| 1 | Litterfall production and nutrient return in different-aged Chinese fir(Cunninghamia lanceolata) plantations in South China显示文摘The amounts of litter produced and nutrients returned play a fundamental role in the productivity and biogeochemical and nutrient cycling of forest ecosystems.We monitored annual litterfall production, nutrient return,and monthly dynamics over a one-year period in Chinese fir plantations aged 10, 22, and 34 years. Our objective was to quantify litterfall and nutrient return over a complete harvest rotation of Chinese fir. Annual litterfall production increased with stand age and was recorded as(3,294.6 ± 360.4),(3,733.9 ± 211.2), and(4,876.1 ± 212.8) kg ha-1a-1in stands aged 10, 22 and 34 years, respectively. Total litter production was significantly greater in the stand aged34 years than in the stand aged 10 years(p \ 0.05). With the exception of miscellaneous components, needle litterfall constituted the highest proportion(27.5–43.6 %), followed by branches/twigs(9.5–16.6 %). In all three plantations,annual total nutrient return to soil was in the order of C(1,119.95–2,709.05 kg ha-1a-1) [ N(39.32–62.04 kg ha-1a-1) [ K(15.95–22.44 kg ha-1a-1) [ P(1.30–1.63 kg ha-1a-1). C, N, K and P input to soil was significantly lower in the 10-year-old stand in comparison to the 22- and34-year-old stands(p \ 0.05). Litterfall production and nutrient return(C, N and K) followed similar patterns, and C and N input to soil was significantly related to litterfall production(needle, branch and total litterfall). | 2 | Celastrus orbiculatus extract induces mitochondrial-mediated apoptosis in human hepatocellular carcinoma cells显示文摘OBJECTIVE:To investigate the apoptotic effects and underlying molecular mechanisms of Celastrus orbiculatus(C.orbiculatus) extract in human hepatocellular carcinoma cells.METHODS:Human hepatocellular carcinoma cells(HCCLM6) were treated with C.orbiculatus extract(COE) at different nontoxic concentrations(10,20,40,80,and 160 μg/mL).The effect of COE on HC-CLM6 viability was examined using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide(MTT) assays.Cellular apoptosis following COE treatment was assessed by flow cytometry and western blot analysis.RESULTS:COE significantly inhibited cell viability and induced apoptosis of HCCLM6 cells in a dose-dependent manner.Apoptosis was accompanied by increased Bax expression and decreased Bcl-2 expression.In addition,COE treatment led to the release of cytochrome c,activation of caspase-3,and cleavage of poly(ADP-ribose) polymerase(PARP).Furthermore,activation of extracellular signal-regulated kinase(ERK),p38 kinase,and c-Jun N-terminal kinase(JNK) phosphorylation,and down-regulation of Akt phosphorylation was observed.CONCLUSION:COE induces mitochondrial-mediated,caspase-dependent apoptosis in HCCLM6 cells,which might be attributed to the activation of mitogen-activated protein kinase(MAPK) and inhibition of Akt signaling pathways.These data suggest that COE may be a potential treatment for human hepatocellular carcinoma. | Hua Zhang Yayun Qian Yanqing Liu Guoqing Li Pingfang Cui Yaodong Zhu Hui Ma Xue Ji Shiyu Guo Hisamits Tadashi | 2012 | Journal of Traditional Chinese Medicine2012,32,4: | 18 |

| 5 | Transcription factor ZmNAC126 plays an important role in transcriptional regulation of maize starch synthesis-related genes显示文摘Maize(Zea mays L.)is one of the most important food crops in the world,and starch is the main component of its endosperm.Transcriptional regulation plays a vital role in starch biosynthesis.However,it is not well understood in maize.We report the identification of the transcription factor ZmNAC126 and its role in regulation of starch synthesis in maize.Transcriptional expression of ZmNAC126 was higher in maize endosperm and kernels than in roots or stems.ZmNAC126 shared a similar expression pattern with starch synthesis genes during seed development,and its expression pattern was also consistent with the accumulation of starch.ZmNAC126 is a typical transcription factor with a transactivation domain between positions 201 and 227 of the amino acid sequence,is located in the nucleus,and binds to CACG repeats in vitro.Yeast one-hybrid assay revealed that ZmNAC126 bound the promoters of ZmGBSSI,ZmSSIIa,ZmSSIV,ZmISA1,and ZmISA2.Transient overexpression of ZmNAC126 in maize endosperm increased the activities of promoters pZmSh2,pZmBt2,pZmGBSSI,pZmSSIIIa,and pZmBT1 but inhibited the activities of pZmISA1 and pZmISA2.ZmNAC126 thus acts in starch synthesis by transcriptionally regulating targeted starch synthesis-related genes in maize kernels. | Qianlin Xiao Yayun Wang Hui Li Chunxia Zhang Bin Wei Yongbin Wang Huanhuan Huang Yangping Li Guowu Yu Hanmei Liu Junjie Zhang Yinghong Liu Yufeng Hu Yubi Huang | 2021 | The Crop Journal2021,9,1: | 5 |
| 6 | Establishment and Characterization of a Cell Based Artificial Antigen-Presenting Cell for Expansion and Activation of CD8^+ T Cells Ex Vivo显示文摘Artificial antigen-presenting cells are expected to stimulate the expansion and acquisition of optimal therapeutic features of T cells before infusion. Here CD32 that binds to a crystallizable fragment of IgG monoclonal antibody was genetically expressed on human K562 leukemia cells to provide a ligand for T-cell receptor. CD86 and 4-1BBL,which are ligands of co-stimulating receptors of CD28 and 4-1BB,respectively,were also expressed on K562 cells. Then we accomplished the artificial antigen-presenting cells by coupling K32/CD86/4-1BBL cell with OKT3 monoclonal antibody against CD3,named K32/CD86/4-1BBL/OKT3 cells. These artificial modified cells had the abilities of inducing CD8+ T cell activation,promoting CD8+ T cell proliferation,division,and long-term growth,inhibiting CD8+ T cell apoptosis,and enhancing CD8+ T cell secretion of IFN-γ and perforin. Furthermore,antigen-specific cytotoxic T lymphocytes could be retained in the culture stimulated with K32/CD86/4-1BBL/OKT3 cells at least within 28 days. | 7 | Stable Lithium-Carbon Composite Enabled by Dual-Salt Additives显示文摘Lithium metal is regarded as the ultimate negative electrode material for secondary batteries due to its high energy density.However,it suffers from poor cycling stability because of its high reactivity with liquid electrolytes.Therefore,continuous efforts have been put into improving the cycling Coulombic efficiency(CE)to extend the lifespan of the lithium metal negative electrode.Herein,we report that using dual-salt additives of LiPF_(6) and LiNO_(3) in an ether solvent-based electrolyte can significantly improve the cycling stability and rate capability of a Li-carbon(Li-CNT)composite.As a result,an average cycling CE as high as 99.30% was obtained for the Li-CNT at a current density of 2.5 mA cm^(-2) and an negative electrode to positive electrode capacity(N/P)ratio of 2.The cycling stability and rate capability enhancement of the Li-CNT negative electrode could be attributed to the formation of a better solid electrolyte interphase layer that contains both inorganic components and organic polyether.The former component mainly originates from the decomposition of the LiNO_(3) additive,while the latter comes from the LiPF_(6)-induced ring-opening polymerization of the ether solvent.This novel surface chemistry significantly improves the CE of Li negative electrode,revealing its importance for the practical application of lithium metal batteries. | Lei Zheng Feng Guo Tuo Kang Yingzhu Fan Wei Gu Yayun Mao Ya Liu Rong Huang Zhiyun Li Yanbin Shen Wei Lu Liwei Chen | 2021 | Nano-Micro Letters2021,13,7: | 3 |

| 18 | Gene amplification-driven RNA methyltransferase KIAA1429 promotes tumorigenesis by regulating BTG2 via m6A-YTHDF2-dependent in lung adenocarcinoma显示文摘Background:Epigenetic alterations have been shown to contribute immensely to human carcinogenesis.Dynamic and reversible N6-methyladenosine(m6A)RNA modification regulates gene expression and cell fate.However,the reasons for activation of KIAA1429(also known as VIRMA,an RNA methyltransferase)and its underlying mechanism in lung adenocarcinoma(LUAD)remain largely unexplored.In this study,we aimed to clarify the oncogenic role of KIAA1429 in the tumorigenesis of LUAD.Methods:Whole-genome sequencing and transcriptome sequencing of LUAD data were used to analyze the gene amplification of RNA methyltransferase.The in vitro and in vivo functions of KIAA1429 were investigated.Transcriptome sequencing,methylated RNA immunoprecipitation sequencing(MeRIP-seq),m6A dot blot assays and RNA immunoprecipitation(RIP)were performed to confirm the modified gene mediated by KIAA1429.RNA stability assays were used to detect the half-life of the target gene.Results:Copy number amplification drove higher expression of KIAA1429 in LUAD,whichwas correlatedwith poor overall survival.Manipulating the expression of KIAA1429 could regulate the proliferation and metastasis of LUAD.Mechanistically,the target genes of KIAA1429-mediated m6A modification were confirmed by transcriptome sequencing and MeRIP-seq assays.We also revealed that KIAA1429 could regulate BTG2 expression in an m6A-dependent manner.Knockdown of KIAA1429 significantly decreased the m6A levels of BTG2 mRNA,leading to enhanced YTH m6A RNA binding protein 2(YTHDF2,the m6A“reader”)-dependent BTG2 mRNA stability and promoted the expression of BTG2;thus,participating in the tumorigenesis of LUAD.Conclusions:Our data revealed the activation mechanism and important role of KIAA1429 in LUAD tumorigenesis,which may provide a novel view on the targeted molecular therapy of LUAD. | Chang Zhang Qi Sun Xu Zhang Na Qin Zhening Pu Yayun Gu Caiwang Yan Meng Zhu Juncheng Dai ChengWang Ni Li Guangfu Jin Hongxia Ma Zhibin Hu Erbao Zhang Fengwei Tan Hongbing Shen | 2022 | Cancer Communications2022,42,7: | 1 |
