2Phytophthora sojae Effector PsAvh240 Inhibits Host Aspartic Protease Secretion to Promote Infection显示文摘Plants secrete defense molecules into the extracellular space (the apoplast) to combat attacking microbes. However, the mechanisms by which successful pathogens subvert plant apoplastic immunity remain poorly understood. In this study, we show that PsAvh240, a membrane-localized effector of the soybean pathogen Phytophthora sojae, promotes P. sojae infection in soybean hairy roots. We found that PsAvh240 interacts with the soybean-resistant aspartic protease GmAP1 in planta and suppresses the secretion of GmAP1 into the apoplast. By solving its crystal structure we revealed that PsAvh240 contain six a helices and two WY motifs. The first two a helices of PsAvh240 are responsible for its plasma membrane-localization and are required for PsAvh240's interaction with GmAP1. The second WY motifs of two PsAvh240 molecules form a handshake arrangement resulting in a handshake-like dimer. This dimerization is required for the effector's repression of GmAP1 secretion. 5Effects of Different Extraction and Purification Methods on the Acquisition of Phycoerythrin from Porphyridium purpureum显示文摘Phycoerythrin, as the main light-harvesting antenna in Porphyridium purpureum, exists at the outermost end of the phycobilisome. It has advantages of good fluorescence intensity, anti-oxidation, scavenging free radicals, and high chroma, so it has been widely used in food, cosmetics, pharmaceuticals, and other industries. In this study, the effects of different extraction(ultrasonic breaking method, bead grinding method, liquid nitrogen grinding method, and freezing-thawing method) and purification methods(salting out method, ultrafiltration method, and combination of salting out and ultrafiltration method) on the acquisition of phycoerythrin from P. purpureum were studied, and the characteristics of phycoerythrin in the P. purpureum were identified. The results showed that the freezing-thawing method could extract phycoerythrin from the powder of P. purpureum to the utmost extent, and the concentration of the extracted phycoerythrin was up to 0.036 g/L. The salting out method could most effectively purify phycoerythrin, and the purity index was 2.216. The identification of phycoerythrin by ultraviolet absorption spectroscopy and fluorescence spectroscopy indicated that the phycoerythrin had the maximum absorption peak at 545 nm, and the maximum Stokes shift was up to 79 nm. 6Zeta Potential and Rheological Behavior of ULC Castables Matrix显示文摘Effects of starting materials and four dispersants(STP,SHP,FDN and FS60) on Zeta potential and rheological behavior of alumina based ULC castables matrix were investigated.The results show that:characteristics of silica fume and alumina cements play a very important role in Zeta potential and viscosity of suspensions of the castables matrix;the dispersants STP and SHP can change Zeta potential values of the matrix suspensions remarkably;the four dispersants can effectively improve the rheological properties of matrix suspensions.For the point of lower viscosity of the matrix suspensions,the suitable additions of the three dispersants(SHP,FDN and FS60) are about 0.2% while that of STP is about 0.3%.XU Yuanchao WANG Zhanmin CAO Xiying 2008China's Refractories2008,17,2:1

8Recent advances in RNA structurome显示文摘RNA structures are essential to support RNA functions and regulation in various biological processes. Recently, a range of novel technologies have been developed to decode genome-wide RNA structures and novel modes of functionality across a wide range of species. In this review, we summarize key strategies for probing the RNA structurome and discuss the pros and cons of representative technologies. In particular, these new technologies have been applied to dissect the structural landscape of the SARS-CoV-2 RNA genome. We also summarize the functionalities of RNA structures discovered in different regulatory layers-including RNA processing, transport, localization, and mRNA translation-across viruses, bacteria, animals, and plants. We review many versatile RNA structural elements in the context of different physiological and pathological processes(e.g., cell differentiation, stress response, and viral replication). 9Fusarium-produced vitamin B6 promotes the evasion of soybean resistance by Phytophthora sojae显示文摘Plants can be infected by multiple pathogens concurrently in natural systems. However,pathogen–pathogen interactions have rarely been studied. In addition to the oomycete Phytophthora sojae, fungi such as Fusarium spp. also cause soybean root rot. In a 3-year field investigation, we discovered that P. sojae and Fusarium spp. frequently coexisted in diseased soybean roots. Out of 336 P. sojae–soybean–Fusarium combinations,more than 80% aggravated disease. Different Fusarium species all enhanced P. sojae infection when co-inoculated on soybean. Treatment with Fusarium secreted non-proteinaceous metabolites had an effect equal to the direct pathogen coinoculation. By screening a Fusarium graminearum mutant library, we identified Fusarium promoting factor of Phytophthora sojae infection 1(Fpp1),encoding a zinc alcohol dehydrogenase. Fpp1 is functionally conserved in Fusarium and contributes to metabolite-mediated infection promotion, in which vitamin B6(VB6) produced by Fusarium is key. Transcriptional and functional analyses revealed that Fpp1 regulates two VB6 metabolism genes, and VB6 suppresses expression of soybean disease resistance-related genes. 11Flat-Panel Laser Displays Based on Liquid Crystal Microlaser Arrays显示文摘Laser displays,benefiting from the characteristic merits of lasers,have led to the revolution of next-generation display technologies owing to their superior color expression.However,the acquisition of pixelated laser arrays as self-emissive panels for flat-panel laser displays remains challenging.Liquid crystal(LC)materials with excellent processability and optoelectronic properties offer considerable potential for the construction of highly ordered multicolor laser arrays.Here,we demonstrate flat-panel laser displays on LC microlaser pixel arrays through a microtemplate-assisted inkjet printing method.Individual organic red-green-blue(RGB)microlaser pixel arrays were obtained by doping dyes into LCs with photonic band edges to obtain single-mode RGB lasing,leading to a much broader color gamut,compared with the standard RGB color space.Then we acquired periodically patterned RGB pixel matrices by positioning LC microlasers precisely into highly ordered arrays,according to the well-organized geometry of the microtemplates.Subsequently,we demonstrated full-color flat-panel laser displays using the LC microlaser pixel matrices as self-emissive panels.These results provide valuable enlightenment for the construction of next-generation flat-panel laser display devices.Fa Feng Xu Yong Jun Li Yuanchao Lv Haiyun Dong Xianqing Lin Kang Wang Jiannian Yao Yong Sheng Zhao 2020CCS Chemistry2020,2,6:1
12Chromosome-scale genome assembly of Cucumis hystrix-a wild species interspecifically cross-compatible with cultivated cucumber显示文摘Cucumis hystrix Chakr.(2n=2x=24)is a wild species that can hybridize with cultivated cucumber(C.sativus L.,2n=2x=14),a globally important vegetable crop.However,cucumber breeding is hindered by its narrow genetic base.Therefore,introgression from C.hystrix has been anticipated to bring a breakthrough in cucumber improvement.Here,we report the chromosome-scale assembly of C.hystrix genome(289 Mb).Scaffold N50 reached 14.1 Mb.Over 90%of the sequences were anchored onto 12 chromosomes.A total of 23,864 genes were annotated using a hybrid method.Further,we conducted a comprehensive comparative genomic analysis of cucumber,C.hystrix,and melon(C.melo L.,2n=2x=24).Whole-genome comparisons revealed that C.hystrix is phylogenetically closer to cucumber than to melon,providing a molecular basis for the success of its hybridization with cucumber.Moreover,expanded gene families of C.hystrix were significantly enriched in“defense response,”and C.hystrix harbored 104 nucleotide-binding site-encoding disease resistance gene analogs.Furthermore,121 genes were positively selected,and 12(9.9%)of these were involved in responses to biotic stimuli,which might explain the high disease resistance of C.hystrix.The alignment of whole C.hystrix genome with cucumber genome and self-alignment revealed 45,417 chromosome-specific sequences evenly distributed on C.hystrix chromosomes.Finally,we developed four cucumber-C.hystrix alien addition lines and identified the exact introgressed chromosome using molecular and cytological methods.The assembled C.hystrix genome can serve as a valuable resource for studies on Cucumis evolution and interspecific introgression breeding of cucumber.Xiaodong Qin Zhonghua Zhang Qunfeng Lou Lei Xia Ji Li Mengxue Li Junguo Zhou Xiaokun Zhao Yuanchao Xu Qing Li Shuqiong Yang Xiaqing Yu Chunyan Cheng Sanwen Huang Jinfeng Chen 2021Horticulture Research2021,8,1:0

14Hormones and carbohydrates synergistically regulate the formation of swollen roots in a Chinese cabbage translocation line显示文摘The genus Brassica contains a rich diversity of species and morphological types,including leaf,root,and oil crops,all of which show substantial phenotypic variation.Both Chinese cabbage and cabbage are typical leaf-type crops with normal roots.We created translocation lines based on interspecific crosses between Chinese cabbage and cabbage and identified qdh225,which exhibited a swollen-root phenotype.The swollen root of qdh225 contained a large number of granular substances,and the formation of its irregular morphological tissue was caused by a thickening of the phloem.Transcriptomic and metabolomic data suggested that differential expression of genes encoding nine types of enzymes involved in starch and sucrose metabolism caused changes in starch synthesis and degradation in the swollen root.These genes jointly regulated sucrose and starch levels,leading to significant enrichment of starch and soluble proteins in the swollen root and a reduction in the content of soluble sugars such as d-glucose and trehalose 6-phosphate.A significant increase in auxin(IAA)and abscisic acid(ABA)contents and a decrease in gibberellin(GA)content in the swollen root likely promoted the differential expression of genes associated with hormone signal transduction,thereby regulating the development of the swollen root.Taken together,our data suggest that accumulation of IAA and ABA and reduction in GA promote swollen root formation by regulating hormone-mediated signaling,leading to a thickening of phloem,root enlargement,and substantial accumulation of starch and soluble proteins.The latter provide materials,energy,and nutrient sources for the development of swollen roots.Xiaojing Ren Wei Ma Shuxin Xuan Dandan Li Yanhua Wang Yuanchao Xu Daling Feng Jianjun Zhao Xueping Chen Shuangxia Luo Shuxing Shen Aixia Gu 2023Horticulture Research2023,10,8:0

16The Phytophthora effector Avh94 manipulates host jasmonic acid signaling to promote infection显示文摘The oomycete pathogen Phytophthora sojae is a causal agent of soybean root rot.Upon colonization of soybeans,P.sojae secretes various RXLR effectors to suppress host immune responses,supporting successful infection.Previous research has demonstrated that the RXLR effector Avh94 functions as a virulence effector,but the molecular mechanism underlying its role in virulence remains unknown.Here,we demonstrate that Avh94 overexpression in plants and pathogens promotes Phytophthora infection.Avh94 interacts with soybean JAZ1/2,which is a repressor of jasmonic acid(JA)signaling.Avh94 stabilizes JAZ1/2 to inhibit JA signaling and silencing of JAZ1/2 enhances soybean resistance against P.sojae.Moreover,P.sojae lines overexpressing Avh94 inhibit JA signaling.Furthermore,exogenous application of methyl jasmonate improves plant resistance to Phytophthora.Taken together,these findings suggest that P.sojae employs an RXLR effector to hijack JA signaling and thereby promote infection.Yao Zhao Bo Yang Huawei Xu Jinbin Wu Zhiyang Xu Yuanchao Wang 2022Journal of Integrative Plant Biology2022,64,11:0

18Genetic map construction and functional characterization of genes within the segregation distortion regions(SDRs)in the F_(2:3) populations derived from wild cotton species of the D genome显示文摘Background:Segregation distortion(SD)is a common phenomenon among stable or segregating populations,and the principle behind it still puzzles many researchers.The F2:3 progenies developed from the wild cotton species of the D genomes were used to investigate the possible plant transcription factors within the segregation distortion regions(SDRs).A consensus map was developed between two maps from the four D genomes,map A derived from F2:3 progenies of Gossypium klotzschianum and G.davidsonii while Map B from G.thurberi and G.trilobum F2:3 generations.In each map,188 individual plants were used.Results:The consensus linkage map had 1492 markers across the 13 linkage groups with a map size of 1467.445 cM and an average marker distance of 1.0370 cM.Chromosome D502 had the highest percentage of SD with 58.6%,followed by Chromosome D507 with 47.9%.Six thousand and thirty-eight genes were mined within the SDRs on chromosome D502 and D507 of the consensus map.Within chromosome D502 and D507,2308 and 3730 genes were mined,respectively,and were found to belong to 1117 gourp out of which 622 groups were common across the two chromosomes.Moreover,genes within the top 9 groups related to plant resistance genes(R genes),whereas 188 genes encoding protein kinase domain(PF00069)comprised the largest group.Further analysis of the dominant gene group revealed that 287 miRNAs were found to target various genes,such as the gra-miR398,gramiR5207,miR164a,miR164b,miR164c among others,which have been found to target top-ranked stress-responsive transcription factors such as NAC genes.Moreover,some of the stress-responsive cis-regulatory elements were also detected.Furthermore,RNA profiling of the genes from the dominant family showed that higher numbers of genes were highly upregulated under salt and osmotic stress conditions,and also they were highly expressed at different stages of fiber development.Conclusion:The results indicated the critical role of the SDRs in the evolution of the key regulatory genes in plants.KIRUNGU Joy Nyangasi MAGWANGA Richard Odongo SHIRAKU Margaret Linyerera LU Pu MEHARI Teame Gereziher XU Yuanchao HOU Yuqing AGONG Stephen Gaya ZHOU Yun CAI Xiaoyan ZHOU Zhongli WANG Kunbo LIU Fang 2020Journal of Cotton Research2020,3,4:0
