group(non-PDR arm),112 of whom had completed the 96-week followup.At the 96th week after ART initiation,7 patients(6.5%,7/108)in the PDR arm and 1 patient(0.9%,1/112)in the non-PDR arm developed VF,exhibiting a significant difference(χ^(2)=4.901,P=0.029).Meanwhile,3 patients(5.8%,3/52)in the con-PDR arm developed VF;the rate was also higher than that in the non-PDR arm,but without a significant difference(χ^(2)=3.549,P=0.095).The CD4^(+)T-cell count in the non-PDR arm increased more than the PDR arm(386.6 vs.319.1 cells/μL,t=2.448,P=0.015)or the con-PDRarm(386.6 vs.325.1 cells/μL,t=1.821,P=0.070)at 12weeks afterART.However,no significant differenceswere observed in the CD4^(+)T-cell count from the 24th week after ART onward.Conclusions:Pretreatment drug resistance was moderately prevalent in Guangdong,China,and could affect the antiretroviral efficacy during a 96-week observation period,indicating the need to closely monitor PDR before ART initiation.Pengle Guo Yun Lan Quanmin Li Xuemei Ling Junbin Li Xiaoping Tang Fengyu Hu Weiping Cai Linghua Li 2022Infectious Diseases & Immunity2022,2,4:0
