| 3 | Atorvastatin attenuates surgery-induced BBB disruption and cognitive impairment partly by suppressing NF-κB pathway and NLRP3 inflammasome activation in aged mice显示文摘In clinic,perioperative neurocognitive disorder is becoming a common complication of surgery in old patients.Neuroinflammation and blood-brain barrier(BBB)disruption are important contributors for cognitive impairment.Atorvastatin,as a strong HMG-CoA reductase inhibitor,has been widely used in clinic.However,it remains unclear whether atorvastatin could prevent anesthesia and surgery-induced BBB disruption and cognitive injury by its anti-inflammatory property.In this study,aged C57BL/6J mice were used to address this question.Initially,the mice were subject to atorvastatin treatment for 7 days(10 mg/kg).After a simple laparotomy under 1.5%isoflurane anesthesia,Morris water maze was performed to assess spatial learning and memory.Western blot analysis,immunohistochemistry,and enzyme-linked immunosorbent assay were used to examine the inflammatory response,BBB integrity,and cell apoptosis.Terminal-deoxynucleotidyl transferase mediated nick end labeling assay was used to assess cell apoptosis.The fluorescein sodium and transmission electron microscopy were used to detect the permeability and structure of BBB.The results showed that anesthesia and surgery significantly injured hippocampal-dependent learning and memory,which was ameliorated by atorvastatin.Atorvastatin could also reverse the surgery-induced increase of systemic and hippocampal cytokines,including IL-1β,TNF-α,and IL-6,accompanied by inhibiting the nuclear factor kappa-B(NF-κB)pathway and Nucleotide-Binding Oligomerization Domain,or Leucine Rich Repeat and Pyrin Domain Containing 3(NLRP3)inflammasome activation,as well as hippocampal neuronal apoptosis.In addition,surgery triggered an increase of BBB permeability,paralleled by a decrease of the ZO-1,occludin,and Claudin 5 proteins in the hippocampus.However,atorvastatin treatment could protect the BBB integrity from the impact of surgery,by up-regulating the expressions of ZO-1,occludin,and Claudin 5.These findings suggest that atorvastatin exhibits neuroprotective effects on cognition in aged mice undergoing surgery. | Pengfei Liu Quansheng Gao Lei Guan Yanting Hu Jingwen Jiang Teng Gao Weixuan Sheng Xinying Xue Hui Qiao Tianzuo Li | 2021 | Acta Biochimica et Biophysica Sinica2021,53,5: | 6 |

| 5 | Research on the Necessity of Building National-level Grape and Cherry Germplasm Resources Bank in Shandong Province显示文摘At present,the grape and cherry industries in Shandong are in the leading position in China and have driven the development of related high-end manufacturing industries such as wine,brandy,preserved fruit processing,fruit juice processing and health products. Therefore,vigorously developing the grape and cherry industries in Shandong Province and carrying out development and innovation are important parts of Shandong Province in responding to the strategy of national new and old kinetic energy conversion. However,currently the germplasm resources preserved in the fruit tree resources banks in China are only 45% of those in the US and 27. 2% of those in the EU. Moreover,the development of fruit trees resources banks in Shandong is relatively backward in China,and there is still no banks related with grape and cherry germplasm resources in Shandong. Therefore,importance can be attached to the agriculture,forestry,and animal husbandry to build germplasm resources banks for grape and cherry industries in Shandong Province. Building the national-level grape and cherry germplasm resources bank in Shandong Province can also promote the utilization of wild and farm germplasm resources in the future; advance the research on the genes related to disease resistance,stress resistance and quality of grapes and cherries; push forward the construction and development of cherry and grape mutants banks. It is conducive to the research on the agronomic traits of grapes and cherries,and can provide the parents resources for planting innovation and improving the quality of grapes and cherries,as well as promote the development and application of molecular markers of grapes and cherries,including the identification of lines and crossbreeding. | 6 | A survey on hearing acuity of centenarians in Hainan Province显示文摘Objective: This study aimed to investigate hearing acuity of centenarians in Hainan Province and provide basis for interventional considerations.Method: Door-to-door follow-up was conducted to investigate hearing acuity of centenarians(aged 100 years or above) in Hainan Province. Physical examination, pure tone audiometry and acoustic impedance test were performed, and the results were analyzed.Result: A total of 460 centenarians(920 ears) were tested by pure tone audiometry. The rate of normal hearing was 0.2%, the rate of mild hearing loss was 1.5%, 12% for moderate hearing loss, 33.9% for moderate to severe hearing loss, 37.4% for severe hearing loss is and 15% for profound hearing loss. Acoustic impedance test was performed in 340 centenarians(668 ears). Tympanogram was type A in 41.2% of the ears, type As in 34.1% ears,type Ad in 6.4%, type B in 11.8%, and type C in 4.9% of the ears, while no response was elicited in 1.5% of the ears.Conclusion: Age related sensorineural hearing loss is prevalent among centenarians in Hainan Province. | 7 | Neuropeptide Y gene transfection inhibits post-epileptic hippocampal synaptic reconstruction显示文摘Exogenous neuropeptide Y has antiepileptic effects; however, the underlying mechanism and optimal administration method for neuropeptide Y are still unresolved. Previous studies have used intracerebroventricular injection of neuropeptide Y into animal models of epilepsy. In this study, a recombinant adeno-associated virus expression vector carrying the neuropeptide Y gene was injected into the lateral ventricle of rats, while the ipsilateral hippocampus was injected with kainic acid to establish the epileptic model. After transfection of neuropeptide Y gene, mossy fiber sprouting in the hippocampal CA3 region of epileptic rats was significantly suppressed, hippocampal synaptophysin (p38) mRNA and protein expression were inhibited, and epileptic seizures were reduced. | 8 | Identification of Differentially Expressed Genes in Grape Skin at Veraison and Maturity and Construction of Co-expression Network显示文摘The ripening process of grape is an important stage during grape growth and development. During this process, color of grape skin is the most obvious change. The molecular mechanism for the ripening of grape(a non-climacteric fruit, which ripens without ethylene and respiration bursts) is still unclear. Although numerous studies have been done on the changes in the contents of metabolites during grape ripening, the differentially expressed genes at veraison and maturity stages have not been systematically analyzed. In this study, 1 524 genes that are significantly differentially expressed in grape(Pinot Noir) skin at veraison and maturity stages were identified, and a co-expression network of these genes was built. Some of the eight co-expression modules we identified may be closely related to the synthesis or metabolism of anthocyanins, sugar acids, and other flavor substances. The transcription factor families WRKY, b ZIP, HSF and WOX may play an important role in the regulation of anthocyanin synthesis or metabolism. | 11 | Somatic mutation contributing to clonal haematopoiesis is a risk factor of recurrent stroke in first-ever acute ischaemic stroke:a prospective cohort study显示文摘Background Somatic mutation contributes to clonal haematopoiesis of indeterminate potential(CHIP)is related to age and associated with a higher risk of stroke and atherosclerotic cardiovascular disease.Here,we investigated the prognostic significance of CHIP in a large first-ever acute ischaemic stroke(AIS)cohort and explored the underlying mechanisms.Methods We studied a prospective cohort of 6016 patients who had a first-ever AIS in China.Whole-genome sequencing was performed to identify CHIP.High-sensitivity C reactive protein(hs-CRP)levels above 3 mg/L at baseline were defined as hyperinflammation.Recurrent stroke during the 3-month follow-up was the primary outcome.Results Among the 6016 patients who had a first-ever AIS,with a median age was 62 years(IQR,54.0‒70.0),3.70%were identified as CHIP carriers.The most common mutations occurred in the DNMT3A(30.0%)and TET2(11.4%)genes.During a follow-up of 3 months,the presence of CHIP was associated with recurrent stroke(HR 1.62,95%CI 1.04 to 2.51,p=0.03),recurrent ischaemic stroke(HR 1.64,95%CI 1.04 to 2.58,p=0.03)and combined vascular events(HR 1.58,95%CI 1.02 to 2.44,p=0.04)after adjusting for hsCRP levels at baseline in patients who had a first-ever AIS.Subgroup analysis demonstrated that CHIP was only associated with recurrent stroke when patients under hyperinflammation(OR 3.10,95%CI 1.92 to 5.00,p<0.001)but not in those without hyperinflammation(OR 0.18,95%CI 0.03 to 1.04,p=0.06,Pinteraction=0.002).Conclusion Our results suggest that somatic mutations contributing to CHIP increase the risk of short-term recurrent stroke in patients who had a first-ever AIS.Hyperinflammation may be important in the relationship between CHIP and recurrent stroke. | Xin Qiu Yalun Dai Si Cheng Hong-Qiu Gu Yong Jiang Xia Meng Yilong Wang Xingquan Zhao Yingyu Jiang Zhe Xu Xinying Huang Meng Wang Tian Jie Lyu Yubo Wang Jiaxu Weng Lingyun Cui Yi Shangguan Hao Li Yongjun Wang Zixiao Li | 2023 | Stroke & Vascular Neurology2023,8,2: | 1 |

| 14 | Numerical Investigation on Convective Heat Transfer of Supercritical Carbon Dioxide in a Mini Tube Considering Entrance Effect显示文摘There are more and more researches on heat transfer characteristics and prediction of supercritical CO_(2).The method of adding adiabatic section before and after heating section is usually adopted in these researches to ensure that the fluid entering the heating section is no longer affected by boundary layer,but the appropriate length range of adiabatic section and the influence of entrance effect are not discussed.However,some studies show that the entrance effect would affect the heat transfer in mini tubes.This paper uses the commercial CFD code FLUENT 19.0 to numerically study the heat transfer of supercritical CO_(2) in a mini tube under different working conditions(such as Re_(in),P_(in),q_(w) and flow direction)and the lengths of the adiabatic section(l_(as)/d).The entrance effects on heat transfer is more pronounced when Re_(in) is within the transition state and wall heat flux is relatively high,the resulting heat transfer deterioration causes T_(w,x) and h_(w,x) to rise sharply.As the adiabatic section increases,the location at which the heat exchange deteriorates moves to the entrance of the heating section and eventually leaves.The buoyancy effect and flow acceleration effect caused by the sharp change of physical properties are analyzed,and the dimensionless velocity distribution at the inlet of the heating section in different adiabatic sections is compared.It is proved that the entrance effect has an influence on the convection heat transfer of supercritical CO_(2) in mini tubes.The interaction reflected by wall shear stress between boundary layer development and drastic changes in physical properties is the cause of heat transfer deterioration. | LIU Meng JIANG Xinying FANG Yufeng GUO Menglei DING Chen | 2021 | Journal of Thermal Science2021,30,6: | 0 |

RNA-directed DNA methylation( RdD M) pathway is considered as the most important mechanism of DNA methylation. A microRNA( miRNA) is processed from a 70-90-nucleotide-long single-stranded RNA precursor that forms a hairpin structure by Dicer enzyme,which plays an important role in plant development,regulation of plant gene expression and silencing of transposons. In recent years,a large number of studies have confirmed that DNA methylation and miRNAs play an important role in disease resistance mechanisms of plants.
