3BCRP基因在非小细胞肺癌组织和非癌肺组织中的表达及意义显示文摘目的:进一步研究从人类乳腺癌细胞株 (MCF-7/AdrVp)中克隆出的、对蒽环类抗癌药耐药的 BCRP基因在非癌肺组织及非小细胞肺癌组织中的表达。方法:从术后即刻获得的正常肺组织和有活力的非小细胞肺癌组织新鲜标本中及时提取细胞总 RNA,通过 RT-PCR及 PCR法获得并扩增 BCRP基因的 cDNA产物,再经凝胶电泳分离 BCRP基因 cDNA带,然后再通过转膜技术把这些 BCRP基因 cDNA产物转移到杂交膜上,用 Southern blot杂交技术检测 BCRP基因的表达程度。结果:细胞总 RNA分别从 8个只有肺癌组织标本和 12对同时获取的肺癌组织及非癌肺组织标本中成功提取,然后用变性凝胶电泳鉴定提取 RNA的质量。其中 4例标本因术中缺血时间太长,或有坏死炎症或术前放化疗等原因,所提 RNA有降解而放弃进一步的检测。通过 RT-PCR及 PCR方法从 16例可用标本中获得并扩增 BCRP基因的 cDNA产物,再通过转膜及 Southern blot杂交技术,最终发现 BCRP基因在正常肺组织中均有不同程度的表达( 10/10),而在非小细胞肺癌组织标本中只有一半病人的标本有不同程度的表达 (8/16)。结论: BCRP基因可能是一个在非癌肺组织中常规表达的基因,而在部分病人的肺癌组织中可能此基因有缺失或被抑制。因此。毛友生 Austin Doyle Weidong Yang Yuetong Wei Mark J.Krasna Douglas D.Ross 2001癌症2001,20,3:9
4MXene-Integrated Microneedle Patches with Innate Molecule Encapsulation for Wound Healing显示文摘Wound healing is a complex physiological process that involves coordinated phases such as inflammation and neovascularization.Attempts to promote the healing process tend to construct an effective delivery system based on different drugs and materials.In this paper,we propose novel MXene-integrated microneedle patches with adenosine encapsulation for wound healing.Owing to the dynamic covalent bonding capacity of boronate molecules with adenosine,3-(acrylamido)phenylboronic acid-(PBA-)integrated polyethylene glycol diacrylate(PEGDA)hydrogel is utilized as the host material of microneedle patches.Benefitting from photothermal conversion capacity of MXene,the release of loaded adenosine could be accelerated under NIR irradiation for maintaining the activation signal around injury site.In vitro cell experiments proved the effect of MXene-integrated microneedle patches with adenosine encapsulation in enhancing angiogenesis.When applied for treating animal models,it is demonstrated that the microneedle patches efficiently promote angiogenesis,which is conductive to wound healing.These features make the proposed microneedle patch potential for finding applications in wound healing and other biomedical fields.Lingyu Sun Lu Fan Feika Bian Guopu Chen Yuetong Wang Yuanjin Zhao 2021Research2021,,1:6
5Smart Microneedles for Therapy and Diagnosis显示文摘Microneedles represent a cutting-edge and idea-inspiring technology in biomedical engineering,which have attracted increasing attention of scientific researchers and medical staffs.Over the past decades,numerous great achievements have been made.The fabrication process of microneedles has been simplified and becomes more precise,easy-to-operate,and reusable.Besides,microneedles with various features have been developed and the microneedle materials have greatly expanded.In recent years,efforts have been focused on generating smart microneedles by endowing them with intriguing functions such as adhesion ability,responsiveness,and controllable drug release.Such improvements enable the microneedles to take an important step in practical applications including household drug delivery devices,wearable biosensors,biomedical assays,cell culture,and microfluidic chip analysis.In this review,the fabrication strategies,distinctive properties,and typical applications of the smart microneedles are discussed.Recent accomplishments,remaining challenges,and future prospects are also presented.Xiaoxuan Zhang Yuetong Wang Junjie Chi Yuanjin Zhao 2020Research2020,,1:4
6A novel quality evaluation method for magnolia bark using electronic nose and colorimeter data with multiple statistical algorithms显示文摘Background:Magnolia bark(Magnolia Officinalis REHD.&WILS.and Magnolia officinalis REHD.&WILS.VAR.biloba REHD.&WILS,Hou Po in Chinese),is widely applied in clinical prescriptions and Chinese patent medicines.Origin place is a crucial factor affecting the quality of Hou Po,and chemical composition is an important index for evaluating its quality,which is closely related to its clinical efficacy.This study aims to develop a novel method for rapidly,accurately and comprehensively identifying the origin places of Hou Po and predicting the contents of its important chemical components.Methods:High performance liquid chromatography was used to analyze the contents of magnolol and honokiol and ultra-performance liquid chromatography the contents of magnocurarine and magnoflorine.The cold soak method was used to determine the contents of water-soluble extracts.The E-nose and colorimeter were used to determine the odor and color characteristics,respectively,of the collected Hou Po samples.Results:Using several statistical algorithms,different discriminant models based on the E-nose and colorimeter data were established to distinguish the origin place of Hou-Po and predict the chemical components of honokiol,magnolol,magnocurarine,magnoflorine and water-soluble extracts.The results showed that the Random Forest classifier combined with the ten-fold cross-validation method provided the highest classification accuracy for origin place,accounting for 99.53%among these models.The correlation coefficients between predicted and experimental values of the five chemical components were all higher than 0.96.Conclusion:This study has indicated that the electronic nose and colorimeter are promising methods for evaluating the quality of Chinese herbal medicines both qualitatively and quantitatively.Jiahui Li Yuanyang Shao Yuebao Yao Yuetong Yu Guangzhao Cao Huiqin Zou Yonghong Yan 2020Journal of Traditional Chinese Medical Sciences2020,7,2:4
7Biosynthesis of selenosubtilisin: A novel way to target selenium into the active site of subtilisin显示文摘Glutathione peroxidase (GPx, EC1.11.1.9), an important anti-oxidative selenoenzyme, can catalyze the reduction of harmful hydroperoxides with concomitant glutathione, thereby protecting cells and other biological issues against oxidative damage. It captures considerable interest in redesign of its function for either the mechanism study or the pharmacological development as an antioxidant. In order to de- velop a general strategy for specifically targeting and operating selenium in active sites of enzymes, the catalytically essential residue selenocysteine (Sec) was first successfully bioincorporated into the catalytic center of subtilisin by using an auxotrophic expression system. The studies of the catalytic activity and the steady-state kinetics demonstrated that selenosubtilisin is an excellent GPx-like bio- catalyst. In comparison with the chemically modified method, biosynthesis exhibits obvious advan- tages: Sec could be site-directly incorporated into active sites of enzymes to overcome the non-speci- ficity generated by chemical modification. 8Functional characterization and key residues engineering of a regiopromiscuity O-methyltransferase involved in benzylisoquinoline alkaloid biosynthesis in Nelumbo nucifera显示文摘Lotus(Nelumbo nucifera),an ancient aquatic plant,possesses a unique pharmacological activity that is primarily contributed by benzylisoquinoline alkaloids(BIAs).However,only few genes and enzymes involved in BIA biosynthesis in N.nucifera have been isolated and characterized.In the present study we identified the regiopromiscuity of an O-methyltransferase,designated NnOMT6,isolated from N.nucifera;NnOMT6 was found to catalyze the methylation of monobenzylisoquinoline 6-O/7-O,aporphine skeleton 6-O,phenylpropanoid 3-O,and protoberberine 2-O.We further probed the key residues affecting NnOMT6 activity via molecular docking and molecular dynamics simulation.Verification using site-directed mutagenesis revealed that residues D316,N130,L135,N176A,D269,and E328 were critical for BIA O-methyltransferase activities;furthermore,N323A,a mutant of NnOMT6,demonstrated a substantial increase in catalytic efficiency for BIAs and a broader acceptor scope compared with wild-type NnOMT6.To the best of our knowledge,this is the first study to report the O-methyltransferase activity of an aporphine skeleton without benzyl moiety substitutions in N.nucifera.The study findings provide biocatalysts for the semisynthesis of related medical compounds and give insights into protein engineering to strengthen O-methyltransferase activity in plants.Yuetong Yu Yan Liu Gangqiang Dong JinZhu Jiang Liang Leng XianJu Liu Jun Zhang An Liu Sha Chen 2023Horticulture Research2023,10,2:3
9Rapid, automated, and reliable antimicrobial susceptibility test from positive blood culture by CAST‐R显示文摘Antimicrobial susceptibility tests(ASTs)are pivotal in combating multidrug resistant pathogens,yet they can be time‐consuming,labor‐intensive,and unstable.Using the AST of tigecycline for sepsis as the main model,here we establish an automated system of Clinical Antimicrobials Susceptibility Test Ramanometry(CAST‐R),based on D2O‐probed Raman microspectroscopy.Featuring a liquid robot for sample pretreatment and a machine learning‐based control scheme for data acquisition and quality control,the 3‐h,automated CAST‐R process accelerates AST by>10‐fold,processes 96 paralleled antibiotic‐exposure reactions,and produces high‐quality Raman spectra.The Expedited Minimal Inhibitory Concentration via Metabolic Activity is proposed as a quantitative and broadly applicable parameter for metabolism‐based AST,which shows 99%essential agreement and 93%categorical agreement with the broth microdilution method(BMD)when tested on 100 Acinetobacter baumannii isolates.Further tests on 26 clinically positive blood samples for eight antimicrobials,including tigecycline,meropenem,ceftazidime,ampicillin/sulbactam,oxacillin,clindamycin,vancomycin,and levofloxacin reveal 93%categorical agreement with BMD‐based results.The automation,speed,reliability,and general applicability of CAST‐R suggest its potential utility for guiding the clinical administration of antimicrobials.Pengfei Zhu Lihui Ren Ying Zhu Jing Dai Huijie Liu Yuli Mao Yuandong Li Yuehui He Xiaoshan Zheng Rongze Chen Xiaoting Fu Lili Zhang Lijun Sun Yuanqi Zhu Yuetong Ji Bo Ma Yingchun Xu Jian Xu Qiwen Yang 2022mLife2022,1,3:3
10Bioinspired structural color particles with multi-layer graphene oxide encapsulated nanoparticle components显示文摘Because of the unique features of spherical symmetry,angle-independency,good monodispersity,controllable components and morphologies,structural color particles(SCPs)have found great significances in various fields such as sensing,monitoring,biological assays,etc.Here,inspired by the melanosome-derived bright structural colors and the self-adhesivity of mussels,we present a kind of bioinspired SCPs assembled from polydopamine(PDA)-adhered multi-layer graphene oxide(GO)encapsulated silica nanoparticles(SNs).It is demonstrated that compared with traditional SCPs,the designed particles possess brighter and more vibrant structural colors,and no complicated modification is required during the functionalization process due to the abundant inherent functional groups of GO.The resultant SCPs are verified to be capable for direct hybridization chain reaction and multiplexed nucleic acid assays.These properties indicate the promising prospects of our designed SCPs.Yuxiao Liu Yuetong Wang Yu Wang Yilai Shu Hui Tan Yuanjin Zhao 2020Bioactive Materials2020,5,4:2
11Hydrogen peroxide and applications in green hydrocarbon nitridation and oxidation显示文摘Basic organic chemicals and high value–added products are mainly produced by hydrocarbon nitridation and oxidation.However,several drawbacks limit the application of the traditional oxidation and nitridation technologies in the future,such as complex processes,poor intrinsic safety,low atom utilization,and serious environmental pollution.The green nitridation and oxidation technologies are urgently needed.Hydrogen peroxide,a well–known green oxidant,is widely used in green hydrocarbon oxidation and nitridation.But its industrial production in China adopts fixed–bed technology,which is fall behind slurry–bed technology adopted by advanced foreign chemical companies,limiting the development of hydrogen peroxide industry and green hydrocarbon nitridation or oxidation industry.This article reviews the industrial production technologies of hydrogen peroxide and basic organic chemicals such as caprolactam,aniline,propene oxide,epichlorohydrin,phenol,and benzenediol,especially introduces the green production technologies of basic organic chemicals related with H_(2)O_(2).The article also emphasis on the efforts of Chinese researchers in developing its own slurry–bed technology of hydrogen peroxide production,and corresponding green hydrocarbon nitridation or oxidation technologies with hydrogen peroxide.Compared with traditional nitridation or oxidation technologies,green production technologies of caprolactam,propene oxide,epichlorohydrin,and benzenediol with hydrogen peroxide promote the nitrogen atom utilization from 60%to near 100%and the carbon atom utilization from 80%to near 100%.The waste emissions and environmental investments are reduced dramatically.Technological blockade against the green chemical industry of China are partially broken down,and technological upgrade in the chemical industry of China is guaranteed.Yanqiang Shi Yuetong Xia Guangtong Xu Langyou Wen Guohua Gao Baoning Zong 2022Chinese Journal of Chemical Engineering2022,35,1:2
12Modeling and Dynamic Analysis for the Foundation of a Blade-Rotor System显示文摘In order to achieve the model-based fault monitoring and diagnosis,an accurate model for the rotor system is necessary to locate and quantify faults.Since the dynamic characteristics of a blade-rotor system is influenced by foundation flexibility,the modeling and dynamic analyses on the foundation were sequentially investigated.Firstly,the effect of element size on the model convergence was investigated using the forward difference quotient as the slope of the frequency difference,which found that the model converged when the element size refined to 4mm.Secondly,a modal analysis and a harmonic response analysis were performed to obtain the dynamic characteristics of the foundation structure.Finally,an optimization to the foundation utilizing an additional stiffener was conducted to reduce the foundation response and make the critical speed far away from the working frequency band of 20—50Hz.Liang Anyang Huang Guoyuan Zhang Yuetong Yue Lin 2017Transactions of Nanjing University of Aeronautics and Astronautics2017,34,3:2
13Boston Ivy-Inspired Disc-Like Adhesive Microparticles for Drug Delivery显示文摘Microparticles with strong adherence are expected as efficient drug delivery vehicles.Herein,we presented an ingenious hydrogel microparticle recapitulating the adhesion mechanism of Boston ivy tendrils adhesive discs(AD)for durable drug delivery.The particles were achieved by replicating a silica colloidal crystal aggregates assembled in a droplet template after rapid solvent extraction.Due to their unique shape,the nanostructure,and the sticky hydrogel component,such novel microparticles exhibited prominent adhesive property to the wet tissue environment.It was demonstrated that the bioinspired microcarriers loading with dexamethasone had a good therapeutic effect for ulcerative colitis due to the strong adhesion ability for prolonging the maintenance of drug availability.These virtues make the biomimetic microparticles potentially ideal for many practical clinical applications,such as drug delivery,bioimaging,and biodiagnostics.Lijun Cai Guopu Chen Yuetong Wang Cheng Zhao Luoran Shang Yuanjin Zhao 2021Research2021,,1:2

16Revealing CO_(2)-Fixing SAR11 Bacteria in the Ocean by Raman-Based Single-Cell Metabolic Profiling and Genomics显示文摘The majority of marine microbes remain uncultured,which hinders the identification and mining of CO_(2)-fixing genes,pathways,and chassis from the oceans.Here,we investigated CO_(2)-fixing microbes in seawater from the euphotic zone of the Yellow Sea of China by detecting and tracking their ^(13)C-bicarbonate(^(13)C-HCO_(3)^(-))intake via single-cell Raman spectra(SCRS)analysis.The target cells were then isolated by Raman-activated Gravity-driven Encapsulation(RAGE),and their genomes were amplified and sequenced at one-cell resolution.The single-cell metabolism,phenotype and genome are consistent.We identified a notyet-cultured Pelagibacter spp.,which actively assimilates ^(13)C-HCO_(3)^(-),and also possesses most of the genes encoding enzymes of the Calvin-Benson cycle for CO_(2) fixation,a complete gene set for a rhodopsin-based light-harvesting system,and the full genes necessary for carotenoid synthesis.The four proteorhodopsin(PR)genes identified in the Pelagibacter spp.were confirmed by heterologous expression in E.coli.These results suggest that hitherto uncultured Pelagibacter spp.uses light-powered metabolism to contribute to global carbon cycling.Xiaoyan Jing Yanhai Gong Teng Xu Paul ADavison Craig MacGregor-Chatwin CNeil Hunter La Xu Yu Meng Yuetong Ji Bo Ma Jian Xu Wei E.Huang 2022BioDesign Research2022,,1:1
