1Prostate cancer identification: quantitative analysis of T2-weighted MR images based on a back propagation artificial neural network model显示文摘Computer-aided diagnosis(CAD) systems have been proposed to assist radiologists in making diagnostic decisions by providing helpful information. As one of the most important sequences in prostate magnetic resonance imaging(MRI), image features from T2-weighted images(T2WI) were extracted and evaluated for the diagnostic performances by using CAD. We extracted 12 quantitative image features from prostate T2-weighted MR images. The importance of each feature in cancer identification was compared in the peripheral zone(PZ) and central gland(CG), respectively. The performance of the computer-aided diagnosis system supported by an artificial neural network was tested. With computer-aided analysis of T2-weighted images, many characteristic features with different diagnostic capabilities can be extracted. We discovered most of the features(10/12) had significant difference(P<0.01) between PCa and non-PCa in the PZ, while only five features(sum average, minimum value, standard deviation, 10 th percentile, and entropy) had significant difference in CG. CAD prediction by features from T2 w images can reach high accuracy and specificity while maintaining acceptable sensitivity. 2Quantitative analysis of diffusion-weighted magnetic resonance images:differentiation between prostate cancer and normal tissue based on a computer-aided diagnosis system显示文摘Diffusion-weighted imaging(DWI) is considered to be one of the dominant modalities used in prostate cancer(PCa) detection and the assessment of lesion aggressiveness,especially for peripheral zone(PZ) PCa.Computer-aided diagnosis(CAD),which is capable of automatically extracting and evaluating image features,can integrate multiple parameters and improve the detection of PCa.In this study,13 quantitative image features were extracted from DWI by CAD,and diagnostic efficacy was analyzed in both the PZ and transition zone(TZ).The results demonstrated that there was a significant difference(P<0.05) between PCa and non-PCa for nine of the 13 features in the PZ and five of the 13 features in the TZ.Besides,the prediction outcome of CAD had a strong correlation with the DWI scores that were graded by experienced radiologists according to the Prostate Imaging-Reporting and Data System Version 2(PI-RADS v2).Ge Gao Chengyan Wang Xiaodong Zhang Juan Hu Xuedong Yang He Wang Jue Zhang Xiaoying Wang 2017Science China(Life Sciences)2017,60,1:13
3Plume aerodynamic effects of cushion engine in lunar landing显示文摘During the second period of China 'Tanyue' Project, the explorer will softland on the moon. The cushion engines are used to decelerate the explorer and reduce the impact on the lunar ground. It is necessary to study its plume effects on the explorer component. The self-developed PWS (Plume WorkStation) software based on direct simulation Monte Carlo (DSMC) method is used to simulate the plume effects of two 150 N engines. Due to the complex structure of the explorer, PWS uses a decoupling method to treat the boundary mesh, which mainly interacts with simulation particles, and has no relation with the computational grids. After the analytical expressions of plane surfaces and curved surfaces of each boundary block are given, the particle position within or without the boundary blocks can be easily determined. Finally the 3D plume field of two 150 N engines is simulated. The pressure, temperature and velocity distributions of plume field are clearly presented by three characteristic slices. The aerodynamic effects on the explorer bottom, the landfall legs and antenna are separately shown. 4FKF1 F-box protein promotes flowering in part by negatively regulating DELLA protein stability under long-day photoperiod in Arabidopsis^FA显示文摘FLAVIN-BINDING KELCH REPEAT F-BOX 1(FKF1)encodes an F-box protein that regulates photoperiod flowering in Arabidopsis under long-day conditions(LDs).Gibberellin(GA)is also important for regulating flowering under LDs.However,how FKF1 and the GA pathway work in concert in regulating flowering is not fully understood.Here,we showed that the mutation of FKF1 could cause accumulation of DELLA proteins,which are crucial repressors in GA signaling pathway,thereby reducing plant sensitivity to GA in flowering.Both in vitro and in vivo biochemical analyses demonstrated that FKF1 directly interacted with DELLA proteins.Furthermore,we showed that FKF1 promoted ubiquitination and degradation of DELLA proteins.Analysis of genetic data revealed that FKF1 acted partially through DELLAs to regulate flowering under LDs.In addition,DELLAs exerted a negative feedback on FKF1 expression.Collectively,these findings demonstrate that FKF1 promotes flowering partially by negatively regulating DELLA protein stability under LDs,and suggesting a potential mechanism linking the FKF1 to the GA signaling DELLA proteins.Jindong Yan Xinmei Li Bingjie Zeng Ming Zhong Jiaxin Yang Piao Yang Xin Li Chongsheng He Jianzhong Lin Xuanming Liu Xiaoying Zhao 2020Journal of Integrative Plant Biology2020,62,11:8

6Efficacy and safety of low-dose interleukin-2 in combination with methotrexate in patients with active rheumatoid arthritis:a randomized,double-blind,placebo-controlled phase 2 trial显示文摘Rheumatoid arthritis(RA)is an aggressive autoimmune arthritis,and current therapies remain unsatisfactory due to low remission rate and substantially adverse effects.Low-dose interleukin-2(Ld-IL2)is potentially a therapeutic approach to further improve the disease.This randomized,double-blind,placebo-controlled trial was undertaken to evaluate the efficacy and safety of Ld-IL2 in patients with active RA.Patients were randomly assigned(1:1)to receive Ld-IL2,defined as a dose of 1 million IU,or placebo in a 12-week trial with a 12-week follow-up.Three cycles of Ld-IL2 or placebo were administered subcutaneously every other day for 2 weeks(a total of 7 doses),followed by a 2-week break.All patients received a stable dose of methotrexate(MTX).The primary outcomes were the proportion of patients achieving the ACR20,DAS28-ESR<2.6,and the change from baseline in CDAI or SDAI at week 24.Secondary endpoints included other clinical responses and safety.The primary outcomes were achieved in the perprotocol population.The improvements from baseline in CDAI and SDAI were significantly greater across time points for the LdIL2+MTX group(n=17)than for the placebo+MTX group(n=23)(P=0.018 and P=0.015,respectively).More patients achieved ACR20 response in the Ld-IL2+MTX group than those in the placebo+MTX group at week 12(70.6%vs 43.5%)and at week 24(76.5%vs 56.5%)(P=0.014).In addition,low Treg and high IL-21 were associated with good responses to Ld-IL2.Ld-IL-2 treatment was well-tolerated in this study.These results suggested that Ld-IL2 was effective and safe in RA.ClinicalTrials.gov number:NCT 02467504.Xiaoying Zhang Miao Miao Ruijun Zhang Xu Liu Xiaozhen Zhao Miao Shao Tian Liu Yuebo Jin Jiali Chen Huixin Liu Xia Zhang Yun Li Yunshan Zhou Yue Yang Ru Li Haihong Yao Yanying Liu Chun Li Yuhui Li Limin Ren Yin Su Xiaolin Sun Jing He Zhanguo Li 2022Signal Transduction and Targeted Therapy2022,7,4:7
7Human ACE2-Functionalized Gold“Virus-Trap”Nanostructures for Accurate Capture of SARS-CoV-2 and Single-Virus SERS Detection显示文摘The current COVID-19 pandemic urges the extremely sensitive and prompt detection of SARS-CoV-2 virus.Here,we present a Human Angiotensin-converting-enzyme 2(ACE2)-functionalized gold“virus traps”nanostructure as an extremely sensitive SERS biosensor,to selectively capture and rapidly detect S-protein expressed coronavirus,such as the current SARS-CoV-2 in the contaminated water,down to the single-virus level.Such a SERS sensor features extraordinary 106-fold virus enrichment originating from high-affinity of ACE2 with S protein as well as“virus-traps”composed of oblique gold nanoneedles,and 109-fold enhancement of Raman signals originating from multi-component SERS effects.Furthermore,the identification standard of virus signals is established by machine-learning and identification techniques,resulting in an especially low detection limit of 80 copies mL^(−1) for the simulated contaminated water by SARS-CoV-2 virus with complex circumstance as short as 5 min,which is of great significance for achieving real-time monitoring and early warning of coronavirus.Moreover,here-developed method can be used to establish the identification standard for future unknown coronavirus,and immediately enable extremely sensitive and rapid detection of novel virus.Yong Yang Yusi Peng Chenglong Lin Li Long Jingying Hu Jun He Hui Zeng Zhengren Huang Zhi-Yuan Li Masaki Tanemura Jianlin Shi John R.Lombardi Xiaoying Luo 2021Nano-Micro Letters2021,13,7:6
8Evolution of permafrost in China during the last 20 ka显示文摘The formation and evolution of permafrost in China during the last 20 ka were reconstructed on the basis of large amount of paleo-permafrost remains and paleo-periglacial evidence, as well as paleo-glacial landforms, paleo-flora and paleofauna records. The results indicate that, during the local Last Glacial Maximum(LLGM) or local Last Permafrost Maximum(LLPMax), the extent of permafrost of China reached 5.3×106-5.4×106 km2, or thrice that of today, but permafrost shrank to only0.80×106-0.85×106 km2, or 50% that of present, during the local Holocene Megathermal Period(LHMP), or the local Last Permafrost Minimum(LLPMin). On the basis of the dating of periglacial remains and their distributive features, the extent of permafrost in China was delineated for the two periods of LLGM(LLPMax) and LHMP(LLPMin), and the evolution of permafrost in China was divided into seven periods as follows:(1) LLGM in Late Pleistocene(ca. 20000 to 13000-10800 a BP)with extensive evidence for the presence of intensive ice-wedge expansion for outlining its LLPMax extent;(2) A period of dramatically changing climate during the early Holocene(10800 to 8500-7000 a BP) when permafrost remained relatively stable but with a general trend of shrinking areal extent;(3) The LHMP in the Mid-Holocene(8500-7000 to 4000-3000 a BP)when permafrost degraded intensively and extensively, and shrank to the LLPMin;(4) Neoglaciation during the late Holocene(4000-3000 to 1000 a BP, when permafrost again expanded;(5) Medieval Warming Period(MWP) in the late Holocene(1000-500 a BP) when permafrost was in a relative decline;(6) Little Ice Age(LIA) in the late Holocene(500-100 a BP), when permafrost relatively expanded, and;(7) Recent warming(during the 20 th century), when permafrost continuously degraded and still is degrading. 10Construction of Soluble Mamu-B~*1703,a Class I Major Histocompatibility Complex of Chinese Rhesus Macaques,Monomer and Tetramer Loaded with a Simian Immunodeficiency Virus Peptide显示文摘Chinese-descent rhesus macaques have become more prevalent for HIV infection and vaccine investigation than Indian-origin macaques. Most of the currently available data and reagents such as major histocompatibility complex (MHC) class I tetramers, however, were derived from Indian-origin macaques due to the dominant use of these animals in history. Although there are significant differences in the immunogenetic background between the two macaque populations, they share a few of common MHC class I alleles. We reported in this study the procedure for preparation of a soluble Mamu-B*1703 (a MHC class I molecule of Chinese macaques) monomer and tetramer loaded with a dominant simian immunodeficiency virus (SIV) epitope IW9 (IRYPKTFGW) that was identified to be Mamu-B*1701-restricted in Indian macaques. The DNA fragment encoding the Mamu-B*1703 extracellular domain fused with a BirA substrate peptide (BSP) was amplified from a previously cloned cDNA and inserted into a prokaratic expression vector. In the presence of the antigenic peptide IW9 and light chain β2-microglobulin, the expressed heavy chain was refolded into a soluble monomer. After biotinylation, four monomers were polymerized as a tetramer by phycoerythrin-conjugated streptavidin. The tetramer, having been confirmed to have the right conformation, was a potential tool for investigation of antigen-specific CD8+ T-lymphocytes in SIV vaccine models of Chinese macaques. 12An injectable and self-healing hydrogel with controlled release of curcumin to repair spinal cord injury显示文摘The harsh local micro-environment following spinal cord injury(SCI)remains a great challenge for neural regeneration.Local reconstitution of a favorable micro-environment by biocompatible scaffolds with desirable functions has thus been an area of concern.Herein,a hybrid hydrogel was developed using Fmoc-grafted chitosan(FC)and Fmoc peptide(FI).Dynamic reversibleπ-πstacking interactions of the fluorenyl rings enabled the FC/FI hybrid hydrogel to exhibit excellent injectable and self-healing properties,as characterized by visual appearances and rheological tests.Furthermore,the FC/FI hybrid hydrogel showed a slow and persistent release of curcumin(Cur),which was named as FC/FI-Cur hydrogel.In vitro studies confirmed that with the support of FC/FI-Cur hydrogel,neurite outgrowth was promoted,and Schwann cell(SC)migration away from dorsal root ganglia(DRG)spheres with enhanced myelination was substantiated.The FC/FI-Cur hydrogel well reassembled extracellular matrix at the lesion site of rat spinal cord and exerted outstanding effects in modulating local inflammatory reaction by regulating the phenotypes of infiltrated inflammatory cells.In addition,endogenous SCs were recruited in the FC/FI-Cur graft and participated in the remyelination process of the regenerated nerves.These outcomes favored functional recovery,as evidenced by improved hind limbs movement and enhanced electrophysiological properties.Thus,our study not only advanced the development of multifunctional hydrogels but also provided insights into comprehensive approaches for SCI repair.Jinghua Luo Xueshuang Shi Liming Li Zan Tan Feng Feng Jun Li Mao Pang Xiaoying Wang Liumin He 2021Bioactive Materials2021,6,12:4
13Optimization of broadband omnidirectional antireflection coatings for solar cells显示文摘Broadband and omnidirectional antireflection coating is generally an effective way to improve solar cell efficiency, because the destructive interference between the reflected and incident light can maximize the light transmission into the absorption layer. In this paper, we report the incident quantum efficiency ηin, not incident energy or power, as the evaluation function by the ant colony algorithm optimization method, which is a swarm-based optimization method. Also, SPCTRL2 is proposed to be incorporated for accurate optimization because the solar irradiance on a receiver plane is dependent on position, season, and time. Cities of Quito, Beijing and Moscow are selected for two-and three-layer antireflective coating optimization over λ = [300,1100] nm and θ = [0°, 90°]. 14Machine-learning-driven on-demand design of phononic beams显示文摘The development of phononic crystals, especially their interaction with topological insulators, allows exploration of the anomalous properties of acoustic/elastic waves for various applications. However, rapidly and inversely exploring the geometry of specific targets remains a major challenge. In this work, we show how machine learning can address this challenge by studying phononic crystal beams using two different inverse design schemes. We first develop the theory of phononic beams using the transfer matrix method. Then, we use the reinforcement learning algorithm to effectively and inversely design the structural parameters to maximize the bandgap width. Furthermore, we employ the tandem-architecture neural network to solve the training-difficulty problem caused by inconsistent data and complete the task of inverse structure design with the targeted topological properties. The two inverse-design schemes have different adaptabilities, and both are characterized by high efficiency and stability. 15Extension of the low diffusion particle method for near-continuum two-phase flow simulations显示文摘The low diffusion (LD) particle method, proposed by Burt and Boyd, is modified for the near-continuum two-phase flow simulations. The LD method has the advantages of easily coupling with the direct simulation Monte Carlo (DSMC) method for multi-scale flow simulations and dramatically reducing the numerical diffusion error and statistical scatter of the equilibrium particle methods. Liquidor solid-phase particles are introduced in the LD method. Their velocity and temperature updating are respectively, calculated from the motion equation and the temperature equation according to the local gas properties. Coupling effects from condensed phase to gas phase are modeled as momentum and energy sources, which are respectively, equal to the negative values of the total momentum and energy increase in liquid or solid phase. The modified method is compared with theoretical results for unsteady flows, and good agreements are obtained to indicate the reliability of the one-way gas-to-particle coupling models. 16Two haplotype-resolved,gap-free genome assemblies for Actinidia latifolia and Actinidia chinensis shed light on the regulatory mechanisms of vitamin C and sucrose metabolism in kiwifruit显示文摘Kiwifruit is a recently domesticated horticultural fruit crop with substantial economic and nutritional value,especially because of the high content of vitamin C in its fruit.In this study,we de novo assembled two telomere-to-telomere kiwifruit genomes from Actinidia chinensis var.‘Donghong’(DH)and Actinidia latifolia‘Kuoye’(KY),with total lengths of 608327852 and 640561626 bp for 29 chromosomes,respectively.With a burst of structural variants involving inversion,translocations,and duplications within 8.39 million years,the metabolite content of DH and KY exhibited differences in saccharides,lignans,and vitamins.A regulatory ERF098 transcription factor family has expanded in KY and Actinidia eriantha,both of which have ultra-high vitamin C content.With each assembly phased into two complete haplotypes,we identified allelic variations between two sets of haplotypes,leading to protein sequence variations in 26494 and 27773 gene loci and allele-specific expression of 4687 and 12238 homozygous gene pairs.Synchronized metabolome and transcriptome changes during DH fruit development revealed the same dynamic patterns in expression levels and metabolite contents;free fatty acids and flavonols accumulated in the early stages,but sugar substances and amino acids accumulated in the late stages.The AcSWEET9b gene that exhibits allelic dominance was further identified to positively correlate with high sucrose content in fruit.Compared with wild varieties and other Actinidia species,AcSWEET9b promoters were selected in red-flesh kiwifruits that have increased fruit sucrose content,providing a possible explanation on why red-flesh kiwifruits are sweeter.Collectively,these two gap-free kiwifruit genomes provide a valuable genetic resource for investigating domestication mechanisms and genome-based breeding of kiwifruit.Xue Han Yilin Zhang Qiong Zhang Ni Ma Xiaoying Liu Wenjing Tao Zhiying Lou Caihong Zhong Xing Wang Deng Dawei Li Hang He 2023Molecular Plant2023,16,2:3
17Cytochrome b5 reductase 2 suppresses tumor formation in nasopharyngeal carcinoma by attenuating angiogenesis显示文摘Background:Cytochrome b5 reductase 2(CYB5R2) is a potential tumor suppressor that inhibits cell proliferation and motility in nasopharyngeal carcinoma(NPC).Inactivation of CYB5R2 is associated with lymph node metastasis in NPC.This study aimed to explore the mechanisms contributing to the anti-neoplastic effects of CYB5R2.Methods:Polymerase chain reaction(PCR) assays were used to analyze the transcription of 84 genes known to be involved in representative cancer pathways in the NPC cell line HONE1.NPC cell lines CNE2 and HONE1 were transiently transfected with CYB5R2,and data was validated by real-time PCR.A chick chorioallantoic membrane(CAM)embryo model was implanted with CYB5R2-expressing CNE2 and HONE1 cells to evaluate the effect of CYB5R2 on angiogenesis.An immunohistochemical assay of the CAM model was used to analyze the protein expression of vascular endothelial growth factor(VEGF).Results:In CYB5R2-transfected NPC cells,PCR assays revealed up-regulated mRNA levels of Fas cell surface death receptor(FAS),FBJ murine osteosarcoma viral oncogene homolog(FOS),phosphoinositide-3-kinase regulatory subunit 1(PIK3R1),integrin beta 3(ITGB3),metastasis suppressor 1(MTSSl),interferon beta 1(IFNB1),and cyclin-dependent kinase inhibitor 2A(CDKN2A) and down-regulated levels of integrin beta 5(ITGB5),insulin-like growth factor 1(IGF1),TEK tyrosine kinase(TEK),transforming growth factor beta receptor 1(TGFBRl),and VEGF.The angiogenesis in the CAM model implanted with CYB5R2-transfected NPC cells was inhibited.Down-regulation of VEGF by CYB5R2 in NPC cells was confirmed by immunohistochemical staining in the CAM model.Conclusion:CYB5R2 up-regulates the expression of genes that negatively modulate angiogenesis in NPC cells and down-regulates the expression of VEGF to reduce angiogenesis,thereby suppressing tumor formation.Huixin Ming Ying Lan Feng He Xue Xiao Xiaoying Zhou Zhe Zhang Ping Li Guangwu Huang 2015Chinese Journal of Cancer2015,34,10:3

20Impact of damping on high speed 850 nm VCSEL performance显示文摘High speed VCSELs are important optical devices in short-reach optical communication links and interconnects because of their low cost and high modulation speeds. In this paper, the impact of damping on the their static and dynamic characteristics is analyzed and demonstrated. Through the shallow corrosion of the top layer DBR, the VCSELs with different damping is designed and fabricated. With the increase of the surface etch depth from 0 to ~55 nm for 9 μm oxide-aperture VCSEL, the K factor related with the damping is reduced from0.31 to 0.23 ns^(-1). When the etch depth of the VCSEL with 9 μm oxide-aperture is decreased to ~25 nm,output power is increased from 4.03 to 4.70 mW and small signal modulation bandwidth is also increased from15.46 to 16.37 GHz.
