1CapOX as neoadjuvant chemotherapy for locally advanced operable colon cancer patients: a prospective single-arm phase Ⅱ trial显示文摘Objective: The aim of this prospective, single-arm phase II trial was to confirm the safety and efficacy of neoadjuvant chemotherapy(NAC) using oxaliplatin plus capecitabine(Cap OX) for patients with operable locally advanced colon cancer(CC).Methods: Patients with computed tomography-defined T4 or lymph node-positive CCs were enrolled. After radiological staging, patients were treated with at least 2 cycles of NAC consisting of 130 mg/m2 oxaliplatin on d 1,plus 1,000 mg/m2 capecitabine twice daily for 14 d every 3 weeks, followed by surgery, and then with the rest cycles of adjuvant chemotherapy. Radiological response was evaluated after 2 cycles of NAC. Tumor response, treatment toxicity, and surgical complications were recorded. The pathological response to therapy was evaluated according to the tumor regression grade(TRG) score. The primary endpoint was pathologic tumor response. This trial is registered in Clinical Trials.gov(No: NCT02415829).Results: Forty-seven patients were enrolled in the study. Forty-two patients completed the planned treatments.The total radiological response rate was 68%(32/47), including complete and partial response rates of 2%(1/47)and 66%(31/47), respectively. Stable disease was observed in 32%(15/47) and progressive disease was observed in none. Complete pathologic response, major regression, and at least moderate regression were achieved in 1(2%), 2(4%), and 29(62%) patients, respectively. Four patients developed grade 3 treatment toxicities. One patient with wound infection occurred after operation(1/47, 2%). 2Combination of anti-PD-1 antibody with P-GEMOX as a potentially effective immunochemotherapy for advanced natural killer/T cell lymphoma显示文摘Advanced natural killer/T cell lymphoma(NKTL)has demonstrated poor prognosis with currently available therapies.Here,we report the efficacy of anti-programmed death 1(PD-1)antibody with the P-GEMOX(pegaspargase,gemcitabine,and oxaliplatin)regimen in advanced NKTL.Nine patients underwent six 21-day cycles of anti-PD-1 antibody(day 1),pegaspargase 2000 U/m^(2)(day 1),gemcitabine 1 g/m^(2)(days 1 and 8)and oxaliplatin 130 mg/m^(2)(day 1),followed by anti-PD-1 antibody maintenance every 3 weeks.Programmed death-ligand 1(PD-L1)expression and genetic alterations were determined in paraffin-embedded pretreatment tissue samples using immunohistochemistry and next-generation sequencing(NGS)analysis.Responses were assessed using 18F-fluorodeoxyglucose positron emission tomography(18FDG-PET)and computed tomography or magnetic resonance imaging.Eight patients exhibited significant responses,comprising of seven complete remissions and one partial remission(overall response rate:88.9%).After a median follow-up of 10.6 months,6/9 patients(66.7%)remained in complete remission.The most common grade 3/4 adverse events were anemia(33.3%),neutropenia(33.3%),and thrombocytopenia(33.3%);all of which were manageable and resolved.Immunochemotherapy produced a high response rate in patients with positive PD-L1 expression(5/6,83.3%).NGS analysis suggested that STAT3/JAK3/PD-L1 alterations and ARID1A mutation were associated with immunochemotherapy efficacy.Mutation in DDX3X and alteration in epigenetic modifiers of KMT2D,TET2,and BCORL1 might indicate a poor response to immunochemotherapy.In conclusion,the anti-PD-1 antibody plus P-GEMOX regimen demonstrated promising efficacy in advanced NKTL.PD-L1 expression combined with specific genetic alterations could be used as potential biomarkers to predict therapeutic responses to immunochemotherapy.Jun Cai Panpan Liu Huiqiang Huang Yajun Li Shuyun Ma Hui Zhou Xiaopeng Tian Yuchen Zhang Yan Gao Yi Xia Xuanye Zhang Hang Yang Lirong Li Qingqing Cai 2020Signal Transduction and Targeted Therapy2020,5,1:6
3Vertical Distribution Characteristics of PM2.5 Observed by a Mobile Vehicle Lidar in Tianjin, China in 2016显示文摘We present mobile vehicle lidar observations in Tianjin, China during the spring, summer, and winter of 2016. Mobile observations were carried out along the city border road of Tianjin to obtain the vertical distribution characteristics of PM_(2.5). Hygroscopic growth was not considered since relative humidity was less than 60% during the observation experiments. PM_(2.5) profile was obtained with the linear regression equation between the particle extinction coefficient and PM_(2.5) mass concentration. In spring, the vertical distribution of PM_(2.5) exhibited a hierarchical structure. In addition to a layer of particles that gathered near the ground, a portion of particles floated at 0.6–2.5-km height. In summer and winter, the fine particles basically gathered below 1 km near the ground. In spring and summer, the concentration of fine particles in the south was higher than that in the north because of the influence of south wind. In winter, the distribution of fine particles was opposite to that measured during spring and summer. High concentrations of PM_(2.5) were observed in the rural areas of North Tianjin with a maximum of 350 μg m–3 on 13 December2016. It is shown that industrial and ship emissions in spring and summer and coal combustion in winter were the major sources of fine particles that polluted Tianjin. 4Genome-wide CRISPR screen identifies synthetic lethality between DOCK1 inhibition and metformin in liver cancer显示文摘Metformin is currently a strong candidate anti-tumor agent in multiple cancers.However,its anti-tumor effectiveness varies among different cancers or sub-populations,potentially due to tumor heterogeneity.It thus remains unclear which hepatocellular carcinoma(HCC)patient subpopulation(s)can benefit from met-formin treatment.Here,through a genome-wide CRISPR-Cas9-based knockout screen,we find that DOCK1 levels determine the anti-tumor effects of met-formin and that DOCK1 is a synthetic lethal target of metformin in HCC.Mechanistically,metformin promotes DOCK1 phosphorylation,which activates RAC1 to facilitate cell survival,leading to metformin resistance.The DOCK1-selective inhibitor,TBOPP,potentiates anti-tumor activity by metformin in vitro in liver cancer cell lines and patient-derived HCC organoids,and in vivo in xenografted liver cancer cells and immunocompetent mouse liver cancer models.Notably,metformin improves overall survival of HCC patients with low DOCK1 levels but not among patients with high DOCK1 expression.This study shows that metformin effective-ness depends on DOCK1 levels and that combining metformin with DOCK1 inhibition may provide a promising personalized therapeutic strategy for met-formin-resistant HCC patients.Junru Feng Hui Lu Wenhao Ma Wenjing Tian Zhuan Lu Hongying Yang Yongping Cai Pengfei Cai Yuchen Sun Zilong Zhou Jiaqian Feng Jiazhong Deng Ying Shu Kun Qu Weidong Jia Ping Gao Huafeng Zhang 2022Protein & Cell2022,13,11:5
5Controllable synthesis of nitrogen-doped porous carbon from metal-polluted miscanthus waste boosting for supercapacitors显示文摘High-value reclamation of metal-polluted plants involved in phytoremediation is a big challenge.In this study,nitrogen-doped nanoporous carbon with large specific area of 2359.1 m^(2)g^(-1) is facilely fabricated from metal-polluted miscanthus waste for efficient energy storage.The synergistic effect of KOH,urea and ammonia solution greatly improve the nitrogen quantity and surface area of the synthesized carbon.Electrodes fabricated with this carbon exhibit the excellent capacitance performance of 340.2 F g^(-1) at 0.5 A g^(-1) and a low combined resistance of 0.116Ω,which are competitive with most of previously reported carbon-based electrodes.In addition,the as-obtained carbon electrode shows a high specific capacitance retention of over 99.6%even after 5000 cycles.Furthermore,the symmetric supercapacitor fabricated using the synthesized carbon achieves a superior energy density of 25.3 Wh kg^(-1)(at 400 W kg^(-1))in 1 mol L^(-1) Na_(2)SO_(4)aqueous solution.This work provides an efficient route to upcycle metal-polluted plant waste for supercapacitor applications.Zuo Chen Man Zhang Yuchen Wang Zhiyu Yang Di Hu Yetao Tang Kai Yan 2021Green Energy & Environment2021,6,6:4
6Probabilistic Anomaly Detection Approach for Data-driven Wind Turbine Condition Monitoring显示文摘Continuous monitoring of wind turbine(WT)opera-tion can improve the reliability of the wind turbine and lower the operation and maintenance costs.To improve the condition mon-itoring(CM)and fault detection performance on WTs,this paper proposes an artificial intelligence-based probabilistic anomaly detection approach that can not only provide a deterministic estimation of the WT condition but also evaluate the uncertainties associated with the estimation.An abnormal WT condition is detected based on the evaluated uncertainties,to provide a noise-free incipient fault indication.Compared to the conventional deterministic CM approaches with a residual-based anomaly detection criterion,the proposed probabilistic approach tends to accurately detect the faults earlier,which allows more time for maintenance scheduling to prevent WT component failure.The early fault detection ability of the proposed approach was verified on an operational WT in China.Yuchen Zhang Meng Li Zhao Yang Dong Ke Meng 2019CSEE Journal of Power and Energy Systems2019,5,2:3

8Shape and position of Earth's bow shock near-lunar orbit based on ARTEMIS data显示文摘Earth's bow shock is the result of interaction between the supersonic solar wind and Earth's magnetopause. However, data limitations mean the model of the shape and position of the bow shock are based largely on near-Earth satellite data. The model of the bow shock in the distant magnetotail and other factors that affect the bow shock, such as the interplanetary magnetic field(IMF) B_y, remain unclear. Here, based on the bow shock crossings of ARTEMIS from January 2011 to January 2015, new coefficients of the tail-flaring angle a of the Chao model(one of the most accurate models currently available) were obtained by fitting data from the middle-distance magnetotail(near-lunar orbit, geocentric distance -20R_E>X>-50R_E). In addition, the effects of the IMF B_y on the flaring angle a were analyzed. Our results showed that:(1) the new fitting coefficients of the Chao model in the middle-distance magnetotail are more consistent with the observed results;(2) the tail-flaring angle a of the bow shock increases as the absolute value of the IMF B_y increases. Moreover, positive IMF B_y has a greater effect than negative IMF B_y on flaring angle. 10Preparation and characterization of pure SiC ceramics by high temperature physical vapor transport induced by seeding with nano SiC particles显示文摘High temperature physical vapor transport(HTPVT) was employed to grow polycrystalline SiC ceramics with high density and purity, induced by nano SiC particles as seeds. The obtained SiC ceramics were identified as 6 H-SiC with a mainly preferred orientation along the(0 0 0 6) plane, and an obvious refinement of grain size was demonstrated for the SiC seeds. Mean grain sizes of the obtained SiC ceramics were112 μm and 314 μm, by using the SiC seeds with the mean particle size of 50 nm and 500 nm, respectively,which were obviously smaller than that of the seed-free sample(960 μm). The samples obtained with the seeds also demonstrated enhanced flexural strength and hardness, attributing to the reduced mean grain size. Furthermore, SiC ceramics without seeds via HTPVT exhibited a high thermal conductivity of242 W·(m·K)^-1 at room temperature due to the highly preferred orientation. While the degree of preferred orientation of seed-induced samples was lower, the thermal conductivity of SiC ceramics induced by seeding still maintained at least 200 W·(m·K)^-1 at room temperature, this level was much higher than most other methods. 11Polydopamine-modified collagen sponge scaffold as a novel dermal regeneration template with sustained release of platelet-rich plasma to accelerate skin repair:A one-step strategy显示文摘Although employed to release growth factors(GFs)for regenerative medicine,platelet-rich plasma(PRP)has been hindered by issues like burst effect.Based on collagen sponge scaffolds(CSSs)modified with polydopamine(pDA),a novel dermal regeneration template(DRT)was designed.However,whether it could efficiently deliver PRP and even foster wound healing remained unclear.In this work,after PRP was prepared and pDA-modified CSSs(pDA-CSSs)were fabricated,microscopic observation,GFs release assay and in-vitro biological evaluations of pDA-CSSs with PRP(pDA-CSS@PRP)were performed,followed by BALA-C/nu mice full-thickness skin defects implanted with pDA-CSS@PRP covered by grafted skins(termed as a One-step strategy).As a result,scanning electron microscope demonstrated more immobilized platelets on pDA-CSS′surface with GFs’controlled release via enzyme-linked immunosorbent assay,compared with CSSs.In line with enhanced in-vitro proliferation,adhesion and migration of keratinocytes&endothelial cells,pDA-CSS@PRP were histologically revealed to accelerate wound healing with less scar via rapid angiogenesis,arrangement of more mature collagen,guiding cells to spread,etc.In conclusion,pDA-CSSs have potential to serve as a novel DRT capable of delivering PRP,which may foster full-thickness skin defect healing by means of a One-step strategy.Zijun Zheng Minxiong Li Pengwei Shi Yanbin Gao Jun Ma Yuchen Li Lei Huang Zhangfeifan Yang Lei Yang 2021Bioactive Materials2021,6,8:2
12Kidney injury molecule-1 is a potential receptor for SARS-CoV-2显示文摘COVID-19 patients present high incidence of kidney abnormalities,which are associated with poor prognosis and mortality.The identification of SARS-CoV-2 in the kidney of COVID-19 patients suggests renal tropism of SARS-CoV-2.However,whether there is a specific target of SARS-CoV-2 in the kidney remains unclear.Herein,by using in silico simulation,coimmunoprecipitation,fluorescence resonance energy transfer,fluorescein isothiocyanate labeling,and rational design of antagonist peptides,we demonstrate that kidney injury molecule-1(KIM1),a molecule dramatically upregulated upon kidney injury,binds with the receptor-binding domain(RBD)of SARS-CoV-2 and facilitates its attachment to cell membrane,with the immunoglobulin variable Ig-like(Ig V)domain of KIM1 playing a key role in this recognition.The interaction between SARS-CoV-2 RBD and KIM1 is potently blockaded by a rationally designed KIM1-derived polypeptide AP2.In addition,our results also suggest interactions between KIM1 Ig V domain and the RBDs of SARS-CoV and MERS-CoV,pathogens of two severe infectious respiratory diseases.Together,these findings suggest KIM1 as a novel receptor for SARS-CoV-2 and other coronaviruses.We propose that KIM1 may thus mediate and exacerbate the renal infection of SARS-CoV-2 in a‘vicious cycle’,and KIM1 could be further explored as a therapeutic target.Chen Yang Yu Zhang Xia Zeng Huijing Chen Dong Yang Yuchen Chen Ziwei Shen Xiaomu Wang Xinran Liu Mingrui Xiong Hong Chen Kun Huang 2021Journal of Molecular Cell Biology2021,13,3:2
13Self-Sensing Test Method for the Temperature of Piezoelectric Stacks显示文摘A self-sensing test method for the temperature of piezoelectric stack,based on the high correlation between the static capacitance and the stack temperature,is proposed in order to construct a self-sufficient methodology of temperature measurement. Firstly,a theoretical model of static capacitance of the piezoelectric stack under preload was set up,and the influence of preload on the static capacitance was analyzed. Secondly,the correctness of the model was verified by static capacitance test experiments under various preloading conditions. Finally, the temperature measurement experiments at low-temperature stage for two kinds of piezoelectric stacks,namely the lowtemperature-resistant piezoelectric stack and conventional piezoelectric stack, were conducted under various preloading conditions using a polynomial fitting method. The results,which validate the accuracy of the test method,show that the maximum temperature deviations of the two kinds of piezoelectric stack are 3.9 ℃ and 2.8 ℃,respectively,when the preload force is close to the specified value. The test method uses the piezoelectric stack itself as a temperature sensor,which does not require additional equipment for temperature sensing,so that the space and equipment cost could be economized. 14Broad temperature adaptability of vanadium redox flow battery-part4:Unraveling wide temperature promotion mechanism of bismuth for V^(2+)/V^(3+) couple显示文摘Vanadium flow battery(VFB)is a fast going and promising system for large-scale stationary energy storage.However,drawbacks such as low power density and narrow temperature window caused by poor catalytic activity of graphite felt(GF)electrodes limit its worldwide application.In this paper,bismuth,as a low-cost,no-toxic and high-activity electrocatalyst,is used to modify the thermal activated GF(TGF)via a facile hydrothermal method.Bismuth can effectively inhibit the side reaction of hydrogen evolution in wide temperature range,while promoting the V^(2+)/V^(3+)redox reaction.As a result,the VFB assembled with Bi/TGF as negative electrode demonstrates outstanding rate performance under the current density up to 400 mA cm^(-2),as well as a long-term stability over 600 charging/discharging cycles at a high current density of 150 mA cm^(-2).Moreover,it also shows excellent temperature adaptability from-10°C to50°C and high durability for life test at the temperature of 50°C.Yuchen Liu Feng Liang Yang Zhao Lihong Yu Le Liu Jingyu Xi 2018Journal of Energy Chemistry2018,27,5:2
15Native point defects and oxygen migration of rare earth zirconate and stannate pyrochlores显示文摘Various excellent properties of rare earth zirconate and stannate pyrochlores are close related with their native point defects.First-principles calculations are performed to systematically investigate the point defect mechanism and the oxygen diffusion behavior of A_(2)B_(2)O_(7)(A=La,Ce,Pr,Nd,Pm,Sm,Eu,Gd;B=Zr,Sn).The possible defect complexes and their associated reactions under stoichiometric and nonstoichiometric conditions are explored.The O Frenkel pairs are the most stable defect structure in stoichiometric zirconates,whereas the cation antisite defects are the predominant one in stoichiometric stannates.In the case of BO_(2) excess zirconates and stannates,the BA cation antisite defect with the A vacancy and/or the oxygen interstitial is energetically favorable,whereas the ABantisite defect together with the oxygen vacancy and/or the A interstitial is preferable under the A_(2)O_(3) excess condition.Meanwhile,the maximum point defect concentrations of zirconates are much higher than those of stannates.Furthermore,the oxygen migration barriers are similar in these compounds,ranging in 0.68 eV–0.80 eV.The predicted point defects and oxygen diffusion mechanisms play the critical role in their engineering applications and are expected to guide the future property improvement of pyrochlores through the control of point defects and/or composition.Juanli Zhao Yuchen Liu Yun Fan Wei Zhang Chengguan Zhang Guang Yang Hongfei Chen Bin Liu 2021Journal of Materials Science & Technology2021,,14:2
16Insights Into the Interfacial Degradation of High-Voltage All-Solid-State Lithium Batteries显示文摘Poly(ethylene oxide)(PEO)-based solid polymer electrolyte(SPE)is considered as a promising solid-state electrolyte for all-solid-state lithium batteries(ASSLBs).Nevertheless,the poor interfacial stability with high-voltage cathode materials(e.g.,LiCoO_(2))restricts its application in high energy density solid-state batteries.Herein,high-voltage stable Li_(3)AlF_(6) protective layer is coated on the surface of LiCoO_(2) particle to improve the performance and investigate the failure mechanism of PEO-based ASSLBs.The phase transition unveils that chemical redox reaction occurs between the highly reactive LiCoO_(2) surface and PEO-based SPE,resulting in structure collapse of LiCoO_(2),hence the poor cycle performance of PEO-based ASSLBs with LiCoO_(2) at charging voltage of 4.2 V vs Li/Li+.By sharp contrast,no obvious structure change can be found at the surface of Li_(3)AlF_(6)-coated LiCoO_(2),and the original layered phase was well retained.When the charging voltage reaches up to 4.5 V vs Li/Li+,the intensive electrochemical decomposition of PEO-based SPE occurs,leading to the constant increase of cell impedance and directly causing the poor performance.This work not only provides important supplement to the failure mechanism of PEO-based batter-ies with LiCoO_(2),but also presents a universal strategy to retain structure stability of cathode-electrolyte interface in high-voltage ASSLBs.Jiawen Li Yuchen Ji Haoran Song Shiming Chen Shouxiang Ding Bingkai Zhang Luyi Yang Yongli Song Feng Pan 2022Nano-Micro Letters2022,14,12:2
17亚热带丘陵区稻田周边氨气干沉降研究显示文摘农业源在氨(NH3)排放清单中占比较大,但农业排放的NH3是否全部进入大气亟待评估。本文选择亚热带丘陵区双季稻田为研究对象,研究了农田排放NH3的去向和沉降过程。结果表明,稻田(0.6 ha)下风向100 m范围较高的NH3浓度主要出现在施用基肥和追肥后15 d内。随着距稻田距离的增加,下风向大气中NH3浓度呈指数下降。稻田下风向100 m范围内NH3沉降量约占挥发量的80%,表明近源沉降是亚热带丘陵区稻田排放NH3的重要汇机制。该机制大幅减少了农业排放NH3进入大气的总量,应该在清单编制过程中予以考虑,以期客观评价农业排放对大气污染的潜在影响。YI Yuchen SHEN Jianlin YANG Chaodong WANG Juan LI Yong WU Jinshui 2020Atmospheric and Oceanic Science Letters2020,13,3:2

20Thermal evaluation of the injectable liquid metal bone cement in orthopedic treatment显示文摘The thermophysical properties of bone cement are important parameters for its application in the orthopedic treatment. This article focused on the thermal evaluation of the low-melting-point metal(Bi In Sn alloy), which has been proved to be an excellent bone cement. Firstly, the basic thermophysical properties of Bi In Sn alloys with different melting points were measured.Secondly, 15 fresh porcine femurs placed in the saline bath, bone cements with different melting points and amounts were injected into the bone cavities, respectively. Thermocouples were used to measure the temperature changes of the bone-cement interface and peripheral bone tissue. The possibility of thermal necrosis was evaluated. Moreover, a three-dimensional human knee model was built to numerically assess the effects of thermal parameters, such as melting point and latent heat on tissue temperature distribution. All the experimental and numerical results implied the heat distribution in the tissue depended on the thermal performances of liquid metal bone cement(LMBC). For LMBC of the same melting point, with increased amounts, the damage to the bone tissue is more severe, while for the same amount of different melting point LMBCs, with the higher melting point, which will lead to more serious damage to the tissue. Also, higher latent heat of LMBC has distinct longer solidification process, which may cause irreversible damage to surrounding tissues.
