2A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 3Blockade of cholecystokinin-2 receptor and cyclooxygenase-2 synergistically induces cell apoptosis, and inhibits the proliferation of human gastric cancer cells in vitro显示文摘Gastrin and cyclooxygenase-2(COX-2) playimportant roles in the carcinogenesis and progression ofgastric cancer.However,it remains unknown whether the combination of cholecystokinin-2(CCK-2) receptor antagonist plus COX-2 inhibitor exerts synergistic anti-tumor effects on human gastric cancer.Here,we demonstrated that the combination of AG-041R(a CCK-2 receptor antagonist) plus NS-398(a selective COX-2 inhibitor) treatment had synergistic effects on proliferation inhibition,apoptosis induction,down-regulation of Bcl-2 and up-regulation of Bax expression in MKN-45 cells.These results indicate that simultaneous targeting of CCK-2 receptor and COX-2 may inhibit gastric cancer development more effectively than targeting either molecule alone.(C)2008 Elsevier Ireland Ltd.All rights reserved.Sun, W. 4The global image of the Moon obtained by the Chang'E-1:Data processing and lunar cartography显示文摘The global lunar image of the first phase of Chinese Lunar Exploration Program is the first image that covered all over the surface of the Moon. It will serve as a critical foundation for succeeding exploration and scientific research. In this paper, the acquisition, characteristics, and data quality of Chang'E-1 CCD image data are described in detail. Also described are the methodology and procedure of data processing. According to rule of planetary cartography, the image data have been processed, geometrically corrected, and then mosaicked and merged in a scale of 1:2.5 million. The results of data processing and charting show that the image data of Chang'E-1 CCD and their geometric precision meet the demand of charting a map in the scale of 1:2.5 million. The relative geometric positioning precision of the global image is better than 240 m, and the absolute geometric positioning precision is slightly better than that of the ULCN2005 and Clementine lunar basemap (V2.0). The plane positioning precision is approximately 100-1500 m. 6Laser altimetry data of Chang'E-1 and the global lunar DEM model显示文摘The Laser AltiMeter (LAM), as one of the main payloads of Chang'E-1 probe, is used to measure the topography of the lunar surface. It performed the first measurement at 02:22 on November 28th, 2007. Up to December 4th 2008, the total number of measurements was approximately 9.12 million, covering the whole surface of the Moon. Using the LAM data, we constructed a global lunar Digtal Elevation Model (DEM) with 3 km spatial resolution. The model shows pronounced morphological characteristics, legible and vivid details of the lunar surface. The plane positioning accuracy of the DEM is 445 m (1σ), and the vertical accuracy is 60 m (1σ). From this DEM model, we measured the full range of the altitude difference on the lunar sur-face, which is about 19.807 km. The highest point is 10.629 km high, on a peak between crater Korolev and crater Dirichlet-Jackson at (158.656°W, 5.441°N) and the lowest point is -9.178 km in height, inside crater Antoniadi (172.413°W, 70.368°S) in the South Pole-Aitken Basin. 8Finite-time adaptive consensus of a class of multi-agent systems显示文摘Multi-agent systems(MASs) are ubiquitous in natural and artificial systems. This paper aims to establish the finite-time adaptive consensus criterion for a class of MASs with nonlinear dynamics. Traditionally, the finite-time consensus criterion is often established based on the prior information on Lipschitz constants and the eigenvalues of Laplacian matrix. However, it is difficult to acquire the above prior information for most real-world engineering systems. To overcome the above difficulty, this paper develops the finite-time consensus criteria for a class of MASs with nonlinear dynamics via adaptive technique. In detail, we design the finite-time distributed node-based and edge-based adaptive consensus protocols for a class of MASs with fixed and switching topologies. 9A 2.8 Ma record of environmental evolution and tectoni events inferred from the Cuoe core in the middle of Tibetan Plateau显示文摘Based on a multi-proxy investigation into the deep core of the Cuoe Lake in the middle of Tibetan Plateau, a 2.8 Ma paleoclimatic and paleoenvironmental evolution is recon- structed. The result of magnetic stratum indicates that the lake basin was formed at about 2.8 MaBP, while the multi-proxy analyses of lithology, grain size, magnetic susceptibility and geo- chemical elements reveal that there have been three major environmental evolution stages and at least two intensive uplifts of the Tibetan Plateau in the lake basin area, i.e. during 2.8—2.5 MaBP, the lake basin came into being as a result of the disaggregation of the planation surface and rapid rising of the Tibetan Plateau. During 2.5—0.8 MaBP, with gradual uplift of the Tibetan Plateau, the environment of this area was more effectively controlled by the climatic cycle of the alternative glacial-interglacial stages. 11Relationship Between Programmed Death-ligand 1 and Clinicopathological Characteristics in Non-small Cell Lung Cancer Patients显示文摘Objective To evaluate the correlation between programmed death-ligand 1(PD-L1)expression in primary lung cancer cells,tumor associated macrophages(TAM)and patients’clinicopathological characteristics.Methods From 2008 to 2010,208 non-small cell lung cancer patients who underwent surgery or CT-guided biopsy were recruited from Huadong Hospital,Fudan University.Immunohistochemistry staining was performed to evaluate the PD-L1 expression in both primary lung cancer cells and CD68 positive TAM.The relationship between PD-L1 expression and the clinical pathology was evaluated usingχ2test.Spearman’s rank correlations were used to determine the correlation between PD-L1 expression in tumor cells and macrophages.Results Positive PD-L1 expression in primary cancer cells was found in 136(65.3%)patients,which were negatively correlated with lymph node metastasis(P=0.009)and smoking history(P=0.036).Besides,TAM with PD-L1 expression(found in 116 patients)was positively associated with smoking history(P=0.034),well-differentiation(P=0.029)and negative lymph node metastasis(P=0.0096).A correlation between PD-L1 expression in primary tumor cells and non-small cell lung cancer associated macrophages was found(r=0.228,P=0.021).Conclusion PD-L1,secreted from TAM,might induce cancer cells apoptosis,and decrease lymph node metastasis.Yan-yan Chen Liu-bo Wang Hui-li Zhu Xiang-yang Li Yan-ping Zhu Yu-lei Yin Fan-zhen Lü Zi-li Wang Jie-ming Qu 2013Chinese Medical Sciences Journal2013,28,3:14

13Primary scientific results of Chang'E-1 lunar mission显示文摘The strategic plan for the development of the unmanned Chinese Lunar Exploration Program is characterized by three distinct stages: 'orbiting around', 'landing on' and 'returning from' the Moon. The first Chinese lunar probe, Chang'E-1, which was successfully launched on October 24th, 2007 at Xichang Satellite Launch Center, and guided to crash on the Moon on March 1st, 2009, at 52.36°E, 1.50°S, in the north of Mare Fecunditatis, is the first step towards the 'orbiting around' stage. The Chang'E-1 mission lasted 495 days, exceeding the expected life-span by about four months. A total of 1.37 TB raw data was received from Chang'E-1. It was then processed into 4 TB scientific data products at various levels. Many scientific results have been obtained by analyzing these data, including especially the 'global lunar image from the first Chinese lunar explora- tion mission'. All scientific goals of Chang'E-1 have been achieved. It provides much useful materials for further advances of lunar sciences and planetary chemistry. 14Modeling of Microstructure Evolution in 22MnB5 Steel during Hot Stamping显示文摘Automobile manufacturers have been increasingly adopting hot-stamped parts for use in newly designed vehicles to improve crash worthiness and fuel efficiency.However,the simulation of hot stamping is rather complex and challenging,and further research still needs to be done on hot stamping hardening mechanism.The microstructure evolution and hardening mechanisms during hot stamping of 22MnB5steel were thoroughly investigated,using information provided in the literatures as well as experimental results.New models were developed to predict the grain growth during heating and the flow stress of a manganese boron steel(22MnB5)with high hardenability by the Gleeble simulation experimental results.The deformed austenite decomposition during stamping and quenching was emphatically quantified based on the transformation thermodynamic and kinetic theories,and the relationship of microstructure to properties was analyzed.The results showed that the optimal process to obtain homogeneous and small lath martensite is heating at 900-950℃for 5min and then quenching at 50℃/s with a pressing time about 8s.Li-juan ZHU Zheng-wei GU Hong XU Yi L Jiang CHAO 2014Journal of Iron and Steel Research(International)2014,21,2:10
15Protective effect of early enteral feeding on postburn impairment of liver function and its mechanism in rats显示文摘AIM To study the protective effect of earlyenteral feeding(EEF)on the postburnimpairment of liver function and its mechanism.METHODS Wistar rats with 30% of total bodysurface area(TBSA)full-thickness burn wereemployed.The effects of EEF on the postburnchanges of gastric intramucosal pH,endotoxinlevels in portal vein,water contents of hepatictissue,blood concentrations of tumor necrosisfactor(TNF-α),plasma activities of alanineaminotransferase(ALT)and asparateaminotransferase(AST),as well as the bloodcontents of total(TB)and direct bilirubin(DB),total protein(TP)and albumin(ALB)wereserially determined within 48h postburn.RESULTS EEF could significantly improvegastric mucosal acidosis,reduce portal veinendotoxin level and water content of hepatictissue,as well as plasma concentrations of TNF-α at all timepoints after severe burns(P<0.01);postburn elevation of the plasma activities ofALT,AST and the contents of TB,DB wereeffectively prevented,whereas the plasmaconcentrations of TP and ALB were markedlyincreased 24 h and 48 h posturn in EEF groupcompared with that of the burn without EEFgroup(P<0.01).CONCLUSION EEF has significant beneficialeffects on the improvement of hepatic function in rats after severe burn,and is probably relatedwith an increase in splanchnic blood flow,reduction of the absorption of gut-originendotoxin and the consequent release ofinflammatory mediators.Zhu L Yang ZC Li A Cheng DC 2000World Journal of Gastroenterology2000,6,1:10
16Use of a Multiplex RT-PCR Assay for Simultaneous Detection of the North American Genotype Porcine Reproductive and Respiratory Syndrome Virus,Swine Influenza Virus and Japanese Encephalitis Virus显示文摘A multiplex reverse transcriptase-polymerase chain reaction(multiplex RT-PCR) assay was developed and subsequently evaluated for its efficacy in the detection of multiple viral infections simultaneously,in swine.Specific primers for each of the 3 RNA viruses,North American genotype porcine reproductive and respiratory syndrome virus,Japanese encephalitis virus,and swine influenza virus,were used in the testing procedure.The assay was shown to be highly sensitive because it could detect as little as 10-5 ng of each of the respective amplicons in a single sample containing a composite of all 3 viruses.The assay was also effective in detecting one or more of the same viruses in various combinations in specimens,including lymph nodes,lungs,spleens,and tonsils,collected from clinically ill pigs and in spleen specimens collected from aborted pig fetuses.The results from the multiplex RT-PCR were confirmed by virus isolation.The relative efficiency(compared to the efficiency of separate assays for each virus) and apparent sensitivity of the multiplex RT-PCR method show that this method has potential for application in routine molecular diagnostic procedures.CHEN Hong-ying WEI Zhan-yong ZHANG Hong-ying LüXiao-li ZHENG Lan-lan CUI Bao-an LIU Jinpeng ZHU Qian-lei WANG Zi-xin 2010Agricultural Sciences in China2010,9,7:10
17Leaf nutrient dynamics and nutrient resorption:a comparison between larch plantations and adjacent secondary forests in Northeast China显示文摘Aims Conversion of secondary forests to pure larch plantations is a common management practice driven by the increasing demand for timber production in Northeast China,resulting in a reduction in soil nutrient availability after a certain number of years following conversion.Nutrient resorption prior to leaf senescence was related to soil fertility,an important nutrient conservation strategy for plants,being especially significant in nutrient-poor habitats.However,the seasonal dynamics of leaf nutrients and nutrient resorption in response to secondary forest conversion to larch plantations is not well understood.Methods A comparative experiment between larch plantations(Larix spp.)and adjacent secondary forests(dominant tree species including Quercus mongolica,Acer mono,Juglans mandshurica and Fraxinus rhynchophylla)was conducted.We examined the variations in leaf nutrient(macronutrients:N,P,K,Ca and Mg;micronutrients:Cu and Zn)concentrations of these tree species during the growing season from May to October in 2013.Nutrient resorption efficiency and proficiency were compared between Larix spp.and the broadleaved species in the secondary forests.Important Findings Results show that the seasonal variation of nutrient concentrations in leaves generally exhibited two trends,one was a downward trend for N,P,K,Cu and Zn,and another was an upward trend for Ca and Mg.The variations in foliar nutrient concentrations were mainly controlled by the developmental stage of leaves rather than by tree species.Resorption of the observed seven elements varied among the five tree species during leaf senescence.Nutrient resorption efficiency varied 6–75%of N,P,K,Mg,Cu and Zn,while Ca was not retranslocated in the senescing leaves of all species,and Mg was not retranslocated in Larix spp.Generally,Larix spp.tended to be more efficient and proficient(higher than 6–30%and 2–271%of nutrient resorption efficiency and resorption proficiency,respectively)in resorbing nutrients than the broadleaved species in the secondary forests,indicating that larch plantations had higher leaf nutrient resorption and thus nutrient use efficiency.Compared with Larix spp.,more nutrients would remain in the leaf litter of the secondary forests,indicating an advantage of secondary forests in sustaining soil fertility.In contrast,the larch plantation would reuse internal nutrients rather than lose nutrients with litter fall and thus produce a positive feedback to soil nutrient availability.In summary,our results suggest that conversion from secondary forests to pure larch plantations would alter nutrient cycling through a plantmediated pathway.Tao Yan Xiaotao Lü Kai Yang Jiaojun Zhu 2016Journal of Plant Ecology2016,9,2:10
18U-Pb zircon geochronology, geochemistry and kinetics of the Huaniushan A-type granite in Northwest China显示文摘The Huaniushan granite is located at the Beishan orogenic belt, northwestern China. At the contact zone between the granite and marble, a hydrothermal Pb-Zn and skarn Au deposit is formed. LA-ICP-MS zircon U-Pb dating yielded a weighted mean 206Pb/238U age of 229.5±2.6 Ma (MSDW=0.93) for the Huaniushan granite, imply-ing its Late Triassic intrusion. Geochemistry analyses show that the Huaniushan granite is enriched in Si, K, Na, and REE, and depleted in Mg and Ca, with contents of SiO2 (70.8% to 74.4%), Na2O+K2O (8.8% to 10.2%), CaO (0.93% to 1.44%), and MgO (0.14% to 0.48%). REE is characterized by obvious negative Eu anomaly. Rb, Th, U, K, Pb, Nb, Zr and Hf elements are rich in the granite while Ba, Sr, P, Ti and Eu are deplete. The granite has a high (Zr+Nb+Ce+Y) abundance and 104 Ga/Al ratios. Petrology, major and trace elements data all indicate that the Hua-niushan granite is A-type granite which intruded in a post-collisional extensional tectonic setting. The magma was dominantly sourced from partial melting of crustal intermediate-felsic igneous rocks. 19Recovery of alkali and alumina from Bayer red mud by the calcification–carbonation method显示文摘Red mud produced in the Bayer process is a hazardous solid waste because of its high alkalinity; however, it is rich in valuable components such as titanium, iron, and aluminum. In this study, a novel calcification–carbonation method was developed to recover alkali and alumina from Bayer red mud under mild reaction conditions. Batch experiments were performed to evaluate the potential effects of important parameters such as temperature, amount of CaO added, and CO_2 partial pressure on the recovery of alkali and alumina. The results showed that 95.2% alkali and 75.0% alumina were recovered from red mud with decreases in the mass ratios of Na_2 O to Fe_2O_3 and of Al_2O_3 to Fe_2O_3 from 0.42 and 0.89 to 0.02 and 0.22, respectively. 20Reduced EGFR signaling enhances cartilage destruction in a mouse osteoarthritis model显示文摘Osteoarthritis(OA) is a degenerative joint disease and a major cause of pain and disability in older adults. We have previously identified epidermal growth factor receptor(EGFR) signaling as an important regulator of cartilage matrix degradation during epiphyseal cartilage development. To study its function in OA progression, we performed surgical destabilization of the medial meniscus(DMM) to induce OA in two mouse models with reduced EGFR activity, one with genetic modification(EgfrWa5/1mice) and the other one with pharmacological inhibition(gefitinib treatment). Histological analyses and scoring at 3 months post-surgery revealed increased cartilage destruction and accelerated OA progression in both mouse models. TUNEL staining demonstrated that EGFR signaling protects chondrocytes from OA-induced apoptosis, which was further confirmed in primary chondrocyte culture. Immunohistochemistry showed increased aggrecan degradation in these mouse models, which coincides with elevated amounts of ADAMTS5 and matrix metalloproteinase 13(MMP13), the principle proteinases responsible for aggrecan degradation, in the articular cartilage after DMM surgery. Furthermore, hypoxia-inducible factor 2a(HIF2a), a critical catabolic transcription factor stimulating MMP13 expression during OA, was also upregulated in mice with reduced EGFR signaling.
