3A rare Waxy allele coordinately improves rice eating and cooking quality and grain transparency显示文摘In rice(Oryza sativa), amylose content(AC) is the major factor that determines eating and cooking quality(ECQ). The diversity in AC is largely attributed to natural allelic variation at the Waxy(Wx)locus. Here we identified a rare Wx allele, Wx^(mw) ,which combines a favorable AC, improved ECQ and grain transparency. Based on a phylogenetic analysis of Wx genomic sequences from 370 rice accessions, we speculated that Wx^(mw) may have derived from recombination between two important natural Wx alleles, Wx^(in) and Wx^(b). We validated the effects of Wx^(mw) on rice grain quality using both transgenic lines and near-isogenic lines(NILs). When introgressed into the japonica Nipponbare(NIP) background, Wx^(mw) resulted in a moderate AC that was intermediate between that of NILs carrying the Wx^(b)allele and NILs with the Wx^(mp) allele. Notably, mature grains of NILs fixed for Wx^(mw) had an improved transparent endosperm relative to soft rice. Further, we introduced Wx^(mw) into a high-yielding japonica cultivar via molecular marker-assisted selection: the introgressed lines exhibited clear improvements in ECQ and endosperm transparency. 4Distribution and geological significance of 17α(H)-diahopanes from different hydrocarbon source rocks of Yanchang Formation in Ordos Basin显示文摘Based on GC-MS testing data of many saturated hydrocarbon samples, 17α(H)-C30 diahopanes (C30*) are extensively distributed in the lacustrine hydrocarbon source rocks of the Yanchang Formation in Ordos Basin, but show remarkable differences in relative abundance among various source rocks. Generally, Chang 7 high-quality source rock (oil shale) developed in deep lake anoxic environment shows lower C30* content, whereas Chang 6-9 dark mudstone developed in shallow to semi-deep lake, sub-oxidizing environment shows relatively high to high C30* value. Particularly, Chang 7 and Chang 9 black mudstones in Zhidan region in the northeast of the lake basin show extremely high C30* value. A comparative analysis was made based on lithology, organic types and various geochemical parameters indicative of redox environment, and the results indicate that environmental factors such as redox settings and lithology are key factors that control the C30* relative abundance, while organic types and maturity may be minor factors. High to extremely high C30* values are indicative of sub-oxidizing environment of fresh-brackish water and shallow to semi-deep lake. Therefore, research on C30* relative content and distribution in lacustrine hydrocarbon source rocks in the Yanchang Formation, especially on the difference in C30* between Chang 7 high-quality source rocks (oil shale) and Chang 6-91 source rocks (dark mudstone), will provide an important approach for classification of Mesozoic lacustrine crudes and detailed oil-source correlation in the basin.ZHANG WenZheng YANG Hua HOU LiHui LIU Fei 2009Science China Earth Sciences2009,52,7:16
5Down.regulation of E.cadherin enhances prostate cancer chemoresistance via Notch signaling显示文摘Background:The chemoresistance of prostate cancer(PCa)is invariably associated with the aggressiveness and metastasis of this disease.New emerging evidence indicates that the epithelial-to-mesenchymal transition(EMT)may play pivotal roles in the development of chemoresistance and metastasis.As a hallmark of EMT,E-cadherin is suggested to be a key marker in the development of chemoresistance.However,the molecular mechanisms underlying PCa chemoresistance remain unclear.The current study aimed to explore the association between EMT and chemoresistance in PCa as well as whether changing the expression of E-cadherin would affect PCa chemoresistance.Methods:Parental PC3 and DU145 cells and their chemoresistant PC3-Tx R and DU145-Tx R cells were analyzed.PC3-Tx R and DU145-Tx R cells were transfected with E-cadherin-expressing lentivirus to overexpress E-cadherin;PC3 and DU145 cells were transfected with small interfering RNA to silence E-cadherin.Changes of EMT phenotype-related markers and signaling pathways were assessed by Western blotting and quantitative real-time polymerase chain reaction.Tumor cell migration,invasion,and colony formation were then evaluated by wound healing,transwell,and colony formation assays,respectively.The drug sensitivity was evaluated using MTS assay.Results:Chemoresistant PC3-Tx R and DU145-Tx R cells exhibited an invasive and metastatic phenotype that associated with EMT,including the down-regulation of E-cadherin and up-regulation of Vimentin,Snail,and N-cadherin,comparing with that of parental PC3 and DU145 cells.When E-cadherin was overexpressed in PC3-Tx R and DU145-Tx R cells,the expression of Vimentin and Claudin-1 was down-regulated,and tumor cell migration and invasion were inhibited.In particular,the sensitivity to paclitaxel was reactivated in E-cadherin-overexpressing PC3-Tx R and DU145-Tx R cells.When E-cadherin expression was silenced in parental PC3 and DU145 cells,the expression of Vimentin and Snail was up-regulated,and,particularly,the sensitivity to paclitaxel was decreased.Interestingly,Notch-1 expression was up-regulated in PC3-Tx R and DU145-Tx R cells,whereas the E-cadherin expression was down-regulated in these cells comparing with their parental cells.The use ofγ-secretase inhibitor,a Notch signaling pathway inhibitor,significantly increased the sensitivity of chemoresistant cells to paclitaxel.Conclusion:The down-regulation of E-cadherin enhances PCa chemoresistance via Notch signaling,and inhibiting the Notch signaling pathway may reverse PCa chemoresistance.Wenchu Wang Lihui Wang Atsushi Mizokami Junlin Shi Chunlin Zou Jinlu Dai Evan T. 7Synergistic effect between Sn and K promoters on supported platinum catalyst for isobutane dehydrogenation显示文摘Catalytic dehydrogenation of isobutane has recently received considerable attention because of the increasing demand for isobutene.In this study,the synergistic effect between Sn and K on PtSnK/γ-Al2O3 catalysts has been investigated by changing the content of Sn.It was found that with the presence of potassium,suitable addition of Sn could not only increase the metal dispersion,but also reduce the catalyst acidity.In these cases,the synergistic effect could also strengthen the interactions between the metal and support,which resulted in an increase in both catalytic activity and stability.In our experiments,Pt-0.6SnK/Al catalyst exhibited the lowest deactivation rate (12.4%) and showed a selectivity to isobutene higher than 94% at the isobutane conversion of about 45.3% after running the reaction for 6 h.However,with the excessive loading of Sn,surface property of active sites and the interactions between metal and support were changed.As a result,the initial optimal ratio between the metallic function and acid function would be destroyed,which was disadvantageous to the reaction.Yiwei Zhang Yuming Zhou Lihui Wan Mengwei Xue Yongzheng Duan Xuan Liu 2011Journal of Natural Gas Chemistry2011,20,6:12

11Genetic diversity and DNA fingerprinting in jute(Corchorus spp.) based on SSR markers显示文摘Genetic diversity analysis and DNA finger printing are very useful in breeding programs,seed conservation and management. Jute(Corchorus spp.) is the second most important natural fiber crop after cotton. DNA fingerprinting studies in jute using SSR markers are limited. In this study, 58 jute accessions, including two control varieties(Huangma 179 and Kuanyechangguo) from the official variety registry in China were evaluated with 28 pairs of SSR primers. A total of 184 polymorphic loci were identified. Each primer detected 3 to 15 polymorphic loci, with an average of 6.6. The 58 jute accessions were DNA-fingerprinted with 67 SSR markers from the 28 primer pairs. These markers differentiated the 58 jute accessions from one another, with Co SSR305-120 and Co SSR174-195 differentiating Huangma 179 and Kuanyechangguo, respectively. NTSYS-pc2.10 software was used to analyze the genetic diversity in the 58 jute accessions. Their genetic similarity coefficients ranged from 0.520 to 0.910 with an average of 0.749, indicating relatively great genetic diversity among them. The 58 jute accessions were divided into four groups with the coefficient 0.710 used as a value for classification, consistent with their species and pedigrees. 12Effect of Ga Addition on Catalytic Performance of PtSnNa/ZSM-5 Catalyst for Propane Dehydrogenation显示文摘PtSnNaGa/ZSM-5 catalysts with different contents of Ga were prepared and characterized by X-ray diffraction (XRD), nitrogen adsorption, hydrogen chemisorption, ammonia temperature-programmed desorption (NH3-TPD), hydrogen temperature-programmed reduction (H2-TPR), and temperature-programmed oxidation (TPO) techniques. The performances of these catalysts for propane dehydrogenation were investigated. The test results indicated that the addition of Ga not only could improve the catalytic stability and propene selectivity, but also could effectively prevent the catalysts from coking. It was found that the PtSnNaGa(0.5 m%)/ZSM-5 catalyst exhibited the best performance in terms of propene selectivity and propane conversion. 14Vertical Distribution Characteristics of PM2.5 Observed by a Mobile Vehicle Lidar in Tianjin, China in 2016显示文摘We present mobile vehicle lidar observations in Tianjin, China during the spring, summer, and winter of 2016. Mobile observations were carried out along the city border road of Tianjin to obtain the vertical distribution characteristics of PM_(2.5). Hygroscopic growth was not considered since relative humidity was less than 60% during the observation experiments. PM_(2.5) profile was obtained with the linear regression equation between the particle extinction coefficient and PM_(2.5) mass concentration. In spring, the vertical distribution of PM_(2.5) exhibited a hierarchical structure. In addition to a layer of particles that gathered near the ground, a portion of particles floated at 0.6–2.5-km height. In summer and winter, the fine particles basically gathered below 1 km near the ground. In spring and summer, the concentration of fine particles in the south was higher than that in the north because of the influence of south wind. In winter, the distribution of fine particles was opposite to that measured during spring and summer. High concentrations of PM_(2.5) were observed in the rural areas of North Tianjin with a maximum of 350 μg m–3 on 13 December2016. It is shown that industrial and ship emissions in spring and summer and coal combustion in winter were the major sources of fine particles that polluted Tianjin. 17Generation of tooth-like structures from integration-free human urine induced pluripotent stem cells显示文摘Background:Tooth is vital not only for a good smile,but also good health.Yet,we lose tooth regularly due to accidents or diseases.An ideal solution to this problem is to regenerate tooth with patients’own cells.Here we describe the generation of tooth-like structures from integration-free human urine induced pluripotent stem cells(ifhU-iPSCs).Results:We first differentiated ifhU-iPSCs to epithelial sheets,which were then recombined with E14.5 mouse dental mesenchymes.Tooth-like structures were recovered from these recombinants in 3 weeks with success rate up to 30%for 8 different iPSC lines,comparable to H1 hESC.We further detected that ifhU-iPSC derived epithelial sheets differentiated into enamel-secreting ameloblasts in the tooth-like structures,possessing physical properties such as elastic modulus and hardness found in the regular human tooth.Conclusion:Our results demonstrate that ifhU-iPSCs can be used to regenerate patient specific dental tissues or even tooth for further drug screening or regenerative therapies.Jinglei Cai Yanmei Zhang Pengfei Liu Shubin Chen Xuan Wu Yuhua Sun Ang Li Ke Huang Rongping Luo Lihui Wang Ying Liu Ting Zhou Shicheng Wei Guangjin Pan Duanqing Pei 2013Cell Regeneration2013,2,1:4

19Optimization of Ultrasonic Extraction Conditions of Polyphenols from the Needles of Black Pine (Pinus thunbergii) Using Response Surface Methodology (RSM)显示文摘Ultrasonic extraction conditions of polyphenols from the needles of black pine(Pinus thunbergii) were optimized by single factors analysis and Box-Behnken experiment design. The results showed that both of ethanol concentration and ultrasonic time had a significant effect on polyphenol extraction, and there was significant interaction between any two variables in the three parameters of ethanol concentration, ultrasonic temperature and ultrasonic time. The optimal parameters for polyphenol extraction were the ethanol concentration of 52.67%, ultrasonic temperature at 49.21 ℃and ultrasonic time of 30.76 min. Under these conditions, the actual content of polyphenols was close to the predicted value. 20Using an Improved Clustering Method to Detect Anomaly Activities显示文摘In this paper, an improved k-means based clustering method (IKCM) is proposed. By refining the initial cluster centers and adjusting the number of clusters by splitting and merging procedures, it can avoid the algorithm resulting in the situation of locally optimal solution and reduce the number of clusters dependency. The IKCM has been implemented and tested. We perform experiments on KDD-99 data set. The comparison experiments with H-means+also have been conducted.
