1Liensinine induces gallbladder cancer apoptosis and G2/M arrest by inhibiting ZFX-induced PI3K/AKT pathway显示文摘Gallbladder carcinoma (GBC) is the most comm on and aggressive can cer of the biliary tract. Liensinine has been proved to have hypotensive effect. However, the effect of liensinine on GBC is still unknown. The aim of this study is to investigate the effect and mechanism of liensinine in human GBC cells. Cell viability assay and colony formation assay were performed to assess cell growth and proliferation. Flow cytometry analysis was used to investigate cell cycle apoptosis in vitro. Hoechst 33342 staining was also used to evaluate cell apoptosis. Western blot analysis was used to determine the expression of proteins corresponding to the related cell cycle and apoptosis.The effect of liensinine treatment in vivo was experimented with xenografted tumors. We found that liensinine significantly inhibited the growth of GBC cells both in vivo and in vitro. In vitro, cell growth and proliferation were significantly suppressed by liensinine in a dose- and time-dependent manner. In vivo, liensinine inhibited tumor growth. Liensinine could induce GBC cells G2/M phase arrest by up-regulating the levels of Cyclin B1 and CDK1 proteins. Liensinine also affected GBC cell cycle progress!on and induced apoptosis by down-regulating phosphorylated protein kinase B (AKT), phosphorylated protein kinase B (p-AKT), phosphatidylinositol 3-kinase (PI3K), and Zinc finger X-chromosomal protein (ZFX) proteins. 2Morphology and magnetic properties of Sm_(2)Co_(7)/α-Fe nanocomposite magnets produced by high energy ball milling and spark plasma sintering显示文摘In this article,the Sm_(2)Co_(7)/α-Fe nanocomposite magnets were prepared by high energy ball milling and spark plasma sintering method.The effect of soft phase content on the magnetic properties was studied.Up to 30 wt% α-Fe was added into Sm_(2)Co_(7) matrix without the decrease of remanence.Optimal energy product(BH)max of 9.2 MGOe was obtained with 20 wt% α-Fe.TEM observation shows that the grain size of α-Fe is 20-50 nm which ensures a good coupling effect between soft and hard phase.One more thing needs to be mentioned is that there exists inter-diffusion between Sm-Co phase and α-Fe phase.Moreover,our results can also illustrate that the Sm_(2)Co_(7)/α-Fe nanocomposite magnets are able to acquire better magnetic properties than the SmCo_(5)/α-Fe magnets prepared by the same process due to the large domain width of Sm_(2)Co_(7) phase.Yitong Zhuge Yuqing Li Xiaochang Xu Dongtao Zhang Hongguo Zhang Weiqiang Liu Ming Yue 2021Journal of Rare Earths2021,39,3:3
3A High Spatiotemporal Iontronic Single-Cell Viscometer显示文摘Ideal single-cell viscometer has remained unachieved,leaving a gap in current palette of single-cell nanotools.Information of single-cell viscosity could contribute to our knowledge of fundamental biological processes,e.g.,mass diffusion,biochemical interaction,and cellular responses to many diseases and pathologies.Although advances have been made to this end,existing methods generally suffer from limitations,e.g.,low spatiotemporal resolution.Here,we describe a high spatiotemporal iontronic single-cell viscometer that operates upon a patch clamp integrated with double-barreled nanopores separated by a septum of ca.32 nm.The system enables reversible electroosmotic manipulation of the adjacent small fluid bridging two nanopores,the viscous alternation of which could be sensitively monitored by the ionic responses.In practical cellular studies,significantly,our findings reveal not only the less deviated medium viscosities than those of lysosomes and mitochondria but also the highest viscosities in the near-nuclear region than those of mitochondrion-dense and lysosome-dense regions.This work has provided an accessible single-cell viscometer and enriched the armory of single-cell nanotools.Tianyang Zhang Siyuan Yu Bing Wang Yitong Xu Xiaomei Shi Weiwei Zhao Dechen Jiang Hongyuan Chen Jingjuan Xu 2022Research2022,,4:1
4Nanoparticle accumulation in liver may induce resistance to immune checkpoint blockade therapy显示文摘Despite immune checkpoint blockade(ICB)therapy has transformed cancer treatment,only 20.2%of these patients achieved a response.Understanding resistance mechanisms to ICB is important for the treatment of a wider population.In this work,we occasionally found that the silica nanoparticles(SiO_(2)NPs)accumulated in the liver can induce resistance to following ICB therapy to a subcutaneous tumor in mice.By analysis of T cells frequency,we uncovered that SiO_(2)NPs in the liver resulted in a siphoning of T cells from circulation to the liver by produced chemokines.In addition,liver immunosuppressive cells further inhibit the function and induce apoptosis of recruited T cells,leading to a systemic loss and reduced tumor infiltration of T cells,which contributes to poor responses to ICB therapy.However,such effect is not observed in poly(lactic-co-glycolic acid)(PLGA)NPs treated mice under the same conditions,likely due to their much lower immunogenicity in perturbing the liver immune microenvironment,indicating that cancer is not a local disease but an ecosystem that is linked to the distal environment.We further provide a new mechanism insight into ICB resistance induced by liver accumulation of nanoparticles.Huaxing Dai Qianyu Yang Rong Sun Yue Zhang Qingle Ma Yifan Shen Beilei Wang Yitong Chen Jialu Xu Bo Tian Fang Xu Chao Wang 2023Nano Research2023,16,4:1
5Nitrogen-doped Carbon Coated Na3V2(PO4)3 with Superior Sodium Storage Capability显示文摘Cathodes with high cycling stability and rate capability are required for ambient temperature sodium ion batteries in renewable energy storage application.Na3V2(PO4)3 is an attractive cathode material with excellent electrochemical stability and fast ion diffusion coefficient within the 3D NASICON structure.Nevertheless,the practical application of Na3V2(PO4)3 is seriously hindered by its intrinsically poor electronic conductivity.Herein,solvent evaporation method is presented to obtain the nitrogen-doped carbon coated Na3V2(PO4)3 cathode material,delivering cnhanced clectrochemical performances.N-Doped carbon layer coating servfes as a highly conducting pathway,and creates numerous extrinsic defects and active sites,which can facilitate the storage and diffusion of Na^+.Morcover,the N-doped carbon layer can provide a stable framework to accommodate the agglomeration of the electrode upon electrode cycling.N-Doped carbon coated Na3V2(PO4)3(NC-NVP)exhibits excellent long cycling life and superior rate performances than bare Na3V2(PO4)3 without carbon coating.NC-NVP delivers a stable capacity of 95.9mA·h/g after 500 cycles at 1C rate,which corresponds to high capacity retention(94.6%)with respect to the initial capacity(101.4mA·h/g).Over 91.3% of the initial capacity is retained after 500 cycles at 5C,and the capacity can reach 85mA·h/g at 30C rate.XU Laiqiang LI Jiayang LI Yitong CAI Peng LIU Cheng ZOU Guoqiang HOU Hongshuai HUANG Lanping JI Xiaobo 2020Chemical Research in Chinese Universities2020,36,3:1

8Antioxidant,Anti-inflammatory,and Antibacterial Activities of Different Extracts from Polygonum capitatum显示文摘[Objectives]To investigate the antioxidant,anti-inflammatory and antibacterial activities of different extracts(aqueous,ethanol,ethyl acetate and n-butanol extracts)of Miao medicine Polygonum capitatum.[Methods]Eleven batches of P.capitatum in Guizhou province were collected,and water,ethanol,ethyl acetate and n-butanol extracts were prepared by reflux extraction.Antioxidant activity was determined by radical scavenging capacity of 1,1 diphenyl-2-picyl hydrazine(DPPH),anti-inflammatory activity was screened by lipopolysaccharide(LPS)induced RAW264.7 cells to produce NO,and the minimum inhibitory concentration(MIC)was screened by broth microdilution method.[Results]When the concentration of ethyl acetate extract was 10 mg/L,the scavenging rate of DPPH ranged from 90%to 99%.The(MIC of the ethyl acetate extract against Staphylococcus aureus(SA),Pseudomonas aeruginosa(PA)and Escherichia coli(EC)was 0.18-0.65,0.13-0.82,and 0.15-0.78 g/L,respectively.In the anti-inflammatory activity,ethyl acetate extract inhibited NO production with inhibition rate of 70%.[Conclusions]The ethyl acetate extract and ethanol extract of Miao medicine P.capitatum have strong antioxidant,anti-inflammatory and antibacterial activities.Bo TU Minghui HE Yitong SHEN Xu ZHANG Yan LIN 2023Medicinal Plant2023,14,2:0

10Assessment of the Capability of ENSEMBLES Hindcasts in Predicting Spring Climate in China显示文摘Using the hindcasts provided by the Ensemble-Based Predictions of Climate Changes and Their Impacts(ENSEMBLES) project for the period of 1980–2005, the forecast capability of spring climate in China is assessed mainly from the aspects of precipitation, 2-m air temperature, and atmospheric circulations. The ENSEMBELS can reproduce the climatology and dominant empirical orthogonal function(EOF) modes of precipitation and 2-m air temperature, with some differences arising from different initialization months. The multi-model ensemble(MME) forecast of interannual variability is of good performance in some regions such as eastern China with February initialization.The spatial patterns of the MME interannual and inter-member spreads for precipitation and 2-m air temperature are consistent with those of the observed interannual spread, indicating that internal dynamic processes have major impacts on the interannual anomaly of spring climate in China. We have identified two coupled modes between intermember anomalies of the 850-hPa vorticity in spring and sea surface temperature(SST) both in spring and at a lead of 2 months, of which the first mode shows a significant impact on the spring climate in China, with an anomalous anticyclone located over Northwest Pacific and positive precipitation and southwesterly anomalies in eastern China.Our results also suggest that the SST at a lead of two months may be a predictor for the spring climate in eastern China. 13Most similar maximal clique query on large graphs显示文摘This paper studies the most similar maximal clique query(MSMCQ).Given a graph G and a set of nodes Q,MSMCQ is to find the maximal clique of G having the largest similarity with Q.MSMCQ has many real applications including advertising industry,public security,task crowdsourcing and social network,etc.MSMCQ can be studied as a special case of the general set similarity query(SSQ).However,the MCs of G has several specialties from the general sets.Based on the specialties of MCs,we propose a novel index,namely MCIndex.MCIndex outperforms the state-of-the-art SSQ method significantly in terms of the number of candidates and the query time.Specifically,we first construct an inverted indexⅠfor all the MCs of G.Since the MCs in a posting list often have a lot of overlaps,MCIndex selects some pivots to cluster the MCs with a small radius.Given a query Q,we compute the distance from the pivots to Q.The clusters of the pivots assured not answer can be pruned by our distance based pruning rule.Since it is NP-hard to construct a minimum MCIndex,we propose to construct a minimal MCIndex onⅠ(v)with an approximation ratio 1+ln|Ⅰ(v)|.Since the MCs have properties that are inherent of graph structure,we further propose a S Index within each cluster of a MCIndex and a structure based pruning rule.S Index can significantly reduce the number of candidates.Since the sizes of intersections between Q and many MCs need to be computed during the query evaluation,we also propose a binary representation of MCs to improve the efficiency of the intersection size computation.Our extensive experiments confirm the effectiveness and efficiency of our proposed techniques on several real-world datasets.Yun PENG Yitong XU Huawei ZHAO Zhizheng ZHOU Huimin HAN 2020Frontiers of Computer Science2020,14,3:0
14Purpurolide C-based microneedle promotes macrophage-mediated diabetic wound healing via inhibiting TLR4-MD2 dimerization and MYD88 phosphorylation显示文摘Delayed wound healing in diabetes is a global challenge,and the development of related drugs is a clinical problem to be solved.In this study,purpurolide C(PC),a small-molecule secondary metabolite of the endophytic fungus Penicillium purpurogenum,was found to promote diabetic wound healing.To investigate the key regulation targets of PC,in vitro RNA-seq,molecular docking calcula-tions,TLR4-MD2 dimerization SDS-PAGE detection,and surface plasmon resonance(SPR)were per-formed,indicating that PC inhibited inflammatory macrophage activation by inhibiting both TLR4-MD2 dimerization and MYD88 phosphorylation.Tlr4 knockout in vivo attenuated the promotion effect of PC on wound healing.Furthermore,a delivery system consisting of macrophage liposome and GelMA-based microneedle patches combined with PC(PC@MLIP MN)was developed,which overcame the poor water solubility and weak skin permeability of PC,so that successfully punctured the skin and delivered PC to local tissues,and accurately regulated macrophage polarization in diabetic wound management.Overall,PC is an anti-inflammatory small molecule compound with a well-defined structure and dualtarget regulation,and the PC@MLIP MN is a promising novel biomaterial for the management of diabetic wound.Yitong Liu Guiyang Xia Yingyi Chen Huan Xia Junji Xu Lijia Guo Sheng Lin Yi Liu 2023Acta Pharmaceutica Sinica B2023,13,12:0

16Single-Atom Ce-N_(4)-C-(OH)_(2)Nanozyme-Catalyzed Cascade Reaction to Alleviate Hyperglycemia显示文摘The enzyme-mimicking catalytic activity of single-atom nanozymes has been widely used in tumor treatment.However,research on alleviating metabolic diseases,such as hyperglycemia,has not been reported.Herein,we found that the single-atom Ce-N_(4)-C-(OH)_(2)(SACe-N_(4)-C-(OH)_(2))nanozyme promoted glucose absorption in lysosomes,resulting in increased reactive oxygen species production in HepG2 cells.Furthermore,the SACe-N_(4)-C-(OH)_(2)nanozyme initiated a cascade reaction involving superoxide dismutase-,oxidase-,catalase-,and peroxidase-like activity to overcome the limitations associated with the substrate and produce•OH,thus improving glucose intolerance and insulin resistance by increasing the phosphorylation of protein kinase B and glycogen synthase kinase 3β,and the expression of glycogen synthase,promoting glycogen synthesis to improve glucose intolerance and insulin resistance in high-fat diet-induced hyperglycemic mice.Altogether,these results demonstrated that the novel nanozyme SACeN_(4)-C-(OH)_(2)alleviated the effects of hyperglycemia without evident toxicity,demonstrating its excellent clinical application potential.Guangchun Song Jia Xu Hong Zhong Qi Zhang Xin Wang Yitong Lin Scott PBeckman Yunbo Luo Xiaoyun He Jin-Cheng Li Kunlun Huang Nan Cheng 2023Research2023,,4:0
17A MODEL OF TWO-PHASE EQUILIBRIUM FOR FORMATION OF KINK-BANDS IN ROCKS显示文摘Kink-bands in rocks have been widely observed in nature and imitated in the labora- tory, and the mechanism of their formation has attracted much attention from various researchers for many years. In this paper, a two-phase equilibrium model is presented in which the kink-bands are considered as a high-strain phase and the other regions outside kink-bands as a low-strain phase and the discontinuity of the deformation gradient and stresses is permitted across the interface between those two phases. Based on the present model, we conduct the analysis for the rocks under plane strain compression by finding the minimum value of the compressive loading at which the governing equations have real, physically acceptable solutions. 18Fabrication of Durable Superhydrophobic Stainless Steel Mesh with Nano/Micro Flower-Like Morphologies for Self-Cleaning and Efficient Oil/Water Separation显示文摘How to fabricate durable superhydrophobic materials for cyclic oil/water separation to solve oil-induced pollutions is still a problem.Herein,we fabricated a durable Superhydrophobic Stainless Steel Mesh(SH-SSM)with nano/micro flower-like sheets by self-assembly of zinc stearate for the cyclic separation of various oil/water mixtures.The SH-SSM exhibits durable superhydrophobicity against static or dynamic treatments.In virtue of durable superhydrophobicity of the SH-SSM,the dusts on surface of SH-SSM can be removed completely by running water immediately showing excellent self-cleaning performance.Moreover,the SH-SSM can be used to perform gravity-driven separation of heavy oil/water and light oil/water mixtures by utilizing its superhydrophobicity that oil phase penetrates the mesh and water phase is restricted.Separation efficiencies of the SH-SSM can reach 99.99%.After 20 cycles of separation,the separation efficiencies for are>97.00%and>98.25%for heavy oil/water and light oil/water mixtures,respectively.Flux values of dichloromethane,hexane and petroleum ether are 2.5×10^(5) L m^(−2) h^(−1),1.7×10^(5) L m^(−2) h^(−1),and 1.8×10^(5) L m^(−2) h^(−1),respectively.This study proposes a facial approach to prepare durable superhydrophobic and self-cleaning material for cyclic and fast separation of oil/water.Changlian Xu Yitong Luo Lu Zhou Yiwen Bi Hao Sun 2022Journal of Bionic Engineering2022,19,6:0
19Development of plant cytosine base editors with the Cas12a system显示文摘Base editors of the Cas9 system have been widely used for precise nucleotide substitution in crops. In this study, Cas12a was applied to construct plant cytosine base editors(CBEs). The main elements of Cas12aCBEs were engineered and their efficiency was evaluated in stably transformed rice cells. An optimized ttCas12a-hyA3Bctd editor, consisting of a LbCas12a variant carrying catalytic inactive D832A and temperature-tolerance D156R double mutations, a truncated human APOBEC3B deaminase, a human RAD51 single-stranded DNA-binding domain, and double copies of UGI, outperformed other Cas12aCBEs in base editing efficiency. In T0transgenic rice plants, ttCas12a-hyA3Bctd edited an average of42.01% and a maximum of 68.75% of lines at six genomic targets. A-to-G conversions were generated in rice by an adenine base editor with a similar architecture to the optimized CBE.
