| 2 | Study on gastric cancer blood plasma based on surface-enhanced Raman spectroscopy combined with multivariate analysis显示文摘A surface-enhanced Raman spectroscopy (SERS) method combined with multivariate analysis was developed for non-invasive gastric cancer detection. SERS measurements were performed on two groups of blood plasma samples: one group from 32 gastric patients and the other group from 33 healthy volunteers. Tentative assignments of the Raman bands in the measured SERS spectra suggest interesting cancer-specific biomolecular changes, including an increase in the relative amounts of nucleic acid, collagen, phospholipids and phenylalanine and a decrease in the percentage of amino acids and saccharide in the blood plasma of gastric cancer patients as compared with those of healthy subjects. Principal components analysis (PCA) and linear discriminant analysis (LDA) were employed to develop effective diagnostic algorithms for classification of SERS spectra between normal and cancer plasma with high sensitivity (79.5%) and specificity (91%). A receiver operating characteristic (ROC) curve was employed to assess the accuracy of diagnostic algorithms based on PCA-LDA. | 3 | A New Species of the Genus Xenophrys(Amphibia: Anura: Megophryidae) from Libo County, Guizhou, China显示文摘A new species of the genus Xenophrys is described from a karst cave environment of Libo County, Guizhou Province, Southern China. The new species, Xenophrys liboensis sp. nov., is distinguished from its congeners by a combination of the following characters: 1) tympanum distinct; 2) vomerine teeth present; 3) the length of lower arm and hand larger than the half of snout-vent; 4) heels overlapped slightly when the flexed legs are held at right angles to the body axis; 5) toe tips with rudimentary webs and without grooves; 6) dermal fringes moderate; 7) tubercles on the dorsum forming an X-shaped weak ridge; 8) horn-like tubercle at the edge of the upper eyelid distinct; 9) color of the iris in life is brown. In Bayesian phylogenetic analysis of 22 species of Xenophrys, all the individuals of X. liboensis sp. nov. clustered into a monophyletic clade with high posterior probabilities. In addition, the ranges of genetic divergences of X. liboensis sp. nov. with other species were interspecific rather than intraspecific. | 9 | The temporal dynamics of antimicrobial-resistant Salmonella enterica and predominant serovars in China显示文摘Salmonella enterica is one of the most common bacterial pathogens in humans and animals.Systematic studies on the trends and geographical distribution of antimicrobial-resistant Salmonella and dominant serovars have been well studied in European and American countries while not in China.Here,taking the One-Health strategy,we used>35000 Salmonella enterica isolates to explore the temporal and spatial dynamics of dominant serovars in China.We found that Salmonella Typhimurium was the dominant serovar causing human infection in China,which was consistent with Australia but inconsistent with North American and European countries.The proportion of Salmonella serovars Typhimurium,London,Rissen,Corvallis,Meleagridis,Kentucky,and Goldcoast showed an increasing trend during 2006-2019.We randomly selected 1962 isolates for comparative genomics and antimicrobial resistance studies and found that the number of antibiotic resistance genes(ARGs)per isolate increased 1.84 and 2.69 times of human and non-human origins,respectively,spanning 14 years.The proportion of antimicrobial-resistant Salmonella isolates had an increasing trend during 2006-2019,especially beta-lactam,quinolone,tetracycline,and rifampicin resistance.Moreover,we found that higher diversity of sequence types(STs)in S.Typhimurium than in other serovars,ST34 from pig and ST 19 from chicken origin,were mainly associated with isolates causing child and adult gastro-infection,respectively.Our results fill in the data gap on the trends of dominant serovars and antimicrobial resistance of Salmonella enterica in China.These data provide useful information for public health decision-makers prioritizing interventions for foodborne diseases and food safety. | Yanan Wang Yue Liu Na Lyu Zhiyuan Li Sufang Ma Demin Cao Yuanlong Pan Yongfei Hu Hua Huang George F.Gao Xuebin Xu Baoli Zhu | 2023 | National Science Review2023,10,3: | 4 |
| 10 | Research progress in artificial upwelling and its potential environmental effects显示文摘Artificial upwelling, as a geoengineering tool, has received worldwide attention because it may actualize ocean fertilization in a sustainable way, which could potentially alleviate the pressures on the fish stocks and human-driven climate change in the ocean. We reviewed the current knowledge on the development of an artificial upwelling system and its potential environmental effects. Special attention was given to the research progress on the air-lift concept artificial upwelling by Zhejiang University. The research on artificial upwelling over the past few decades has generated a range of devices that have been successfully applied in the field for months. Based on field experiments and the associated modeling results, part of them reported positive effects on increasing primary production and enhancing CO2 sequestration. However, as a significant disturbance to the environment, especially for large-scale applications, the uncertainties related to the potential effects on ecosystem remain unsolved. Zhejiang University has overcome the technical challenges in designing and fabricating a robust and high efficiency artificial upwelling device which has been examined in two field experiments in Qiandao Lake and one sea trial in the East China Sea. It was investigated that cold and hypoxic deep ocean water(DOW) could be uplifted to the euphotic layer, which could potentially change the nutrient distribution and adjust the N/P ratio. Both simulation and field experiments results confirmed that utilizing self-powered energy to inject compressed air to uplift DOW was a valid and efficient method. | 18 | A small molecule inhibitor of the UBE2F-CRL5 axis induces apoptosis and radiosensitization in lung cancer显示文摘Protein neddylation is catalyzed by a neddylation activating enzyme(NAE,E1),an E2 conjugating enzyme,and an E3 ligase.In various types of human cancers,the neddylation pathway is abnormally activated.Our previous study validated that the neddylation E2 UBE2F is a promising therapeutic target in lung cancer.Although the NAE inhibitor MLN4924/pevonedistat is currently under clinical investigation as an anti-cancer agent,there are no small molecules available that selectively target UBE2F.Here,we report,for the first time,the discovery,via structure-based virtual screen and chemical optimization,of such a small molecule,designated as HA-9104.HA-9104 binds to UBE2F,reduces its protein levels,and consequently inhibits cullin-5 neddylation.Blockage of cullin-5 neddylation inactivates cullin-RING ligase-5(CRL5)activity,leading to accumulation of the CRL5 substrate,NOXA,to induce apoptosis.Moreover,HA-9104 appears to form the DNA adduct via its 7-azaindole group to induce DNA damage and G2/M arrest.Biologically,HA-9104 effectively suppresses the growth and survival of lung cancer cells and confers radiosensitization in both in vitro cell culture and in vivo xenograft tumor models.In summary,we discovered a small molecule,designated HA-9104,that targets the UBE2F-CRL5 axis with anti-cancer activity alone or in combination with radiation. | Tiantian Xu Qisheng Ma Yanan Li Qing Yu Peichen Pan Yawen Zheng Zhijian Li Xiufang Xiong Tingjun Hou Bin Yu Hongmin Liu Yi Sun | 2022 | Signal Transduction and Targeted Therapy2022,7,11: | 2 |
| 19 | Marine bacteria inhibit corrosion of steel via synergistic biomineralization显示文摘Metal corrosion often results in incalculable economic loss and significant safety hazards. Although numerous traditional methods have been used to mitigate the issue, such as coating and corrosion inhibitors, they are environmentally unfriendly and difficult to maintain. Therefore, in this study, an environmental approach was taken to protect steels from corrosion in a multi-species bacterial environment via synergistic biomineralization. The marine bacterium Pseudoalteromonas lipolytica mixed with Bacillus subtilis or Pseudomonas aeruginosa strains offered extraordinary corrosion protection for steel.The surface characterization and electrochemical tests showed that the biomineralized film generated by the mixed bacteria was more compact and protective than that induced by a single bacterium. Herein,we found that the synergistic mechanisms were rather different for the different bacterial groups. For Pseudoalteromonas lipolytica and Bacillus subtilis group, the related mechanisms were due to the increase of pH in the medium, secretion of carbonic anhydrase. As for Pseudoalteromonas lipolytica and Pseudomonas aeruginosa group, the synergistic mechanism was attributed to the inhibiting corrosive bacteria in biofilm by the growth advantage of Pseudoalteromonas lipolytica.
