| 3 | Exosomal piRNA profiling revealed unique circulating piRNA signatures of cholangiocarcinoma and gallbladder carcinoma显示文摘Cholangiocarcinoma(CCA)and gallbladder carcinoma(GBC)are biliary tract cancers with poor five-year survival and high recurrence rates.Both CCA and GBC patients suffer from lack of circulating diagnostic biomarkers at the early stage.Extracellular vesicles,especially exosomes,have been emerged as promising diagnostic sources for cancers due to easy and quick accessibility.Hence,identification of exosomal biomarkers provides a novel strategy for CCA and GBC diagnosis.Here,five CCA patients and four GBC patients were enrolled for exosomal small RNA sequencing.Our data showed that exosomal piwi-interacting RNA(piRNA)populations were altered in the plasma of CCA and GBC patients.In comparison to healthy individuals,694 and 323 piRNAs were upregulated in CCA and GBC,respectively,while 36 and 191 piRNAs were down regulated.Interestingly,sequencing results predicted that piR-2660989,piR-10506469,piR-20548188,piR-10822895,piR-hsa-23209,and piR-18044111 were upregulated in both CCA and GBC plasma.Importantly,we further included blood samples from 50 health individuals,40 CCA patients,and 25 GBC patients and found that piR-10506469 were significantly increased in the exosomes of plasma from both CCA and GBC patients.Moreover,we analyzed the expression levels of differentially expressed exosomal piRNAs in the plasma of CCA and GBC patient before and after surgeries and found that piR-10506469 and piR-20548188 were significantly decreased in patients underwent surgeries.Taken together,our data revealed that exosomal piRNAs those are differentially expressed in CCA and GBC plasma may serve as potential biomarkers for the diagnosis of CCA and GBC. | Xinjin Gu Chen Wang Hui Deng Chong Qing Rong Liu Sanhong Liu Xinying Xue | 2020 | Acta Biochimica et Biophysica Sinica2020,52,5: | 7 |
| 4 | Risk identification and risk mitigation during metro station construction by enlarging shield tunnel combined with cut-and-cover method显示文摘Constructing a metro station by enlarging shield tunnels combined with a mining/cut-and-cover method provides a new method to solve the contradictions of construction time limits of shield tunnels and stations. As a new-style construction method, there are several specific risks involved in the construction process. Based on the test section of Sanyuanqiao station on Beijing metro line 10, and combined with the existing methods of risk identification at present, including a review of world-wide operational experience of similar projects, the study of generic guidance on hazards associated with the type of work being undertaken, and discussions with qualified and experienced staff from the project team, etc., the specific risks during the construction process of the metro station constructed by enlarging shield tunnels combined with the cut-and-cover method are identified. The results show that the specific risks mainly come from three construction processes which include constructing upper enclosure structures, excavating the soil between shield tunnels and demolishing shield segments. Then relevant risk mitigation measures are put forward. | 5 | Shale pore characteristics of Shahejie Formation: Implication for pore evolution of shale oil reservoirs in Dongying sag, north China显示文摘The pore characteristics of shale reservoirs in the lower submember of Member 3 to upper submember of Member 4 of Shahejie Formation in Dongying sag are analyzed,influences of mineral content and organic matter content on porosity and pore size are also investigated,and through the diagenetic thermal simulation experiment,the main pore evolution is further discussed.The results show that the pore structure of shale reservoirs is complex,the micron-nanometer pores can storage liquid hydrocarbons,and the free-phase crude oil is mainly distributed in intergranular dissolution pores of calcite,recrystallized intergranular pores,intergranular shrinkage fractures of clay mineral which have large pore size.Framework minerals and organic matter content directly influence porosity and pore size of shale reservoirs,relationship between porosity and content of felsic mineral as well as content of organic matter content is linear and positive,while relationship between content of carbonated mineral is negative.At the buried depth from 2500 to 3500 m,concentration of organic acid from hydrocarbon generation and expulsion of organic matter,increasing range of pressure coefficient,are well corresponding to highporosity intervals;pore formation in shale oil reservoirs are almost controlled by diagenetic evolution of clay minerals;framework storage spaces formed by carbonate grain crystals as well as intergranular and intergranular dissolution pores of carbonate increases porosity of shale oil reservoirs;local increase of porosity at the depth of 3500e3800 m is mainly caused by coupling of hydrocarbon-generating overpressure and dissolution,and size,distribution and connectivity of pores are enhanced obviously. | Shun Zhang Huimin Liu Min Wang Xinjin Liu Huilai Liu Youshu Bao Weiqing Wang Runze Li Xin Luo Zhengwei Fang | 2019 | Petroleum Research2019,4,2: | 2 |

| 14 | Fungal-derived selenium nanoparticles and their potential applications in electroless silver coatings for preventing pin-tract infections显示文摘Pin-tract infections(PTIs)are a common complication of external fixation of fractures and current strategies for preventing PTIs have proven to be ineffective.Recent advances show that the use of anti-infection coatings with local antibacterial activity may solve this problem.Selenium has been considered as a promising anti-infection agent owing to its antibacterial and antibiofilm activities.In this study,selenium nanoparticles(SeNPs)were synthesized via a cost-effective fungi-mediated biorecovery approach and demonstrated excellent stability and homogeneity.To investigate their anti-infection potential,the SeNPs were doped in silver coatings through an electroless plating process and the silver–selenium(Ag–Se)coatings were tested for antibacterial and antibiofilm properties against Staphylococcus aureus F1557 and Escherichia coli WT F1693 as well as corrosion resistance in simulated body fluid.It was found that the Ag–Se coating significantly inhibited S.aureus growth and biofilm formation on the surface,reducing 81.2%and 59.7%of viable bacterial adhesion when compared with Ag and Ag–PTFE-coated surfaces after 3 days.The Ag–Se coating also exhibited improved corrosion resistance compared with the Ag coating,leading to a controlled release of Ag^(+),which in turn reduced the risk of cytotoxicity against hFOBs.These results suggest that the fungal-derived SeNPs may have potential in use as implant coatings to prevent PTIs. | Xinjin Liang Shuai Zhang Geoffrey Michael Gadd John McGrath David W.Rooney Qi Zhao | 2022 | Regenerative Biomaterials2022,9,1: | 0 |
| 15 | 高强度聚焦超声对中晚期胰腺癌的止痛作用(英文)显示文摘Objective: The aim of this study was to evaluate the analgesic effect of high intensity focused ultrasound (HIFU) in patients with advanced pancreatic cancer. Methods: A total of 106 patients with advanced pancreatic cancer accompanied by abdominal pain were treated by HIFU. Pain intensities and quantities of morphine consumption before and after treatment were observed and compared. Results: The average pain intensities before treatment, and at d3, d7 after treatment were 5.80 ± 2.14, 2.73 ± 2.68, 2.45 ± 2.43 respectively (P < 0.01). Fifty-nine cases (55.7%) got to extremely effective, and 29 cases (27.4%) effective. Total efficient rate was 83.0 %. The average quantities of morphine consumption before and after treatment in the patients with grade III pain were 114.9 ± 132.5 mg, 16.8 ± 39.7 mg each person everyday respectively (P < 0.01). Conclusion: HIFU can relieve pain suffered by patients with pancreatic cancer effectively. | 18 | ANXA2 Facilitates Enterovirus 71 Infection by Interacting with 3D Polymerase and PI4KB to Assist the Assembly of Replication Organelles显示文摘Similar to that of other enteroviruses, the replication of enterovirus 71(EV71) occurs on rearranged membranous structures called replication organelles(ROs). Phosphatidylinositol 4-kinase Ⅲ(PI4KB), which is required by enteroviruses for RO formation, yields phosphatidylinositol-4-phosphate(PI4P) on ROs. PI4P then binds and induces conformational changes in the RNA-dependent RNA polymerase(Rd Rp) to modulate Rd Rp activity. Here, we targeted 3D polymerase, the core enzyme of EV71 ROs, and found that the host factor Annexin A2(ANXA2) can interact with 3 D polymerase and promote the replication of EV71. Then, an experiment showed that the annexin domain of ANXA2, which possesses membranebinding capacity, mediates the interaction of ANXA2 with EV71 3 D polymerase. Further research showed that ANXA2 is localized on ROs and interacts with PI4KB. Overexpression of ANXA2 stimulated the formation of PI4P, and the level of PI4P was decreased in ANXA2-knockout cells. Furthermore, ANXA2, PI4KB, and 3D were shown to be localized to the viral RNA replication site, where they form a higher-order protein complex, and the presence of ANXA2 promoted the PI4 KB-3D interaction. | 20 | Gas-assisted microfluidic step-emulsification forgenerating micron- and submicron-sized droplets显示文摘Micron- and submicron-sized droplets have extensive applications in biomedical diagnosis and drug delivery.Moreover, accurate high-throughput analysis requires a uniform droplet size distribution and high production rates.Although the previously reported microfluidic coflow step-emulsification method can be used to generate highlymonodispersed droplets, the droplet diameter (d) is constrained by the microchannel height (b), d\3b, while theproduction rate is limited by the maximum capillary number of the step-emulsification regime, impedingemulsification of highly viscous liquids. In this paper, we report a novel, gas-assisted coflow step-emulsificationmethod, where air serves as the innermost phase of a precursor hollow-core air/oil/water emulsion. Air graduallydiffuses out, producing oil droplets. The size of the hollow-core droplets and the ultrathin oil layer thickness bothfollow the scaling laws of triphasic step-emulsification. The minimal droplet size attains d 1:7b, inaccessible instandard all-liquid biphasic step-emulsification. The production rate per single channel is an order-of-magnitudehigher than that in the standard all-liquid biphasic step-emulsification and is also superior to alternative emulsificationmethods.
