1A new type of internal solitary waves with a re-appearance period of 23 h observed in the South China Sea显示文摘The South China Sea (SCS), in particular the northern SCS, is one of ocean areas where energetic internal solitary waves (ISWs)occur most frequently (Cai et al., 2012; Zheng, 2017). Based on the re-appearance period (RP) at an observation station, Ramp et al.(2004) divided the ISWs into two types:Type-a and Type-b. Type-a ISWs arrive regularly at the same time every day, i.e., the RP is about 24 h, and Type-b ISWs arrive about one hour late every day, i.e., the RP is about 25 h. 2Pioneering function of Isl1 in the epigenetic control of cardiomyocyte cell fate显示文摘Generation of widely differing and specialized cell types from a single totipotent zygote involves large-scale transcriptional changes and chromatin reorganization. Pioneer transcription factors play key roles in programming the epigenome and facilitating recruitment of additional regulatory factors during successive cell lineage specification and differentiation steps. Here we show that Isl1 acts as a pioneer factor driving cardiomyocyte lineage commitment by shaping the chromatin landscape of cardiac progenitor cells. Using an Isl1 hypomorphic mouse line which shows congenital heart defects, genome-wide profiling of Isl1 binding together with RNA- and ATAC-sequencing of cardiac progenitor cells and their derivatives, we uncover a regulatory network downstream of Isl1 that orchestrates cardiogenesis. Mechanistically, we show that Isl1 binds to compacted chromatin and works in concert with the Brg1-Baf60c-based SWI/SNF complex to promote permissive cardiac lineage-specific alterations in the chromatin landscape not only of genes with critical functions in cardiac progenitor cells, but also of cardiomyocyte structural genes that are highly expressed when Isl1 itself is no longer present. 3An open conformation determined by a structural switch for 2A protease from coxsackievirus A16显示文摘Coxsackievirus A16 belongs to the family Picornaviridae,and is a major agent of hand-foot-and-mouth disease that infects mostly children,and to date no vaccines or antivi-ral therapies are available.2A protease of enterovirus is a nonstructural protein and possesses both self-cleavage activity and the ability to cleave the eukaryotic translation initiation factor 4G.Here we present the crystal structure of coxsackievirus A162A protease,which interestingly forms hexamers in crystal as well as in solution.This structure shows an open conformation,with its active site accessible,ready for substrate binding and cleav-age activity.In conjunction with a previously reported“closed”state structure of human rhinovirus 2,we were able to develop a detailed hypothesis for the conforma-tional conversion triggered by two“switcher”residues Glu88 and Tyr89 located within the bll2-cII loop.Substrate recognition assays revealed that amino acid residues P1′,P2 and P4 are essential for substrate specificity,which was verifi ed by our substrate binding model.In addition,we compared the in vitro cleavage effi ciency of 2A pro-teases from coxsackievirus A16 and enterovirus 71 upon the same substrates by fl uorescence resonance energy transfer(FRET),and observed higher protease activity of enterovirus 71 compared to that of coxsackievirus A16.In conclusion,our study shows an open conformation of coxsackievirus A162A protease and the underlying mechanisms for conformational conversion and substrate specifi city.These new insights should facilitate the future rational design of effi cient 2A protease inhibitors.Yao Sun Xiangxi Wang Shuai Yuan Minghao Dang Xuemei Li Xuejun CZhang Zihe Rao 2013Protein & Cell2013,4,10:3
4Mooring observed mode-2 internal solitary waves in the northern South China Sea显示文摘The mode-2 internal solitary waves(ISWs)generated by mode-2 internal tide(IT)are identified by mooring observations in the northern South China Sea(SCS)from 2016 to 2017.Two mode-2 ISWs with a re-appearance period of 24.9 h observed on 29 and 30 July 2016 are characterized by type-b ISWs.They occurred when the isotherms compressed obviously in the vertical direction.Modal decomposition of IT horizontal currents shows that the vertical compression of the isotherms is mainly caused by diurnal mode-2 IT.The analysis of the role of the density stratification reveals that a deeper and thinner pycnocline is favorable for generation of mode-2 ISWs rather than pycnocline intensity.By comparing the mode-2 nonlinear,dispersion coefficients and the Ursell numbers calculated based on the stratification associated with different kinds of ITs with the observation results,it is shown that the diurnal mode-2 IT plays a crucial role in the generation of the mode-2 ISWs.When the diurnal mode-2 IT interacts with the semidiurnal IT and causes a deeper and thinner pycnocline,the mode-2 ISWs are easily excited.Liang Chen Xuejun Xiong Quanan Zheng Yeli Yuan Long Yu Yanliang Guo Guangbing Yang Xia Ju Jia Sun Zhenli Hui 2020Acta Oceanologica Sinica2020,39,11:3
5HER3 intracellular domains play a crucial role in HER3/HER2 dimerization and activation of downstream signaling pathways显示文摘Dimerization among the EGFR family of tyrosine kinase receptors leads to allosteric activation of the kinase domains of the partners.Unlike other members in the family,the kinase domain of HER3 lacks key amino acid residues for catalytic activity.As a result,HER3 is suggested to serve as an allosteric activator of other EGFR family members which include EGFR,HER2 and HER4.To study the role of intracellular domains in HER3 dimerization and activation of downstream signaling pathways,we constructed HER3/HER2 chimeric receptors by replacing the HER3 kinase domain(HER3-2-3)or both the kinase domain and the C-terminal tail(HER3-2-2)with the HER2 counterparts and expressed the chimeric receptors in Chinese hamster ovary(CHO)cells.While over expression of the intact human HER3 transformed CHO cells with oncogenic properties such as AKT/ERK activation and increased proliferation and migration,CHO cells expressing the HER3-2-3 chimeric receptor showed significantly reduced HER3/HER2 dimerization and decreased phosphorylation of both AKT and ERK1/2 in the presence of neuregulin-1(NRG-1).In contrast,CHO cells expressing the HER3-2-2 chimeric receptor resulted in a total loss of downstream AKT activation in response to NRG-1,but maintained partial activation of ERK1/2.The results demonstrate that the intracellular domains play a crucial role in HER3’s function as an allosteric activator and its role in downstream signaling.Byung-Kwon Choi Xiumei Cai Bin Yuan Zhao Huang Xuejun Fan Hui Deng Ningyan Zhang Zhiqiang An 2012Protein & Cell2012,3,10:3
6Olfactory and gustatory dysfunctions of COVID-19 patients in China:A multicenter study显示文摘Introduction:With the spread of the epidemic worldwide,an increasing number of doctors abroad have observed the following atypical symptoms of coronavirus disease 2019(COVID-19):olfactory or taste disorders.Therefore,clarifying the incidence and clinical characteristics of olfactory and taste disorders in Chinese COVID-19 patients is of great significance and urgency.Materials and Methods:A retrospective study was conducted,which included 229 severe acute respiratory syndrome coronavirus 2 confirmed patients,through face-to-face interviews and telephone follow-up.Following the completion of questionnaires,the patients participating in the study,were categorized according to the degree of olfactory and taste disorders experienced,and the proportion of each clinical type of patient with olfactory and taste disorders and the time when symptoms appeared were recorded.Results:Among the 229 patients,31(13.54%)had olfactory dysfunction,and 44(19.21%)had gustatory dysfunction.For the patients with olfactory dysfunction,6(19.35%)developed severe disease and became critically ill.Olfactory dysfunction appeared before the other symptoms in 21.43%of cases.The proportion of females with olfactory and gustatory dysfunction was higher than that of males(P<0.001).Conclusions:The incidence of olfactory and gustatory dysfunction was much lower than that reported abroad;the prognosis of patients with olfactory dysfunction is relatively favorable;olfactory and gustatory dysfunction can be used as a sign for early screening;females are more prone to olfactory and gustatory dysfunction.Jianhui Li Yi Sun Enqiang Qin Hu Yuan Mingbo Liu Wenqi Yi Zhu Chen Chengcheng Huang Fengjie Zhou Ruiyao Chen Leibo Zhang Ning Yu Qiong Liu Xuejun Zhou Jingjing He Boyu Li Fusheng Wang Changliang Yang Shiming Yang 2022World Journal of Otorhinolaryngology-Head and Neck Surgery2022,8,4:2
7Contrasting effects of nitrogen addition on litter decomposition in forests and grasslands in China显示文摘Nitrogen(N)addition has profound impacts on litter-mediated nutrient cycling.Numerous studies have reported different effects of N addition on litter decomposition,exhibiting positive,negative,or neutral effects.Previous meta-analysis of litter decomposition under N addition was mainly based on a small number of samples to allow comparisons among ecosystem types.This study presents the results of a meta-analysis incorporating data from 53 published studies(including 617 observations)across forests,grasslands,wetlands,and croplands in China,to investigate how environmental and experimental factors impact the effects of N addition on litter decomposition.Averaged across all of the studies,N addition significantly slows litter decomposition by 7.02%.Considering ecosystem types,N addition significantly accelerates litter decomposition by 3.70%and 11.22%in grasslands and wetlands,respectively,clearly inhibits litter decomposition by 14.53%in forests,and has no significant effects on litter decomposition in croplands.Regarding the accelerated litter decomposition rate in grasslands due to N addition,litter decomposition rate increases slightly with increasing rates of N addition.However,N addition slows litter decomposition in forests,but litter decomposition is at a significantly increasing rate with increasing amounts of N addition.The responses of litter decomposition to N addition are also influenced by the forms of N addition,experiential duration of N addition,humidity index,litter quality,and soil pH.In summary,N addition alters litter decomposition rate,but the direction and magnitude of the response are affected by the forms of N addition,the rate of N addition,ambient N deposition,experimental duration,and climate factors.Our study highlights the contrasting effects of N addition on litter decomposition in forests and grasslands.This finding could be used in biogeochemical models to better evaluate ecosystem carbon cycling under increasing N deposition due to the differential responses of litter decomposition to N addition rates and ecosystem types.SU Yuan MA Xiaofei GONG Yanming LI Kaihui HAN Wenxuan LIU Xuejun 2021Journal of Arid Land2021,13,7:2

15Syntheses of CuO nanostructures in ionic liquids显示文摘A simple and efficient approach is developed to fabricate single-crystalline CuO nanostructures through an ionic liquid assisted one-step low-temperature solid-state route.Both nanoparticles(5 nm in size)and nanorods(5-10 nm in diameter and 50-100 nm in length)of monoclinic CuO were obtained. 18Kinetic and thermodynamic studies of adsorption of gallium(Ⅲ) on nano-TiO_2显示文摘Nano-TiO2 was employed for the adsorption of gallium from aqueous solution in batch equilibrium experiments to investigate its adsorption properties. It was found that the adsorption efficiency of Ga(Ⅲ) was more than 96% at pH 3.0. The adsorption capacities and rates of Ga(Ⅲ) onto nano-TiO2 were evaluated as a function of solution concentration and temperature. The results were analyzed using the Langmuir adsorption isotherms. Adsorption isothermal data could be well interpreted by the Langmuir model. The mean energy of adsorption, 15.81 kJ·mol-1, was calculated from the D-R adsorption isotherm. The kinetic experimental data properly correlate with the pseudo-second-order kinetic model. The thermodynamic parameters for the process of adsorption have been estimated.
