1Family-level diversity of extracellular proteases of sedimentary bacteria from the South China Sea显示文摘Protease-producing bacteria and their extracellular proteases are key players in degrading organic nitrogen to drive marine nitrogen cycling and yet knowledge on both of them is still very limited. This study screened protease-producing bacteria from the South China Sea sediments and analyzed the diversity of their extracellular proteases at the family level through N-terminal amino acid sequencing. Results of the 16 S rRNA gene sequence analysis showed that all screened protease-producing bacteria belonged to the class Gammaproteobacteria and most of them were affiliated with different genera within the orders Alteromonadales and Vibrionales. The Nterminal amino acid sequence analysis for fourteen extracellular proteases from fourteen screened bacterial strains revealed that all these proteases belonged to the M4 family of metalloproteases or the S8 family of serine proteases. 2Mechanics analysis of axisymmetric thin-walled part in warm sheet hydroforming显示文摘To obtain the influence of fluid pressure and temperature on warm hydroforming of5A06-O aluminum alloy sheet, the unified mechanics equilibrium equations, which take throughthickness normal stress and friction into account, were established in spherical coordinate system.The distribution of through-thickness normal stress in the thickness direction was determined. The relation between through-thickness normal stress and fluid pressure was also analyzed in different regions of cylindrical cup. Based on the method of subtracting one increasing function from another, the constitutive equation of 5A06-O applied to warm hydroforming was established and in a good agreement with uniaxial tensile data. Based on whether the thickness variation was taken into account, two mechanic models were established to do the comparative study. The results for the studied case show that the calculated stress values are pretty close according to the two models and consistent with results of finite element analysis; the thickness distribution in flange computed by the second model conforms to the experimental data. 3The establishment of polypeptide PSMA-targeted chimericantigen receptor-engineered natural killer cells forcastration-resistant prostate cancer and the induction of ferroptosis-related cell death显示文摘Background:The mortality of castration-resistant prostate cancer(CRPC)is high due to lack of an effective treatment.Chimeric antigen receptor(CAR)-based therapy is a promising immunotherapeutic strategy.Here,we aimed to design a novel CAR-natural killer(NK)cells with a clinically significant tumoricidal effect on CRPC.Methods:We constructed novel CAR-NK92MI cells with a CD244-based recombinant lentiviral vector.Different intracellular segments(CD244,NKG2D,or CD3ζ)were screened to identify the best candidate according to cell lysis assay and CD107a expression levels.To enhance the affinity of the CAR to the tumor antigen,we compared an antibody specific for prostate-specific membrane antigen(anti-PSMA)with PSMA-targeted polypeptide(p-PSMA),which was screened by phage display combinatorial library.Then,CAR-NK92MI cells with both a high affinity for PSMA and a strong tumoricidal capacity were generated.In addition,we verified their tumor-killing effect in vitro and in vivo.The release of cytokine by NK92MI cells was compared with that by CAR-NK92MI cells through flow cytometry and enzyme-linked immunosorbent assay.Moreover,ferroptosis-related cell death was explored as a possible underlying mechanism.Results:Three different CAR intracellular regions CAR1(CD244),CAR2(CD244,NKG2D)and CAR3(CD244,NKG2D,and CD3ζ)were constructed.CAR2 was chosen to confer a stronger tumoricidal ability on CAR-NK92MI cells.Compared with anti-PSMA,p-PSMA exhibited enhanced affinity for the tumor antigen.Thus,p-PSMA-CAR-NK92MI cells,which expressed CAR with a polypeptide-based antigen-binding region,an intracellular CD244 and a NKG2D costimulatory domain,were generated.They could selectively and successfully kill PSMA+target cells and exhibited specific lysis rate of 73.19%for PSMA-positive C4-2 cells and 33.04%for PSMA-negative PC3 cells.Additionally,p-PSMA-CAR-NK92MI cells had significantly higher concentrations of IFN-γ,TNF-αand granzyme B than NK92MI cells.In a CRPC cancer xenograft model,p-PSMA-CAR-NK92MI cells significantly inhibited tumor growth and exerted a more consistent killing effect than NK92MI cells.Moreover,ferroptosis is a potential mechanism through which CAR-NK92MI cells attack cancer cells,and is triggered by IFN-γ.Conclusions:p-PSMA-CAR-NK92MI cells can effectively kill CRPCPSMA+cells in vitro and in vivo.This strategy may provide additional treatment options for patients with CRPC.Liyuan Wu Fei Liu Le Yin Fangming Wang Hui Shi Qinxin Zhao Feiya Yang Dong Chen Xiying Dong Yuchun Gu Nianzeng Xing 2022Cancer Communications2022,42,8:2

6NudCL2 is an autophagy receptor that mediates selective autophagic degradation of CP110 at mother centrioles to promote ciliogenesis显示文摘Primary cilia extending from mother centrioles are essential for vertebrate development and homeostasis maintenance.Centriolar coiled-coil protein 110(CP110)has been reported to suppress ciliogenesis Initiation by capping the distal ends of mother centrioles.However,the mechanism underlying the specific degradation of mother centriole-capping CP110 to promote cilia initiation remains unknown.Here,we find that autophagy is crucial for CP110 degradation at mother centrioles after serum starvation in MEF cells.We further identify NudC-like protein 2(NudCL2)as a novel selective autophagy receptor at mother centrioles,which contains an LC3-interacting region(UR)motif mediating the association of CP110 and the autophagosome marker LC3.Knockout of NudCL2 induces defects in the removal of CP110 from mother centrioles and ciliogenesis,which are rescued by wild-type NudCL2 but not its UR motif mutant.Knockdown of CP110 significantly attenuates ciliogenesis defects in NudCL2-deficient cells.In addition,NudCL2 morphants exhibit ciliation-related phenotypes in zebrafish,which are reversed by wild-type NudCL2,but not its UR motif mutant.Importantly,CP110 depletion significantly reverses these ciliary phenotypes in NudCL2 morphants.Taken together,our data suggest that NudCL2 functions as an autophagy receptor mediating the selective degradation of mother centriole-capping CP110 to promote ciliogenesis,which is indispensable for embryo development in vertebrates.Min Liu Wen Zhang Min Li Jiaxing Feng Wenjun Kuang Xiying Chen Feng Yang Qiang Sun Zhangqi Xu Jianfeng Hua Chunxia Yang Wei Liu Qiang Shu Yuehong Yang Tianhua Zhou Shanshan Xie 2021Cell Research2021,31,11:1
7Exosomal transfer of microRNA-590-3p between renal tubular epithelial cells after renal ischemia-reperfusion injury regulates autophagy by targeting TRAF6显示文摘Background:Acute kidney injury(AKI)is a common complication in patients,especially elderly patients,who undergo cardiac surgery with cardiopulmonary bypass.Studies have indicated a protective role of autophagy in AKI.However,the mechanisms underlying the regulatory effect of autophagy in AKI among patients undergoing cardiac surgeries are poorly understood.In this study,we aimed to test the hypothesis that exosomal microRNAs(miRNAs)regulate autophagy in tubular epithelial cells after AKI.Methods:Plasma exosomal RNA was extracted from young and elderly AKI patients undergoing cardiac surgery,and the miRNAs expression during the perioperative period were analyzed using next-generation sequencing.The screened miRNAs and their target genes were subjected to gene oncology function and Kyoto Encyclopedia of Genes and Genome enrichment analyses.Renal tubular epithelial cell line(HK-2 cells)was cultured and hypoxia/reoxygenation(H/R)model was established,which is an in vitro renal ischemia/reperfusion(I/R)model.We used Western blot analysis,cell viability assay,transfection,luciferase assay to investigate the mechanisms underlying the observed increases in the levels of renal I/R injury-mediated exosomal miRNAs and their roles in regulating HK-2 cells autophagy.Results:miR-590-3p was highly enriched in the plasma exosomes of young AKI patients after cardiac surgery.Increased levels of miR-590-3p led to the increases in the expression of autophagy marker proteins,including Beclin-1 and microtubule associated protein 1 light chain 3 beta(LC3II),and prolonged the autophagic response in HK-2 cells after H/R treatment.These effects were achieved mainly via increases in the exosomal miR-590-3p levels,and the tumor necrosis factor receptor-associated factor 6 protein was shown to play a key role in I/R injury-mediated autophagy induction.Conclusion:Exosomes released from HK-2 cells after renal I/R injury regulate autophagy by transferring miR-590-3p in a paracrine manner,which suggests that increasing the miR-590-3p levels in HK-2 cell-derived exosomes may increase autophagy and protect against kidney injury after renal I/R injury.Yimeng Chen Congya Zhang Yingjie Du Xiying Yang Min Liu Wenjing Yang Guiyu Lei Guyan Wang 2022Chinese Medical Journal2022,135,20:1

12Effect of Treated Graphite on Properties of Al_2O_3-SiC-C Castables显示文摘Al2O3-SiC-C castables with 2% pitch or 2%,4% and 6% treated graphite,respectively,were prepared based on the basic formulation of traditional Al2O3-SiC-C castables. The flowability of castables,and bulk density,volume stability,strength,oxidation resistance and slag resistance of specimens fired at 110 ℃,1 100 ℃ and 1 500 ℃ respectively were comparatively studied. 13Effects of Different Protein Energy Ratios on Fattening Performance,Slaughter Performance and Meat Quality of Pigs显示文摘[Objectives]Protein energy ratio refers to the proportional relationship between protein and energy levels in animal diets,i.e.,the grams of crude protein corresponding to every megacalorie of energy,which is generally expressed as CP:ME or CP:DE.This study was conducted to investigate the effects of different diets on fattening and slaughter performance and meat quality traits for'L(Large Yorkshire)×L(Landrace)'crossbred pigs.[Methods]Eighteen piglets of L×L crossbred with similar body weights about 51 kg were selected.The piglets were divided into 3 groups randomly and each group was assigned to 3 replicates with 2 piglets in a replicate.Group A was fed diet Ⅰ (control diet),group B was fed the same diet of group A in the first month of the trial but fed diet Ⅱ in second month,and group C consumed diet Ⅲ.All the pigs were fed in the same feeding condition for two months except the different diets during the trial.One pig in similar body weight from each replicate was slaughtered for the determination of slaughter performance and meat quality traits in the end of the trial.[Results]The average daily feed intake(ADFI)of pigs from group B was only(2.32±0.52)kg and significantly lower than those from group A and group C(P<0.05),but no difference was found in average daily gain(ADG) and feed/gain(P>0.05).Also,no differences occurred in the carcass length,back fat thickness,longissimus muscle (LM) area and dressing percentage(P>0.05).In addition,no differences were found in the meat quality traits of shear force,meat color,pH_(45 min),pH_(24 h) and cooking loss(P>0.05).However,the water-holding capacity of meat from group C was(2.58±0.02)ms and significantly lower than that of(2.80±0.20)ms from group A(P<0.05).Although the contents of glutamic acid and cystine in LM from group B was significantly lower than those from group A and group C(P<0.05),no differences occurred in the contents of other amino acids,the total amino acid and total flavor amino acid among the three groups(P>0.05).However,the inosine monophosphate content of LM from group C was only(331.80±11.53)mg/100 g and significantly lower than those of(361.00±6.36)and(366.37±4.80)mg/100 g from group A and B(P<0.05).Even though no differences were found in the contents of DM and CP in LM among the three groups of pigs,the content of intramuscular fat(IMF)in LM from group B and group C was increased by 45.6%and 46.58%respectively from that of group A(P<0.05),but no difference occurred between group B and C(P>0.05).[Conclusions]DietsⅡ and Ⅲ in this study caused no differences in fattening and slaughter performance of L×L crossbred pigs,but the effects on some meat traits were still significant,especially on the improvement of intramuscular fat in experimental pigs.Therefore,they could improve the meat quality of crossbred pigs to a certain extent.Cui YANG Rongsheng JIANG Yongshao WU Xiying YANG Haihong LIAO Qian QIN Shishan QIN Ying ZHANG Jiahuang YANG Limei QIN Fulin WANG 2023Agricultural Biotechnology2023,12,2:0

15The influence of Bi content on dielectric properties of Bi_(4-x)Ti_(3)O_(12-1.5x) ceramics显示文摘A kind of lead-free dielectric materials,such as the bismuth layered perovskite-type structure of Bi4-xTi3O12-1.5x(x=0.04,0.02,0,-0.02,-0.04),was prepared by the conventional solid-state method at 800C and sintered at 1100C.The variation of structure and electrical properties with different Bi concentration was studied.All the Bi4-xTi3O12-1.5x(x=0.04,0.02,0,-0.02,-0.04)samples exhibited a single structured phase.SEM could be a better approach to present the microstructure of Bi4-xTi3O12-1.5x(x=0.04,0.02,0,-0.02,-0.04)ceramics.It could be found that the grain size of Bi4.02Ti3O12.03 sintered at 1100C was smaller than that of others among the five samples through grain size mechanics.Through impedance spectra analysis,we knew,when the Bi content was fixed,that the dielectric constant and the loss values increased with the decrease of frequency.The Curie temperature of the five samples was about 670C.In particular,while at the frequency of 100 kHz,the lowest loss was 0.001 when Bi content was 3.98.The Bi4.02Ti3O12.03 ceramics with the minimum grain size had highest dielectric constant and the relatively low loss.Due to its high Curie temperature,high permittivity and low loss,the Bi_(4)Ti_(3)O_(12)(BIT)ceramics have a broad application prospect in high density memory,generator,sensor,ferroelectric tunnel junctions and so on.Hui Gong Xiying Ke Shuqin Yang Zhaozhi Li Huyin Su Cancan Zhang Qian Luo Qi Chen Lingfang Xu Yong Chen Wei Wang 2017Journal of Advanced Dielectrics2017,7,3:0
16Identification and characterization of stem cells in mammalian esophageal stratified squamous epithelia显示文摘Somatic stem cells are essential for the maintenance of tissue homeostasis. Despite its importance, how the esophageal stratified squamous epithelium executes its self-renewal and maintenance remains elusive. In this study, using 5-bromo-2′-deoxyuridine label-chase in rats in vivo and rat esophageal organoids in vitro together with genome-wide DNA methylation and single-cell RNA sequencing, we identified a slow-cycling/quiescent stem cell population that contained high levels of hemidesmosomes (HDs) and low levels of Wnt signaling localized spatially and randomly at the basal layer of the esophageal epithelium. Pseudotime cell trajectory analysis indicated that tissue cells originated from quiescent basal stem cells in the basal layer. Perturbations of HD component expression and/or Wnt signaling reduced the stem cell population in the basal layer of esophageal keratinocyte organoids, resulting in alterations in the organoid formation rate, size, morphogenesis, and proliferation–differentiation homeostasis. Furthermore, not only high levels of HDs and low levels of Wnt signaling but also an interplay between HD and Wnt signaling defined the stem cells of the basal layer.
