| 2 | Current status of severe fever with thrombocytopenia syndrome in China显示文摘Severe fever with thrombocytopenia syndrome(SFTS) is an emerging infectious disease caused by SFTS virus(SFTSV). SFTSV is associated with a high mortality rate and has been reported in China, South Korea and Japan. SFTSV undergoes rapid changes owing to evolution, gene mutations, and reassortment between different strains of SFTSV. In this review, we summarize the recent cases and general properties of SFTS, focusing on the epidemiology, genetic diversity,clinical features, and diagnostics of SFTSV in China. From 2010 to October 2016, SFTS cases were reported in 23 provinces of China, with increased numbers yearly. Infection and death cases are mainly found in central China, where the Haemaphysalis longicornis ticks are spread. The national average mortality rate of SFTS infection was 5.3%, with higher risk to elder people. The main epidemic period was from May to July, with a peak in May. | 6 | AFLP analysis revealed differences in genetic diversity of four natural populations of Manila clam (Ruditapes philippinarum) in China显示文摘The amplified fragment length polymorphism (AFLP) technology was used to analyze the genetic diversities in four natural populations of Manila clam (Ruditapes philippinarum), distributed in four sea areas of China, i.e., the Bohai Sea, the Huanghai Sea, the East China Sea and the South China Sea. Two hundred and sixty-four AFLP loci were analysed in 195 individuals and revealed high levels of genetic diversity. The percentage of polymorphic loci ranged from 92.13% to 96.06% and the Shannon’s information index was from 0.256 8 to 0.275 6. By analyzing molecular variance (AMOVA), it was found that there were high levels of genetic differentiation between populations of Qingdao and the other three sea areas. Cluster analysis by Nei’s pairwise distances grouped specimens by geographical origin, except the population of Qingdao. A conclusion can be drawn that there are high genetic diversities in the four natural populations of Manila clam in China and some distinct differences existed among and between the four populations. | 8 | Improvement of Mono-window Algorithm for Retrieving Land Surface Temperature from HJ-1B Satellite Data显示文摘The thermal infrared channel (IRS4) of HJ-1B satellite obtains view zenith angles (VZA) up to ±33°. The view angle should be taken into account when retrieving land surface temperature (LST) from IRS4 data. This study aims at improving the mono-window algorithm for retrieving LST from IRS4 data. Based on atmospheric radiative transfer simulations,a model for correcting the VZA effects on atmospheric transmittance is proposed. In addition,a generalized model for calculating the effective mean atmospheric temperature is developed. Validation with the simulated dataset based on standard atmospheric profiles reveals that the improved mono-window algorithm for IRS4 obtains high accuracy for LST retrieval,with the mean absolute error (MAE) and root mean square error (RMSE) being 1.0 K and 1.1 K,respectively. Numerical experiment with the radiosonde profile acquired in Beijing in winter demonstrates that the improved mono-window algorithm exhibits excellent ability for LST retrieval,with MAE and RMSE being 0.6 K and 0.6 K,respectively. | 9 | Complement C3 activation regulates the production of tRNA-derived fragments Gly-tRFs and promotes alcohol-induced liver injury and steatosis显示文摘Complement is known to play a role in alcoholic fatty liver disease (AFLD), but the underlying mechanisms are poorly understood, thereby constraining the development of a rational approach for therapeutic intervention in the complement system. C3 deficiency has been shown to impart protective effects against ethanol-induced hepatic steatosis and inflammation. Here we demonstrate a protection effect in wild-type mice by treatment with CR2-Crry, a specific inhibitor of C3 activation. The expression of glycine transfer (t) RNA-derived fragments (Gly-tRFs) is upregulated in ethanol-fed mice and inhibition of Gly-tRFs in vivo decreases chronic ethanol feeding-induced hepatosteatosis without affecting inflammation. The expression of Gly-tRF was downregulated in C3-deficient or CR2-Crry-treated mice, but not in C5-deficient mice;Gly-tRF expression was restored by the C3 activation products C3a or Asp (C3a-des-Arg) via the regulation of CYP2E1. Transcriptome profiling of hepatic tissues showed that Gly-tRF inhibitors upregulate the expression of sirtuin1 (Sirt1) and subsequently affect downstream lipogenesis and β-oxidation pathways. Mechanistically, Gly-tRF interacts with AGO3 to downregulate Sirt1 expression via sequence complementarity in the 3′ UTR. Notably, the expression levels of C3d, CYP2E1 and Gly-tRF are upregulated, whereas Sirt1 is decreased in AFLD patients compared to healthy controls. Collectively, our findings suggest that C3 activation products contribute to hepatosteatosis by regulating the expression of Gly-tRF. Projectile fragments produced by the 78Kr ions at 481.88 MeV/u were separated with the new radioactive beam line in Lanzhou (RIBLL2), and injected into and stored in the experimental storage ring (CSRe). By operating the CSRe as an isochronous mass spectrometry, a typical mass resolution around 2.0×10?5 has been achieved. The masses for 63Ge, 65As and 67Se were measured for the first time. The measured masses are compared with theoretical predictions and the location of the proton drip-line for As isotopes is discussed. | 16 | Protective effects of transplanted neural stem cells on the brain of Alzheimer’s disease rats显示文摘BACKGROUND: To date, no drugs are able to halt the progression of Alzheimer's disease (AD). Neural stem cells (NSCs) transplantation has been widely used to treat AD, but the mechanism of AD treatment remains unclear. OBJECTIVE: To observe changes in protein and factors in the hippocampus and frontal lobe of AD rats following NSCs transplantation, and to understand mechanism of action of NSCs transplantation in AD treatment. DESIGN, TIME AND SETTING: A randomized, controlled animal study was conducted at the First Clinical Hospital, Jilin University, China from July 2007 to March 2009. MATERIALS: NSCs were harvested from the hippocampus of 10 E16 Wistar rats. METHODS: A total of 57 male adult Wistar rats were equally and randomly divided into normal control, AD model and NSCs groups. AD models were established in the AD model and NSCs groups by bilateral removal of hippocampus. At 2 weeks postsurgery, NSCs were transplanted into the hippocampus of rats from the NSCs group. MAIN OUTCOME MEASURES: Protein levels were measured in the hippocampus of rats from normal control, NSCs and AD model groups using proteomics. Expression of choline acetyl transferase mRNA, glial fibrillary acidic protein and S100β was measured in the hippocampus and frontal lobe of rats using in situ hybridization and immunohistochemistry. RESULTS: Expression of choline acetyl transferase mRNA, heat shock protein 70, heat shock protein 90, F-actin and actin was significantly higher in the NSCs group compared with AD model group. Glial fibrillary acidic protein and S100β expression was less in the NSCs group compared with AD model group. CONCLUSIOIN: NSCs implanted into the brain may generate new neural cells, which can relieve damage to the cholinergic system and resist apoptosis. | 17 | OsSPL18 controls grain weight and grain number in rice显示文摘Grain weight and grain number are two important traits directly determining grain yield in rice. To date,a lot of genes related to grain weight and grain number have been identified; however, the regulatory mechanism underlying these genes remains largely unknown. In this study, we studied the biological function of OsSPL18 during grain and panicle development in rice. Knockout (KO) mutants of OsSPL18exhibited reduced grain width and thickness, panicle length and grain number, but increased tiller number. Cytological analysis showed that OsSPL18 regulates the development of spikelet hulls by affecting cell proliferation. qRT-PCR and GUS staining analyses showed that OsSPL18 was highly expressed in developing young panicles and young spikelet hulls, in agreement with its function in regulating grain and panicle development. Transcriptional activation experiments indicated that OsSPL18is a functional transcription factor with activation domains in both the N-terminus and C-terminus, and both activation domains are indispensable for its biological functions. Quantitative expression analysis showed that DEP1, a major grain number regulator, was significantly down-regulated in OsSPL18 KO lines.Both yeast one-hybrid and dual-luciferase (LUC) assays showed that OsSPL18 could bind to the DEP1promoter, suggesting that OsSPL18 regulates panicle development by positively regulating the expression of DEP1. Sequence analysis showed that OsSPL18 contains the OsmiR156k complementary sequence in the third exon; 5?RLM-RACE experiments indicated that OsSPL18 could be cleaved by OsmiR156k. | 19 | Successful Production of an All-Female Common Carp (Cyprinus carpio L.) Population Using cyp17a1-Deficient Neomale Carp显示文摘Due to sexual dimorphism in the growth of certain cultured fish species,the production of monosex fishes is desirable for the aquaculture industry.Nowadays,the most widely practiced technique available for the mass production of monosex fish populations is sex steroid-induced sex reversal.Here,a novel strategy for the successful production of all-female(AF)common carp(Cyprinus carpio L.),to take advantage of the sexual dimorphism in growth documented in this species,has been developed using genetic engineering via single gene-targeting manipulation without any exogenous hormone treatments.Male and female heterozygous cyp17a1-deficient common carp were first obtained using the clustered regularly interspaced short palindromic repeats/CRISPR-associated endonuclease 9(CRISPR/Cas9)technique.An all-male phenotype for homozygous cyp17a1-deficient carp,regardless of the individuals’sexdetermination genotypes(XY or XX),has been observed.A male-specific DNA marker newly identified in our laboratory was used to screen the neomale carp population with the XX genotype from the cyp17a1-deficient carp.These neomale carp develop a normal testis structure with normal spermatogenesis and sperm capacity.The neomale common carp were then mated with wild-type(WT)females(cyp17a1^(+/+)XX genotype)using artificial fertilization.All the AF offspring sample fish from the neomale-WT female mating were confirmed as having the cyp17a^(1+/-)XX genotype,and normal development of gonads to ovaries was observed in 100.00%of this group at eight months post-fertilization(mpf).A total of 1000 carp fingerlings,500 from the WT male and female and 500 from the neomale and WT female mating,were mixed and reared in the same pond.The average body weight of cyp17a1^(+/-)XX females was higher by 6.60%(8 mpf)and 32.66%(12 mpf)than that of the control common carp.Our study demonstrates the first successful production of a monosex teleost population with the advantages of sexual dimorphism in growth using genetic manipulation targeting a single locus. | Gang Zhai Tingting Shu Kuangxin Chen Qiyong Lou Jingyi Jia Jianfei Huang Chuang Shi Xia Jin Jiangyan He Donghuo Jiang Xueqiao Qian Wei Hu Zhan Yin | 2022 | Engineering2022,8,1: | 7 |
| 20 | Effects of Niaoduqing Particles(尿毒清颗粒)on Delaying Progression of Renal Dysfunction:A Post-trial,Open-Label,Follow-up Study显示文摘Objective: To follow up the participants of the randomized clinical trial 'Efficacy and Safety of Niaoduqing Particles(尿毒清颗粒) for Delaying Moderate-to-Severe Renal Dysfunction', and assess the long-term effects of Niaoduqing Particles on delaying the progression of renal dysfunction. Methods: Participants, who had previously been randomly assigned to receive Niaoduqing Particles or placebo for 24 weeks(146 cases in each group), were invited to follow-up and all were administered Niaoduqing Particles 5 g thrice daily and 10 g before bedtime for 24 weeks. The primary endpoints were changes in baseline serum creatinine(Scr) and estimated glomerular filtration rate(e GFR) after completion of the open-label treatment period. Results: After the double-blind period, the median(interquartile range) changes in Scr were 1.1(–13.0–24.1) and 11.7(–2.6–42.9) μmol/L for the Niaoduqing Particle and placebo groups, respectively(P=0.008), and the median changes in e GFRs were –0.2(–4.3–2.7) and –2.21(–5.7–0.8) mL·min^(-1)·1.73 m^(-2), respectively(P=0.016). There were significant differences in the double-blind period changes in renal function between groups. After the open-label period, the median changes in Scr were 9.0(–10.0–41.9) and 17.5(–6.0–50.0) μmol/L for the Niaoduqing Particle and placebo groups according to baseline grouping, respectively(P=0.214), and the median changes in eGFRs were –2.3(–6.4–1.9) and –3.7(–7.5–1.1) mL·min^(-1)·1.73 m^(-2), respectively(P=0.134). There were no statistical differences in the open-label period changes in renal function between groups. The eGFR reduction of participants who accepted Niaoduqing Particle treatment for 48 weeks was projected to 2.5 m L·min^(-1)·1.73 m(-2) per year. Conclusions: Niaoduqing Particles appear to have long-term efficacy for patients with moderate-to-severe renal dysfunction.
