| 5 | Sequential fate-switches in stem-like cells drive the tumorigenic trajectory from human neural stem cells to malignant glioma显示文摘Glioblastoma(GBM)is an incurable and highly heterogeneous brain tumor,originating from human neural stem/progenitor cells(hNSCs/hNPCs)years ahead of diagnosis.Despite extensive efforts to characterize hNSCs and end-stage GBM at bulk and single-cell levels,the de novo gliomagenic path from hNSCs is largely unknown due to technical difficulties in early-stage sampling and preclinical modeling.Here,we established two highly penetrant hNSC-derived malignant glioma models,which resemble the histopathology and transcriptional heterogeneity of human GBM.Integrating time-series analyses of whole-exome sequencing,bulk and single-cell RNA-seq,we reconstructed gliomagenic trajectories,and identified a persistent NSC-like population at all stages of tumorigenesis.Through trajectory analyses and lineage tracing,we showed that tumor progression is primarily driven by multi-step transcriptional reprogramming and fate-switches in the NSC-like cells,which sequentially generate malignant heterogeneity and induce tumor phenotype transitions.We further uncovered stage-specific oncogenic cascades,and among the candidate genes we functionally validated C1QL1 as a new glioma-promoting factor.Importantly,the neurogenic-to-gliogenic switch in NSC-like cells marks an early stage characterized by a burst of oncogenic alterations,during which transient AP-1 inhibition is sufficient to inhibit gliomagenesis.Together,our results reveal previously undercharacterized molecular dynamics and fate choices driving de novo gliomagenesis from hNSCs,and provide a blueprint for potential early-stage treatment/diagnosis for GBM. | Xiaofei Wang Ran Zhou Yanzhen Xiong Lingling Zhou Xiang Yan Manli Wang Fan Li Chuanxing Xie Yiming Zhang Zongyao Huang Chaoqiong Ding Kaidou Shi Weida Li Yu Liu Zhongwei Cao Zhen-Ning Zhang Shengtao Zhou Chong Chen Yan Zhang Lu Chen Yuan Wang | 2021 | Cell Research2021,31,6: | 8 |

| 7 | Microbial and molecular fossils from the Permian Zoophycos in South China显示文摘The trace fossil Zoophycos was enriched in the lower part of the Guadalupian Maokou Formation of Permian in Laibin, Guangxi of South China, and characterized by observable lamellae within its spreiten. Associated with these lamellae are the prolific coccus- and spirillum-shaped microbe fossils. These microbes morphologically identified are 0.4―3.0 μm in diameter or length. Molecular fossils, including normal alkanes (dominated by C18, without an odd-over-even predominance), acyclic isoprenoids (such as pristane (Pr) and phytane (Ph)), extended tricyclic terpanes, pentacyclic triterpanes, steranes, al-kylcyclohexanes, dibenzothiophenes, benzonaphthothiophenes, benzobisbenzothiophenes, were in-strumentally identified in the wackestone characterized by the occurrence of abundant Zoophycos (composite ichnofabric indices are 4) and crowded microbial fossils. The value of the Pr/Ph ratio is less than 1, indicative of a dysoxic condition. The identification of abundant sulfur compounds (the thio-phene series) related to the contribution of reductive sulfur favors the occurrence of sulphate-reducing bacteria. The association of abundant microbial colonies with microbial molecular fossils within the spreiten suggests that trace fossils Zoophycos would be a multifunctional garden carefully constructed by the Zoophycos-producer, where different microbial colonies were orderly and carefully planted and cultured in different minor lamellae within spreiten. | 10 | An Approach for Network Function Combination Based on Least Busy Placement Algorithm显示文摘Recently, integrating Softwaredefined networking(SDN) and network functions virtualization(NFV) are proposed to address the issue that difficulty and cost of hardwarebased and proprietary middleboxes management. However, it lacks of a framework that orchestrates network functions to service chain in the network cooperatively. In this paper, we propose a function combination framework that can dynamically adapt the network based on the integration NFV and SDN. There are two main contributions in this paper. First, the function combination framework based on the integration of SDN and NFV is proposed to address the function combination issue, including the architecture of Service Deliver Network, the port types representing traffic directions and the explanation of terms. Second, we formulate the issue of load balance of function combination as the model minimizing the standard deviations of all servers' loads and satisfying the demand of performance and limit of resource. The least busy placement algorithm is introduced to approach optimal solution of the problem. | 11 | A novel cancer vaccine with the ability to simultaneously produce anti-PD-1 antibody and GM-CSF in cancer cells and enhance Th1-biased antitumor immunity显示文摘Tumor escape from immune-mediated destruction has been associated with immunosuppressive mechanisms that inhibit T-cell activation.A promising strategy for cancer immunotherapy is to disrupt key pathways regulating immune tolerance,such as program death-1(PD-1/PD-L1)pathway in the tumor environment.However,the determinants of response to anti-PD-1 monoclonal antibodies(mAbs)treatment remain incompletely understood.In murine models,PD-1 blockade alone fails to induce effective immune responses to poorly immunogenic tumors,but is successful when combined with additional interventions,such as cancer vaccines.Novel cancer vaccines combined with antibody may offer promising control of cancer development and progression.In this investigation,we generated a novel tumor cell vaccine simultaneously expressing anti-PD-1 mAbs and granulocyte-macrophage colony stimulating factor(GM-CSF)in CT26 colon cancer and B16-F10 melanoma.The antitumor effect of the vaccine was verified by therapeutic and adoptive animal experiments in vivo.The antitumor mechanism was analyzed using Flow cytometry,Elispot and in vivo intervention approaches.The results showed that tumor cell vaccine secreting PD-1 neutralizing antibodies and GM-CSF induced remarkable antitumor immune effects and prolonged the survival of tumor-bearing animals compared with animals treated with either PD-1 mAbs or GM-CSF alone.Antitumor effects and prolonged survival correlated with strong antigen-specific T-cell responses by analyzing CD11c^(+)CD86^(+)DC,CD11b^(+)F4/80^(+)MΦcells,increased ratio of Teff/Treg in the tumor microenvironment,and higher secretion levels of Th1 proinflammatory cytokines in serum.Furthermore,the results of ELISPOT and in vivo blocking strategies further confirmed that the antitumor immune response is acquired by CD4 and CD8 T immune responses,primarily dependent on CD4 Th1 immune response,not NK innate immune response.The combination of PD-1 blockade with GM-CSF secretion potency creates a novel tumor cell vaccine immunotherapy,affording significantly improved antitumor responses by releasing the state of immunosuppressive microenvironment and augmenting the tumor-reactive T-cell responses. | Hongwei Tian Gang Shi Qin Wang Yiming Li Qianmei Yang Chunlei Li Guoyou Yang Min Wu Qian Xie Shuang Zhang Yang Yang Rong Xiang Dechao Yu Yuquan Wei Hongxin Deng | 2016 | Signal Transduction and Targeted Therapy2016,1,1: | 3 |

This technique is a combi-nation of the usage of polarimetric information of SAR images and the unsupervised classification method based on fuzzy set theory. Image quantization and image enhancement are used to preprocess the POLSAR data. Then the polarimetric information and Fuzzy C-Means (FCM) clustering algorithm are used to classify the preprocessed images. The advantages of this algorithm are the automated classification, its high classifica-tion accuracy, fast convergence and high stability.
