| 8 | Molecular subtyping and genomic profiling expand precision medicine in refractory metastatic triple-negative breast cancer:the FUTURE trial显示文摘Triple-negative breast cancer(TNBC)is a highly heterogeneous disease,and molecular subtyping may result in improved diagnostic precision and targeted therapies.Our previous study classified TNBCs into four subtypes with putative therapeutic targets.Here,we conducted the FUTURE trial(ClinicalTrials.gov identifer:NCT03805399),a phaseⅠb/Ⅱsubtyping-based and genomic biomarker-guided umbrella trial,to evaluate the efficacy of these targets.Patients with refractory metastatic TNBC were enrolled and stratifed by TNBC subtypes and genomic biomarkers,and assigned to one of these seven arms:(A)pyrotinib with capecitabine,(B)androgen receptor inhibitor with CDK4/6 inhibitor,(C)anti PD-1 with nab-paclitaxel,(D)PARP inhibitor included,(E)and(F)anti-VEGFR included,or(G)mTOR inhibitor with nab paclitaxel.The primary end point was the objective response rate(ORR).We enrolled 69 refractory metastatic TNBC patients with a median of three previous lines of therapy(range,1-8).Objective response was achieved in 20(29.0%,95%confidence interval(CI):18.7%-41.2%)of the 69 intention-to-treat(ITT)patients.Our results showed that immunotherapy(arm C),in particular,achieved the highest ORR(52.6%,95%CI:28.9%-75.6%)in the ITT population.Arm E demonstrated favorable ORR(26.1%,95%C:10.2%-48.4%in the ITT population)but with more high grade(23)adverse events.Somatic mutations of TOP2A and CD8 immunohistochemical score may have the potential to predict immunotherapy response in the immunomodulatory subtype of TNBC.In conclusion,the phaseⅠb/ⅡFUTURE trial suggested a new concept for TNBC treatment,demonstrating the clinical beneft of subtyping-based targeted therapy for refractory metastatic TNBC. | Yi-Zhou Jiang Yin Liu Yi Xiao Xin Hu Lin Jiang Wen-ia Zuo Ding Ma Jiahan Ding Xiaoyu Zhu Jianjun Zou Claire Verschraegen Daniel G.Stover Virginia Kaklamani Zhong-Hua Wang Zhi-Ming Shao | 2021 | Cell Research2021,31,2: | 37 |
| 9 | SlMYB75,an MYB-type transcription factor,promotes anthocyanin accumulation and enhances volatile aroma production in tomato fruits显示文摘Genetic manipulation of genes to upregulate specific branches of metabolic pathways is a method that is commonly used to improve fruit quality.However,the use of a single gene to impact several metabolic pathways is difficult.Here,we show that overexpression of the single gene SlMYB75(SlMYB75-OE)is effective at improving multiple fruit quality traits.In these engineered fruits,the anthocyanin content reached 1.86mg g−1 fresh weight at the red-ripe stage,and these SlMYB75-OE tomatoes displayed a series of physiological changes,including delayed ripening and increased ethylene production.In addition to anthocyanin,the total contents of phenolics,flavonoids and soluble solids in SlMYB75-OE fruits were enhanced by 2.6,4,and 1.2 times,respectively,compared to those of wild-type(WT)fruits.Interestingly,a number of aroma volatiles,such as aldehyde,phenylpropanoid-derived and terpene volatiles,were significantly increased in SlMYB75-OE fruits,with some terpene volatiles showing more than 10 times higher levels than those in WT fruits.Consistent with the metabolic assessment,transcriptomic profiling indicated that the genes involved in the ethylene signaling,phenylpropanoid and isoprenoid pathways were greatly upregulated in SlMYB75-OE fruits.Yeast one-hybrid and transactivation assays revealed that SlMYB75 is able to directly bind to the MYBPLANT and MYBPZM cis-regulatory elements and to activate the promoters of the LOXC,AADC2 and TPS genes.The identification of SlMYB75 as a key regulator of fruit quality attributes through the transcriptional regulation of downstream genes involved in several metabolic pathways opens new avenues towards engineering fruits with a higher sensory and nutritional quality. | Wei Jian Haohao Cao Shu Yuan Yudong Liu Juanfang Lu Wang Lu Ning Li Jianhui Wang Jian Zou Ning Tang Chan Xu Yulin Cheng Yanqiang Gao Wanpeng Xi Mondher Bouzayen Zhengguo Li | 2019 | Horticulture Research2019,6,1: | 39 |
| 10 | Lime and Phosphate Could Reduce Cadmium Uptake by Five Vegetables Commonly Grown in South China显示文摘A pot experiment was conducted in artificially Cd-contaminated (5 mg Cd kg-1) soils to investigate the feasibility of using lime (3 g kg-1) or phosphate (80 mg P kg-1) to mitigate uptake of Cd by vegetables.Five common vegetables in South China,including lettuce (Lactuca sativa L.var.ramosa Hort.),Chinese cabbage [Brassica rapa L.subsp.Chinensis (L.) var.parachinensis (L.H.Bailey) Hanect],Chinese broccoli (Brassica oleracea L.var.albiflora Kuntze),white amaranth (Amaranthus tricolor L.) and purslane (Amaranthus viridis L.),were grown in the soils and harvested after 60 d.The results showed that liming significantly reduced Cd uptake by most vegetables by 40%-50% (or a maximum of 70%),mainly due to immobilization of soil Cd.Increased availability of Ca in the soil might also contribute to the Cd uptake reduction as a result of absorption competition between Ca and Cd.Liming caused biomass reduction in white amaranth and purslane,but did not influence growth of the other vegetables.Phosphate decreased Cd uptake by vegetables by 12%-23%.Compared with lime,phosphate decreased,to a smaller extent,the bioavailability of Cd in most cases.Phosphate markedly promoted growth of vegetables.Changes in soil chemistry by adding lime or phosphate did not markedly influence nutrient uptake of vegetables except that lime increased Ca content and phosphate increased P content in shoots of the vegetables.The results suggested that a proper application of lime could be effective in reducing Cd uptake of vegetables,and phosphate could promote growth of the vegetables as well as alleviate the toxicity of Cd. | TAN Wan-Neng LI Zhi-An QIU Jing ZOU Bi LI Ning-Yu ZHUANG Ping WANG Gang | 2011 | Pedosphere2011,21,2: | 34 |
| 11 | Berberine inhibits hepatic gluconeogenesis via the LKB1-AMPK-TORC2 signaling pathway in streptozotocin-induced diabetic rats显示文摘AIM: To investigate the molecular mechanisms of berberine inhibition of hepatic gluconeogenesis in a diabetic rat model.METHODS: The 40 rats were randomly divided into five groups. One group was selected as the normal group. In the remaining groups(n = 8 each), the rats were fed on a high-fat diet for 1 mo and received intravenous injection of streptozotocin for induction of the diabetic models. Berberine(156 mg/kg per day)(berberine group) or metformin(184 mg/kg per day)(metformin group) was intragastrically administered to the diabetic rats and 5-aminoimidazole-4-carboxamide1-β-D-ribofuranoside(AICAR)(0.5 mg/kg per day)(AICAR group) was subcutaneously injected to the diabetic rats for 12 wk. The remaining eight diabetic rats served as the model group. Fasting plasma glucose and insulin levels as well as lipid profile were tested.The expressions of proteins were examined by western blotting. The nuclear translocation of CREB-regulated transcription co-activator(TORC)2 was observed by immunohistochemical staining. RESULTS: Berberine improved impaired glucose tolerance and decreased plasma hyperlipidemia. Moreover, berberine decreased fasting plasma insulin and homeostasis model assessment of insulin resistance(HOMA-IR). Berberine upregulated protein expression of liver kinase(LK)B1, AMP-activated protein kinase(AMPK) and phosphorylated AMPK(p-AMPK). The level of phophorylated TORC2(p-TORC2) protein in the cytoplasm was higher in the berberine group than in the model group, and no significant difference in total TORC2 protein level was observed. Immunohistochemical staining revealed that more TORC2 was localized in the cytoplasm of the berberine group than in the model group. Moreover, berberine treatment downregulated protein expression of the key gluconeogenic enzymes(phosphoenolpyruvate carboxykinase and glucose-6-phosphatase) in the liver tissues. | 12 | Carbon Dioxide, Methane, and Nitrous Oxide Emissions from a Rice-Wheat Rotation as Affected by Crop Residue Incorporation and Temperature显示文摘Field measurements were made from June 2001 to May 2002 to evaluate the e?ect of crop residue application and temperature on CO2, CH4, and N2O emissions within an entire rice-wheat rotation season. Rapeseed cake and wheat straw were incorporated into the soil at a rate of 2.25 t hm?2 when the rice crop was transplanted in June 2001. Compared with the control, the incorporation of rapeseed cake enhanced the emissions of CO2, CH4, and N2O in the rice-growing season by 12.3%, 252.3%, and 17.5%, respectively, while no further e?ect was held on the emissions of CO2 and N2O in the following wheat- growing season. The incorporation of wheat straw enhanced the emissions of CO2 and CH4 by 7.1% and 249.6%, respectively, but reduced the N2O emission by 18.8% in the rice-growing season. Signi?cant reductions of 17.8% for the CO2 and of 12.9% for the N2O emission were observed in the following wheat- growing season. A positive correlation existed between the emissions of N2O and CO2 (R2 = 0.445,n = 73,p < 0.001) from the rice-growing season when N2O was emitted. A trade-o? relationship between the emissions of CH4 and N2O was found in the rice-growing season. The CH4 emission was signi?cantly correlated with the CO2 emission for the period from rice transplantation to ?eld drainage, but not for the entire rice-growing season. In addition, air temperature was found to regulate the CO2 emissions from the non-waterlogged period over the entire rice-wheat rotation season and the N2O emissions from the non- waterlogged period of the rice-growing season, which can be quantitatively described by an exponential function. | 13 | Diagenetic fluids evolution and genetic mechanism of tight sandstone gas reservoirs in Upper Triassic Xujiahe Formation in Sichuan Basin, China显示文摘The reservoirs of the Upper Triassic Xujiahe Formation in Sichuan Basin have the characteristics of low compositional maturity, low contents of cements and medium textural maturity. The general physical properties of the reservoirs are poor, with low porosity and low permeability, and there are only a few reservoirs with medium porosity and low permeability in local areas. Based on the diagenetic mineral association, a diagenetic sequence of cements is established: early calcites (or micrite siderites) →first quartz overgrowth→chlorite coatings→dissolution of feldspars and debris→chlorite linings→ second quartz overgrowth (quartz widen or filled in remain intergranular pores and solution pores)→dissolution→third quartz overgrowth (quartz filled in intergranular and intragranular solution pores)→intergrowth (ferro) calcites→dolomites→ferro (calcites) dolomites→later dissolution→veins of quartz and calcites formation. Mechanical compaction is the main factor in making the reservoirs tight in the basin, followed by the second and third quartz overgrowth. In a long-term closed system, only feld-spars and some lithic fragments are dissolved by diagenetic fluids, while intergranular cements such as quartz and calcit are not dissolved and thus have little influence on the porosity of the Xujiahe Formation. This is the third factor that may have kept the sandstones of Xujiahe Formation tight finally. | 15 | Engineering Broad-Spectrum Bacterial Blight Resistance by Simultaneously Disrupting Variable TALE-Binding Elements of Multiple Susceptibility Genes in Rice显示文摘Xanthomonas oryzae pv.oryzae(Xoo),the causal agent of bacterial blight of rice,employs the transcription activator-like effectors(TALEs)to induce the expression of the OsSWEET family of putative sugar transporter genes,which function in conferring disease susceptibility(S)in rice plants.To engineer broadspectrum bacterial blight resistance,we used CRISPR/Cas9-mediated gene editing to disrupt the TALEbinding elements(EBEs)of two S genes,OsSWEETH and OsSWEET14,in rice cv.Kitaake,which harbors the recessive resistance allele of Xa25/OsSWEET13.The engineered rice line MS14K exhibited broadspectrum resistance to most Xoo strains with a few exceptions,suggesting that the compatible strains may contain new TALEs.We identified two PthXo2-like TALEs,Tal5LN18 and Tal7PX061,as major virulence factors in the compatible Xoo strains LN18 and PX061,respectively,and found that Xoo encodes at least five types of PthXo2-like effectors.Given that PthXo2/PthXo2.1 target OsSlVEETf3 for transcriptional activation,the genomes of 3000 rice varieties were analyzed for EBE variationsin the OsSWEET13 promoter,and 10Xa25-like haplotypes were identified.We found that Tal5LN18 and Tal7PX〇6i bind slightly different EBE sequences in the OsSWEET13 promoter to activate its expression.CRISPR/Cas9 technology was then used to generate InDels in the EBE of the OsSWEET13 promoter in MS14K to creat a new germplasm with three edited OsSWEET EBEs and broad-spectrum resistance against all Xoo strains tested.Collectively,our findings illustrate how to disarm TALE-S co-evolved loci to generate broad-spectrum resistance through the loss of effector-triggered susceptibility in plants. | Zhengyin Xu Xiameng Xu Qiang Gong Ziyang Li Ying Li Sai Wang Yangyang Yang Wenxiu Ma Longyu Liu Bo Zhu Lifang Zou Gongyou Chen | 2019 | Molecular Plant2019,12,11: | 29 |

Here, wecarried out cDNA microarray-based global transcription profiling of HeLa cells in response to As2O3/emodin cotreatment,comparing with As2O3–only treatment. The results showed that the expression of a number of genes was substantiallyaltered at two time points. These genes are involved in different aspects of cell function. | 17 | Effect of lifestyle intervention on non-alcoholic fatty liver disease in Chinese obese children显示文摘AIM: To investigate the effect of lifestyle intervention on non-alcoholic fatty liver disease (NAFLD) in Chinese obese children. METHODS: Seventy-six obese children aged from 10 to 17 years with NAFLD were enrolled for a one-month intervention and divided randomly into three groups. Group1, consisting of 38 obese children, was an untreated control group without any intervention. Group 2, consisting of 19 obese children in summer camp, was strictly controlled only by life style intervention. Group 3, consisting of 19 obese children, received oral vitamin E therapy at a dose of 100 mg/d. The height, weight, fasting blood glucose (FBG), fasting serum insulin (FINS), plasma alanine aminotransferase (ALT), aspartate aminotransferase (AST), triglyceride (TG), total cholesterol (TCHO) and homeostasis model assent- insulin resistance (HOMA-IR) were measured at baseline and after one month. All patients were underwent to an ultrasonographic study of the liver performed by one operator who was blinded to the groups. RESULTS: The monitor indices of BMI, ALT, AST, TG, TCHO and HOMA-IR were successfully improved except in group 1. BMI and ALT in group 2 were reduced more significantly than in group 3 (2.44 ± 0.82 vs 1.45 ± 0.80, P = 0.001; 88.58 ± 39.99 vs 63.69 ± 27.05, P = 0.040, respectively).CONCLUSION: Both a short-term lifestyle intervention and vitamin E therapy have an effect on NAFLD in obese children. Compared with vitamin E, lifestyle intervention is more effective. | 19 | Effects of intracoronary injection of nicorandil and tirofiban on myocardial perfusion and short-term prognosis in elderly patients with acute ST-segment elevation myocardial infarction after emergency PCI显示文摘BACKGROUND:This study investigated the effects of the intracoronary injection of nicorandil and tirofiban on myocardial perfusion and short-term prognosis in elderly patients with acute ST-segment elevation myocardial infarction(STEMI)after emergency percutaneous coronary intervention(PCI).METHODS:Seventy-eight STEMI patients with age>65 years who underwent emergency PCI were consecutively enrolled.These patients received conventional PCI and were randomly divided into a control group and a treatment group(n=39 per group).The control group received an intracoronary injection of tirofiban followed by a maintenance infusion for 36 hours after surgery.The treatment group received intracoronary injection of tirofiban and nicorandil,and then intravenous infusion of tirofiban and nicorandil 36 hours after surgery.The following parameters were measured:TIMI grade,corrected TIMI frame count(c TFC),TIMI myocardial perfusion grade(TMPG),STsegment resolution(STR)rate 2 hours post-operatively,resolution of ST-segment elevation(STR)at 2 hours postoperatively,peak level of serum CK-MB,left ventricular end diastolic diameter(LVEDD)and left ventricular ejection fraction(LVEF)at 7–10 days postoperatively,and major adverse cardiac events(MACEs)in-hospital and within 30 days post-operatively.RESULTS:Compared with the control group,more patients in the treatment group had TIMI 3 and TMPG 3,and STR after PCI was significantly higher.The treatment group also had significantly lower c TFC,lower infarction relative artery(IRA),lower peak CK-MB,and no reflow ratio after PCI.The treatment group had significantly higher LVEDD and LVEF but lower incidence of MACEs than the control group.CONCLUSION:The intracoronary injection of nicorandil combined with tirofiban can effectively improve myocardial reperfusion in elderly STEMI patients after emergency PCI and improve shortterm prognoses. | Guo-xiong Chen Hong-na Wang Jin-lin Zou Xiao-xu Yuan | 2020 | World Journal of Emergency Medicine2020,11,3: | 27 |
